Monoamine oxidase activity, lipid peroxidation and membrane structure were tested after chronic stress and 7-day treatment with drugs in the brain cortex synaptosomal membranes of rats. The most potent corrector of stress-induced changes, as compared to hydrophobic antioxidant dibunol and tranquilizer phenazepam, was compound 3-OP, a hydrophilic antioxidant.

Comparative experimental investigation of albino male rats 3 and 16 months old has been performed. 16-month-old rats demonstrated age-dependent functional insufficiency in some behavioural tests. 2 month injection of 3-hydroxypyridine antioxidant (50 mg/kg/day) normalized memory function, motor skill learning and motor coordination function in 16-month-old rats.

The experiments on rats, using conflict situation method, have established that anxiolytic effect of 3-hydroxypyridine derivative is removed by bicucullin and picrotoxin, but not by Ro 5-3663, specific antagonist of benzodiazepine receptors Ro 15-1788 and inversive antagonist ethyl beta-carboline-3-carboxylate. The data obtained suggest that GABA-chloriontophor complex is involved into the realization of 3-hydroxypyridine anxiolytic effect.

Influence of membrane-active derivative of 3-hydroxypyridine on psychotropic activity of drugs with different chemical structure and type of action--neuroleptics (chlorpromazine, trifluoperazine, reserpine), tranquillizers (diazepam, phenazepam, calcium valproate), hypnotics (Hexobarbital), was studied in the experiment on animals. Application of psychotropic drugs after preliminary 3-hydroxypyridine administration was established to induce a considerable increase of their pharmacological activity--anxiolytic, sedative and hypnotic. Mechanism of action of 3-hydroxypyridine on pharmacological effects of psychotropic drugs is supposed to be caused by its membrane-modulating action, precisely by its influence on physical-chemical properties and phospholipid composition of synaptic biomembranes.

Experiments on mice and rats were made to study the effect of inhibitors-antioxidants (3-hydroxypyridine derivatives) on the central nervous system. It was established that 3-hydroxypyridine derivatives (50 to 200 mg/kg) possess a broad spectrum of psychotropic effects. They cause normalization of behavior in conflict situations, exert an antiaggressive and anticonvulsant action (antagonism with corazol), are capable of potentiating the hypnotic effect of barbiturates, and of suppressing the animals' motor activity if given in high doses. Most compounds under study have antihypoxic and antiamnestic effects.
