RESUMO

The objective was to evaluate the behavior of melon genotypes (Cucumis melo L.) in the physical, chemical and biochemical quality of melon fruits as a function of electrical conductivity irrigation water levels (ECw). The experimental design adopted was randomized blocks in a 5 x 3 factorial scheme with five replications. The first factor was represented by five salinity levels (0.5, 1.5, 3.0, 4.5, and 6.0 dS m-1) and the second factor by accessions A35, and A24, and the hybrid Sancho. The physical, chemical and biochemical variables showed a reduction in production, with smaller fruits, with less weight, smaller cavity, with increased pulp thickness for Sancho. Vitamin C and yellow flavonoids increased indicating antioxidant power against ROS. The genotypes showed similar post-harvest behavior, however, the hybrid Sancho stood out over the others, possibly because it is an improved material. Accession A24 presented physiological and biochemical responses that classify it as intolerant.

Anticarsia gemmatalis Hünber, 1818 is one of the main defoliating species in the soybean crop. Bacillus thuringiensis Berliner, 1915, is a bacterium used in the biological control of this pest species. Resistant populations and their sublethal effects caused by the use of the bacteria have already been reported; however, there are no studies on phenotypic plasticity in adulthood exposed to Bt-based bioinsecticide sub-doses. This study aimed to evaluate the morphometry of A. gemmatalis adults under laboratory conditions submitted to the Bt-based bioinsecticide Dipel® over the three generations. The body segments mensuread were width, length, and area of the anterior and posterior wings, the weight of the integument, chest, abdomen, wings, and the whole adult of males and females. Among the treatments, LC5 in the first generation and LC10 in the second generation were those with lower thresholds in relation to the weight of the chest and abdomen, considering the proportions of the body smaller than the females. The female's weight adulthood was reduced by 10% about males, and, only in the first generation. Males have larger body size and more pronounced phenotypic plasticity than females. Here, we demonstrate the first study assessing the phenotypic plasticity of A. gemmatalis adults.

Bacillus thuringiensis Berliner, 1915, é uma bactéria utilizada no controle biológico dessa espécie de praga. Populações resistentes e seus efeitos subletais causados pelo uso da bactéria já foram relatados, no entanto, não há estudos sobre a plasticidade fenotípica na idade adulta exposta a subdoses de bioinseticida à base de Bt. Este trabalho teve como objetivo avaliar a morfometria de adultos de A. gemmatalis em condições de laboratório submetidos ao bioinseticida Dipel® ao longo de três gerações. Os segmentos corporais mensuráveis eram largura, comprimento e área das asas anterior e posterior, o peso do tegumento, tórax, abdômen, asas e todo o adulto de machos e fêmeas. Dentre os tratamentos, CL5 na primeira geração e CL10 na segunda geração foram aqueles com limiares mais baixos em relação ao peso do tórax e abdômen, considerando as proporções do corpo menores que as do sexo feminino. O peso da fêmea na idade adulta foi reduzido em 10% em relação aos machos e, apenas na primeira geração. Os machos têm tamanho corporal maior e plasticidade fenotípica mais pronunciada do que as fêmeas. Este estudo demonstra o primeiro estudo avaliando a plasticidade fenotípica de adultos de A. gemmatalis.

This experiment evaluated reproductive and productive responses of beef cows receiving self-fed low-moisture blocks (LMB) enriched or not with Ca salts of soybean oil (CSSO) throughout the breeding season. Non-pregnant, suckled multiparous Angus-influenced cows were assigned to a fixed-time artificial insemination (AI) protocol (day -10 to 0) followed by natural service (day 15-70). Cows were managed in 12 groups (46 ± 4 cows/group) maintained in individual pastures, and groups received LMB enriched with 25 % (as-fed basis) of CSSO or ground corn (CON) from day - 10 to 100. Both treatments were designed to yield a daily LMB intake of 0.454 kg/cow (as-fed basis). Cows receiving CSSO had greater (P < 0.01) mean concentrations of ω-6 fatty acids in plasma samples collected on days 0 and 55. Cows receiving CSSO had greater (P = 0.05) pregnancy rate to fixed-time AI (67.2 vs. 59.3 %), whereas final pregnancy rate did not differ (P = 0.92) between treatments. Pregnancy loss was less (P = 0.03) in CSSO cows (4.50 vs. 9.04 %), which also calved earlier during the calving season (treatment × week; P = 0.04). Weaning rate tended to be greater (P = 0.09) in CSSO (84.8 vs. 79.4 %), although calf weaning age and weight did not differ (P ≥ 0.72) between treatments. Kilos of calf weaned/cow exposed was greater (P = 0.04) in CSSO cows (234 vs. 215 kg). Therefore, supplementing CSSO to beef cows via LMB during the breeding season improved their reproductive and overall productivity during a cow-calf cycle.

This research was carried out aiming at evaluating the effects of nitrate and ammonium ions on nutrient accumulation, biochemical components and yield of Italian zucchini (cv. Caserta) grown in a hydroponic system under salt stress conditions. The experiment was carried out in a greenhouse utilizing an experimental design in randomized blocks, arranged in a 2 x 5 factorial scheme, with 4 replications. The treatments consisted of two forms of nitrogen (nitrate - NO3- and ammonium - NH4+) and 5 electrical conductivity levels of irrigation water (ECw) (0.5, 2.0, 3.5, 5.0 and 6.5 dS m-1). The analysis of the results indicated that supply of N exclusively in NH4+ form promotes greater damage to the leaf membrane and reduction in accumulation of macronutrients and higher Na+/K+, Na+/Ca++ and Na+/Mg++ ratios in the shoots of zucchini plants. Electrical conductivity of irrigation water above 2.0 dS m-¹ reduces the accumulation of nutrients in shoot and yield of Italian zucchini plant. The toxicity of NH4+ under Italian zucchini plants overlap the toxicity of the salinity, since its fertilization exclusively with this form of nitrogen inhibits its production, being the NO3- form the most suitable for the cultivation of the species.

Caserta) cultivada em sistema hidropônico sob estrese salino. O experimento foi conduzido em casa de vegetação utilizando o delineamento experimental em blocos casualizados, arranjados em esquema fatorial 2 x 5, com 4 repetições. Os tratamentos foram constituídos de duas formas de nitrogênio (nitrato - NO3- e amônio - NH4+) e cinco níveis de condutividade elétrica da água de irrigação (CEa) (0,5; 2,0; 3,5; 5,0 e 6,5 dS m-¹). The objective of this study was to characterize plasma concentrations of pregnancy-associated glycoprotein (PAG) and progesterone (P4) as predictors of twins and pregnancy loss in Holstein cows with high-risk pregnancies. High-risk pregnancy was characterized using transrectal ultrasonography 37 d after artificial insemination (AI) based on the following criteria: small embryo size <15 mm, n = 10), slow heartbeat (<60 beats per minute, n = 11), and extra-amniotic membrane (additional amniotic membrane, n = 3). A cohort of twins (n = 41) diagnosed at d 37 post-AI was also enrolled. Each high-risk and twin pregnancy cow was paired with a cow of the same parity carrying a normal singleton at d 37 post-AI (control, n = 65). Blood samples were collected to measure PAG and P4 at d 37, 44, and 51 post-AI. Statistical analysis was performed using ANOVA, logistic regression, and receiver operator characteristic (ROC) curve with JMP software (SAS Institute Inc., Cary, NC). Pregnancy loss at d 51 post-AI was greater in high-risk pregnancies than in controls and twins (control = 1.5%; high-risk = 87.5%; twins = 12.2%). Concentration of PAG at d 37 post-AI did not differ among groups (control = 5.3 ± 0.7; high-risk = 4.8 ± 1.2; twins = 4.0 ± 0.9 ng/mL). Concentration of P4 at d 37 post-AI was greater in twins than in high-risk pregnancies and control, and lesser in high-risk pregnancies than in controls (control = 7.0 ± 0.3; high-risk = 5.9 ± 0.4; twins = 8.4 ± 0.3 ng/mL). Regression and ROC analysis for PAG at d 37 post-AI did not find a threshold to predict pregnancy loss or twins. Regression and ROC analysis for P4 at d 37 post-AI found that a threshold of 6.5 ng/mL predicted pregnancy loss with an area under the curve (AUC) of 0.64, and a threshold of 7.2 ng/mL predicted twins with AUC of 0.71. In summary, pregnancy loss and twins were predicted with only moderate accuracy by P4 concentration at d 37 post-AI, and the variability in PAG concentrations at d 37 post-AI was not sufficient to generate a threshold to predict pregnancy loss and twins in Holstein lactating cows.

Insecticidal gene silencing by RNA interference (RNAi) involves a post-transcriptional mechanism with great potential for insect control. Here, we aim to summarize the progress on RNAi research toward control of insect pests in the Neotropical region and discuss factors determining its efficacy and prospects for pest management. We include an overview of the available RNAi information for Neotropical pests in the Lepidoptera, Coleoptera, Diptera, and Hemiptera orders. Emphasis is put on significant findings in the use of RNAi against relevant Neotropical pests, including diamondback moth (Plutella xylostella L.), Asian citrus psyllid (Diaphorina citri Kuwayama), and the cotton boll weevil (Anthonomus grandis Boheman). We also examine the main factors involved in insecticidal RNAi efficiency and major advances to improve screening of lethal genes, formulation, and delivery. Few studies detail resistance mechanisms to RNAi, demonstrating a need for more research. Advances in formulation, delivery, and resistance management tools for insecticidal RNAi in the Neotropics can provide a basis for efficient field application.

The main aim of this study was to define the optimal adsorption and elution conditions for the purification of human immunoglobulin G (IgG) by mixed-mode chromatography using the multimodal resin Capto MMC. To this end, Central Composite Experimental Design (ED) was performed for both the adsorption and desorption stages. In the first case, the conditions were systematically studied in batch mode while in the latter case, these were performed in column. For both studies, the experimental design was conducted using high-purity human IgG samples. Buffer pH and concentration as well as the salt concentration were the parameters under study in the ED. Adsorption kinetics and equilibrium experiments were performed under the best conditions defined in the ED (phosphate buffer 60 mmol/L, pH 6.75, no salt). The equilibrium experimental data were fit to the Langmuir equation, with maximum uptake qmax equal to 549.2 mg/g. The qmax value found for IgG in Capto MMC was quite high as compared to other chromatographic techniques that employ single modes of interaction. Regarding elution, the best conditions were obtained with acetate buffer (56.40 mmol/L), pH 5.2 and 0.2 mol/L NaCl. An ultimate recovery of 46.96% for high-purity IgG was achieved. Thus, the effectiveness of Capto MMC for IgG adsorption and recovery could be confirmed. Moreover, electrophoretic runs in the human serum indicated that although co-elution of HSA and IgG proteins occurs, substantial HSA removal and a high IgG recovery were achieved in the elution step.

Abstract Fruit flies (Diptera: Tephritidae) are considered the main fruit pests worldwide. In Brazil, two species are predominant: the South American fruit fly, Anastrepha fraterculus and the Mediterranean fruit fly, Ceratitis capitata. In this study, we evaluated the effect of artificial diets with variable pH in their larval development and adult performance. The experiments were carried out in the laboratory at 25 ± 2 °C, 70 ± 10% RH and 12:12h (L:D) photoperiod. Semisolid diets with pH values of 6.0, 5.0, 4.0, 3.0, 2.0, 1.5, and 1.0, adjusted by adding hydrochloric acid were tested. Results indicated that the diet with pH 6.0 did not support larval development of both species of fruit fly. Diets with greater acidic pH values did not allow egg, larvae or pupae development and adult reproduction of A. fraterculus. For C. capitata , the pH of artificial diet exerts greater influence compared to A. fraterculus on the duration and viability of the larval stage, number of pupae, sex ratio and longevity of males.


Resumo As moscas-das-frutas (Diptera: Tephritidae) são consideradas as principais pragas da fruticultura mundial. No Brasil, duas espécies são predomindantes: a mosca-das-frutas Sul-americana, Anastrepha fraterculus e a mosca-do-Mediterrâneo, Ceratitis capitata. Neste estudo avaliamos o efeito de dietas artificiais com pH variável no seu desenvolvimento larval e performance de adultos. Os experimentos foram realizados em laboratório a 25 ± 2 °C, 70 ± 10% UR e fotoperíodo de 12:12 horas. Foram testadas dietas semi-sólidas com valores de pH de 6,0; 5,0; 4,0; 3,0; 2,0; 1,5 e 1,0, ajustados pela adição de ácido clorídrico. Os resultados indicaram que a dieta com pH 6,0 não suportou o desenvolvimento larval de ambas as espécies de mosca-das-frutas. As dietas com pH ácido não permitiram o desenvolvimento de ovos, larvas ou pupas e a reprodução de adultos de A. fraterculus. Para C. capitata o pH da dieta artificial exerceu maior influência do que para A. fraterculus nos parâmetros de duração e viabilidade do estágio larval, número de pupas, razão sexual e longevidade de machos.

Emerging data highlighting gut microbiome influences on health support evaluation of how microbial fermentation end-products influence postabsorptive systems. This study aimed to investigate the effect of increased propionate status on progesterone profiles and insulin sensitivity in dairy heifers. Eleven Holstein heifers, synchronized in estrus, were assigned to one of two continuous, 5-day IV treatments: sodium propionate (PRO; n = 5) or saline (CON; n = 6). These infusions culminated in a hyperglycemic clamp with daily blood samples for an additional 7 days. Plasma propionate concentrations increased over the first 9 h in PRO heifers, then decreased until day 3 when they matched CON heifers. Maximum plasma progesterone concentrations tended to be greater in PRO heifers than CON heifers (4.19 vs 3.73 ng/mL; P = 0.087). Plateau insulin concentrations in CON animals were significantly greater than those in PRO animals (249.4 ± 25.1 vs 123.9 ± 35.8; P = 0.008) with a trend for an increased insulin sensitivity index in PRO heifers compared to CON heifers (P = 0.06). These changes in plasma propionate clearance leading to increased progesterone response and changes in insulin sensitivity suggest a role for SCFA metabolism in reproductive hormone regulation.

This study investigated oviposition of Anastrepha fraterculus (Wiedemann, 1830) and Ceratitis capitata (Wiedemann, 1824) and its relation with the pericarp of citrus fruits. We evaluated the relationship between depth of oviposition of A. fraterculus and C. capitata and epicarp thickness of orange [Citrus sinensis (L.) Osbeck)] 'Navelina' and tangerine [C. reticulata (L.)] 'Clemenules' and the influence of fruit mesocarp of tangerine 'Clemenules' on oviposition of these species. The study was conducted under controlled conditions of temperature (25 ± 2 °C), relative humidity (70 ± 10% RH) and photophase (12 h). A. fraterculus and C. capitata laid their eggs in the flavedo region of orange 'Navelina' and between the albedo and flavedo of tangerine 'Clemenules'. When fruits with mesocarp exposed were offered, there was no oviposition by both fruit fly species. The results show that epicarp thickness of citrus fruits did not influence oviposition of A. fraterculus and C. capitata as oviposition did not occur only in the presence of the mesocarp, suggesting that other factors are involved in oviposition of these species.

Este estudo investigou a oviposição de Anastrepha fraterculus (Wiedemann, 1830) e Ceratitis capitata (Wiedemann, 1824) e sua relação com o pericarpo de frutos cítricos. Foi avaliada a relação entre a profundidade de oviposição de A. fraterculus e de C. capitata e a espessura do epicarpo dos frutos de laranjeira [Citrus sinensis (L.) Osbeck)] 'Navelina' e tangerineira [C. reticulata (L.)] 'Clemenules' e a influência do mesocarpo de frutos de tangerineira 'Clemenules' na oviposição destas espécies. O estudo foi conduzido em condições controladas de temperatura (25 ± 2 °C), umidade relativa (70 ± 10%) e fotofase (12 horas). A. fraterculus e C. capitata depositaram ovos no flavedo de frutos de laranjeira 'Navelina' e entre o flavedo e o albedo de frutos de tangerineira 'Clemenules'. Quando oferecido frutos com mesocarpo exposto, não houve oviposição por ambas as espécies de mosca. Os resultados demonstram que a espessura do epicarpo de frutos cítricos não influenciou a oviposição de A. fraterculus e de C. capitata, a qual não ocorreu na presença apenas do mesocarpo, sugerindo que outros fatores estão envolvidos na oviposição por estas espécies.

Lipopolysaccharide-induced tumor necrosis factor-α (LITAF) is a membrane protein that is highly dependent on correct location to exert transcription factor activity and protein quality control. In humans, LITAF, PIG7 (p53-inducible gene 7), and SIMPLE (small integral membrane protein of the lysosome/late endosome) refer to the same gene, which acts as a tumor suppressor. Several studies have shown that the transcription factor activity and nuclear translocation of LITAF protein are critical for the induction of several immune cells via classical pathways. In plants, LITAF protein corresponds to the plasma membrane protein AtGILP (Arabidopsis thaliana GSH-induced LITAF domain protein). The conservation of LITAF proteins across species and their putative role is still unclear. In this study, we investigate the LITAF-containing proteins, which we call GILP proteins, in Viridiplantae. We identified a total of 59 genes in 46 species, whose gene copies range from one to three. Phylogenetic analysis showed that multiple copies were originated via block duplication posteriorly to monocot and eudicot separation. Analysis of the LITAF domain of GILP proteins allowed the identification of a putative domain signature in Viridiplantae, containing a CXXCX41HXCPXC motif. The subcellular location for the majority of GILP proteins was predicted to be in the plasma membrane, based on a transmembrane domain positioned within the LITAF domain. In silico analysis showed that the GILP genes are neither tissue-specific nor ubiquitously expressed, being responsive to stress conditions. Finally, investigation of the GILP protein network resulted in the identification of genes whose families are known to be involved with biotic and/or abiotic stress responses. Together, the expression modulation of GILP genes associated with their plasma membrane location suggests that they could act in the signaling of biotic/abiotic stress response in plants.
