The impact of the COVID-19 pandemic on the social, economic, and health levels is unprecedented. In Chile alone until July 2020, more than 340,000 people have tested positive and around 9,000 have died because of it. This health crisis has led to different adaptations in the health system and extraordinary measures have been taken to meet these needs. The physiotherapist as a health professional constitutes a fundamental piece in the care of these patients. Its role has been reflected in all disease's stages, from the emergency room to the intensive care unit and after hospital discharge. In addition to his clinical-attendance role, the physiotherapist may contribute in different areas such as education, health promotion, health management, and scientific research, aspects that could be essential in managing the pandemic. As physiotherapist responsibility, we should consider this opportunity and assume the multiple challenges derived from the pandemic to commit and provide answers to current health needs.

RESUMO

The presence of sleep disordered breathing (SDB) in pediatric population has been associated with several and relevant health problems. Unfortunatly, most of SDB cases remain under diagnosed due to diferent factors incluiding lack of available and good diagnostic tools. Sleep questionnaires have emerged as an alternative screening tool for SDB. Depending on specific measurement purpose, sleep questionnaires may assess different aspects that are related to SDB or sleep duration and quality. Among these questionnaires, the "Pediatric Sleep Questionnaire" (PSQ) is a validated diagnostic tool for SDB worldwide. in Chile, PSQ's diagnostic accuracy was evaluated in 83 children between 0 and 15 years old, showing an area under curve (AUC) 0.687 (CI 95 percent 0.567-0.808), a sensibility 0.78 and specificity 0.72. Based on information described in this review, utlization of sleep screening questionnaire is feasible, leading to an alternative tool to detect SDB in our national clinical context and during research process in sleep medicine

The present work describes reversed-phase high performance liquid chromatographic methodologies (RP-HPLC) for the qualitative and quantitative analysis of the human glycoprotein hormones thyrotropin (hTSH), follitropin (hFSH), choriogonadotropin (hCG) and lutropin (hLH) in the presence of a large excess (up to 250:1) of human serum albumin (HSA). Chromatographic profiles with a good separation between the hormone and HSA were obtained by using a C4 column and specific gradient elution conditions for each hormone. Parameters such as resolution factor, tailing factor and relative retention time, were determined, and are useful for the evaluation of the quality of the separation obtained between the active pharmaceutical ingredient and the excipient present in the final formulation. The potential of each method for quantification of both HSA and the hormone was also demonstrated. Besides furnishing chromatographic quantifications that can substitute for in vivo bioassays and animal use, the chromatograms also provide a direct panorama of the quality and heterogeneity of the protein of interest.

RESUMO

Reversed-phase high-performance liquid chromatography (RP-HPLC) was compared with the classical Steelman-Pohley bioassay (BA), based on animal use, for the determination of human follicle-stimulating hormone (hFSH) biological activity. A linear relationship (BA(IU)=0.9925 RP-HPLC(IU)-1.3165) with a highly significant correlation (r=0.9371; p<0.0001; n=24) was found for these two methods for six hFSH preparations of different origins. The mean difference between the bioactivity predicted from RP-HPLC data via this equation and the mean of the bioactivities obtained with the two methods for six other hFSH preparations was -1.4%, with a 95% confidence interval of -9.3 to +6.6%. The precision of these parameters was 1.63% and 2.82%, respectively. These results demonstrate that RP-HPLC is a viable physical-chemical alternative to the use of an in vivo bioassay for hFSH potency determination, applicable also to hFSH Standards containing large amounts of human serum albumin.

RESUMO

A reptile specimen from the Lystrosaurus Assemblage Zone of the Beaufort Group, lowermost Triassic of South Africa, represents a new procolophonoid parareptile. Sauropareion anoplus gen. et sp. nov. is identified as the sister taxon of Procolophonidae in a phylogenetic analysis of procolophonoids. Stratigraphic calibration of the most parsimonious tree reveals that four of the six procolophonoid lineages originating in the Permian Period extended into the succeeding Triassic Period. This relatively high taxic survivorship (67%) across the Permo-Triassic boundary strongly suggests that procolophonoids were little if at all affected by the mass extinction event that punctuated the end of the Palaeozoic Era (ca. 251 million years ago).

RESUMO

The roles that human immunodeficiency virus (HIV) and Epstein-Barr virus (EBV) play in the genesis of acquired immune deficiency syndrome (AIDS)-related lymphomas are not understood. A human B cell line (B-HIV), developed to study AIDS-related lymphomagenesis, contains EBV and HIV genomes and is malignantly transformed. This line was produced by exposing B cells from an EBV-seropositive donor to HIV. To investigate the number of independent transformation events that took place at the time of HIV infection, we sought to determine how many transformed lineages are present in this cell line. B-HIV was found to have multiple different sites of HIV integration (> or = 25) as determined by fluorescence in situ hybridization. As a control, we analyzed a clonal cell line of HIV-infected human T cells, 8E5, and found HIV sequences located exclusively at band q22 in chromosome 13. We conclude that B-HIV is polyclonal, and viral sequences are located at multiple variable chromosomal sites in different B-HIV cells.

RESUMO

The R/B genes of maize encode a family of basic helix-loop-helix proteins that determine where and when the anthocyanin-pigment pathway will be expressed in the plant. Previous studies showed that allelic diversity among family members reflects differences in gene expression, specifically in transcription initiation. We present evidence that the R gene Lc is under translational control. We demonstrate that the 235-nt transcript leader of Lc represses expression 25- to 30-fold in an in vivo assay. Repression is mediated by the presence in cis of a 38-codon upstream open reading frame. Furthermore, the coding capacity of the upstream open reading frame influences the magnitude of repression. It is proposed that translational control does not contribute to tissue specificity but prevents overexpression of the Lc protein. The diversity of promoter and 5' untranslated leader sequences among the R/B genes provides an opportunity to study the coevolution of transcriptional and translational mechanisms of gene regulation.

RESUMO

The problem of cell survival as well as the biochemical and histological effects on skin of low temperature storage in liquid nitrogen has already received attention. However, little is known of the submicroscopic alterations induced by such treatment. Our study shows that rapid or slow freezing, direct or mediate contact with the liquid nitrogen, pretreatment before freezing, type and concentration of cryoprotectants, all have various effects on the fine morphology of epidermal cells. Glycerol is shown as having better cryoprotective activity than DMSO at concentrations of 15% or less. Our data are examined taking into account all available reports on the problem of cryopreservation and the use of cryoprotectants.
