RESUMO

Malayan pangolin SARS-CoV-2-related coronavirus (SARSr-CoV-2) is closely related to SARS-CoV-2. However, little is known about its pathogenicity in pangolins. Using CT scans we show that SARSr-CoV-2 positive Malayan pangolins are characterized by bilateral ground-glass opacities in lungs in a similar manner to COVID-19 patients. Histological examination and blood gas tests are indicative of dyspnea. SARSr-CoV-2 infected multiple organs in pangolins, with the lungs the major target, and histological expression data revealed that ACE2 and TMPRSS2 were co-expressed with viral RNA. Transcriptome analysis indicated that virus-positive pangolins were likely to have inadequate interferon responses, with relative greater cytokine and chemokine activity in the lung and spleen. Notably, both viral RNA and viral proteins were detected in three pangolin fetuses, providing initial evidence for vertical virus transmission. In sum, our study outlines the biological framework of SARSr-CoV-2 in pangolins, revealing striking similarities to COVID-19 in humans.

Halogenated methanesulfonic acids (HMSAs) are an important new class of organic compounds as they were universal in the water cycle and drinking water sources. However, no study has investigated the presence of HMSAs in surface water and sediment from China. The present study reports the occurrence and spatiotemporal distribution of seven HMSAs in water and sediment samples from Hangzhou Bay, China. Trifluoromethanesulfonic acid (TFMSA) was the main contributor to the concentrations of HMSAs in water and sediment samples from spring, summer, autumn and winter which were 30.8-541 ng/L, n. d.-86.6 ng/L, 4.22-70.9 ng/L and 8.86-192 ng/L, separately, while in sediment samples were n. d.-11.1 ng/g, n. d.-12.9 ng/g, n. d.-22.5 ng/g, n. d.-4.60 ng/g, respectively. The levels of HMSAs in water from winter and spring were higher than those in summer and autumn, and the concentrations of the target HMSAs in water presents a seasonal pattern affected by the temperature, the precipitation and river flow variations. Nevertheless, the levels of HMSAs in sediment were highest in the area near the industrial area and the confluences of rivers. Correlation analysis revealed that the concentrations of TFMSA were significantly positively correlated with total organic carbon (TOC) in water samples. Although TFMSA is regarded as low toxic based on the EC50 value of acute toxicity, the potential risks to aquatic ecology should be paid more attention due to its high concentrations in the aquatic system and the environmental persistency.

Opportunistic feeding and multiple other environment factors can modulate the gut microbiome, and bias conclusions, when wild animals are used for studying the influence of phylogeny and diet on their gut microbiomes. Here, we controlled for these other confounding factors in our investigation of the magnitude of the effect of diet on the gut microbiome assemblies of nonpasserine birds. We collected fecal samples, at one point in time, from 35 species of birds in a single zoo as well as 6 species of domestic poultry from farms in Guangzhou city to minimize the influences from interfering factors. Specifically, we describe 16S rRNA amplicon data from 129 fecal samples obtained from 41 species of birds, with additional shotgun metagenomic sequencing data generated from 16 of these individuals. Our data show that diets containing native starch increase the abundance of Lactobacillus in the gut microbiome, while those containing plant-derived fiber mainly enrich the level of Clostridium Greater numbers of Fusobacteria and Proteobacteria are detected in carnivorous birds, while in birds fed a commercial corn-soybean basal diet, a stronger inner-connected microbial community containing Clostridia and Bacteroidia was enriched. Furthermore, the metagenome functions of the microbes (such as lipid metabolism and amino acid synthesis) were adapted to the different food types to achieve a beneficial state for the host. In conclusion, the covariation of diet and gut microbiome identified in our study demonstrates a modulation of the gut microbiome by dietary diversity and helps us better understand how birds live based on diet-microbiome-host interactions.IMPORTANCE Our study identified food source, rather than host phylogeny, as the main factor modulating the gut microbiome diversity of nonpasserine birds, after minimizing the effects of other complex interfering factors such as weather, season, and geography. Adaptive evolution of microbes to food types formed a dietary-microbiome-host interaction reciprocal state. The covariation of diet and gut microbiome, including the response of microbiota assembly to diet in structure and function, is important for health and nutrition in animals. Our findings help resolve the major modulators of gut microbiome diversity in nonpasserine birds, which had not previously been well studied. The diet-microbe interactions and cooccurrence patterns identified in our study may be of special interest for future health assessment and conservation in birds.

Novel additives of lanthanum aminopolycarboxylates with inorganic anions, Na12n[La(edta)L]4n·8nNaCl·4nH2O (1: L = HPO32-; 2: L = CO32-) and K12n[La(cdta)(CO3)]4n·35nH2O (3) (H4edta = ethylenediaminetetraacetic acid; H4cdta = cyclohexanediaminetetraacetic acid), were obtained in alkaline solution. Structural analyses reveal that 1 and 2 are isomorphous and contain interesting square structures. HPO32- (CO32-) was encaged in the constructed tetranuclear frameworks. Tetranuclear lanthanum ethylenediaminetetraacetate was further encaged in superstructures of sodium chloride. 3 has a similar square structure, in which edta is replaced by cdta. All complexes are fully characterized via elemental, FT-IR, NMR, thermogravimetric and structural analyses. Solution 13C NMR spectra show that 1 and 2 dissociate into mononuclear units in water. Interestingly, 2 possesses 3.7 Å diameter holes inside its crystals, which can adsorb a small amount of O2 or CO2 selectively. The amounts of O2 and CO2 adsorbed increase gradually from 0.32 and 0.38 mg g-1 at 0.4 bar to 15.90 and 10.54 mg g-1 at 29.9 bar, respectively.

An automated analyzer was developed to achieve fast, precise, and accurate measurements of seawater total alkalinity (AT) based on single-point titration and spectrophotometric pH detection. The single-point titration was carried out in a circulating loop, which allowed the titrant (hydrochloric acid and bromocresol green solution) and a seawater sample to mix at a constant volume ratio. The dissolved CO2 in the sample-titrant mixture was efficiently removed by an inline CO2 remover, which consists of a gas-permeable tubing (Teflon AF2400) submerged in a sodium hydroxide (NaOH) solution. The pH of the mixture was then measured with a custom-made spectrophotometric detection system. The analyzer was calibrated against multiple certified reference materials (CRMs) with different AT values. The analyzer features a sample throughput time of 6.5 min with high precision (±0.33-0.36 µmol kg(-1); n = 48) and accuracy (-0.33 ± 0.99 µmol kg(-1); n = 10). Intercomparison to a traditional open-cell AT titrator showed overall good agreement of 0.88 ± 2.03 µmol kg(-1) (n = 22). The analyzer achieved excellent stability without recalibration over 11 days, during which time 320 measurements were made with a total running time of over 40 h. Because of its small size, low power consumption requirements, and its ability to be automated, the new analyzer can be adapted for underway and in situ measurements.
