We report the results of taxonomic studies on members of the family Micrococcaceae that, according to the 16S rRNA, internal transcribed spacer 1 (ITS1), average nucleotide identity (ANI), and average amino acid identity (AAI) tests, are related to Kocuria rosea strain RCAM04488, a plant-growth-promoting rhizobacterium (PGPR) isolated from the rhizosphere of potato (Solanum tuberosum L.). In these studies, we used whole-genome phylogenetic tests and pangenomic analysis. According to the ANI > 95 % criterion, several known members of K. salina, K. polaris, and K. rosea (including K. rosea type strain ATCC 186T) that are related most closely to isolate RCAM04488 in the ITS1 test should be assigned to the same species with appropriate strain verification. However, these strains were isolated from strongly contrasting ecological and geographical habitats, which could not but affect their genotypes and phenotypes and which should be taken into account in evaluation of their systematic position. This contradiction was resolved by a pangenomic analysis, which showed that the strains differed strongly in the number of accessory and strain-specific genes determining their individuality and possibly their potential for adaptation to different ecological niches. Similar results were obtained in a full-scale AAI test against the UniProt database (about 250 million records), by using the AAI-profiler program and the proteome of K. rosea strain ATCC 186T as a query. According to the AAI > 65 % criterion, members of the genus Arthrobacter and several other genera belonging to the class Actinomycetes, with a very wide geographical and ecological range of sources of isolation, should be placed into the same genus as Kocuria. Within the paradigm with vertically inherited phylogenetic markers, this could be regarded as a signal for their following taxonomic reclassification. An important factor in this case may be the detailing of the gene composition of the strains and the taxonomic ratios resulting from analysis of the pangenomes of the corresponding clades.

It was shown for the first time that the treatment of winter wheat (Triticum aestivum L.) seeds with gold nanoparticles (average diameter 15.3 nm; solution concentration 20 µg/mL) increases plant tolerance to low temperature. It was found that an increase in tolerance under the influence of nanoparticles is accompanied by a number of changes depending on temperature conditions. In optimal temperature conditions, gold nanoparticles stimulated plant growth and the activity of the photosynthetic apparatus, whereas in conditions of low-temperature hardening (2°C, 7 days) they inhibited growth but maintained photosynthetic activity, contributing to the accumulation of soluble sugars (cryoprotectants) in the leaves. It is concluded that gold nanoparticles can be considered as adaptogens that increase plant tolerance to low temperatures; however, their effectiveness in this role and the subtle mechanisms of action require further study.

PURPOSE: The aim of this study was to isolate Babesia canis soluble antigens and to investigate the effect of their conjugates with gold nanoparticles on the immunogenicity in laboratory animals. METHODS: A procedure was developed for isolating and purifying B. canis antigens. The isolated culture antigen of B. canis 495 was coupled to gold nanoparticles, and the conjugate was used to immunize laboratory mice. RESULTS: Western blotting showed that the resultant antiserum specifically recognized the proteins of the B. canis strains isolated from naturally infected dogs. The antibody titer, the respiratory activity of peritoneal macrophages, the proliferative activity of splenocytes, and the production of cytokines were maximal when the animals were immunized with the antigen-nanoparticle conjugate emulsified in complete Freund's adjuvant. Without adjuvant, the babesial antigen was weakly immunogenic. CONCLUSION: Therefore, the use of gold nanoparticles as an antigen carrier induced a broad immune response involving both cellular and humoral responses. The antibodies raised by the proposed procedure are potentially effective at immunodetection of Babesia canis infections in dogs.

Gold nanoparticle conjugates with Vibrio cholerae antigens were synthesized. The animals were immunized with the obtained conjugates. Rabbit polyclonal antibodies to the antigens were obtained, which showed high specific activity. On the model of white laboratory mice, the protective activity of conjugates of cholera antigens with nanoparticles during infection of vaccinated animals was evaluated using a commercial vaccine as a control. It was shown that in terms of immunogenicity, the created prototypes of cholera vaccine using gold nanoparticles as a carrier and adjuvant complied with the production regulations for the Russian national cholera chemical vaccine.

RESUMO

Functionalized gold nanoparticles (GNPs) with controlled geometrical and optical properties have been the subject of intense research and biomedical applications. This review summarizes recent data and topical problems in nanomedicine that are related to the use of variously sized, shaped, and structured GNPs. We focus on three topical fields in current nanomedicine: (1) use of GNP-based nanoplatforms for the targeted delivery of anticancer and antimicrobial drugs and of genes; (2) GNP-based cancer immunotherapy; and (3) combined chemo-, immuno-, and phototherapy. We present a summary of the available literature data and a short discussion of future work.

The electro-acoustical sensor for the real-time identification of the bacteriophage FAl-SR65 is described. The polyclonal antibodies specific to the bacteriophage FAl-SR65 were for the first time obtained, and the possibility of their application to the bacteriophage detection by using the electro-acoustical method of analysis was also studied for the first time. The electro-acoustical sensor was based on the lateral electric field excited piezoelectric resonator. The frequency dependences of the real and imaginary parts of the electrical impedance of such a resonator, loaded with the virus-antibody suspension, significantly differed from those measured for the same resonator with the same virus suspension but without antibodies (control). The limit for the reliable bacteriophage detection during their interaction with the antibodies (sensitivity level) was determined to be 106 phages ml-1. The obtained results demonstrate the possibility to record the bacteriophage-antibody interactions and can be used as the basis for the development of the biological sensor for the quantitative virus detection.

The aim of this research was to design a method of immobilization of high-purity human butyrylcholinesterase on the surface of gold nanoparticles preserving the activity of the enzyme. In order to achieve this aim, the method of fractionation and purification of human butyrylcholinesterase from plasma was modified. The synthesis of 15-nm gold nanoparticles was carried out by citrated method. A method of conjugation of the high-purity butyrylcholinesterase with gold nanoparticles was developed. It was found that the Immobilization of butyrylcholinesterase on the surface of gold nanoparticles resulted in a significant (to 23%) increase in the specific activity of the enzyme.

It has been demonstrated that electroacoustic analysis with polyclonal antibodies can be used for bacteriophage detection. The frequency dependences of the real and imaginary parts of electrical impedance of a resonator with a viral suspension with antibodies were shown to be essentially different from the dependences of a resonator with control viral suspension without antibodies. It was shown that ΦAl-Sp59b bacteriophages were detected with the use of antibodies in the presence of foreign virus particles. The ΦAl-Sp59b bacteriophage content in the analyzed suspension was ~1010­106 phages/mL; the time of analysis was no more than 5 min. The optimally informative parameter for obtaining reliable information was the change in the real or imaginary part of electrical impedance at a fixed frequency near the resonance upon the addition of specific antibodies to the analyzed suspension. It was demonstrated that the interaction between bacteriophages and antibodies can be recorded, offering good prospects for the development of a biological sensor for liquid-phase identification and virus detection.

Active application of gold nanoparticles for various diagnostic and therapeutic purposes started in recent decades due to the emergence of new data on their unique optical and physicochemical properties. In addition to colloidal gold conjugates, growth in the number of publications devoted to the synthesis and application of multifunctional nanocomposites has occurred in recent years. This review considers the application in biomedicine of multifunctional nanoparticles that can be produced in three different ways. The first method involves design of composite nanostructures with various components intended for either diagnostic or therapeutic functions. The second approach uses new bioconjugation techniques that allow functionalization of gold nanoparticles with various molecules, thus combining diagnostic and therapeutic functions in one medical procedure. Finally, the third method for production of multifunctional nanoparticles combines the first two approaches, in which a composite nanoparticle is additionally functionalized by molecules having different properties.

The changes in the electro-acoustic parameters of cell suspension due to the interaction of cells with bacteriophages both in a pure. culture and in the presence of extraneous microflora were investigated. It has been found that the specific changes in the electroacoustic parameters of cell suspension under the action of bacteriophage occur only in microbial cells which are sensitive to the bacteriophage studied. It has been established that a sensor unit allows of distinguishing a situation when the bacterial cells are infected with specific bacteriophages of the control experiments and a situation with no introduction of infection. An approximate criterion of the presence of specific interactions of bacteriophages and cells in suspension was developed. In accordance with this criterion the change in electrical impedance of the sensor unit must not be less than - 1%. In control experiments a standard microbiological technique, plating the cells infected with bacteriophages on solid nutrient medium, was used. For the first time the possibility of using the method of electroacoustic analysis for determination of a spectrum of lytic activity of bacteriophages was shown. The results obtained may be used for development of a new express method for determining the sensitivity to bacteriophages of the microbial cells.

Penetration of pegylated (enveloped in polyethylene glycol) gold nanoparticles 5 and 30 nm in diameter through the placental barrier was studied in pregnant rats injected intravenously with these particles in a dose of about 0.8 mg Au/kg on day 10 of gestation. The particles were visualized in tissues by silver nitrate autometallography; the total content of gold in the fetuses was evaluated by atomic adsorption spectroscopy. Gold nanoparticles were detected in the fetal liver macrophages and in the spleens; high total content of gold in the fetuses was demonstrated for particles of both sizes. The data suggest that gold nanoparticles penetrate through rat placental barrier in vivo. No morphological changes were detected in the liver, kidneys, spleen, and brain of fetuses.

This is the first study to demonstrate that the medicinal basidiomycete Lentinula edodes can reduce gold (III) ions from hydrogen tetrachloaurate (chloroauric acid) H[AuCl4] to the elementary state with the formation of spherical nanoparticles (nanospheres). When a culture was grown under submerged conditions in the presence of chloroauric acid, the appearance of an intense purple-red color of L. edodes filamentous hyphae was recorded, which indicates that gold ions were reduced to gold nanoparticles. Using transmission electron microscopy and X-ray fluorescence, we observed accumulation of colloidal gold by the fungal mycelium in the form of electron-dense nanospheres of 5 to 50 nm in diameter on the surface and inside fungal cells.

The phage mini-antibodies to bacterial cells of strain Azospirillum brasilense Sp245 were obtained and the possibility of using them for detection of microbial cells by means of a lateral field excited piezoelectric resonator was studied. It has been found that the frequency dependencies of the real and imaginary parts of the electrical impedance of the resonator loaded by the cell suspension A. brasilense Sp245 with the mini-antibodies, significantly differ from those of the resonator with the control cell suspension without mini-antibodies. The concentration limit of possible determination of the microbial cells in their interaction with the mini-antibodies is equal to 10(3) cells/ml. It has been ascertained that detection of A. brasilense Sp245 cells using the mini-antibodies is possible even in the presence of other cultures, for example, E. coli BL-Ril and A. brasilense Sp7 cells. Therefore, it has been shown for the first time that detection of microbial cells by an electro-acoustic sensor is feasible.

We introduce a new genosensing approach employing CTAB (cetyltrimethylammonium bromide)-coated positively charged colloidal gold nanoparticles (GNPs) to detect target DNA sequences by using absorption spectroscopy and dynamic light scattering. The approach is compared with a previously reported method employing unmodified CTAB-coated gold nanorods (GNRs). Both approaches are based on the observation that whereas the addition of probe and target ssDNA to CTAB-coated particles results in particle aggregation, no aggregation is observed after addition of probe and nontarget DNA sequences. Our goal was to compare the feasibility and sensitivity of both methods. A 21-mer ssDNA from the human immunodeficiency virus type 1 HIV-1 U5 long terminal repeat (LTR) sequence and a 23-mer ssDNA from the Bacillus anthracis cryptic protein and protective antigen precursor (pagA) genes were used as ssDNA models. In the case of GNRs, unexpectedly, the colorimetric test failed with perfect cigar-like particles but could be performed with dumbbell and dog-bone rods. By contrast, our approach with cationic CTAB-coated GNPs is easy to implement and possesses excellent feasibility with retention of comparable sensitivity--a 0.1 nM concentration of target cDNA can be detected with the naked eye and 10 pM by dynamic light scattering (DLS) measurements. The specificity of our method is illustrated by successful DLS detection of one-three base mismatches in cDNA sequences for both DNA models. These results suggest that the cationic GNPs and DLS can be used for genosensing under optimal DNA hybridization conditions without any chemical modifications of the particle surface with ssDNA molecules and signal amplification. Finally, we discuss a more than two-three-order difference in the reported estimations of the detection sensitivity of colorimetric methods (0.1 to 10-100 pM) to show that the existing aggregation models are inconsistent with the detection limits of about 0.1-1 pM DNA and that other explanations should be developed.

Functionalized gold nanoparticles with controlled geometrical and optical properties are the subject of intensive studies and biomedical applications, including genomics, biosensorics, immunoassays, clinical chemistry, laser phototherapy of cancer cells and tumors, the targeted delivery of drugs, DNA and antigens, optical bioimaging and the monitoring of cells and tissues with the use of state-of-the-art detection systems. This work will provide an overview of the recent advances and current challenges facing the biomedical application of gold nanoparticles of various sizes, shapes, and structures. The review is focused on the application of gold nanoparticle conjugates in biomedical diagnostics and analytics, photothermal and photodynamic therapies, as a carrier for delivering target molecules, and on the immunological and toxicological properties. Keeping in mind the huge volume and high speed of the data update rate, 2/3 of our reference list (certainly restricted to 250 Refs.) includes publications encompassing the past 5 years.

RESUMO

Animals were immunized with transmissible gastroenteritis virus conjugated with gold nanoparticles. The resultant antibodies had a higher titer than antibodies produced in response to native virus. Immunization with the antigen-colloidal gold complex led to a significant increase of the peritoneal macrophages respiratory activity and of plasma IFN-γ level in immunized animals.

The electrooptical abilities of the microbial suspensions during a cells interaction with antibodies (ABs) of a different specificity have been studied on the example of the Azospirillum brasilense Sp245 cells and their interaction with the polyclonal monospecific and polyspecific antibodies. Measuring of the orientational spectra of the cells has been performed using the ELUS electrooptical analyzer. A discrete frequency set of an orienting electric field (740, 1000, 1450, 2000, and 2800 kHz) was used. It has been shown that an interaction of the polyspecific AB with the investigated cells redoubles the value of an electrooptical signal of the cells' suspension as compared with the monospecific antibodies. These findings can be used for a development a new method of microorganism detection.

The bacterial action of gentamicin and that of a mixture of gentamicin and 15-nm colloidal-gold particles on Escherichia coli K12 was examined by the agar-well-diffusion method, enumeration of colony-forming units, and turbidimetry. Addition of gentamicin to colloidal gold changed the gold color and extinction spectrum. Within the experimental errors, there were no significant differences in antibacterial activity between pure gentamicin and its mixture with gold nanoparticles (NPs). Atomic absorption spectroscopy showed that upon application of the gentamicin-particle mixture, there were no gold NPs in the zone of bacterial-growth suppression in agar. Yet, free NPs diffused into the agar. These facts are in conflict with the earlier findings indicating an enhancement of the bacterial activity of similar gentamicin-gold nanoparticle mixtures. The possible causes for these discrepancies are discussed, and the suggestion is made that a necessary condition for enhancement of antibacterial activity is the preparation of stable conjugates of NPs coated with the antibiotic molecules.

This work was undertaken to examine the possibility of preparing polyclonal antibodies to diminazene by using colloidal gold nanoparticles as an antigen carrier and adjuvant. The antibodies prepared by us were used for immunodot determination of diminazene in calf-blood plasma. The immunochemical results agreed with those obtained by high-performance liquid chromatographic estimation of diminazene content in animal biological fluids.

The effectiveness of an in vivo synthesis of antibodies to clenbuterol with the use of gold nanoparticles as a carrier was evaluated. For comparison, conjugates of clenbuterol with bovine serum albumin were used in immunization. The serum titer was determined by an ELISA. The antibodies were tested by an immunodot assay with immunogold markers. With both techniques we obtained specific and relatively high-titer antibodies to clenbuterol. It was found that the antibodies obtained with the use of gold nanoparticles were not inferior in titer to those obtained to the conjugates of clenbuterol with the protein but surpassed them in specificity.
