RESUMO

Understanding the role of 3CLpro protease for SARS-CoV-2 replication and knowing the potential of flavonoid molecules like rutin, myricetin, and baicalein against 3CLpro justify an investigation into their inhibition. This study investigates possible bonds and reactivity descriptors of rutin, myricetin, and baicalein through conformational and electronic properties. Density functional theory was used to determine possible interactions. Analyses were carried out through the molecular electrostatic potential, electron localization function, Fukui function descriptors based on frontier orbitals, and non-covalent interactions. A docking study was performed using a resolution of 1.55 Å for 3CLpro to analyze the interactions of rutin, myricetin, and baicalein. Scores of structures showed that rutin is the best ligand, followed by myricetin and baicalein. Docking studies showed that baicalein and rutin can establish effective interactions with residues of the catalytic dyad (Cys145 and His41), but just rutin forms a hydrogen bond. Myricetin, in turn, could not establish an effective interaction with Cys145. Baicalein interaction arose with active residues such as Arg188, Val186, Gln189, and Gln192. Interactions of rutin and myricetin with Arg188 and Gln189 were also found. A critical interaction was observed only for rutin with the hydroxyls of ring A with His41, and also for Cys145 with rings B and C, which is probably related to the highest score of rutin.

RESUMO

(1) Measuring usual iodine intake is a complex task due to the food consumption variability and its natural concentration in food. Therefore, the use of covariates to adjust statistical methods to estimate usual intake could improve the estimates obtained through dietary surveys. This study aims to evaluate the influence of salt and seasoning usage covariates on the estimates of usual iodine intake and the prevalence of its inadequacy. (2) A cross-sectional study was conducted with Brazilian pregnant women's food consumption data obtained with 24-h recall (n = 2247). The usual iodine intake was adjusted for intraindividual variability, supplement use, temporal effects, data collection methods, and sociodemographic characteristics with the tool UCD/NCI SIMPLE in the SAS software. Then, salt and seasoning usage covariates were used to adjust the distribution. The harmonized intake reference values for populations were used to assess intake adequacy. (3) The adjustments for salt and seasoning usage yielded a higher mean of usual iodine intakes. The only exception was the adjustment for the "habit of adding salt to meals after preparing/cooking", which produced a lower mean of usual intake and increased the prevalence of insufficient intake. (4) Salt and seasoning usage covariates affect the estimates evaluated. However, more studies are necessary to evaluate the influence observed.

RESUMO

Metabolic syndrome (MetS) has been prevalent among adolescents. The association between the concentration of inflammatory markers and the individual components of the metabolic syndrome indicates that inflammation, when there is no recent or ongoing disease, mediated by an inflammatory process, is an event that may precede the development of metabolic disorders in teenagers. The objective of this study is to verify the association of inflammatory biomarkers with the components of metabolic syndrome in adolescents. From a search of 3 databases, 13 articles met the study inclusion criteria. Two investigators independently extracted data from included studies. The evaluated inflammatory biomarkers are related to the components of MetS (insulin resistance, central and visceral obesity, arterial hypertension, dyslipidemia), which may increase the risk of developing the syndrome in adolescents. The results of this review are of clinical relevance, since the evaluation of inflammatory biomarkers in the presence of metabolic alterations can help to identify the risk factors that lead to the progression of MetS in adolescents.

RESUMO

OBJECTIVE: To investigate the effect of iodine supplementation during gestation on the neurocognitive development of children in areas where iodine deficiency is common. METHODS: Based on the PRISMA methodology, we conducted the search for articles in the PubMed, LILACS and Scopus databases, between March and April 2020, without limitation of dates. We used descriptors in English, Portuguese, and Spanish, without filters. Four clinical trials and four cohort articles were included in the review. RESULTS: The maximum supplementation was 300 µg of potassium iodide per day. The Bayley scale and Children's Communication Checklist-Short were used to assess neurodevelopment in children. There was no significant improvement in the children's mental development index and behavioural development index in the supplemented group; however, the psychomotor development index (PDI) showed improvement in the poorer gross motor skills. We found differences in the response time to sound in the supplemented group living in mild deficiency areas. CONCLUSION: Daily supplementation with iodine can improve poor psychomotor development of children living in mild to moderate iodine deficiency areas. Thus, it is necessary to perform further studies to assess the effect of supplementation on neurodevelopment before, during and after gestation in mild to moderate iodine deficiency areas.

RESUMO

OBJECTIVE: To determine the prevalence of insufficient iodine intake in pregnant women. MATERIALS AND METHODS: The search was performed in the electronic databases Medline (PubMed), Latin American and Caribbean Literature in Health Sciences (Lilacs) and Scopus. Review studies, experimental studies, those with adolescent pregnant women (< 20 years) and iodine supplementation were excluded. The selection followed the steps of identifying the articles in the databases, deleting the duplicates, and reading the titles, abstracts, and then the entire article. The search for the articles occurred in September 2017, using the descriptors "pregnant" and "iodine deficiency" NOT "supplementation" in English, Portuguese and Spanish. RESULTS: Thirteen articles were included, the deficiency prevalence ranged from 16.1% to 84.0%, and the median of iodine intake was insufficient in 75% of the studies. There is no classification for mild, moderate or severe levels of iodine deficiency in pregnant women, which makes it impossible to know the real dimension of the problem. CONCLUSION: The high prevalence of insufficient iodine intake in pregnant women, observed worldwide, shows the need for a population classification in order to direct public policies. RESUMO

Nanomaterials based on graphene oxide nanoribbons (GNR) and nanohydroxyapatite (nHAp) serve as attractive materials for bone tissue engineering. Herein, we evaluated the potential of nHAp/GNR toward in vitro analysis of specific genes related to osteogenesis and in vivo bone regeneration using animal model. Three different concentrations of nHAp/GNR composites were analyzed in vitro using a cytotoxicity assay, and osteogenic potential was determined by ALP, OPN, OCN, COL1, and RUNX2 genes and alkaline phosphatase assays. In vivo bone neoformation using a well-established in vivo rat tibia defect model was used to confirm the efficiency of the optimized composite. The scaffolds were nontoxic, and the osteogenesis process was dose-dependent (at 200 µg mL-1 of nHAp/GNR) compared to controls. The in vivo results showed higher bone neoformation after 15 days of nHAp/GNR implantation compared to all groups. After 21 days, both nHAp/GNR composites showed better lamellar bone formation compared to control. We attributed this enhanced bone neoformation to the high bioactivity and surface area presented by nHAp/GNR composites, which was systematically evaluated in previous studies. These new in vivo results suggest that nHAp/GNR composites can be exploited for a range of strategies for the improved development of novel dental and orthopedic bone grafts to accelerate bone regeneration.

Caprine Arthritis Encephalitis is a multisystemic infectious disease, caused by a lentivirus. The objective of this study was to evaluate the transmissibility of caprine lentivirus to goats and their offspring, through experimentally infected semen. Therefore, eleven free-CAEV goats were artificially inseminated using semen from a free-CAEV buck experimentally infected with CAEV-Cork strain (experimental group one). Pregnancy was confirmed in only six goats and their offspring (n=6) constituted the experimental group two. Two free-CAEV females were artificially inseminated with semen from the same seronegative buck, without viral inoculum to constitute the control group. The diagnosis of caprine lentivirus infection was performed using AGID, cELISA and nested-PCR. All females were monitored for 210 days after artificial insemination. Kids were immediately separated from their mothers after birth, and monitored at zero time, 15 days old and monthly until 12 months old. Regarding goat samples, 56.96% (9/159) were positive in cELISA, 24.05% (38/158) were positive in IDGA and none was positive in nested-PCR. Regarding to the offspring samples, 11.28% (15/133) and 5.26% (7/133) were positive in nested-PCR and IDGA, respectively, while no sample was positive in cELISA. The control group showed no positives in the three techniques. The positivity observed to nested-PCR may show its importance to identify infected, but seronegative animals, in late seroconversion situations. According to results, the transmission of caprine lentivirus to offspring and their mothers through infected semen is possible.(AU)

RESUMO

Cancer is currently in the spotlight due to their heavy responsibility as main cause of death in both developed and developing countries. Analysis of the epidemiological situation is required as a support tool for the planning of public health measures for the most vulnerable groups. We analyzed cancer mortality trends in Brazil and geographic regions in the period 1996 to 2010 and calculate mortality predictions for the period 2011 to 2030.This is an epidemiological, demographic-based study that utilized information from the Mortality Information System on all deaths due to cancer in Brazil. Mortality trends were analyzed by the Joinpoint regression, and Nordpred was utilized for the calculation of predictions.Stability was verified for the female (annual percentage change [APC] = 0.4%) and male (APC = 0.5%) sexes. The North and Northeast regions present significant increasing trends for mortality in both sexes. Until 2030, female mortality trends will not present considerable variations, but there will be a decrease in mortality trends for the male sex. There will be increases in mortality rates until 2030 for the North and Northeast regions, whereas reductions will be verified for the remaining geographic regions. This variation will be explained by the demographic structure of regions until 2030.There are pronounced regional and sex differences in cancer mortality in Brazil, and these discrepancies will continue to increase until the year 2030, when the Northeast region will present the highest cancer mortality rates in Brazil.

OBJECTIVE: To investigate the determinants of dyslipidemia in preschoolers. METHODS: A total of 227 preschoolers residing in an urban area of the city of Diamantina, Minas Gerais, Brazil were evaluated at age 5 years, using a cross-sectional design. Dietary intake from a food frequency questionnaire, anthropometric/biochemical parameters, and socioeconomic/behavioral information from a questionnaire were evaluated. 'Mixed diet', 'snack', and 'unhealthy' dietary patterns were identified using principal component analysis. The determinants of dyslipidemia were examined using Poisson regression analysis. RESULTS: The prevalence of dyslipidemia in this study was 65.19%. Preschoolers who less frequently consumed foods in the 'mixed diet' dietary pattern had a higher risk of high concentrations of low-density lipoprotein cholesterol (PR = 2.30; p = 0.004) when compared with those with more frequent consumption of the 'mixed diet' dietary pattern. Preschoolers whose mothers had lower levels of education presented a lower risk of high concentrations of low-density lipoprotein cholesterol (PR = 0.43; p = 0.003), and preschoolers who were overweight/obese presented with greater risk of high concentrations of low-density lipoprotein cholesterol (PR = 2.23; p = 0.003). CONCLUSION: The determinants of dyslipidemia identified in this study were less frequent consumption of foods in the 'mixed diet' dietary pattern, higher body mass index, and higher level of maternal education. This study shows that despite the young age of the group under study, they already present a high prevalence of dyslipidemia, which is an important risk factor for cardiovascular disease.

RESUMO

AIMS: This study aimed to describe the expression of oestrogen receptor (ER)α, ERß and aromatase in salivary gland adenoid cystic carcinoma (ACC). METHODS AND RESULTS: ERα, ERß and aromatase expression was analysed by immunohistochemistry in tissue microarray blocks from 38 cases of ACC and seven normal salivary glands. The intracellular localization and amount of total protein expression were investigated by immunofluorescence and western blotting in an ACC cell line. Western blotting analysis showed overexpression of ERα, ERß and aromatase in the ACC cell line; however, with immunofluorescence, only ERß was shown to be expressed in the nucleus. Immunohistochemistry revealed positive nuclear expression of ERß, positive cytoplasmic expression of aromatase and a lack of ERα expression as compared with normal salivary glands. CONCLUSIONS: The nuclear expression of ERß indicates that oestrogen may be active in ACC and possibly able to mediate E2-targeted gene transcription. This study strongly suggests that ERß may be involved in tumour progression, playing a role in tumour development, and thus corroborating the indication for ER antagonists in the clinical control of ACC. This study opens a new perspective on the potential use of anti-oestrogens and aromatase inhibitors as therapeutic agents against ACC.

RESUMO

The aim of this study was to evaluate the relationships between fasting glucose levels, hs-CRP, BMI and serum retinol and diet in elderly people. For this study, a cross-sectional, epidemiological, population-based format was adopted. One hundred and sixty-three individuals between 60 and 90 years of age participated; the participants were from different socioeconomic and health backgrounds, except for the individuals diagnosed with diabetes. Most subjects had serum concentrations in the reference ranges for all variables. There was not a significant relationship between fasting glucose and serum retinol or diet. There was a significant correlation between hs-CRP and BMI (r=0.22, p=0.0082) and hs-CRP and total calorie intake (p=0.0091), which reinforces the notion that an increased intake of calories results in being overweight and/or obese. Considering the importance of vitamin A in various physiological processes, more studies on these relationships are necessary to establish a recommendation of vitamin A for the treatment and prevention of changes in fasting glucose and obesity in the elderly population.

RESUMO

OBJECTIVE: To compare the effects of different prophylactic iron doses on the growth and nutritional status of non-anemic infants. METHODS: Prospective randomized study. Infants aged 5.0 to 6.9 months who met the inclusion criteria and showed capillary hemoglobin >or= 11 g/dL were randomly allocated into three groups who received the following prophylactic doses of iron supplement (ferrous sulfate): 1 mg/kg/day (n = 39); 2 mg/kg/day (n = 36); and 25 mg/week (n = 39). This supplementation was given during 16 weeks. Both weight and length were measured. The nutritional status was evaluated by comparing z scores for weight/age, length/age and weight/length based on the World Health Organization (2006) references. Morbidity information was collected during monthly visits. RESULTS: The groups showed similar nutritional status before supplementation. There were no differences in daily nutrient intake among groups. During the study, weight and length gain, and increments in anthropometric indices did not differ statistically among supplemented groups. The occurrence and duration of morbidity episodes did not differ statistically among groups. In general, improvements were observed in both weight/age and weight/length indices in the population under study, whereas length/age showed no differences before and after supplementation. CONCLUSION: Different prophylactic iron doses had no different effects on the growth and nutritional status of non-anemic infants.

MÉTODOS: Estudo do tipo prospectivo e randomizado. A suplementação durou 16 semanas.

This transversal study is aimed at diagnosing work accidents involving exposure to biological material occurred at the Brasília University Hospital, as well as at analyzing institutional preventive actions. The REPAT Work Accident Prevention Network electronic form was used for data collection. Data were collected during 2003 and 2004 and the statistical analysis was performed through computation of frequencies. Among the 2.000 to 2.011 workers in action at that time, 107 accidents were evidenced. The highest amount of them occurred in the morning, among women, and involved perforating lesion in hands, caused by needles and catheters in the Medical Clinic and emergency units. Due to the characteristics of these accidents, the preventive actions, which included training, visits to work places and individual orientation, need to be revised and expanded.

RESUMO

OBJECTIVE: To investigate risk factors for anemia in infants assisted by public health services. METHODS: In a cross-sectional study carried out in Viçosa, state of Minas Gerais, Brazil, 205 children from 6 to 12 months were evaluated. Socioeconomic, environmental and biological data were collected, as well as information on child's birth, nutritional status, maternal data, child health care practices, feeding practices, and iron supplementation. Diagnosis of anemia was based on hemoglobin levels under 11 g/dL, using a portable Hemocue photometer. To analyze variables associated with anemia, a hierarchical logistic regression model was used. RESULTS: The prevalence of anemia was 57.6%. Family income per capita less than 0.5 minimum wage, frequency of fruit intake less than daily and lack of iron supplementation increased the chance of anemia among infants. CONCLUSION: Adequate health and nutrition support to low income families, promotion of healthy nutritional habits and prescription of iron supplements are of great importance to prevent and manage anemia in infants assisted by public health services.
