RESUMO

The effects of the natural antioxidants-anthocyans and vitamin E (in a solubilized pharmaceutical form) on carbon tetrachloride-induced liver injury in rats are studied. The changes in the activity of serum transaminases (ALAT and ASAT), the content of the reduced glutathione and cytochrome P-450 as well as the intensity of the processes of lipid peroxidation are assessed. The anthocyans exert a protective effect comparable to that of vitamin E on liver cells. The favorable effects of the combination of the antioxidants on the content of the reduced glutathione and on the processes of lipid peroxidation are more intensely expressed. The morphological changes occurring in hepatocytes correlate with the results of the biochemical studies. It is evident that both substances have a marked hepatoprotective activity.

Studies were carried out on male white mice to investigate the influence of cantocyans on pain both singly and in combination with the classic analgetics. The following tests were used: the test for narcotic analgetics "hot plate" and the analgetic morphine, administered subcutaneously in sub- and nonanalgetic doses (10 and 5 mg/kg of body mass respectively); the test for nonnarcotic analgetics-chemical peritoneal stimulation with 3% of acetic acid and the analgetic analgin, administered subcutaneously in a nonanalgetic dose (100 mg/kg of body mass). The results from the experiments showed that antocyans did not possess its own analgetic effect in both of the experimental models. However their combination with morphine and analgin, administered in noneffective analgetic doses, caused manifested analgetic effect of the combinations in all used doses of antocyans (50, 100 and 200 mg/kg of body mass). The possibility for therapeutic usage of analgetic combinations of antocyans and narcotic and nonnarcotic analgetics is discussed.

The effects of an pretreatment course (2 x 5 days) combining vitamin E 20 mg/kg, anthocyans 50 mg/kg, and pyracetam 200 mg/kg on animal survival after lethal irradiation (8 Gy) and blood formation after sublethal irradiation (6 Gy) were studied in mice. Assessments were based on records of an integral criterion, 30-day survival, of spleen weight and cellularity, and of bone-marrow cellularity. Pretreatment with the mixture distinctly raised survival rate (50%) in the protected population. The effect observed was shown to result from potentiating interactions between mixture components. Blood-forming organs experienced less radiation damage, and recovery processes in them were stimulated.

The toxic and antitumor properties of biocysplantinum (cis-diaminodichlo-platinum) as well as a new platinum complex with acetic acid after their single and combined usage with antocyanes were studied on healthy mice and mice with transplanted ascitic tumour of Ehrlich. There was a reduction in total toxicity of biocysplatinum after simultaneous administration with antocyanes (300 mg/kg per os). Protective effect of antocyanes was observed on leukocytopoiesis. Preservation of antitumor activity of platinum complexes was found in the tumour of Ehrlich. The obtained data indicate possibilities for usage of antocyanes as chemioprotectors during therapy with drugs, containing platinum.
