RESUMO

Chlorate and perchlorate compounds, used as herbicides, solid fuel propellants, and explosives, are increasingly recognized as pollutants in groundwater. Stable isotope characterization would permit both environmental monitoring of extent of remediation and forensic characterization. Stoichiometric reduction to chloride (greater than 98% yield), by Fe(II) for chlorate and alkaline fusion-decomposition for perchlorate, allows analysis by standard methods to give highly reproducible and accurate delta37Cl results (0.05/1000, 2 x standard error). Analysis of various compounds from different suppliers yielded delta37Cl values for chlorate samples near to +0.2/1000 (SMOC), but one has within-sample heterogeneity of 0.5/1000, possibly due to crystallization processes during manufacture. Results for perchlorate samples also are generally near +0.2/1000, but one is +2.3/1000 (SMOC). The initial results suggest that both forensic and environmental applications might be feasible.

A primary objective of this study was to determine whether the binary traits heifer pregnancy (HP) and subsequent rebreeding (SR) were heritable in an experimental population of Angus cattle. A second objective was to determine the nature of the additive genetic relationships among HP, SR, and stayability (S(5/1)) in the same population. Heifer pregnancy was defined as the observation of a heifer conceiving and remaining pregnant to palpation at 120 d, given exposure during the breeding season. Subsequent rebreeding was defined as the observation of a 2-yr-old conceiving and remaining pregnant to palpation at 105 d, given pregnancy as a yearling and exposure during the breeding season. Stayability was defined as the probability of a female having at least five calves, given she becomes a dam as a 2 yr old. Data were analyzed using a maximum a posteriori probit threshold model to predict breeding values on the liability scale and Method R procedures to estimate variance components in the determination of heritability (h2). Additive genetic groups were used in determining the additive genetic relationships among these fertility traits. Additive genetic groups were formed on one trait's breeding values and used in the prediction of another trait's breeding values. Analyses yielded h2 estimates that were out of the parameter space 8.5 and 46.3% for HP and SR, respectively, and 5.9% for the reestimation of S(5/1). The majority of point estimates outside the parameter space for SR converged toward 0, whereas those for HP and S(5/1) primarily converged toward 1. From the subsamples producing h2 estimates within the parameter space, average h2 for HP, SR, and S(5/1) were .21, .19, and .15, with standard deviations of .12, .14, and .08, respectively. The estimates of h2 indicate that HP and S(5/1) were heritable and should respond favorably to selection; however, SR did not appear heritable due to the large number of subsamples producing h2 estimates out of the parameter space. Fixed effect estimates for age of dam were significant for HP. From the analyses using additive genetic groups, the relationship among HP and S(5/1) appeared to be nonlinear. This potential nonlinear relationship seen between HP and S(5/1) indicates that selection for improved female fertility would be most effective by having predictions on both traits.

Data from inseminating 1,000 heifers consecutively with sexed sperm and 370 heifers with control sperm in 11 small field trials are summarized. Semen was from 22 bulls of unknown fertility of various beef and dairy breeds, and 6 inseminators participated. Freshly collected sperm were sexed using a MoFlo flow cytometer/cell sorter after staining sperm with the DNA-binding dye Hoechst 33342; the principle is that the bovine X chromosome has 3.8% more DNA than the Y chromosome. Accuracy approaching 90% males or females was achieved. There was little difference in pregnancy rates between sexed, unfrozen and sexed, frozen sperm. In 5 of 6 field trials, there was little difference in pregnancy rates between insemination doses of 1.0 to 1.5 x 10(6) versus 3.0 x 10(6) sexed, frozen sperm. In the most recent trials, pregnancy rates with sexed, frozen sperm were within 90% of unsexed, frozen controls that had 7 to 20 times more sperm/insemination dose; however, in a few trials, control pregnancy rates were substantially higher than with low doses of sexed sperm. There were too few inseminations per bull to test bull differences in pregnancy rates rigorously. Insemination of sexed, frozen sperm bilaterally into the uterine horns produced pregnancy rates similar to insemination into the uterine body in 4 of 5 field trials. Pregnancy rates among inseminators did not differ significantly. There was no excess embryonic death between 1 and 2 months of gestation with pregnancies from sexed sperm, and very few abortions occurred between 2 months of gestation and term. Although rigorous epidemiological studies remain to be done, calves resulting from sexed sperm appear to exhibit no more abnormalities than controls.
