Some Gram-negative bacteria possess a system for the uptake of iron from host's transferrin consisting of two outer-membrane proteins, namely transferrin-binding proteins 1 and 2. Here we present the cloning and sequencing of the gene encoding the transferrin-binding protein 1 from a serotype 1 isolate of the Gram-negative pathogen Actinobacillus pleuropneumoniae. We have also cloned for expression the Tbp1 and Tbp2 encoding sequences. Both proteins were produced in Escherichia coli and further purified. We also report the deduced Tbp1 amino acid sequence comparison to Tbp1s from related species.

The case of a 65-years-old woman with non-productive cough persisting for one year is described. The X-ray showed lower right paratracheal calcification. The diagnosis of broncholithiasis was made by fiberoptic bronchoscopy, and Mycobacterium avium was isolated after analysis of the specimen collected by bronchial biopsy. We discuss the characteristics of the case, emphasizing the association of broncholithiasis and non-tubercular mycobacterial infection. The literature is reviewed.

Two outer-membrane proteins are involved in the uptake of iron from transferrin by certain Gram-negative bacteria, transferrin-binding proteins 1 and 2. The gene encoding transferrin-binding protein 1 from a serotype 1 isolate of the Gram-negative pathogen Actinobacillus pleuropneumoniae was cloned, and a fragment encoding 700 amino acids of Tbp1 was expressed in Escherichia coli. We also report here sequencing of the tbpl gene and a comparison of the deduced amino acid sequence with Tbpls from related species. The predicted polypeptide product of tbpl is a 106 kDa protein with a 22-residue signal peptide.

BACKGROUND: High psychosocial risk pregnancies require specialized multidisciplinary help and follow-up in order to improve the outcome of babies. POPULATION AND METHODS: Thirty pregnancies were selected among 3500 and followed by a multidisciplinary team during 1990 and 1991. Evaluation included risk predictors, neurodevelopmental outcome of children at 9 and 24 months, number and time of judicial notifications. Evaluation of the mother-to-baby relationship by a psychologist as well as the environmental variables were performed. RESULTS: Four families had one or two, 20 families three or five and six families, six or more of the 15 risk predictors studied. Over the babies followed-up until the age of 24 months, four were placed out of the family, three hospitalizations were medically unjustified and there was no hospitalization for child abuse or neglect. Twelve children had development delay and two developed an environmental-related staturo-ponderal delay. Judicial notifications were realized 18 times during pregnancy or just after delivery and six times during the follow-up for child abuse and neglect. CONCLUSIONS: Taking care of high psychosocial risk pregnancies could be improved in special hospitalization units for mother and child. A better awareness of this problem could also improve the outcome of children, which remains very alarming.

Protamine of the archaeogastropod mollusc Monodonta turbinata has been isolated and characterized. With a mass of 13,476 Da, it is the largest known protamine. Amino acid sequence of this protamine (106 residues) was established from data provided by automated sequence analysis and mass spectrometry of the protein and of its fragments. The primary structure of the NH2-terminal region exhibits repetitive sequence motifs "Basic-Ser" (mainly R-S) and both central and COOH-terminal regions are composed by arginine clusters. The amino acid sequence of Monodonta turbinata protamine shows structural similarities with other protamines from invertebrates and from birds and mammals.
