RESUMO

Successful containment strategies for the SARS-CoV-2 pandemic will depend on reliable diagnostic assays. Point-of-care antigen tests (POCT) may provide an alternative to time-consuming PCR tests to rapidly screen for acute infections on site. Here, we evaluated two SARS-CoV-2 antigen tests: the STANDARD™ F COVID-19 Ag FIA (FIA) and the SARS-CoV-2 Rapid Antigen Test (RAT). For diagnostic assessment, we used a large set of PCR-positive and PCR-negative respiratory swabs from asymptomatic and symptomatic patients and health care workers in the setting of two University Hospitals in Munich, Germany, i.e. emergency rooms, patient care units or employee test centers. For FIA, overall clinical sensitivity and specificity were 45.4% (n = 381) and 97.8% (n = 360), respectively, and for RAT, 50.3% (n = 445) and 97.7% (n = 386), respectively. For primary diagnosis of asymptomatic and symptomatic individuals, diagnostic sensitivities were 60.9% (FIA) (n = 189) and 64.5% (RAT) (n = 256). This questions these tests' utility for the reliable detection of acute SARS-CoV-2-infected individuals, in particular in high-risk settings. We support the proposal that convincing high-quality outcome data on the impact of false-negative and false-positive antigen test results need to be obtained in a POCT setting. Moreover, the efficacy of alternative testing strategies to complement PCR assays must be evaluated by independent laboratories, prior to widespread implementation in national and international test strategies.

Vineyard soils show an increased risk of degradation due to being intensively cultivated. The preservation of soil integrity and fertility is a key concept of organic and biodynamic farming. However, both systems are also subject to criticism due to their higher amount of plant protection products used and their increased traffic intensity compared to integrated viticulture, both detrimental to soil quality. The aim of this study was therefore to assess long-term effects of these three management systems on chemical and physical soil quality parameters. For this purpose, topsoil samples were taken in a long-term field trial vineyard at different positions and examined for bulk density, available water capacity (AWC), soil organic carbon (SOC), N, pH, and for total and bioavailable copper (Cu) concentrations. Biodynamic plots had a lower bulk density and higher SOC concentration than the integrated ones, which is probably due to the species-rich cover crop mixture used in the inter-row. However, organic and biodynamic farming showed an accumulation of copper in the under-vine area and in the tractor track, which is problematic for soil fertility in the long-term. Therefore, alternatives for copper in plant protection are necessary to ensure sustainable soil quality through organic and biodynamic viticulture.

An active and diverse soil biota is important for maintaining crop productivity and quality, and preservation of these traits is a major goal of sustainable farming. This study aimed at unravelling the impact of different management practices on soil fungal and bacterial biodiversity in vineyards as a model for permanent crops. Species diversity was assessed using an amplicon sequencing approach in a long-term field experiment in the Rheingau wine region of Germany where integrated, organic and biodynamic management practices had been in place for 10 years. Fungal community composition under integrated management differed significantly from organic and biodynamic management, whereas fungal species richness remained unaffected. Soil under integrated management had a significantly reduced bacterial species richness compared to organic, but community composition was similar to organically and biodynamically managed soils. Highest fungal richness was obtained under cover crop between rows in topsoil, arising from cover cropping and organic carbon supply.

The main objective of this study was to determine growth, yield and fruit quality of grapevines under organic and biodynamic management in relation to integrated viticultural practices. Furthermore, the mechanisms for the observed changes in growth, yield and fruit quality were investigated by determining nutrient status, physiological performance of the plants and disease incidence on bunches in three consecutive growing seasons. A field trial (Vitis vinifera L. cv. Riesling) was set up at Hochschule Geisenheim University, Germany. The integrated treatment was managed according to the code of good practice. Organic and biodynamic plots were managed according to Regulation (EC) No 834/2007 and Regulation (EC) No 889/2008 and according to ECOVIN- and Demeter-Standards, respectively. The growth and yield of the grapevines differed strongly among the different management systems, whereas fruit quality was not affected by the management system. The organic and the biodynamic treatments showed significantly lower growth and yield in comparison to the integrated treatment. The physiological performance was significantly lower in the organic and the biodynamic systems, which may account for differences in growth and cluster weight and might therefore induce lower yields of the respective treatments. Soil management and fertilization strategy could be responsible factors for these changes. Yields of the organic and the biodynamic treatments partially decreased due to higher disease incidence of downy mildew. The organic and the biodynamic plant protection strategies that exclude the use of synthetic fungicides are likely to induce higher disease incidence and might partially account for differences in the nutrient status of vines under organic and biodynamic management. Use of the biodynamic preparations had little influence on vine growth and yield. Due to the investigation of important parameters that induce changes especially in growth and yield of grapevines under organic and biodynamic management the study can potentially provide guidance for defining more effective farming systems.

Grapevine (Vitis vinifera L.) expresses different responses to water stress, depending not only on genotype, but also on the influence of vineyard growing conditions or seasonality. Our aim was to analyse the effects on drought response of two grapevine cultivars growing on two soils, one water draining (WD) containing sand 80% volume and the other water retaining (WR), with no sand. Under these two different water-holding capacities Syrah, displaying a near-anisohydric response to water stress, and Cabernet Sauvignon (on the contrary, near-isohydric) were submitted to water stress in a pot trial. Xylem embolism contributed to plant adaptation to soil water deprivation: in both cultivars during late phases of water stress, however, in Syrah, already at moderate early stress levels. By contrast, Syrah showed a less effective stomatal control of drought than Cabernet Sauvignon. The abscisic acid (ABA) influenced tightly the stomatal conductance of Cabernet Sauvignon on both pot soils. In the near-anisohydric variety Syrah an ABA-related stomatal closure was induced in WR soil to maintain high levels of water potential, showing that a soil-related hormonal root-to-shoot signal causing stomatal closure superimposes on the putatively variety-induced anisohydric response to water stress.
