RESUMO

Soy molasses is rich in oligosaccharides like sucrose, stachyose, and raffinose, with stachyose and raffinose being functional oligosaccharides. Harnessing soy molasses for the production of functional soy oligosaccharides (FSO) can significantly elevate its value. Biological purification, a method leveraging the selective utilization of different carbon sources by microorganisms, allows for the specific removal of sucrose from soy molasses while preserving stachyose and raffinose, thereby increasing the FSO content. This research identified a yeast named YT312 with strong purification capabilities for soy molasses and optimized the purification conditions. The study revealed that yeast YT312 was Wickerhamomyces anomalus, exhibiting a broad range of growth temperatures and pH levels alongside a high tolerance to glucose, sucrose, and NaCl. Through single-factor and orthogonal experiments, it was established that under specific conditions-0.375% inoculum size, 30 °C fermentation temperature, 150 rpm shaking speed, 10-fold dilution ratio, pH of 7, and 12 h of fermentation-sucrose was completely removed from soy molasses, while functional raffinose and stachyose were retained at rates of 96.1% and 90.2%, respectively. Consequently, W. anomalus YT312 displayed exceptional characteristics for the biological purification of soy molasses and the production of FSO.

Introduction: In recent years, research has used psycholinguistic features in public discourse, networking behaviors on social media and profile information to train models for depression detection. However, the most widely adopted approach for the extraction of psycholinguistic features is to use the Linguistic Inquiry Word Count (LIWC) dictionary and various affective lexicons. Other features related to cultural factors and suicide risk have not been explored. Moreover, the use of social networking behavioral features and profile features would limit the generalizability of the model. Therefore, our study aimed at building a prediction model of depression for text-only social media data through a wider range of possible linguistic features related to depression, and illuminate the relationship between linguistic expression and depression. Methods: We collected 789 users' depression scores as well as their past posts on Weibo, and extracted a total of 117 lexical features via Simplified Chinese Linguistic Inquiry Word Count, Chinese Suicide Dictionary, Chinese Version of Moral Foundations Dictionary, Chinese Version of Moral Motivation Dictionary, and Chinese Individualism/Collectivism Dictionary. Results: Results showed that all the dictionaries contributed to the prediction. The best performing model occurred with linear regression, with the Pearson correlation coefficient between predicted values and self-reported values was 0.33, the R-squared was 0.10, and the split-half reliability was 0.75. Discussion: This study did not only develop a predictive model applicable to text-only social media data, but also demonstrated the importance taking cultural psychological factors and suicide related expressions into consideration in the calculation of word frequency. Our research provided a more comprehensive understanding of how lexicons related to cultural psychology and suicide risk were associated with depression, and could contribute to the recognition of depression.

CONTEXT AND RESULTS: 6-Azido-8-nitrotetrazolo[1,5-b]pyridazine-7-amine (3at) is a promising green energetic material, which meets the development requirements of environment-friendly explosives. By discussing the relationship between lattice parameters and pressure, it is found that the compression ratio indicates anisotropy of compressibility. And bond lengths get shorter under pressure, resulting in stronger intermolecular bonds. The N3 group rotates under pressure. And then, the optical properties basically change regularly with the change of pressure. As the pressure increases, the absorption range widens. In the low energy interval, it shows transparency, and then with the increase of energy and pressure, it shows better optical activity. With the increase of pressure and energy, the absorption coefficient increases, representing that the optical activity becomes high. Finally, according to the analysis of mechanical properties, 3at exhibited brittle behavior at 0 GPa and 100 GPa, while at 10 to 90 GPa, the values of ν and B/G are malleable. COMPUTATIONAL AND THEORETICAL TECHNIQUES: Based on density functional theory, the crystal parameters, electronic properties, optical properties, and elastic and mechanical properties of 3at under different pressures were studied theoretically. The GGA-PW91+OBS method was used to calculate the physical parameters under pressure, such as lattice parameters, energy band structures, dielectric function, refractive index, absorption coefficient, and elastic constants. Physical properties under (3at) pressure are predicted.

The band structure, total density of states, and atomic orbit projected density of states analysis were performed to investigate HMX/NMP cocrystal by using the first-principles calculations. Results show that the HMX/NMP cocrystal is equipped with a direct band gap and the interactions between HMX and NMP molecules are rather weak. The O orbits hybridize with H orbits, and the parts of charge transform from H to O atoms by analyzing the DOS. The HMX/NMP cocrystal possesses three types of intermolecular interactions between HMX and NMP; these interactions and the arrangement of two molecules in the structure are the main reasons for the low sensitivity of the cocrystal. The C-H…O type hydrogen bond is the key role in forming the structure, and the strength of the hydrogen bond interaction for C-H…O-N is higher than that of C-H…O-C.

The structure, electronic, and magnetic properties have been investigated by the first-principles calculations for paramagnetic, ferromagnetic, and antiferromagnetic Cu3TeO6 under pressure from 0 to 100 GPa. The calculated lattice parameters at 0 GPa are in excellent agreement with the available calculated and experimental values. With increasing pressure, the lattice parameters and volume decrease, but Cu3TeO6 keeps a stable cubic structure. The electronic calculations show that paramagnetic and ferromagnetic Cu3TeO6 are metallic, and antiferromagnetic Cu3TeO6 is non-metallic with a direct band gap which decreases with the increasing pressure. Under the pressure, their non-locality of density of states enhances and the electrons become more active. Moreover, for antiferromagnetic Cu3TeO6, the spin moments of Cu atoms are affected obviously by pressures, and Te atoms show nonmagnetic performance. The total magnetic moment, which is mainly contributed by Cu, reaches the maximum at 20 GPa, and decreases with the increasing pressure. The knowledge of these properties will provide reference and guidance for the subsequent study of Cu3TeO6.

BACKGROUND: The outbreak of COVID-19 has resulted in serious concerns in China and abroad. To investigate clinical features of confirmed and suspected patients with COVID-19 in west China, and to examine differences between severe versus non-severe patients. METHODS: Patients admitted for COVID-19 between January 21 and February 11 from fifteen hospitals in Sichuan Province, China were included. Experienced clinicians trained with methods abstracted data from medical records using pre-defined, pilot-tested forms. Clinical characteristics between severe and non-severe patients were compared. RESULTS: Of the 169 patients included, 147 were laboratory-confirmed, 22 were suspected. For confirmed cases, the most common symptoms from onset to admission were cough (70·7%), fever (70·5%) and sputum (33·3%), and the most common chest CT patterns were patchy or stripes shadowing (78·0%); throughout the course of disease, 19·0% had no fever, and 12·4% had no radiologic abnormality; twelve (8·2%) received mechanical ventilation, four (2·7%) were transferred to ICU, and no death occurred. Compared to non-severe cases, severe ones were more likely to have underlying comorbidities (62·5% vs 26·2%, P = 0·001), to present with cough (92·0% vs 66·4%, P = 0·02), sputum (60·0% vs 27·9%, P = 0·004) and shortness of breath (40·0% vs 8·2%, P <  0·0001), and to have more frequent lymphopenia (79·2% vs 43·7%, P = 0·003) and eosinopenia (84·2% vs 57·0%, P = 0·046). CONCLUSIONS: The symptoms of patients in west China were relatively mild, and an appreciable proportion of infected cases had no fever, warranting special attention.

According to the analysis of investigation, there are many problems in the current clinical skills training for medical education, such as large demand, high difficulty and less opportunity. With the development and application of computer network technology, it has provided new directions for the training in the medical field. Using modern management techniques, it has combined computer technology, biochemical science and microbiology principles with traditional clinical medical education experience, which has promoted the development of advanced clinical medicine concepts and skill levels. The combination of distance education and medical education is a process of integrating theoretical knowledge into practice, which not only improve the medical management capabilities of medical staff, but also improve the overall level of clinical skills.

In order to promote the risk-based strategy in the investigation, assessment, and remediation of Chinese brownfield sites, the Health and Environmental Risk Assessment (HERA) software was developed. It is vital to validate the HERA model and compare the inter-model differences of HERA model against other available risk assessment tools. This paper discusses the similarities and differences between the Risk-Based Corrective Action (RBCA) Tool Kit and the HERA model by evaluating the health risk of organic contaminated groundwater sources for a chemical works in China for the first time. Consequently, the HERA and RBCA models yielded the identical results for Site-Specific Assessment Criteria (SSAC) under the commercial redevelopment. However, the HERA estimated more conservative and stringent SSACs under the residential scenario based on the different exposure calculations. The inhalation of indoor vapors was the most predominated exposure pathway for all the volatile organic compounds (VOCs) determined using the RBCA and HERA models. According to the HERA model, inhalation of chloroform may cause the highest unacceptable carcinogenic risk at 2.31 × 10(-3) under the residential scenario. Therefore, it is recommended that a risk-based remedial strategy be developed to ensure the safe and sustainable redevelopment of the site.

The nitrogen balance can serve as an indicator of the risk to the environment of nitrogen loss from agricultural land. To investigate the temporal and spatial changes in agricultural nitrogen application and its potential threat to the environment of the Haihe Basin in China, we used a database of county-level agricultural statistics to calculate agricultural nitrogen input, output, surplus intensity, and use efficiency. Chemical fertilizer nitrogen input increased by 51.7% from 1990 to 2000 and by 37.2% from 2000 to 2010, concomitant with increasing crop yields. Simultaneously, the nitrogen surplus intensity increased by 53.5% from 1990 to 2000 and by 16.5% from 2000 to 2010, presenting a continuously increased environmental risk. Nitrogen use efficiency decreased from 0.46 in 1990 to 0.42 in 2000 and remained constant at 0.42 in 2010, partly due to fertilizer composition and type improvement. This level indicates that more than half of nitrogen inputs are lost in agroecosystems. Our results suggest that although the improvement in fertilizer composition and types has partially offset the decrease in nitrogen use efficiency, the environmental risk has still increased gradually over the past 20 years, along with the increase in crop yields and nitrogen application. It is important to achieve a better nitrogen balance through more effective management to significantly reduce the environmental risk, decrease nitrogen surplus intensity, and increase nitrogen use efficiency without sacrificing crop yields.

BACKGROUND AND OBJECTIVE: Endothelial progenitor cells (EPCs) play an important role in hypoxia-triggered tumor vasculogenesis. However, the homing of exogenous EPCs in tumors is still unclear. In this study, we investigated the recruitment of exogenous EPCs in human lung adenocarcinoma model of nude mice. METHODS: EPCs labeled with green fluorescence protein (GFP) were transplanted into nude mice bearing human lung adenocarcinoma. The growth of tumor was observed. After the mice were killed, GFP-EPCs in different tissues were examined by fluorescence. The tumor tissues were stained for CD133, hypoxia-inducible factor-1alpha (HIF-1α), stromal cell-derived factor-1α (SDF-1α), and vascular endothelial growth factor receptor (KDR). Real-time polymerase chain reaction of CD133, HIF-1α, SDF-1α, and VEGF-1 were also performed. RESULTS: The growth of tumor in EPC group was significantly faster than that in saline solution group (P <0.05). Under fluorescence microscope, GFP-EPCs were strongly expressed in both tumor and bone marrow. EPCs were recruited to the tumor periphery to participate in tumor vasculogenesis. The expression of CD133, HIF-1α, and SDF-1 mRNA in tumor and bone marrow were significantly higher than that in the liver, spleen, and skin (P<0.05). CONCLUSIONS: Exogenous EPCs can be recruited to tumor and accelerate tumor growth. Except tumor, bone marrow can also recruit EPCs.

Central nervous system involvement is common in HIV infection. We determined the relationship between T-cell subsets and viral load in the cerebrospinal fluid (CSF) of therapy-naive, asymptomatic HIV-infected individuals. A shift from naive to effector-memory CD8 T cells was observed in the CSF of HIV-infected individuals compared with controls. The HIV load strongly correlated with CD8 T cells in CSF. Effector-memory CD8 T cells were positively and effector CD8 T cells were negatively associated with viral replication in CSF.

CD34 glycoprotein in human hematopoiesis is expressed on a subset of progenitor cells capable of self-renewal, multilineage differentiation, and hematopoietic reconstitution. Nucleolin is an abundant multifunctional phosphoprotein of growing eukaryotic cells, involved in regulation of gene transcription, chromatin remodeling, and RNA metabolism, whose transcripts are enriched in murine hematopoietic stem cells, as opposed to differentiated tissue. Here we show that, in human CD34-positive hematopoietic cells, nucleolin activates endogenous CD34 and Bcl-2 gene expression, and cell surface CD34 protein expression is thereby enhanced by nucleolin. Nucleolin-mediated activation of CD34 gene transcription results from direct sequence-specific interactions with the CD34 promoter region. Nucleolin expression prevails in CD34-positive cells mobilized into peripheral blood (PB), as opposed to CD34-negative peripheral blood mononuclear cells (PBMCs). Therefore, in intact CD34-positive mobilized PB cells, a recruitment of nucleolin to the CD34 promoter region takes place, accompanied by nucleosomal determinants of gene activity, which are absent from the CD34 promoter region in CD34-negative PBMCs. Our data show that nucleolin acts as a component of the gene regulation program of CD34-positive hematopoietic cells and provide further insights into processes by which human CD34-positive hematopoietic stem/progenitor cells are maintained.

OBJECTIVE: To investigate the feasibility and short- term effect of injecting botulinum toxin A through anorectum to treat Hirschsprung disease. METHODS: Eight cases with Hirschsprung disease were proven pathologically or diagnosed by barium enema. At the clock point of 3,6,9 under supine posture,a total dose of 1.5 U/ kg of botulinum toxin A was injected into the internal anal sphincter and the submucosa of rectum. The pressure of anus- rectum was measured before operation and 1 month,1 year after operation,respectively. Barium enema examination was carried out within 3-6 months after operation,then once a year during followed- up. RESULTS: All of the patients recovered well without operative or postoperative complications. In the first year,all patients could defecate spontaneously without constipation and abdominal distention. Three cases had constipation again and slight abdominal distention 1 year after operation,but the symptoms were relieved after anus dilatation. The resting pressure of anus- rectum was lower 1 month after operation than that before operation (P< 0.01) in 8 cases,and still lower 1 year after operation (P< 0.05)in 6 cases. Barium enema examination within 3-6 months after operation revealed that the dilated colon shrank. CONCLUSION: Botulinum toxin A injection is a new,feasible and safe method for Hirschsprung disease,but the long- term efficacy needs to be further studied.
