RESUMO
The enzymic interaction between acetoacetate and glycine which are added outside is established to occur in the tissue homogenates of the liver, kidneys and spleen of rats. The interaction leads to a decrease of the both components in the equivalent quantities. Homogeneous crystalline preparations of the enzyme catalyzing the above mentioned reaction are isolated from the liver tissues and purified. Certain properties of the enzyme are studied.

The blood oxaloacetate, alpha-ketoglutarate, malate and citrate concentrations and citrate excretion with the urine were studied in 86 patients with diabetes mellitus. An enhanced citrate content and lowered oxaloacetate, malate and alpha-ketoglutarate concentration were detected in the blood of patients with diabetes mellitus. Compensation of the disease and combined pathogenetic therapy are factors, promoting normalization of Krebs cycle metabolite content.

The paper deals with studies of the effect of the 10-day complex therapy with carbostimulin application on the content of the tricarboxylic cycle and glycolysis metabolites, total lipids, cholesterol, triglycerides in blood and potassium and sodium in blood serum of patients with diabetes mellitus. It is established that under the effect of carbostimulin the content of oxaloacetate, lactate and alpha-glycerophosphate becomes normal, the lactate/pyruvate ratio elevated in diabetes decreases, which evidences for intensification of reduction processes in the cytoplasm.

Application of carbostimulin, the preparation stimulating CO2 fixation in tissues, in a complex therapy of diabetes mellitus patients rises the CO2 level, restores the total content of alpha-ketoacids and free amino acids in blood, increases (within the physiological normal limits) the urea content in blood and urine of diabetes mellitus patients.

In rabbits with alloxane diabetes the intensity of [I-14C] glycine label incorporation into various tissue components is considerably lowered. Stimulation of carboxylation in animals under these conditions intensifies the label incorporation into proteins of the liver, kidneys, spleen as well as into blood serum albumin and globulin. The level of lipids, glycogen in the liver, the total content of proteins in blood serum and their specific radioactivity increase. All this evidences for intensification of biosynthetic processes and for a possibility to normalize the metabolism with diabetes by stimulating the processes of carboxylation.

Feeding carboxylin and sodium citrate to rabbits with alloxane diabetes, normalizes the disturbed contents of malate, alpha-ketoglutarate, oxaloacetate, citrate and pyruvate in the blood and liver of these animals restores the total content of alpha-keto-and free amino acids, increases considerably the urea content in the liver.

The total content of ketoacids, ketonic bodies and state of acid-base balance in blood of sugar diabetes patients was studied as affected by 10-day complex therapy with application of carbostimulin. It is established that carbostimulin lowers the content of ketonic bodies, increases the total content of ketoacids, concentration of buffer bases, CO2, HCO3-. Application of the complex therapy without the preparation causes only a tendency to study the mentioned indexes for the same period of the treatment.
