RESUMO

The present study aims at exploring the potential of the Daruharidra plant (stem and bark) for inhibition of alpha-amylase. Aqueous and ethanolic extraction yielded the highest total phenolic content (TPC) of 101.4 and 111.8 mcg of gallic acid equivalent. Methanol and ethanol extract had Total flavonoid content (TFC) of 319.6 and 288.3 mcg of quercetin equivalent, respectively. In contrast, petroleum ether extraction resulted in the lowest TPC of 23.6 and TFC of 8.33 mcg, respectively. Methanol (5.554 mg/ml), acetone (6.576 mg/ml), and ethanol (7.321 mg/ml) extract had the lowest IC50 values in alpha-amylase inhibition with the mode of inhibition being non-competitive inhibition. HR-LCMS was used for comprehension of phytoconstituents present in the extract. Amongst hundreds of hits observed 10 ligands of alkaloid nature were used for docking studies. Berbamine, alloxanthine, protopine and benazepril along with reference molecule (Acarbose) were subjected to Molecular dynamics (MD) simulation to analyze the stability of the docked protein-ligand complex. The values of RMSD, RMSF, RG, H-Bond and SASA, the interaction energy of all protein-ligand complexes were calculated after 150 ns of MD simulation. The results of screened complexes revealed good stability as compared to reference Acarbose. These screened ligands used for simulation have the most negative binding energies that interacted with alpha amylase enzyme having -9.28 kcal/mol, -7.51 kcal/mol, -7.73 kcal/mol and -8.00 kcal/mol, energies respectively. The results show significant alpha-amylase inhibitory activity and interaction of ligands targeting this enzyme, which can be used for cross-validation, in vitroCommunicated by Ramaswamy H. Sarma.

The present study explores the potential of the Azadirachta indica (Neem) plant parts (stem and bark) component gedunin for inhibition of alpha-amylase and alpha glucosidase. In contrast, Methanol at 50 mg/ml and 65 mg/ml had the lowest IC50 in alpha glucosidase and alpha amylase with noncompetitive and mixed inhibition, respectively. Azadirachta indica seeds collected from ICAR showed anti-diabetes activity in vitro and in vivo seeds collected were subjected to soxhlet and nonsoxhlet techniques followed by chromatography. HR-LCMS, HPLC, and FTIR to comprehend phytoconstituents present in the extract were used to comprehend phytoconstituents present in the extract and showed the presence gedunin. Among many hits observed, gedunin was used for docking studies using ICM software and for molecular dynamic simulation using gromacs. The results show significant alpha-amylase inhibitory activity and alpha glucosidase inhibitory activity and interaction of ligand targeting these enzymes, which can be used for cross-validation, in vitro using ligplot maps and visualization.

RESUMO

Snakebites have been declared a neglected health problem that is considered a national disease by the WHO (world health organisation). Asian countries like India have high snakebite death rates due to short antidotes and poorly equipped doctors. In today's scenario, local resources like herbs need to be used to prepare cheap antidotes and are often available to victims. Snake bites should be viewed as an emergency problem and require additional national guidelines, doctor training, expertise, and human concentration for effective and timely treatment-measures to be taken to ensure the availability and mass production of antidotes. Currently available, antidotes have problems with storage, manufacture, and aspects of the results. Attention should be paid to the natural compound Gedunin with antitoxic effects. To determine Gedunin's therapeutic efficacy, well-designed clinical research is required. This article emphasizes and proves the therapeutic effectiveness of the herbal plant active ingredient Gedunin against snakebites.

Snakebites are a problem due to the increasing number of deaths and permanent disabilities. There is currently a shortage of antidotes for snakebite. The existing antibody antidote, produced from horse/sheep plasma/sera is expensive, species-dependent, and causes fatal side effects. Therefore, it is of interest use of natural flavonoid named gedunin from the Azadirachta indica (Neem) plant species to combat snakebites. Thus, we show the molecular docking analysis of gedunin (C26H31N2O6F) with enzymes (common in snake species) such as 5-nucleotidase, acetyl cholinesterase, L-aao, metalloproteinase, serine, thrombin and phospholipase A2. The modified gedunin in the enzyme pocket showed improved pharmacological properties for further consideration in combating snakebites.

BACKGROUND: Tumor microenvironment is composed of a largely altered extracellular matrix with different cell types. The complex interplay between macrophages and tumor cells through several soluble factors and signaling is an important factor in breast cancer progression. METHODS: We have extended our earlier studies on monocyte and macrophage conditioned medium (MϕCM) and have carried out proteomic analysis to identify its constituents as well as validation. The 8-gene signature identified through macrophage-breast cancer cell interactions was queried in cBioportal for bioinformatic analyses. RESULTS: Proteomic analysis (MALDI-TOF and LC-MS/MS) revealed integrin and matrix metalloproteinases in MϕCM which activated TGF-ß1, IL-6, TGF- ßRII and EGFR as well as its downstream STAT and SMAD signaling in breast cancer cells. Neutralization of pro-inflammatory cytokines (TNF-α. Il-1ß, IL-6) abrogated the MϕCM induced migration but invasion to lesser extent. The 8- gene signature identified by macrophage-tumor interactions (TNF-α, IL-1ß, IL-6, MMP1, MMP9, TGF-ß1, TGF-ßRII, EGFR) significantly co-occurred with TP53 mutation, WTAPP1 deletion and SLC12A5 amplification along with differential expression of PSAT1 and ESR1 at the mRNA level and TPD52and PRKCD at the protein level in TCGA (cBioportal). Together these genes form a novel 15 gene signature which is altered in 63.6% of TCGA (1105 samples) data and was associated with high risk and poor survival (p<0.05) in many breast cancer datasets (SurvExpress). CONCLUSIONS: These results highlight the importance of macrophage signaling in breast cancer and the prognostic role of the15-gene signature. GENERAL SIGNIFICANCE: Our study may facilitate novel prognostic markers based on tumor-macrophage interaction.

Concentration of heavy metals (Cd, Ni, Zn, Fe, Cu, Mn, Pb, Cr, Hg and As) in the waters of River Yamuna and in the soil of agricultural fields along its course in Delhi are reported from 13 sites, spread through the Delhi stretch of Yamuna, starting from the Wazirabad barrage till the Okhla barrage. Varying concentration of heavy metals was found. Peaks were observed in samples collected downstream of Wazirabad and Okhla barrage, indicating the anthropogenic nature of the contamination. The Wazirabad section of the river receives wastewater from Najafgarh and its supplementary drains, whereas the Shahdara drain releases its pollution load upstream of the Okhla barrage. Average heavy metal concentration at different locations in the river water varied in the order of Fe>Cr>Mn>Zn>Pb>Cu>Ni>Hg>As>Cd. The river basin soil shows higher level of contamination with lesser variation than the water samples among sampling locations, thereby suggesting deposition over long periods of time through the processes of adsorption and absorption. The average heavy metal concentration at different locations in soil varied in the order of Fe>Mn>Zn>Cr>Pb>Ni>Hg>Cu>As>Cd.
