RESUMO

Graphene oxide (GOX) has become attractive due to its unique physicochemical properties. This nanomaterial can associate with other dendrimers, making them more soluble and allowing better interaction with biomacromolecules. The present study aimed to investigate, by real-time microscopy, the behavior of human breast cancer cells exposed to particles of materials based on graphene oxide. The MCF-7 cell line was exposed to GOX, GOX associated with Polypropylenimine hexadecaamine Dendrimer, Generation 3.0-DAB-AM-16 (GOXD) and GOX associated with polypropyleneimine-PAMAM (GOXP) in the presence or absence of fetal bovine serum (FBS). GOX, GOXD and GOXP were taken up by the cells in clusters and then the clusters were fragmented into smaller ones inside the cells. Real-time microscopy showed that the presence of FBS in the culture medium could allow a more efficient internalization of graphene materials. After internalizing the materials, cells can redistribute the clumps to their daughter cells. In conclusion, the present study showed that the particles can adhere to the cell surface, favoring their internalization. The presence of FBS contributed to the formation of smaller aggregates of particles, avoiding the formation of large ones, and thus transmitted a more efficient internalization of the materials through the interaction of the particles with the cell membrane.

Octa-(3-chloropropryl)silsesquioxane was chemically modified with histidine (SSQ-H) and characterized by spectroscopy in the infrared region (FT-IR), X-ray diffraction (XRD), X-ray Dispersive Energy Spectroscopy (EDX), Scanning Electron Microscopy (SEM). The analytical properties of SSQ-H were tested regarding of Cu2+ and Zn2+ adsorption and as an electrochemical sensor for the detection of ascorbic acid. The metal sorption results indicate that maximum amount of Cu2+ and Zn2+ adsorbed (Nfmax) were 1.58 × 10-3 and 5.67 × 10-4 mol g-1, respectively. After Cu2+ and Zn2+ ion adsorption and interaction with potassium hexacyanoferrate (III), the investigated materials displayed electroactivity for ascorbic acid detection. The anodic peak currents responses of a graphite paste electrode containing CuHCFSSQ-H presented a linear response in the concentration range of 4.0 × 10-4 to 4.0 × 10-3 mol L-1 for ascorbic acid detection. The limit of detection was of 2.99 × 10-4 mol L-1, with an amperometric sensitivity of 1.69 µA/mol L-1. The intensity of the anodic peak currents of the graphite paste electrode containing ZnHCFSSQ-H in the concentration range of 9.0 × 10-5 to 9.0 × 10-4 mol L-1 also displayed a linear response. The limit of detection was of 6.76 × 10-5 mol L-1, with an amperometric sensitivity of 0.0206 A/mol L-1.

The aim of this study was to investigate the capacity of the host dendrimer DAB-Am-16 as a drug carrier to reduce the time required for the encapsulated naloxonaxine to establish an irreversible covalent bond with µ(1)-opioid receptor (resulting in a pharmacologically selective effect). The efficacy of dendrimer-naloxonazine nanocomplex (DNC) was studied in antinociception induced by convulsions elicited by intraperitoneal (IP) administration of pentylenetetrazole, and analgesia was measured by the tail-flick test. We found that animals showed increased tail-flick latencies following convulsions. Furthermore, acute pre-treatment (10 minutes) with DNC, but not with naloxonazine alone, antagonized post-ictal analgesia in comparison with control pre-treatment. However, naloxonazine treatment 24 hours before PTZ decreased post-ictal antinociception, but DNC failed to antagonize tonic-clonic seizure-induced analgesia. In addition, according to Racine's index of seizure severity, naloxonazine, DAB-Am-16 dendrimer or DNC did not influence seizure severity when administered either 10 minutes or 24 hours before PTZ. FROM THE CLINICAL EDITOR: This study characterizes the effect of a dendrimer-naloxonazine complex on µ1 receptor-mediated post-ictal antinociception in an animal model of seizure disorder.

In this work, a hydrophilic clay, Na-montmorillonite from Wyoming, USA, was rendered organophilic by exchanging the inorganic interlayer cations for hexadecyltrimethylammonium ions (HDTA), with the formulae of [(CH(3))(3)N(C(16)H(33))](+) ion. Based on fact that organo-clay has high affinities for non-ionic organic molecules, 1,3,4-thiadiazole-2,5-dithiol was loaded on the HDTA-montmorillonite surface, resulting in the 1,3,4-thiadiazole-2,5-dithiol-HDTA-montmorillonite complex (TDD-organo-clay). The following properties of TDD-organo-clay are discussed: selective adsorption of heavy metal ions measured by batch and chromatographic column techniques, and utilization as preconcentration agent in a chemically modified carbon paste electrode (CMCPE) for determination of mercury(II). The main point of this paper is the construction of a selective sensor, a carbon paste electrode modified with TDD-organo-clay, its properties and its application to the determination of mercury(II) ions, as this element belongs to the most toxic metals. The chemical selectivity of this functional group and the selectivity of voltammetry were combined for preconcentration and determination.
