AIMS: Show that tomato leaf phyllosphere bacteria are candidates for biocontrol of tomato leaf diseases. METHODS AND RESULTS: Seven bacterial isolates from surface-sterilized Moneymaker tomato plants were tested for growth inhibition of 14 tomato pathogens on potato dextrose agar. Biocontrol assays were conducted with tomato leaf pathogens, Pseudomonas syringae pv. tomato (Pto) and Alternaria solani (A. solani). Two potential isolates showing the greatest inhibition were identified by 16S rDNA sequencing as Rhizobium sp. (isolate b1) and Bacillus subtilis (isolate b2), both produce protease and isolate b2 cellulase. Both reduced tomato leaf infections by Pto and A. solani in detached leaf bioassays. Both bacteria b1 and b2 reduced pathogen development in a tomato growth trial. Bacteria b2 also induced the tomato plant salicylic acid (SA) immune response pathway. Disease suppression in biocontrol assays with b1 and b2 varied between five commercial tomato varieties. CONCLUSIONS: Tomato phyllosphere bacteria when used as phyllosphere inoculants, inhibited tomato diseases caused by Pto and A. solani.

AIMS: Sub-therapeutic use of antibiotics as a growth promoter in animal diets has either been banned or voluntarily withdrawn from use in many countries to help curb the emergence of antibiotic-resistant pathogens. Probiotics may be an alternative to antibiotics as a growth promoter. We investigated the effects of a novel probiotic strain, Bacillus amyloliquefaciens H57 (H57) on the performance and microbiome-associated metabolic potential. METHODS AND RESULTS: Broiler chickens were fed either sorghum- or wheat-based diets supplemented with the probiotic H57. The growth rate, feed intake, and feed conversion in supplemented birds were compared with those in non-supplemented control. Caecal microbial metabolic functions were studied with shotgun metagenomic sequencing. H57 supplementation significantly increased the growth rate and daily feed intake of meat chickens relative to the non-supplemented controls without any effect on feed conversion ratio. In addition, relative to the non-supplemented controls, gene-centric metagenomics revealed that H57 significantly altered the functional capacity of the caecal microbiome, with amino acid and vitamin synthesis pathways being positively associated with H57 supplementation. CONCLUSIONS: Bacillus amyloliquefaciens H57 improves the performance of meat chickens or broilers and significantly modifies the functional potential of their caecal microbiomes, with enhanced potential capacity for amino acid and vitamin biosynthesis.

This study examined whether the probiotic Bacillus amyloliquefaciens strain H57 (H57) affects ruminal fermentation parameters that exercise post-ingestive feedback appetite control mechanisms. A 4 × 4 Latin square design was used to separate pre- and post-ingestive effects of H57 in four rumen-fistulated steers. The steers were offered a set amount of feedlot pellets, inoculated with H57 or without H57 (control, C). Half of the total amount of pellets fed were introduced intra-ruminally (r), and then the remaining pellets were orally consumed (o) to make four feeding treatments: H57r/H57o, H57r/Co, Cr/H57o and Cr/Co. Rumen fluid was sampled at 2, 4 and 6 h after feeding. Preference behaviour was tested immediately after the 6 h rumen fluid sampling by simultaneously offering the steers 4 kg of each of H57 and C pellets in adjacent troughs for 5 min. Steers preferred the pellets with added H57 over the C pellets (56:44; p < 0.001) and their preferences were not affected by the treatment protocol imposed to separate post- from pre-ingestive effects (p > 0.05). Steers fed H57 pellets had higher ruminal pH, molar proportions of iso-butyrate and iso-valerate (p < 0.05) and tended to have greater ruminal ammonia concentrations compared to those fed C pellets (p < 0.1). However, post-ingestive signals did not affect diet preference more than pre-ingestive signals.

Mould and bacterial contamination releases microbial volatile organic compounds (mVOCs), causing changes in the odour profile of a feed. Bacillus amyloliquefaciens strain H57 (H57) has the potential ability to inhibit microbial growth in animal feeds. This study tested the hypothesis that H57 influences the odour profile of stored feedlot pellets by impeding the production of mVOCs. The emission of volatile organic compounds (VOCs) of un-inoculated pellets and those inoculated with H57, stored either at ambient temperature (mean 22 °C) or at 5 °C, was monitored at 0, 1, 2, and 3 months by gas chromatography-mass spectrometry. Forty VOCs were identified in all the pellet samples analysed, 24 of which were potentially of microbial and 16 of non-microbial origin. A score plot of the principal component analysis (PCA) showed that the VOC profiles of the pellets stored at ambient temperature changed more rapidly over the 3 months than those stored at 5 °C, and that change was greater in the un-inoculated pellets when compared to the inoculated ones. The bi-plot and correlation loading plots of the PCA indicated that the separation of the un-inoculated pellets from the other treatments over the 3 months was primarily due to nine mVOCs. These mVOCs have been previously identified in grains spoiled by fungi, and could be considered potential markers of the types of fungi that H57 can protect pellets against. These data indicate the ability of H57 to maintain the odour profile and freshness of concentrated feed pellets. This protective influence can be detected as early as 3 months into ambient temperature storage.

This study tested the hypothesis that Bacillus amyloliquefaciens strain H57 (H57) improves preference by reducing the development of microbial volatile organic compounds (mVOCs) in feed pellets. Sixteen bull calves were, for 4 weeks, provided equal access to a panel of 8 automated feed bunks in a single paddock with some hay. Each bunk contained pellets with (H57) or without (Control) the H57, each aged for 4 months at either ambient or chiller temperature. Each treatment was changed to a new bunk pair position weekly. Relative preference was determined according to weight of pellets remaining per hour per treatment bunk pair per 24 h. Pellets were analysed for volatile organic compounds (VOCs) and the concentrations tested for correlation with relative preference. Calves showed the lowest preference (p < 0.0001) for the Control/Ambient treatment whereas preference for all other treatments (H57/Ambient; H57/Chiller; Control/Chiller) was similar. The Control/Ambient treatment odour profile grouped differently to the other 3 treatments which grouped similarly to each other. Up to 16 mVOCs were determined to have potential as pre-ingestive signals for the extent of microbial spoilage. Further studies are required to find which combination of these mVOCs, when added to pellets, results in feed aversion.

Local application of the biosphere reserve concept in Kien Giang, Vietnam was examined to see how it compared with other biosphere reserves both in Vietnam and internationally and from that to assess the level of adoption and what could be limiting processes. This was undertaken mainly through qualitative document analysis, field surveys, and extensive interviews of stakeholders. While the designation the Kien Giang Biosphere Reserve and establishment of the management regulation conformed with the conceptual model and criteria outlined by UNESCO, the practical implementation has been inadequate to achieve the desired outcomes of the biosphere reserve concept. There was limited public awareness and understanding of the biosphere reserve approach because of poorly developed communication channels. Top-down, state-control based on a strong sectoral approach to biosphere reserve planning and management hindered stakeholder and community participation. Weak engagement from the Province as the designated lead agency in biosphere reserve governance limited cross-sectoral collaboration in the delivery of the biosphere reserve mandated functions. External projects were perceived by community stakeholders to have only a temporary impact on biosphere reserve operation because of their small, short-term scale with the project maintaining control over funding and design of individual activities. Without proper investment in public awareness and improvement of Biosphere Reserve governance leadership, the desire for development of strategic public-private partnerships to support implementation remains unfulfilled and the Biosphere Reserve model will, as a consequence, contribute little to the long-term biodiversity conservation and socio-economic development in the region.

Biosphere reserves established under the UNESCO Man and the Biosphere Program aim to harmonise biodiversity conservation and sustainable development. Concerns over the extent to which the reserve network was living up to this ideal led to the development of a new strategy in 1995 (the Seville Strategy) to enhance the operation of the network of reserves. An evaluation of effectiveness of management of the biosphere reserve network was called for as part of this strategy. Expert opinion was assembled through a Delphi Process to identify successful and less successful reserves and investigate common factors influencing success or failure. Ninety biosphere reserves including sixty successful and thirty less successful reserves in 42 countries across all five Man and the Biosphere Program regions were identified. Most successful sites are the post-Seville generation while the majority of unsuccessful sites are pre-Seville that are managed as national parks and have not been amended to conform to the characteristics that are meant to define a biosphere reserve. Stakeholder participation and collaboration, governance, finance and resources, management, and awareness and communication are the most influential factors in the success or failure of the biosphere reserves. For success, the biosphere reserve concept needs to be clearly understood and applied through landscape zoning. Designated reserves then need a management system with inclusive good governance, strong participation and collaboration, adequate finance and human resource allocation and stable and responsible management and implementation. All rather obvious but it is difficult to achieve without commitment to the biosphere reserve concept by the governance authorities.

Bacillus amyloliquefaciens H57 is a bacterium isolated from lucerne for its ability to prevent feed spoilage. Further interest developed when ruminants fed with H57-inoculated hay showed increased weight gain and nitrogen retention relative to controls, suggesting a probiotic effect. The near complete genome of H57 is ~3.96 Mb comprising 16 contigs. Within the genome there are 3,836 protein coding genes, an estimated sixteen rRNA genes and 69 tRNA genes. H57 has the potential to synthesise four different lipopeptides and four polyketide compounds, which are known antimicrobials. This antimicrobial capacity may facilitate the observed probiotic effect.

Although Cu is phytotoxic at Cu(2+) activities as low as 1-2 microM, the effect of Cu(2+) on the nodulation of legumes has received little attention. The effect of Cu(2+) on nodulation of cowpea (Vigna unguiculata (L.) Walp. cv. Caloona) was examined in a dilute solution culture system utilising a cation exchange resin to buffer solution Cu(2+). The nodulation process was more sensitive to increasing Cu(2+) activities than both shoot and root growth; whilst a Cu(2+) activity of 1.0 microM corresponded to a 10% reduction in the relative yield of the shoots and roots, a Cu(2+) activity of 0.2 microM corresponded to a 10% reduction in nodulation. This reduction in nodulation with increasing Cu(2+) activity was associated with an inhibition of root hair formation in treatments containing > or =0.77 microM Cu(2+), rather than to a reduction in the size of the Rhizobium population.

Leaf water relations responses to limited water supply were determined in 7-month-old plants of a dry inland provenance of Eucalyptus argophloia Blakely and in a humid coastal provenance (Gympie) and a dry inland provenance (Hungry Hills) of Eucalyptus cloeziana F. Muell. Each provenance of E. cloeziana exhibited a lower relative water content at the turgor loss point, a lower apoplastic water content, a smaller ratio of dry mass to turgid mass and a lower bulk modulus of elasticity than the single provenance of E. argophloia. Osmotic potential at full turgor and water potential at the turgor loss point were significantly lower in E. argophloia and the inland provenance of E. cloeziana than in the coastal provenance of E. cloeziana. There was limited osmotic adjustment in response to soil drying in E. cloeziana, but not in E. argophloia. Between-species differences in water relations parameters were larger than those between the E. cloeziana provenances. Both E. cloeziana provenances maintained turgor under moderate water stress through a combination of osmotic and elastic adjustments. Eucalyptus argophloia had more rigid cell walls and reached lower water potentials with less reduction in relative water content than either of the E. cloeziana provenances, thereby enabling it to extract water from dryer soils.
