RESUMO

AIM: To experimentally assess activity and safety of anti-anthrax intravenous immunoglobulin manufactured on standard technology. MATERIALS AND METHODS: Plasma from selected donors vaccinated with combined anthrax vaccine was tested by enzyme immunoassay. Samples of plasma with increased titer of anti-anthrax antibodies were merged in one manufacturing load and fractionated in ethanol at negative temperature according to standard technology. Formulation of intravenous immunoglobulin was manufactured according to standard technology of acid-enzyme hydrolysis. RESULTS: Proved medical technology of donors immune plasma fractionation provided 4 - 8-fold concentration of anti-anthrax antibodies. The finished product contained 5% of protein and was apyrogenic, non-toxic, thermostable, electrophoretically homogenous, had pH 6.65 and meet the requirements for manufacturing batches of human intravenous immunoglobulin. CONCLUSION: Protective effects of experimental human anti-anthrax immunoglobulin were comparable with control biological--equine anti-anthrax immunoglobulin for intramuscular use.

RESUMO

The influence of nanodiamonds synthesized by detonation on human red and white blood cells in vitro was studied. Nine nanodiamond samples produced by different manufacturers and two samples of modified nanodiamonds with high colloidal stability were used in the study. It was shown that nanoparticles cause the destruction of white cells and the hemolysis of erythrocytes. Possible mechanisms of the desctructive effect of detonation nanodiamonds on blood-forming elements are discussed.

RESUMO

The mice intoxication with intraperitoneal introduction of 1,3,5-trinitrobenzene and 2,4,6-trinitrotoluene causes an increase of methemoglobin and a decrease of glutathione level. But intoxication by polynitrobenzenes exerts no effect on the total level of SH-groups. A small increase in total activity of dehydrogenases of the pentose phosphate way in response to 2,4,6-trinitrotoluene intoxication is also revealed. The level of both lipid peroxidation, superoxide dismutase and catalase activity is not influenced by 2,4,6-trinitrotoluene and 1,3,5-trinitrobenzene.

RESUMO

Intraperitoneal administration of meta-nitrobenzoic, 3,5-dinitrobenzoic, 2,4,6-trinitrobenzoic and 3,5-dinitro-4-methylbenzoic acids to the white mice in a dose equal to LD50 has induced an increase in the methaemoglobin content in their blood. Total activity of dehydrogenases of pentosephosphate pathway, content of 2,3-diphosphoglycerate increase in response to the intoxication evoked by the mentioned acids. The acute intoxication does not practically change the activity of the key enzymes of antioxidant protection: superoxide-dismutase, catalase and glutathione peroxidase.
