Objective: To investigate the feasibility and safety of magnetic resonance guided focused ultrasound surgery (MRgFUS) ablation treatment for uterine fibroids and adenomyosis. Methods: From February 2017 to July 2018, a total of 61 women in Foshan Hospital of Traditional Chinese Medicine with uterine fibroids or adenomyosis (36 cases of fibroids and 25 cases of adenomyosis) were included for treatment of MRgFUS, mean age was 27-48 (39±5) years. The treatment status, treatment effect and complications were recorded, and the differences between myoma and adenomyosis were compared. Results: Thirty-two (88.9%) patients of fibroid group and twenty-one (84.0%) patients of adenomyosis group were completed MRgFUS treatment respectively (P>0.05). The spot energy of adenomyosis group was 1 039-5 698(2 852±991) J, which was higher than 600-6 466(2 485±1 137) J of fibroid group (P<0.01). There was no significant statistical difference in mean temperature of spot and ablation time between the two groups (P>0.05).The non-perfusion volume ratio (NPVR) of the fibroid and adenomyosis group was 54%-99%(84%±15%) and 60%-98%(82%±12%) and there was no significant statistical difference (P>0.05), but the ablation efficiencies of adenomyosis group was less than fibroid group (0.8-4.3(2.1±0.9) cm(3)/min vs 1.3-7.8(3.6±1.5)cm(3)/min, P<0.01).The incidence of complications of adenomyosis group was 47.6%(10/21), it was higher than fibroid group 18.8%(6/32) (P<0.05). Conclusions: MRgFUS is a non-invasive, safe and effective treatment for both uterine fibroids and adenomyosis. Compared with uterine fibroids, MRgFUS treatment of adenomyosis has some disadvantages such as higher energy, lower ablation efficiency and more adverse reactions, and further optimization is needed.

OBJECTIVE: The apoptosis of vascular endothelial cells (VEC) is related to ischemic stroke. Phosphatidylinositol-3 kinase (PI3K)/protein kinase B (AKT/PKB) signaling pathway can upregulate Bcl-2 expression, reduce reactive oxygen species (ROS) production, and induce apoptosis. The level of miR-124 was significantly increased after cerebral ischemia. This study aimed to investigate the role of miR-124 in regulating PI3K expression, brain VEC apoptosis, and ROS production. MATERIALS AND METHODS: The expressions of miR-124, PI3K, p-AKT, and Bcl-2 in brain VEC of rats from the sham group and middle cerebral artery occlusion (MCAO) group were tested. Bioinformatics analysis showed the complementary binding site between miR-124 and PI3K mRNA. ROS content and cell apoptosis were detected by flow cytometry. Rat brain VEC were cultured in vitro and treated by oxygen-glucose deprivation (OGD) for 6 h. VECs were divided into four groups, including miR-NC, miR-124 inhibitor, pIRES2-blank, and pIRES2-PI3K groups, and were further treated by OGD. RESULTS: MiR-124 expression, ROS content, and cell apoptosis were markedly increased, whereas the levels of PI3K, p-AKT, and Bcl-2 were markedly reduced in rat VECs from MCAO group compared with that in the sham group. OGD treatment significantly induced VECs apoptosis, upregulated miR-124 expression and ROS content, and down-regulated the levels of PI3K, p-AKT, and Bcl-2. MiR-124 inhibitor or transfection of pIRES2-PI3K plasmid apparently enhanced PI3K, p-AKT, and Bcl-2 expressions, alleviated cell apoptosis and decreased ROS content in VECs induced by OGD. CONCLUSIONS: Our data demonstrated that miR-124 induced the apoptosis of brain vascular endothelial cells via the down-regulation of PI3K/AKT signaling pathway and promotion of ROS production.

Favorable uterine involution and ovarian activity are very important for the next reproductive cycle of postpartum cows. The objective of this study was to evaluate the effect of parity on uterine involution and resumption of ovarian activity in Chinese Holstein dairy cows after calving under similar postpartum nutritional conditions. Traits of the status of uterus and ovaries detected by ultrasonography, dry matter intake (DMI), milk yield, body condition score (BCS), and estradiol concentration in milk samples were analyzed for 46 Chinese Holstein dairy cows in various parities (primiparous=18; biparous=13; multiparous=15). The results showed that there was no significant difference for DMI, BCS, and milk yield among different parities; all cows were considered to be under similar nutritional conditions. Days of the previous gravid uterine horn involution were significantly greater in primiparous dairy cows than in biparous and multiparous dairy cows. Days from calving to ovulation (first and second) and the number of follicular waves to first ovulation were significantly greater in primiparous cows than in multiparous cows. In summary, there was a significant negative relationship between parity and postpartum uterine involution and resumption of ovarian activity in Chinese Holstein cows under similar body conditions.

To study the frequency of numerical chromosome aberrations in sperm of workers exposed to benzene series, the aneuploidy frequencies of 9 and 18 chromosomes were detected in sperm of workers by two color fluorescence in situ hybridization with digoxingenin labeled 9 chromosome probe (D9z1) and biotin labelled 18 chromosome probe (D18z1). The timeweight average air concentration (TWA) of benzene in the workplace was 86.49 mg/m3, it was two fold higher than the national maximum allowable concentration. The concentration of urinary trans, transmuconic acid (ttMA) in exposed group was significantly higher than that of control group. The sperms counted in 14 exposed workers and 16 control workers were 136,401 and 156,955 respectively. The results showed that the disomy frequencies of sperm for chromosome 9 and 18 (0.168% +/- 0.063% and 0.055% +/- 0.031% respectively) and the frequency of diploidy sperm (0.073% +/- 0.045%) were statistically increased over that of the control group (0.050% +/- 0.030%; 0.033% +/- 0.025% and 0.040% +/- 0.036%, respectively). The nullisomic frequencies of 9 and 18 chromosome in the exposed group (0.206% +/- 0.047%, 0.068% +/- 0.044%) were statistically increased over that of the control group too (0.067% +/- 0.037%, 0.048% +/- 0.034%). The frequency of overall numerical chromosome aberrations in the exposed group was 0.570% +/- 0.144%, when this value was compared with that of control (0.218% +/- 0.071%), a statistically significance was presented. Our experiments showed that exposed to benzene at higher concentration may induce increase in aneuploidy frequency of sperm autosomal chromosome in exposed workers.
