RESUMO

Takotsubo syndrome (TTS) is a stress-induced cardiomyopathy, characterized by an increased concentration of catecholamines, free radicals, and inflammatory cytokines, endothelial dysfunction, and increased apoptotic activity. High doses of isoprenaline are used in animal models to induce Takotsubo (TT)-like myocardial injury. The aim of the study was to investigate the antiapoptotic effects of liraglutide in experimental TTS and its role in the NF-κB pathway. Wistar rats were pretreated with liraglutide for 10 days, and on days 9 and 10, TT-like myocardial injury was induced with isoprenaline. After the sacrifice on day 11, hearts were isolated for histopathological and immunohistochemical analysis. Liraglutide reduced isoprenaline-induced cardiomyocyte apoptosis by decreasing cleaved caspase-3 (CC3), BCL-2-associated X protein (BAX), and NF-κB and increasing B-cell lymphoma/leukemia-2 (BCL-2). An increase in NF-κB in isoprenaline-treated rats was in positive correlation with proapoptotic markers (BAX and CC3) and in negative correlation with antiapoptotic marker BCL-2. Liraglutide increased BCL-2 and decreased NF-κB, BAX, and CC3, preserving the same correlations of NF-κB to apoptotic markers. It is concluded that liraglutide protects cardiomyocytes against isoprenaline-induced apoptosis in experimental TT-like myocardial injury through downregulation of the NF-κB pathway.

Given the complex nature of the handball as a game, players are required to possess a distinct set of physical and physiological attributes to attain peak performance. With the countermovement vertical jump (CVJ) being widely implemented as a non-invasive and time-efficient testing modality in sports settings, the purpose of the present study was twofold: (a) to establish a CVJ profile of professional female handball players and (b) to examine differences in force-time metrics between starters and non-starters. Forty-two professional female handball players (e.g., SuperLeague) volunteered to participate in this study. Each athlete performed three maximum-effort CVJs with no arm swing while standing on a uni-axial force plate system sampling at 1,000 Hz. Independent t-tests were used to examine differences in each variable between starters and non-starters. The results revealed that starters attained superior performance within the eccentric phase of the CVJ when compared to non-starters, particularly in terms of eccentric peak velocity (-0.957 ± 0.242 vs. -0.794 ± 0.177 m·s-1), eccentric mean power (320.0 ± 77.7 vs. 267.1 ± 75.2 W), and eccentric peak power (929.0 ± 388.1 vs. 684.4 ± 214.2 W). While not reaching the level of statistical significance, moderate-to-large effect sizes were observed for concentric impulse, peak velocity, and mean and peak force and power, all in favor of players included in the starting lineup (g = 0.439-0.655). Overall, these findings suggest that at the top-tier level of handball competition, the ability to secure a spot in a starting lineup may be possibly influenced by the athlete's eccentric performance capabilities. Thus, the development of lower-body eccentric strength and power may positively impact on-court athlete performance and ultimately help the team secure the desired game outcome.

BACKGROUND: Nutrition recommendations in patients with type 2 diabetes mellitus (T2DM) are to consume rye or integral bread instead of white bread. A positive effect on glucoregulation has been achieved by enriching food with various biologically active substances of herbal origin, so we formulated an herbal mixture that can be used as a supplement for a special type of bread (STB) to achieve better effects on postprandial glucose and insulin levels in patients with T2DM. AIM: To compare organoleptic characteristics and effects of two types of bread on postprandial glucose and insulin levels in T2DM patients. METHODS: This trial included 97 patients with T2DM. A parallel group of 16 healthy subjects was also investigated. All participants were given 50 g of rye bread and the same amount of a STB with an herbal mixture on 2 consecutive days. Postprandial blood glucose and insulin levels were compared at the 30th, 60th, 90th and 120th min. A questionnaire was used for subjective estimation of the organoleptic and satiety features of the two types of bread. RESULTS: Compared to patients who consumed rye bread, significantly lower postprandial blood glucose and insulin concentrations were found in T2DM patients who consumed STB. No relevant differences were found among the healthy subjects. Subjectively estimated organoleptic and satiety characteristics are better for STB than for rye bread. CONCLUSION: STB have better effects than rye bread on postprandial glucoregulation in T2DM patients. Subjectively estimated organoleptic and satiety characteristics are better for STB than for rye bread. Therefore, STB can be recommended for nutrition in T2DM patients.

Background: Patients with ST-segment elevation myocardial infarction (STEMI) and COVID-19 infection have a worse clinical course and prognosis. The prognostic significance of the timing of STEMI in relation to COVID-19 infection was not investigated. Objectives: To assess whether the time of STEMI development in relation to COVID-19 infection (concurrent or following the infection) influenced the short-term prognosis. Methods: This was an observational study of consecutive COVID-19 patients with STEMI admitted to the COVID-hospital Batajnica (February 2021-March 2022). The patients were divided into the "STEMI first" group: patients with STEMI and a positive polymerase chain reaction test for COVID-19, and the "COVID-19 first" group: patients who developed STEMI during COVID-19 treatment. All patients underwent coronary angiography. The primary endpoint was in-hospital all-cause mortality. Results: The study included 87 patients with STEMI and COVID-19 (Mage, 66.7 years, 66% male). The "STEMI first" group comprised 54 (62.1%) patients, and the "COVID-19 first" group included 33 (37.9%) patients. Both groups shared a comparatively high burden of comorbidities, similar angiographic and procedural characteristics, and high percentages of performed percutaneous coronary interventions with stent implantation (90.7% vs. 87.9%). In-hospital mortality was significantly higher in the "COVID-19 first" group compared to the "STEMI first" group (51.5% vs. 27.8%). Following adjustment, the "COVID-19 first" group had a hazard ratio of 3.22 (95% confidence interval, 1.18-8.75, p = .022) for in-hospital all-cause death, compared with the "STEMI first" group (reference). Conclusion: Clinical presentation with COVID-19 infection, followed by STEMI ("COVID-19 first"), was associated with greater short-term mortality compared to patients presenting with STEMI and testing positive for COVID-19 ("STEMI first").

Background: Isoprenaline (ISO), a synthetic catecholamine and a ß-adrenoceptor agonist, is widely used to develop an experimental model of myocardial injury (MI) in rats. The leading hypothesis for ISO-induced MI in rats is that it results from catecholamine overstimulation, oxidative stress, inflammatory responses, and development of cardiomyopathy during ISO administration. Folic acid (FA) reduces oxidative stress, improves endothelial function and prevents apoptosis, thereby contributing to cardiovascular protection. This study aimed to investigate the potentially protective effect of FA pretreatment on ISO-induced MI in rats. Methods: For 7 days, adult male Wistar albino rats were pretreated with 5 mg/kg/day of FA. On the sixth and seventh days, MI in rats was induced by administering 85 mg/kg/day of ISO. Prooxidant markers in plasma samples, antioxidant capacity in erythrocyte lysates, cardiac damage markers, lipid profile, electrocardiography (ECG) and histopathological analysis were evaluated. Results: FA pretreatment significantly alleviated changes induced by ISO; it decreased the homocysteine and high-sensitivity troponin I level. FA moderately decreased the reactive oxygen species (ROS) levels (superoxide anion radical, hydrogen peroxide and thiobarbituric acid reactive substances) and improved the antioxidant activities of catalase, superoxide dismutase and reduced glutathione. ISO reduced the nitrite level and FA significantly alleviated this change. Conclusion: It can be concluded that FA, as a mild antioxidant, could be an appropriate cardioprotective substance in the rat model of ISO-induced MI.

Pre-post practice changes in countermovement vertical jump force-time metrics in professional male basketball players. J Strength Cond Res 37(11): e609-e612, 2023-Despite the countermovement vertical jump (CVJ) being one of the most popular noninvasive and time-efficient methods for monitoring neuromuscular status, there is a lack of scientific literature focused on examining fatigue-induced alterations in performance in elite athletes. Thus, the purpose of this study was to examine changes in force-time metrics pre-post practice in professional male basketball players. Seventeen athletes competing in first-tier and second-tier national basketball leagues in Europe participated in this study. While standing on a uniaxial force plate sampling at 1,000 Hz, each athlete completed 3 CVJs pre-practice and post-practice. The practice consisted of individual and team shooting drills, position-specific player development drills, 5-on-0 offensive actions, and 5-on-5 play, including full-court transition (∼2 hours). The findings reveal that pre-post practice changes in force-time metrics seem to be phase specific. Despite a trivial increase in eccentric mean force (920.4 ± 100.2, 929.4 ± 100.0 N), most changes were observed within the concentric phase of the CVJ. The concentric phase duration increased pre-post practice (0.233 ± 0.027, 0.242 ± 0.033 seconds), whereas concentric impulse (262.9 ± 18.8, 258.6 ± 21.6 N·s), peak velocity (2.93 ± 0.22, 2.86 ± 0.22 m·s-1), mean force (2052.4 ± 179.2, 2002.7 ± 188.2 N), mean power (3,165.5 ± 269.5, 3,030.9 ± 326.8 W), and peak power (5,523.4 ± 607.3, 5,246.6 ± 663.7 W) experienced a significant decrease. Moreover, alongside longer contraction time (0.663 ± 0.065, 0.686 ± 0.074 seconds), lower vertical jump height (41.0 ± 6.8, 38.9 ± 6.6 cm) and reactive strength index-modified (0.634 ± 0.113, 0.579 ± 0.111 m·s-1) values were observed post-practice. Overall, these findings may allow practitioners to detect fatigue-induced changes in CVJ force-time metrics in professional male basketball players that can ultimately improve the acute and longitudinal training-adaptation monitoring process.

NEW FINDINGS: What is the central question of this study? What are the biggest challenges in performing in vitro studies on isolated human umbilical arteries? What is the main finding and its importance? The protocols presented in this study indicate some potential outcomes important for interpretation of the vascular responsivities of human umbilical arteries and could be useful for planning future in vitro studies with human umbilical arteries. ABSTRACT: Human umbilical artery (HUA) preparations are of particular importance for in vitro studies on isolated blood vessels because their sampling is not risky for the patient, and they can provide the closest possible impression of changes related to the uteroplacental circulation during pre-eclampsia. Using organ bath techniques, useful experimental protocols are provided for measuring some pathophysiological phenomena in the vascular responses of HUAs. Several vasoconstrictors (serotonin, prostaglandin F and phenylephrine) and vasodilators (acetylcholine and minoxidil) were seleted for determination of their vasoactivity in HUAs. The role of L-type voltage-operated calcium channels and different types of potassium channels (KATP , BKCa and KV ) were assessed, as was the impact of homocysteine. Serotonin was confirmed to be the most potent vasoconstrictor, while acetylcholine and phenylephrine caused variability in the relaxation and contraction response of HUA, respectively. The observed increase in serotonin-induced contraction and a decrease in minoxidil-induced relaxation in the presence of homocysteine suggested its procontractile effect on HUA preparations. Using selective blockers, it was determined that KATP and KV channels participate in the minoxidil-induced relaxation, while L-type voltage-dependent Ca2+  channels play an important role in the serotonin-induced contraction. The presented protocols reveal some of the methodological challenges related to HUA preparations and indicate potential outcomes in interpreting the vascular effects of the investigated substances, both in physiological conditions and in the homocysteine-induced pre-eclampsia model.

Takotsubo syndrome (TTS) is an acute heart failure syndrome characterised by catecholamine-induced oxidative tissue damage. Punica granatum, a fruit-bearing tree, is known to have high polyphenolic content and has been proven to be a potent antioxidant. This study aimed to investigate the effects of pomegranate peel extract (PoPEx) pre-treatment on isoprenaline-induced takotsubo-like myocardial injury in rats. Male Wistar rats were randomised into four groups. Animals in the PoPEx(P) and PoPEx + isoprenaline group (P + I) were pre-treated for 7 days with 100 mg/kg/day of PoPEx. On the sixth and the seventh day, TTS-like syndrome was induced in rats from the isoprenaline(I) and P + I groups by administering 85 mg/kg/day of isoprenaline. PoPEx pre-treatment led to the elevation of superoxide dismutase and catalase (p < 0.05), reduced glutathione (p < 0.001) levels, decreased the thiobarbituric acid reactive substances (p < 0.001), H2O2, O2- (p < 0.05), and NO2- (p < 0.001), in the P + I group, when compared to the I group. In addition, a significant reduction in the levels of cardiac damage markers, as well as a reduction in the extent of cardiac damage, was found. In conclusion, PoPEx pre-treatment significantly attenuated the isoprenaline-induced myocardial damage, primarily via the preservation of endogenous antioxidant capacity in the rat model of takotsubo-like cardiomyopathy.

The paper provides the physicochemical analysis of galvanic sludge to determine the presence and concentration of toxic metals. Two sludges sampled from the same factory, but from different technological processes, alkaline galvanic sludge obtained from galvanizing process and acidic sludge generated from the chromium plating process were analyzed. Inductively Coupled Plasma - Optical Emission Spectrometry (ICP-OES) revealed increased concentrations of toxic heavy metal ions Zn2+, Cr3+, Ni2+ and Pb2+ in the sludge from the galvanizing process and Cr3+, Cu2+, Ni2+, Pb2+, Cd2+ and Zn2+ from the chroming process. Moreover, the sludges were further physicochemically characterized by Reflectance Fourier Transform InfraRed Spectrometry (FTIR), Scanning Electron Microscopy with Energy-dispersive X-ray Spectroscopy Analysis (SEM-EDX) and X-ray diffraction (XRD). The results of ICP-OES were corroborated by FTIR. Analysis of FTIR spectra revealed the specific bands indicating the existence of metal oxides in the analyzed sludges, as well as the presence of organic substances, i.e. solvents and surfactants, used in the electroplating process. The analysis was accomplished following international norms and confirmed the increased concentrations of heavy metal ions from both sludges. In line with the regulations of the Environmental Protection Agency (EPA), the results proved the hypothesis that galvanic sludge is hazardous waste.

The interaction of the SARS-CoV-2 spike (S) glycoprotein receptor-binding domain with the host-cell ACE2 receptor is a well-known step in virus infection. Neuropilin-1 (NRP-1) is another host factor involved in virus internalization. The interaction between S-glycoprotein and NRP-1 has been identified as a potential COVID-19 treatment target. Herein, the effectiveness of folic acid and leucovorin in preventing contact between S-glycoprotein and NRP-1 receptors was investigated using in silico studies and then confirmed in vitro. The results of a molecular docking study showed that leucovorin and folic acid had lower binding energies than EG01377, a well-known NRP-1 inhibitor, and lopinavir. Two hydrogen bonds with Asp 320 and Asn 300 residues stabilized the leucovorin, while interactions with Gly 318, Thr 349, and Tyr 353 residues stabilized the folic acid. The molecular dynamic simulation revealed that the folic acid and leucovorin created very stable complexes with the NRP-1. The in vitro studies showed that the leucovorin was the most active inhibitor of the S1-glycoprotein/NRP-1 complex formation, with an IC75 value of 185.95 µg/mL. The results of this study suggest that folic acid and leucovorin could be considered as potential inhibitors of the S-glycoprotein/NRP-1 complex and, thus, could prevent the SARS-CoV-2 virus' entry into host cells.

Type 2 diabetes mellitus (T2DM) increases the risk of cardiovascular disease, especially myocardial injury. Due to their hypoglycemic effects, glucagon-like peptide-1 receptor agonists (GLP-1RAs) are efficiently used for T2DM management. GLP-1RAs also have anti-inflammatory and antioxidative effects and can improve cardiac function. The aim of this study was to investigate the cardioprotective effects of liraglutide, a GLP-1RA, on isoprenaline-induced myocardial injury in rats. The study included four groups of animals. They were pretreated with saline for 10 days + saline on days 9 and 10 (control), saline for 10 days + isoprenaline on days 9 and 10 (isoprenaline group), liraglutide for 10 days + saline on days 9 and 10 (liraglutide group), and liraglutide for 10 days, and on days 9 and 10 isoprenaline was administered. This study evaluated ECG, myocardial injury markers, oxidative stress markers, and pathohistological changes. The results showed that liraglutide mitigated the isoprenaline-induced cardiac dysfunction recorded by ECG. Liraglutide reduced serum markers of myocardial injury such as high-sensitive troponin I, aspartate aminotransferase, alanine aminotransferase, reduced thiobarbituric acid reactive substances, increased catalase and superoxide dismutase activity, increased reduced glutathione level, and improved lipid profile. Liraglutide induced antioxidative protection and alleviated isoprenaline-induced myocardial injury.

The purpose of the present study was (i) to explore the reliability of the most commonly used countermovement jump (CMJ) metrics, and (ii) to reduce a large pool of metrics with acceptable levels of reliability via principal component analysis to the significant factors capable of providing distinctive aspects of CMJ performance. Seventy-nine physically active participants (thirty-seven females and forty-two males) performed three maximal CMJs while standing on a force platform. Each participant visited the laboratory on two occasions, separated by 24-48 h. The most reliable variables were performance variables (CV = 4.2-11.1%), followed by kinetic variables (CV = 1.6-93.4%), and finally kinematic variables (CV = 1.9-37.4%). From the 45 CMJ computed metrics, only 24 demonstrated acceptable levels of reliability (CV ≤ 10%). These variables were included in the principal component analysis and loaded a total of four factors, explaining 91% of the CMJ variance: performance component (variables responsible for overall jump performance), eccentric component (variables related to the breaking phase), concentric component (variables related to the upward phase), and jump strategy component (variables influencing the jumping style). Overall, the findings revealed important implications for sports scientists and practitioners regarding the CMJ-derived metrics that should be considered to gain a comprehensive insight into the biomechanical parameters related to CMJ performance.

INTRODUCTION: This study was created to analyze dynamic alterations in coagulation, hematological and biochemical parameters and their association with mortality of COVID-19 patients. To identify the most sensitive biomarkers as predictors of mortality more research is required. METHODS: The present study was a prospective, one-year-long observational study conducted on all critically ill, COVID-19 patients with respiratory failure. The following data were collected: demographic and clinical characteristics of the study population, comorbidities, coagulation, biochemical and hematological parameters. The primary outcome was the proportion of patients who died. RESULTS: 91 patients with median age 60 (50-67), 76.9% male, met the acute respiratory distress syndrome criteria. It was tested whether dynamic change (delta-Δ) of parameters that were found to be predictors of mortality is independently associated with poor outcome. Adjusted (multivariate) analysis was used, where tested parameters were corrected for basic and clinical patients characteristics. The only inflammatory parameter which dynamic change had statistically significant odds ratio was ΔCRP (p < 0.005), while among coagulation parameters statistically significant OR was found for Δ fibrinogen (p < 0.005) in predicting mortality. CONCLUSION: Monitoring of coagulation, hematological and biochemical parameters abnormalities and their dynamical changes can potentially improve management and predict mortality in critically ill COVID -19 patients.

The study aimed to test the hypothesis that homeostatic microbiome (HM) disorders lead to the increased indirect influence of certain microorganisms (MO) in the gastrointestinal tract, causing a disorder of insulin secretion, insulin resistance, and diabetes. We highlighted Candida and certain types of bacteria since previous in vitro research showed they significantly affect insulin secretion and can cause insulin resistance in obese patients with metabolic syndrome. After determining the type of MO present in the throat swab and the stool, the oral glucose tolerance test (OGTT) test, and analysis of glucose and insulin secretion were performed in patients (n = 38) who were positive for certain types of MO compared to negative patients. Finally, all patients were divided into two groups: overweight patients (body mass index [BMI] < 30) and obese patients (BMI > 30). These two groups were compared for the percentage of certain types of MO to determine which MO can affect an increase in obesity and BMI. The presence of Diphtheroids in the throat (60.5%) reduces insulin secretion in patients compared with the negative group (194.5: 332.4) and the difference was statistically significant (P = .030). The presence of Candida in the throat (10%) increases insulin secretion, but the difference was statistically insignificant. The presence of Candida in the stool (28.9%) also increases insulin secretion and the difference was statistically significant (P = .038). Cumulative results (throat + stool) were similar (180: 332, P = .022). Analysis of BMI showed that the percentage of Diphtheroids in the throat decreases with increased body weight (53.8: 75%) while the percentage of Candida (38.5: 8.3%) and Enterobacter (61.5: 25%) increases, but these differences were statistically insignificant (P > .05). Diphtheroids in the throat can reduce insulin secretion by synthesizing their metabolites. Candida albicans is a conditional pathogen and as a significant indirect factor induces increased insulin secretion and insulin resistance. There are indications that elevated levels of Candida in the intestinal system can cause increased body weight of patients. C albicans should be considered a new factor in the pathogenesis of diabetes.

The main objective of present study was to evaluate inter-rater reliability and concurrent validity of Side Hop Test stopwatch vs. force plates timing, and to determine the number of sessions and trials required to minimize the effects of learning on Side Hop Test total time and limb symmetry index. Fifteen healthy male physical education students (mean ± SD: age, 23 ± 3 years; height, 181 ± 9 cm; and weight 72 ± 6 kg) participated. Side Hop Test total time (stopwatch and force plates) of left and right leg, and limb symmetry index (force plates) were obtained over seven sessions conducted 5-7 days apart. Time recordings of two raters were similar (t = -0.56, p > 0.05) with high reliability (all ICC >0.99 and CV% <0.1) and no systematic bias when compared to force plate data (p > 0.05; for rater 1 and 2, respectively). Total time improved across the Sessions (F = 25.87, p < 0.01, ω 2 = 0.18) and Trials (F = 68.15, p < 0.01, ω 2 = 0.10), with no significant interaction between factors. No between-leg differences were detected (F = 0.52, p > 0.05, ω 2 = 0.001). Limb symmetry index ranged from 0.999 to 1.055 across all sessions and trials (all p > 0.05 and ω 2 < 0.00). Due to low coefficient of correlation, high interclass correlation coefficient, and the lack in heteroscedasticity, stopwatch measurements are valid to measure total time in the Side Hop Test. Moreover, stopwatch measurements could be reliably used to measure total time in the Side Hop Test, while the test could be administrated with only one experienced rater. Unlike total times, findings on limb symmetry index suggest it could be reliably assessed after seven familiarization sessions.

Over the past decade, force-velocity (F-v) profiling has emerged as a promising tool for assessing neuromuscular capacity to design individually tailored interventions in diverse populations. To date, a limited number of studies have addressed the optimization of the linear method for measuring F-v profiles of single-joint isokinetic movements. We aimed to simplify the measurement of knee extension (KE) and knee flexion (KF) isokinetic tasks by evaluating the most appropriate combination of two velocities (i.e., the 2-point method). Twenty-two healthy participants (11 males and 11 females) were included in the study. Isokinetic peak torque was measured at nine angular velocities (30-60-90-120-150-180-210-240-300°/s) and under isometric conditions (at 150° and 120° of KF for KE, and KF, respectively). Maximal theoretical force (F0), maximal theoretical velocity (v0), slope of the relationship (Sfv) and maximal theoretical power (Pmax) were derived from the linear F-v profiles of KE and KF and compared between the 9-point method and all possible combinations (36 in total) of the 2-point methods. The F-v profiles obtained from nine points were linear for KE (R2 = 0.95; 95% CI = 0.94-0.96) and KF (R2 = 0.93; 95% CI = 0.90-0.95), with F0 underestimating isometric force. Further analyses revealed great to excellent validity (range: ICCs = 0.89-0.99; CV = 2.54%-4.34%) and trivial systematic error (range: ES = -0.11-0.24) of the KE 2-point method when force from distant velocities (30°/s, 60°/s or 90°/s combined with 210°/s, 240°/s or 300°/s) was used. Similarly, great to excellent validity and trivial systematic error of the KF 2-point method for F0 and Pmax (range: ICC = 0.90-0.96; CV = 2.94%-6.38%; ES = -0.07-0.14) were observed when using the previously described combinations of velocities. These results suggest that practitioners should consider using more distant velocities when performing simplified isokinetic 2-point single-joint F-v profiling. Furthermore, the F-v profile has the potential to differentiate between the mechanical properties of knee extensors and flexors and could therefore serve as a potential descriptor of performance.

Cardiovascular diseases are the leading cause of death and the main cause of disability. In the last decade, homocysteine has been found to be a risk factor or a marker for cardiovascular diseases, including myocardial infarction (MI) and heart failure (HF). There are indications that vitamin B6 plays a significant role in the process of transsulfuration in homocysteine metabolism, specifically, in a part of the reaction in which homocysteine transfers a sulfhydryl group to serine to form α-ketobutyrate and cysteine. Therefore, an elevated homocysteine concentration (hyperhomocysteinemia) could be a consequence of vitamin B6 and/or folate deficiency. Hyperhomocysteinemia in turn could damage the endothelium and the blood vessel wall and induce worsening of atherosclerotic process, having a negative impact on the mechanisms underlying MI and HF, such as oxidative stress, inflammation, and altered function of gasotransmitters. Given the importance of the vitamin B6 in homocysteine metabolism, in this paper, we review its role in reducing oxidative stress and inflammation, influencing the functions of gasotransmitters, and improving vasodilatation and coronary flow in animal models of MI and HF.

BACKGROUND: Type 2 diabetes mellitus (T2DM) is commonly associated with hyperglycemia, dyslipidemia, oxidative stress and inflammation which are well known cardiovascular risk factors. Pomegranate peel polyphenols have a proven hypolipemic, antioxidant and anti-inflammatory activity. However, there is a lack of clinical studies that would confirm its antioxidant and anti-inflammatory effects in diabetic patients. The potential of pomegranate peel extract (PoPEx) to counteract inflammation and oxidative stress in T2DM patients was investigated. For this purpose, a randomized, double-blind placebo-controlled study involving adult T2DM patients treated with PoPEx or placebo for eight-weeks was conducted. METHODS: Patients were randomly divided into two groups: the first group (n = 30) received capsules containing PoPEx 250 mg twice daily, while the placebo group (n = 30) received placebo capsules twice daily. Plasma concentration of inflammatory factors (interleukin 6 (IL-6), tumor necrosis factor α (TNF-α) and high sensitivity C reactive protein (hsCRP)), oxidative stress biomarkers (thiobarbituric acid reactive substances (TBARS), nitrites (NO2-), superoxide anion radical (O2-), hydrogen peroxide (H2O2), total antioxidant capacity (TAC)), homocysteine and lipid profile were analyzed. RESULTS: The PoPEx treatment showed a significant reduction of inflammatory factors (IL-6, TNF-α, hsCRP), oxidative stress biomarkers (TBARS, NO2-, O2-) and homocysteine, while the TAC was increased. Moreover, a significant improvement in lipid profile was observed in the PoPEx group. Additional analysis showed a significant inverse correlation between the decrements of all measured inflammatory markers and TAC in the PoPEx group. CONCLUSIONS: The study demonstrated that eight-week-long PoPEx administration had favorable effects on inflammatory status and oxidative stress biomarkers in diabetic patients.

The present study aimed to explore the possibility of comprehensively assessing whole-body muscle strength by testing as few muscle groups as possible, using a single testing method (isometric or isokinetic dynamometry) and a single variable (maximal force or rate of force development). Knee, hip, shoulder and elbow extensors and flexors were evaluated in males with high (n = 26) and low strength levels (n = 32). The principal component analysis revealed three factors that explained 62.5% of the total variance, while the main factors were loaded by the different testing methods and strength variables for the muscles acting on the knee (first component), hip (second component) and arm joints (third component). These results were confirmed by a three-way ANOVA which revealed a significant factor of group (P < 0.001) and the interaction test type × group (P = 0.002), but not of test type (P = 0.644), muscle group (P = 0.999), or their interactions (P > 0.205). The correlations of strength outcomes across the muscles ranged from trivial to very large (r range = -0.17, 0.84), being generally higher for the antagonistic muscles. Overall, a comprehensive assessment of whole-body muscle strength can be obtained using isometric dynamometry and maximal force, but it should consider at least one muscle group from the antagonistic pair.
