RESUMO

Effect of seven-day enteral administration of clinkers, cement and dust sedimented on electric filters on DNA and RNA, total protein levels, adenylate desaminase (AD) and alkaline phosphatase (AP) activities in thymus und spleen was shown in rats. Harmful effect of industrial dust on the lymphatic organs was maximal in thymus: diminished lymphatic tissue, decreased DNA, RNA and total protein levels, increased AD and AP activities. Compounds of chromium with lead, copper and mercury appeared to have maximal harmful effect on the lymphatic tissue.

RESUMO

RNA, DNA, total protein, transketolase (TK), alanine aminotransferase (AlAT), aspartate aminotransferase (AsAT), methionyl-tRNA (met-tRNA)- and cysteinyl-tRNA (cys-tRNA)-synthetases in the thymus and spleen of 46 intact rats exposed to hypokinesia and vitamin B1 deficiency were measured. It was found that 15-day hypokinesia induced a significant decrease of the thymus and spleen weight and an increase of AlAT in the thymus. Vitamin B1 deficiency (hydroxy thiamine in drinking water at a dose of 2 mg/kg body weight for 15 days) led to a substantial decrease of TK in blood, AlAT and AsAT in the spleen; it diminished AsAT and increased AlAT in the thymus. Combined exposure to hypokinesia and vitamin B1 deficiency caused a more marked decrease of the weight of lymph organs, a significant loss of body weight, an increase of DNA in the thymus and spleen, and an increase of TK, met-tRNA-synthetase and AlAT in the thymus. These results suggest that vitamin B1 deficiency aggravates disorders in protein and nucleic acid metabolism in the lymph organs of hypokinetic animals.

RESUMO

A decrease in thymus gland mass and an increase in spleen tissue mass, decrease of DNA content in thymus, cervical and submaxillary lymph nodes, increase in DNA and RNA content in axillary lymph nodes and of RNA in spleen were detected within 15-19 days in rats (220-240 g of body mass), sensitized with 0.4 ml of encephalogenous mixture (0.1 ml in each foot) containing 100 mg of homologous brain tissue, liquid petrolatum and 10 mg of heated BCG vaccine. Activity of transketolase was decreased in thymus, submaxillary lymph nodes and increased in spleen tissue of the sensitized animals.

After four administrations of human blood serum gamma-globulin (intraperitoneally, once during 4 days at a dose of 1 mg and the following reimmunization within 60 days at a dose of 3-4 mg) distinct incorporation of 14C-thymidine (specific activity 54 muCi/mumole, by 5 muCi per an animal, 2 hrs before decapitation, intraperitoneally) into rat spleen DNA was detected within 4 days of immunogenesis. Acute oxythiamine B1 avitaminosis (single subcutaneous administration of oxythiamine at a dose of 400 mg/kg) led to a decrease in the label incorporation into spleen DNA of immunized rats by 70% (p less than 0.01) as compared with control immunized animals, while incorporation of 14C-thymidine was unaltered in intact rats. Content of RNA was not distinctly altered in spleen of B1-deficient rats within the 4 days of immunogenesis. Acute deficiency of vitamin B1 thus impaired synthesis of DNA in spleen during development of immunogenesis.

RESUMO

Distinct increase in incorporation of 14C-protein hydrolysate of chlorella (5 microCi per an animal of 150 g body mass) into proteins of rat spleen was observed within 4 days after a single or repeated immunization of rats (180-200 g of body mass) with human blood serum gamma-immunoglobulin at a dose of 30-40 mg per 1 kg of animal mass. Under conditions of acute B1-avitaminosis caused by a single administration of hydroxythiamine at a high dose of 400 mg/kg incorporation of the label into spleen proteins of the immunized rats was decreased within 72 hrs. Thus, acute hydroxythiamine-produced B1-avitaminosis inhibited induction of protein biosynthesis stimulated during immunogenesis.

RESUMO

A study was made of the effect of different doses of hydroxythiamine on activity of alkaline phosphatase and transketolase in rat spleen and thymus during primary and secondary administration of human gamma-globulin. Immunization of rats significantly raised alkaline phosphatase activity in both organs. Administration of a large single dose of hydroxythiamine (400 mg/kg bw) reduced transketolase activity in the thymus and spleen in both immunized and non-immunized animals and did not change alkaline phosphatase activity in these organs.
