Numerous adverse effects on human health have been reported in epidemiological studies of oleoresin capsicum (OC) and other riot control agents (RCAs). Importantly, the daunting risk of such RCAs can be neutralized by optimizing the desired concentration of such agents for mob dispersal. Hence, a nonlethal riot control combinational formulation (NCF) was prepared for dispersing rioters without imparting fatal outcomes. However, for desired utilization of NCF, it is essential to recognize its extent of potential toxicity. Therefore, the current investigation evaluated the dermal toxicity of NCF using experimental animals in compliance with the OECD guidelines. Additionally, few essential metal ions were analyzed and found non -significantly different in the test rats as compared to control rats. Moreover, abnormal dermal morphology and lesions ultrastructural tissue defects were not noticed as evinced by different studies like ultrasonography, histology, and scanning electron microscopy (SEM) respectively. Further, Doppler ultrasonography exhibited non-significantly different blood flow velocity in both groups, whereas miles test demonstrated a significantly increased Evans blue concentration in test rats compared to the control rats, which might be due to an initial increase in blood flow via an instant action of the NCF at the cutaneous sensory nerve endings. However, our results demonstrated NCF can produce initial skin irritating and sensitizing effects in guinea pigs and rabbits without the antecedence of acute toxicity (≤2000 mg/kg) in Wistar rats.

In this study,the water samples were collected from 31 sites of Tawang, Arunachal Pradesh, India (North-Eastern Himalaya), during the winter season to check the suitability of water for drinking and irrigation purposes.The study scientifically demonstrates the estimation of Water quality index (WQI) andhydrogeochemical characteristics of surface water samples by utilizing multivariate statistical methods. The main water quality parameters considered for this study were TDS, conductivity, salinity, pH, hardness, cations and anions. WQI was calculated in order to find out the deviation in the water quality parameters particularly with respect to BIS permissible limits.The major influencing factors responsible for the variation in these parameters were derived by using Principal component analysis (PCA) and Correlation matrix.To check the suitability of water for drinking purpose, hydrogeochemical facies and rock water interaction was derived by using well established methods such as Piper Plot (determine water type), WQI (Quality monitoring), and saturation index (for mineral dissolution). The results revealed that the silicate weathering was the main ionic source in comparison to carbonate weathering which is due to the higher dissolution capacity of silicate minerals.The results of the scattered plot between (Ca2+ + Mg2+)-(HCO3- + SO42-) versus (Na+ + K+)-Cl- (meq/L) highlighted thation exchange occurs between Mg2+ and Ca2+ofsurface water with Na+ and K+of rock /soil. This means that calcium ion was getting adsorbed, and sodium ion was getting released. The Ca2+-Mg2+-HCO3-, Na+-HCO3-and Na+-Cl- type of surface water suggested permanent and temporary hardness respectively in the studied region. The dominant cations of this study were Na+ and Ca2+ while the dominant anions were HCO3- and SO42-. In order to check the suitability of water sources for irrigation, parameters like, Magnesium hazard (MH), Total hardness (TH), Permeability Index (PI), Kelly Index (KI), Sodium adsorption rate (SAR), Sodium percentage (Na%), and Residual sodium carbonate (RSC) were determined. The results showed that 93% of the samples had PI score < 75, which indicates the suitability of the water for irrigation. Also the WQI calculation showed an average WQI value of 82.49, amongst which 61% samples were in the range of 0-50 being considered as good for drinking, while 39% were catageorised as unsuitable for drinking showing a value of > 50. Hence the above findings reveal that geogenic activities play a major role in influencing the water quality of Tawang region. Hence suitable water treatment technologies or methods might be used to eliminate thenon desirable elements and minerals present in surface water.

OBJECTIVE: Development of Frostbite healing hydrogel of Manuka honey and hyaluronic acid. SIGNIFICANCE: Frostbite is a cold-induced ischemic vascular injury non-responsive to most of the wound healing products. Thrombus-induced ischemia is the main cause of frostbite-related necrosis. Hyaluronic acid is known to possess significant antithrombotic and wound healing activity. Moreover, Manuka Honey is also rich in flavonoids and polyphenols with potential antithrombotic activity. These two agents were together utilized to develop a frostbite healing formulation. METHODS: In-silico antithrombotic efficacy of major phytoconstituents of Manuka honey was evaluated using in-silico-docking studies against Tissue plasminogen activator and Cyclooxygenase-1 protein. Further in-vivo frostbite healing evaluation was carried out in Wistar rats, by inducing frostbite with a supercooled rod. RESULTS: The results indicate that major leptosin and other major phytoconstituent of Manuka honey has significant antithrombotic property. The hydrogel formulation of HA and MH possess significant antimicrobial efficacy. The wound contraction studies and histopathological evaluation reveals that the hydrogel also has a good frostbite healing activity showing complete wound healing within an 18-day period. The findings of the western blotting studies suggest that the hydrogel acts by VEGF- NRF-2 pathway. CONCLUSION: This result implies that the prepared hydrogel can serve as an effective frostbite healing formulation.

RESUMO

Frostbite is a severe ischemic injury which occurs due to the tissue vascular damage after sub-zero temperature tissue exposure. Deep frostbite can result in necrosis and may need amputation of affected tissue. Though a serious injury, it is not very well understood, and further scientific exploration is needed. This work explores the current understanding of the pathophysiology of frostbite. We reviewed the current status of the diagnostics, the drugs, the therapies and the surgical practices for prevention and management of frostbite. Advances in nanotechnology and drug delivery had improved the therapeutic outcomes significantly. This review also explored the latest advancements and researches done for development of newer therapeutics and diagnostics for frostbite care.

Aim: The use of oleoresin capsicum (OC) sprays, due to their irreversible health effects has now grown into a matter of heated debate. In the present study, the early phase pulmonary events involving chemotactic and inflammatory mediators after short-exposure duration to OC have been presented.Materials and methods: Female Wistar rats used in the evaluation of respiratory parameters at 1 h, 3 h, and 24 h post-exposure, were sacrificed for the evaluation of blood cell counts, BALF cytokine estimation, lung capillary leakage, study of oxidative stress and histopathology of the lungs.Results: Results confirmed a dose-dependent effect of OC exposure on serum clinical chemistry and hematological parameters. Subsequent upregulation of IL-l and TNF-α indicated lung's responses to acute oxidant-induced injury and inflammation after OC exposure. Significant alterations in the pulmonary levels of reactive oxygen intermediates were seen following the inhalation of OC. Infiltration of polymorphonuclear leukocytes, mostly neutrophils, into the site of infection was evident in the cytocentrifuged samples of BALF. Histological samples of rat lung sections revealed the recruitment of inflammatory cells in the airways and around blood vessels in the subepithelium of conducting airways.Conclusion: Results of the present study demonstrated that, exposure to OC spray may mitigate inflammatory response and development of acute lung injury in rats. However, it can be concluded that although OC spray causes pulmonary hazards in the aforementioned concentrations, it can be used as a non-lethal riot control agent in minimal concentration. Understanding the in-depth mechanism of action in the molecular and receptor level will help in developing effective antagonist against OC.

Tremendous explosion of population has led to about 200% increment of total energy consumptions in last twenty-five years. Apart from conventional fossil fuel as limited energy source, alternative non-conventional sources are being explored worldwide to cater the energy requirement. Lignocellulosic biomass conversion for biofuel production is an important alternative energy source due to its abundance in nature and creating less harmful impacts on the environment in comparison to the coal or petroleum-based sources. However, lignocellulose biopolymer, the building block of plants, is a recalcitrant substance and difficult to break into desirable products. Commonly used chemical and physical methods for pretreating the substrate are having several limitations. Whereas, utilizing microbial potential to hydrolyse the biomass is an interesting area of research. Because of the complexity of substrate, several enzymes are required that can act synergistically to hydrolyse the biopolymer producing components like bioethanol or other energy substances. Exploring a range of microorganisms, like bacteria, fungi, yeast etc. that utilizes lignocelluloses for their energy through enzymatic breaking down the biomass, is one of the options. Scientists are working upon designing organisms through genetic engineering tools to integrate desired enzymes into a single organism (like bacterial cell). Studies on designer cellulosomes and bacteria consortia development relating consolidated bioprocessing are exciting to overcome the issue of appropriate lignocellulose digestions. This review encompasses up to date information on recent developments for effective microbial degradation processes of lignocelluloses for improved utilization to produce biofuel (bioethanol in particular) from the most plentiful substances of our planet.

The present study is aimed at the development of a sunscreen cream for use in high altitude areas which have been found to possess superior sun protection factor (SPF) along with remarkable antioxidant activity. The topical formulation is a standard oil-in-water emulsion of a combination of United States Food and Drug Administration (US FDA) approved ultraviolet filters; along with melatonin and pumpkin seed oil. The in-silico optimized formulation was characterized using established methods and the stability study was carried out as per International Conference on Harmonization guidelines. The formulation was prepared after requisite pre-formulation analysis by Fourier-transform infrared spectroscopy, differential scanning calorimetric and thermogravimetric analyses; followed by characterization based on color, odor, phase separation, spreadability, specific gravity, homogeneicity, centrifugation and sensitivity. For the stability study, a total of three samples from three batches of the finished product were subjected to the stability study. The samples were analyzed for content uniformity, pH, in vitro SPF, rheology, zeta potential, droplet diameter and microbial analysis of the 0th day and also the the end of the storage period. Results obtained from the stability study indicated that the formulation possesses 50+ in vitro SPF value and remained stable for 6 months and 12 months under storage at 40 ± 2 °C and 75 ± 5% relative humidity; and -20 °C ± 5 °C respectively.

The presence of 40-50% more UV radiation in high altitude areas renders the plethora of sunscreen products available in the market virtually ineffective. In this light of event, four US FDA approved UV filters were combined with melatonin and pumpkin seed oil to produce a broad spectrum sunscreen cream, which is envisaged to provide optimum sunprotection along with enhanced antioxidant activity. The objective of this study is to evaluate the protective effect of the sunscreen cream against UV radiation-induced skin photoaging in adult Wistar albino rats and identify its possible underlying mechanism. Wistar rats were exposed to broad spectrum UV radiation for 28 days. The test group received the sunscreen formulation dermally every day prior to UV radiation. The effects of the formulation against UV induced symptoms; viz. skin thickness and edema, in vivo antioxidant activities, inflammatory cytokines, collagen content, histopathological examination and expression of specific genes established the protective activity of the formulation. The test formulation was able to mitigate the harmful effects of UV radiation by increasing in vivo SOD, GSH-Px, CAT and collagen levels; decreasing skin edema, skin thickness and cytokines like IL-6, IL-1ß, TNF-α and TGF-ß1. UV radiation induced changes in histological architecture and arrangement of collagen and elastin fibers were also prevented by the test formulation. Finally, the formulation was able to regulate the expression of COL3A1, COX-2, bFGF, VEGF-C, Smad2, Smad4, Smad7 genes which induced significant photoprotective activity. The sunscreen formulation ameliorated UV induced photoaging by preventing oxidative collagen degradation and augmentation of TGF-ß-Smad-mediated collagen production.

Background & methods We investigated the in vitro antioxidant and antifungal activity by agar disc diffusion assay of leaf extract of some stinging plants namely, Urtica dioica L., Tragia involucrate L., Carduus nutans L. and Mucuna pruriens (L.) DC., against pathogenic fungi causing infections/diseases. Results M. pruriens (Disc 4), T. involucrate (Disc 2), U. dioica (Disc 1) showed significant antifungal activity against all tested pathogens, while C. nutans (Disc 3) showed intermediate activity against only Chaetomium globosum (Cg). The leaf extract of M. Pruriens showed maximum total phenol content (~1004 µg g-1 dry wt) followed by T. involucrate, C. nutans and U. dioica. However, the ascorbate was observed highest in T. involucrate (~10.3 µg g-1 dry wt) followed by M. pruriens (~9.2 µg g-1 dry wt) but the difference was not significant (p ≤ 0.05). Likewise, M. pruriens showed maximum anthocyanin content (~0.3 µg g-1 dry wt). The activity of antioxidant enzymes revealed that M. Pruriens showed maximum ascorbate peroxidase (APX) activity, while the highest guaiacol peroxidase (GPX) and catalase (CAT) activities were observed in C. nutans and U. dioica, respectively. Conclusions M. Pruriens showed potential in vitro antioxidant and antifungal activity against studied pathogens that may be used for ethno-pharmacological uses.
