RESUMO

Transparent protective coatings capable of preventing fog and dust accumulation have broad application prospect in photovoltaic systems, optical devices and consumer electronics. Although a number of superhydrophobic coatings have been developed for self-cleaning purpose over the past three decades, there is still a lack of surfaces that can simultaneously possess high transparency, remarkable superhydrophobicity, and excellent fog and dust resistance. In this study, we have prepared surfaces featuring sub-wavelength nanofiber cluster structures through a facile plasma etching method, and further modified the surface with liquid-like perfluoropolyether (PFPE) brushes. The prepared PFPE modified nanofibrous surface (PFPE-NS) exhibits superior optical transparency (transmittance 90.4 % ± 0.7 %) and water repellency, with a water contact angle as high as 171.0° ± 0.6° and sliding angle down to 0.5° ± 0.1° (5 µL). More importantly, benefitted from the nanofiber cluster structures and the slippery liquid-like surface chemistry, the adhesion and accumulation of fog droplets and dust particles on PFPE-NS is greatly inhibited. As a consequence, PFPE-NS can keep excellent optical clearness after 2 h fogging test and maintain an average transmittance above 87 % after 24 h dusting test. Our study provides a promising strategy through constructing liquid-like nanofibrous coating for optical protection that could be applicable in practical rainy, foggy, and dusty environments.

Super-liquid-repellent surfaces capable of preventing wetting with various liquids have tremendous application. However, high liquid repellency relies on surface texturing to minimize the solid-liquid interfacial contact, which generally results in impaired interface robustness and pressure resistance. Consequently, the surface tends to undergo a Cassie-Baxter to Wenzel wetting transition and loses liquid repellency under high-velocity liquid impact, especially for low-surface-tension liquids. Here, surface design through combining the nanoscale effect and doubly reentrant structure is demonstrated to solve the above challenge. By utilizing a facile colloidal lithography process, robust liquid repellent surfaces featuring nanoscale doubly reentrant (NDR) structures are constructed. The nanoscale features ensure sufficient triple contact line density at a low solid-liquid contact fraction to enhance the capillary force for liquid suspension. In conjunction with the doubly reentrant topography that maximizes the upward component of capillary force, such NDR surfaces enable an extremely robust solid-liquid-gas composite interface. As a result, the prepared NDR surface maintain excellent repellency upon high-velocity impact of various liquids, including ethylene glycol drops with a Weber number (We) above 306 and ethanol drops with a We of 57. The above findings can help the development of super-liquid-repellent surfaces applicable to harsh conditions of high-velocity liquid impact or high hydrostatic pressure.

Photoresist is the key material in the fabrication of micropatterns or microstructures. Tuning the surface wettability of photoresist film is a critical consideration in its application of microfluidics. In this work, the surface wettability tuning of acrylic resin photoresist by oxygen plasma or ultra-violet/ozone, and its aging performance in different atmospheres, were systematically studied. The chemical and physical characterizations of the surfaces before and after modification show a dramatic decrease in the C-C group and increase in surface roughness for oxygen plasma treatment, while a decrease of the C-C group was found for the UV/ozone treatment. The above difference in the surface tuning mechanism may explain the stronger hydrophilic modification effect of oxygen plasma. In addition, we found an obvious fading of the wettability tuning effect with an environment-related aging speed, which can also be featured by the decrease of the C-C group. This study demonstrates the dominated chemical and physical changes during surface wettability tuning and its aging process, and provides basis for surface tuning and the applications in microfluidics.

RESUMO

In the mid-90s, hepatitis B virus (HBV)-directed immune responses were for the first time investigated in detail and revealed suboptimal T-cell responses in chronic HBV patients. Based on these studies, therapeutic vaccination exploiting the antigen presentation capacity of dendritic cells to prime and/or boost HBV-specific T-cell responses was considered highly promising. Now, 25 years later, it has not yet delivered this promise. In this review, we summarise what has been clinically tested in terms of antigen targets and vaccine forms, how the immunological and therapeutic effects of these vaccines were assessed and what major clinical and immunological findings were reported. We combine the lessons learned from these trials with the most recent insights on HBV antigen presentation, T-cell responses, vaccine composition, antiviral and immune-modulatory drugs and disease biomarkers to derive novel opportunities for the next generation of therapeutic vaccines designed to cure chronic HBV either alone or in combination therapy.

Synthetic long peptide (SLP) vaccination is a promising new treatment strategy for patients with a chronic hepatitis B virus (HBV) infection. We have previously shown that a prototype HBV-core protein derived SLP was capable of boosting CD4+ and CD8+ T cell responses in the presence of a TLR2-ligand in chronic HBV patients ex vivo. For optimal efficacy of a therapeutic vaccine in vivo, adjuvants can be conjugated to the SLP to ensure delivery of both the antigen and the co-stimulatory signal to the same antigen-presenting cell (APC). Dendritic cells (DCs) express the receptor for the adjuvant and are optimally equipped to efficiently process and present the SLP-contained epitopes to T cells. Here, we investigated TLR2-ligand conjugation of the prototype HBV-core SLP. Results indicated that TLR2-ligand conjugation reduced cross-presentation efficiency of the SLP-contained epitope by both monocyte-derived and naturally occurring DC subsets. Importantly, cross-presentation was improved after optimization of the conjugate by either shortening the SLP or by placing a valine-citrulline linker between the TLR2-ligand and the long SLP, to facilitate endosomal dissociation of SLP and TLR2-ligand after uptake. HBV-core SLP conjugates also triggered functional patient T cell responses ex vivo. These results provide an import step forward in the design of a therapeutic SLP-based vaccine to cure chronic HBV.

Introducing spacers into pixelated electrowetting displays (EWDs) normally gives mechanical strengthening, while bringing undesired disturbance of water/oil interfacial dynamics. Hence, spacer array is a key pixel structure needs careful consideration in the design and fabrication of electrowetting displays. Here, we propose a spacer array, which is designed standing on the junction of adjacent pixel walls, fabricated by photolithography. The spacer array provides mechanical strength enhancement and reliable oil motion controllability. By optimizing the spacer distribution density, the EWD device may achieve 28% increase in open ratio (white area fraction) and withstand 60 N/mm2 pressure. This design of spacer array reasonably solves the contradiction between mechanical strength enhancement and optoelectronic performance in EWDs, providing potential applications in oil-water two-phase microfluidic devices.

Immunotherapy represents an attractive option for the treatment of chronic hepatitis B virus (HBV) infection. The HBV proteins polymerase (Pol) and HBx are of special interest for antigen-specific immunotherapy because they are essential for viral replication and have been associated with viral control (Pol) or are still expressed upon viral DNA integration (HBx). Here, we scored all currently described HBx- and Pol-derived epitope sequences for viral indispensability and conservation across all HBV genotypes. This yielded 7 HBx-derived and 26 Pol-derived reported epitopes with functional association and high conservation. We subsequently predicted novel HLA-binding peptides for 6 HLA supertypes prevalent in HBV-infected patients. Potential epitopes expected to be the least prone to immune escape were subjected to a state-of-the-art in vitro assay to validate their HLA-binding capacity. Using this method, a total of 13 HLA binders derived from HBx and 33 binders from Pol were identified across HLA types. Subsequently, we demonstrated interferon gamma (IFN-γ) production in response to 5 of the novel HBx-derived binders and 17 of the novel Pol-derived binders. In addition, we validated several infrequently described epitopes. Collectively, these results specify a set of highly potent T cell epitopes that represent a valuable resource for future HBV immunotherapy design.IMPORTANCE Multiple HBV-derived T cell epitopes have been reported, which can be useful in a therapeutic vaccination strategy. However, these epitopes are largely restricted to HLA-A*02, which is not dominantly expressed in populations with high HBV prevalence. Thus, current epitopes are falling short in the development of a global immunotherapeutic approach. Therefore, we aimed to identify novel epitopes for 6 HLA supertypes most prevalent in the infected population. Moreover, established epitopes might not all be equally effective as they can be subject to different levels of immune escape. It is therefore important to identify targets that are crucial in viral replication and conserved in the majority of the infected population. Here, we applied a stringent selection procedure to compose a combined overview of existing and novel HBV-derived T cell epitopes most promising for viral eradication. This set of T cell epitopes now lays the basis for the development of globally effective HBV antigen-specific immunotherapies.

Pegylated IFNα (PEG-IFN) is one of the treatment options for chronic HBV (CHB) patients. However, the high patient treatment burden and limited response rate together clearly ask for biomarkers to predict PEG-IFN response. Soluble CD14 (sCD14) is considered a marker for immune activation and has been shown to predict clinical outcome of HIV infection. However, studies on sCD14 in CHB infection are inconclusive, and its relationship with clinical outcome is largely unknown. Here, we measured sCD14 levels in CHB patients and investigated whether changes in sCD14 level related to PEG-IFN response. Serum sCD14 levels were determined in 15 healthy controls, 15 acute self-limited HBV, 60 CHB patients in different disease phases and 94 HBeAg+ CHB patients at week 0 and week 12 of a 52-week PEG-IFN treatment. Response to PEG-IFN treatment was defined as HBeAg seroconversion or HBeAg loss at 26 weeks post-treatment. The mean sCD14 level in acute HBV patients (3.0 µg/mL) was significantly higher than in CHB patients (2.4 µg/mL) and healthy controls (2.4 µg/mL). In CHB patients receiving PEG-IFN, a significant increase in sCD14 was found after 12-week treatment (median week 0:2.1 µg/mL; week 12:3.7 µg/mL). After 12-week treatment, the fold change (FC = w12/w0) in sCD14 was significantly higher in responders compared to nonresponders (HBeAg seroconversion: median FCresponder  = 2.1 vs FCnonresponder  = 1.6; HBeAg loss: median FCresponder  = 2.2 vs FCnonresponder  = 1.5). Receiver operating characteristic curves demonstrated that FC-sCD14wk12/wk0 levels can be of significant value as a stopping rule to select patients at week 12 who are not likely to benefit from further PEG-IFN treatment.

As a common physicochemical phenomenon, protonation can cause molecules, atoms or ions with lone-pair electrons to become charged, and can further cause some changes in their physical and chemical properties. Our study first focused on the molecular protonation process and accompanying transitions of the oil/water interface properties in an electric field. The relationship between the protonation degree increment and applied voltage was proposed as a guide for controlling the protonation via applying an electric field. Besides the protonation degree, the water solubility of the oily target molecule obviously increased at 30 V for 600 s along with electric field-driven protonation. At the same time, the electrical conductivity and the underwater interface wettability of oil phase transitioned. These property transitions are anticipated to guide the further improvement and updating of promising protonation functions.

Oil motion control is the key for the optical performance of electro-fluidic displays (EFD). In this paper, we introduced an extra pinning structure (EPS) into the EFD pixel to control the oil motion inside for the first time. The pinning structure canbe fabricated together with the pixel wall by a one-step lithography process. The effect of the relative location of the EPS in pixels on the oil motion was studied by a series of optoelectronic measurements. EPS showed good control of oil rupture position. The properly located EPS effectively guided the oil contraction direction, significantly accelerated switching on process, and suppressed oil overflow, without declining in aperture ratio. An asymmetrically designed EPS off the diagonal is recommended. This study provides a novel and facile way for oil motion control within an EFD pixel in both direction and timescale.

Background: Vaccination with synthetic long peptides (SLP) is a promising new treatment strategy for chronic hepatitis B virus (CHB). SLP can induce broad T-cell responses for all HLA types. Here we investigated the ability of a prototype HBV-core (HBc)-sequence-derived SLP to boost HBV-specific T cells in CHB patients ex vivo. Methods: HBc-SLP was used to assess cross-presentation by monocyte-derived dendritic cells (moDC) and BDCA1+ blood myeloid DC (mDC) to engineered HBV-specific CD8+ T cells. Autologous SLP-loaded and toll-like receptor (TLR)-stimulated DC were used to activate patient HBc-specific CD8+ and CD4+ T cells. Results: HBV-SLP was cross-presented by moDC, which was further enhanced by adjuvants. Patient-derived SLP-loaded moDC significantly increased autologous HBcAg18-27-specific CD8+ T cells and CD4+ T cells ex vivo. HBV-specific T cells were functional as they synthesized tumor necrosis factor-alpha and interferon-gamma. In 6/7 of patients blockade of PD-L1 further increased SLP effects. Also, importantly, patient-derived BDCA1+ mDC cross-presented and activated autologous T-cell responses ex vivo. Conclusions: As a proof of concept, we showed a prototype HBc-SLP can boost T-cell responses in patients ex vivo. These results pave the way for the development of a therapeutic SLP-based vaccine to induce effective HBV-specific adaptive immune responses in CHB patients.

Molecular self-assembly has become a popular tool to prepare nanomaterials with potential applications, such as ion-responsive detection of Hg(2+) in aqueous solutions. In this study, FFACD aromatic pentapeptides, whose N-terminuses were protected by carboxyl (Ac-FFACD) or a 9-fluorenylmethoxycarbonyl group (Fmoc-FFACD), were chosen as building blocks to produce nanostructures in solutions. Based on the preliminary determination of the critical aggregation concentration (CAC) of Ac-FFACD and Fmoc-FFACD aromatic pentapeptides in water, the order of magnitude of which is 10(-5) mol·L(-1), self-assembled spiral and networked nanowires can be easily obtained over a range of concentrations. These nanowires were characterized by transmission electron microscopy (TEM), scanning electron microscopy (SEM), and atomic force microscopy (AFM). The self-assembled spiral and networked nanowires were designed to be used as templates for preparing CdS quantum dots (QDs) in-situ at room temperature. The peptide-functionalized, nanowire-encapsulated CdS QDs can be used for rapid, sensitive, and selective detection of trace amounts of mercuric ions (Hg(2+)) in aqueous solutions. This method enables rapid, linear detection (the linear correlation coefficients are 0.9972 of ΔF = 257.09 + 3.58 cHg(2+) for Ac-FFACD and 0.9994 of ΔF = 48.13 + 32.96 cHg(2+) for Fmoc-FFACD) with the Hg(2+) limit of detection at 300.85 ng·L(-1) and 32.09 ng·L(-1) for Ac-FFACD and Fmoc-FFACD, respectively. The supramolecular, self-assembled nanowires, fabricated from the two aromatic pentapeptides and having encapsulated QDs, exhibit superior properties attributable to the large loading capacity and the coordination sites of these peptides with Hg(2+). These structures can serve as novel Hg(2+) sensors and have possible applications for detection of various targets in scientific and engineering systems.

Industrialization of electrofluidic devices requires both high performance coating laminates and efficient material utilization on large area substrates. Here we show that screen printing can be effectively used to provide homogeneous pin-hole free patterned amorphous fluoropolymer dielectric layers to provide both the insulating and fluidic reversibility required for devices. Subsequently, we over-coat photoresist using slit coating on this normally extremely hydrophobic layer. In this way, we are able to pattern the photoresist by conventional lithography to provide the chemical contrast required for liquids dosing by self-assembly and highly-reversible electrofluidic switching. Materials, interfacial chemistry, and processing all contribute to the provision of the required engineered substrate properties. Coating homogeneity as characterized by metrology and device performance data are used to validate the methodology, which is well-suited for transfer to high volume production in existing LCD cell-making facilities.

Honeycomb films with various building units, showing potential applications in biological, medical, physicochemical, photoelectric, and many other areas, could be prepared by the breath figure method. The ordered hexagonal structures formed by the breath figure process are related to the building units, solvents, substrates, temperature, humidity, air flow, and other factors. Therefore, by adjusting these factors, the honeycomb structures could be tuned properly. In this review, we summarized the development of the breath figure method of fabricating honeycomb films and the factors of adjusting honeycomb structures. The organic-inorganic hybrid was taken as the example building unit to discuss the preparation, mechanism, properties, and applications of the honeycomb films.

Influenza B viruses fall in two antigenically distinct lineages (B/Victoria/2/1987 and B/Yamagata/16/1988 lineage) that co-circulate with influenza A viruses of the H3N2 and H1N1 subtypes during seasonal epidemics. Infections with influenza B viruses contribute considerably to morbidity and mortality in the human population. Influenza B virus neutralizing antibodies, elicited by natural infections or vaccination, poorly cross-react with viruses of the opposing influenza B lineage. Therefore, there is an increased interest in identifying other correlates of protection which could aid the development of broadly protective vaccines. blast analysis revealed high sequence identity of all viral proteins. With two online epitope prediction algorithms, putative conserved epitopes relevant for study subjects used in the present study were predicted. The cross-reactivity of influenza B virus-specific polyclonal CD8+ cytotoxic T-lymphocyte (CTL) populations obtained from HLA-typed healthy study subjects, with intra-lineage drift variants and viruses of the opposing lineage, was determined by assessing their in vitro IFN-γ response and lytic activity. Here, we show for the first time, to the best of our knowledge, that CTLs directed to viruses of the B/Victoria/2/1987 lineage cross-react with viruses of the B/Yamagata/16/1988 lineage and vice versa.

Magnetic field effect on the phase transition at high temperature (from 50 °C) inside the magnetic field has been found in C14G2 (N-tetradecyllactobionamide)/C12EO4 (tetraethylene glycol monododecyl ether)/D2O system. The phase was transited quickly from lamellar phase to isotropic phases [bottom, micellar phase (L1 phase) and top, sponge phase (L3 phase)] induced by a magnetic field, which was demonstrated by (2)H NMR and FF-TEM measurements. The isotropic phases induced by magnetic field were not stable, and the upper L3 phase can recover to lamellar phase after being restored in a 55 °C thermostat outside the magnetic field for about one month. During the mechanism study, the C12EO4 molecule was proved to be the dominant component for the phase transition induced by the magnetic field, while the C14G2 molecule was the auxiliary and just affected the transition speed. The breaking and rebuilding of hydrogen bonds could play an important role in the phase transition and recovering. Moreover, the surfactant concentration had an effect on the speed of phase transiting and phase recovering. These observations could provide an understanding of the phase transition and also the applications for the controlled drug delivery system of bilayer membranes driving, induced by the magnetic field.

Spontaneous transformation of lamellar structures, such as multilamellar vesicles from micelles or unilamellar vesicles, is an important challenge in the field of amphiphile molecules, which may serve as models to understand biologically relevant bilayer membranes. Herein, we report a progressive self-assembly progress of N-tetradecyllactobionamide (C14G2) and tetraethylene glycol monododecyl ether (C12EO4) mixtures in aqueous solution. Increasing temperature or surfactant compositions causes spontaneous transformation from simple to high-level aggregates, i.e., from unilamellar vesicles, to coexisting multilamellar vesicles, terraced planar bilayers, and finally terraced planar bilayers. Deuterium nuclear magnetic resonance ((2)H NMR), freeze-fracture transmission electron microscopy (FF-TEM), and small-angle X-ray scattering (SAXS) measurements clearly demonstrate the spontaneously progressive self-assembly process. The interlamellar spacing (d) of the bilayers decreases from unilamellar vesicles to the terraced planar bilayers with an increase of the temperature or surfactant compositions. Lamellar samples consisting of terraced planar bilayers at higher temperature still show viscoelastic properties, being Bingham fluids, and both the viscoelasticity and yield stress increase with the composition and decrease with the temperature. The spontaneous transformation of the progressive self-assembly progress of C14G2 and C12EO4 aqueous mixtures is due to a balance of three driving forces, hydrophobic interactions, hydrogen bonding, and steric effects.

In conventional cationic-anionic (catanionic) surfactant mixtures with excess monovalent salt, two lamellar phase (L(alpha)) regions are usually observed in both the cation-rich and anion-rich solutions, and precipitates form when there is an equal molar ratio of the cationic and anionic surfactants. The phase- and temperature-dependent behavior of the calcium dodecyl sulfate (CDS)-tetradecyltrimethylammonium bromide (TTABr)-water system with excess CaBr(2) is reported. A birefringent L(alpha)-phase is observed in the cation-rich solution while the precipitates in both the anion-rich and the cation-rich regions. The introduction of the Ca(2+) ion is proposed to alter the electrostatic shielding of the surfactant headgroups. Stable vesicles were characterized by TEM and rheology. The formation of the vesicles is driven by electrostatic and hydrophobic interactions between the cationic and anionic surfactants. The results from TEM show that the temperature markedly influences the molecular interactions and changes the structure of molecular bilayers, leading to the instability of the vesicles. The vesicles were used to prepare microcrystals of calcium oxalate monohydrate (CaC(2)O(4) x H(2)O) by adding dimethyl oxalate to L(alpha) solution. X-ray diffraction and scanning electron microscopy indicate that the vesicle phases play an important role in affecting the formation and growth of the CaC(2)O(4) crystals. Namely, the microcrystals formed in the reaction are mainly bricklike (dodecahedrons) and starlike (icositetrahedrons). The two morphologies have not been previously observed in COM. Furthermore, they are larger than those prepared without surfactant, which may also explain a role for surfactants in calcium oxalate biomineralization.
