Chenopodium murale (Syn. Chenopodiastrum murale) (amaranthaceae) is used in the rural Egypt to treat oral ulcers in newborn children. The current study aimed to discover new natural products suitable for treating candidiasis disease with minimal side effects. Characterization of bioactive compounds by LC-QTOF-HR-MS/MS from Chenopodium murale fresh leaves' juice (CMJ) was carried out in order to elucidate their potential anti-fungal and immunomodulatory effects in oral candidiasis in immunosuppressed rats. An oral ulcer candidiasis model was created in three stages: (i) immunosuppression by drinking dexamethasone (0.5 mg/L) for two weeks; (ii) Candida albicans infection (3.00 × 106 viable cell/mL) for one week; and (iii) treatment with CMJ (0.5 and 1.0 g/kg orally) or nystatin (1,000,000 U/L orally) for one week. Two doses of CMJ exhibited antifungal effects, for example, through a significant reduction in CFU/Petri (236.67 ± 37.86 and 4.33 ± 0.58 CFU/Petri), compared to the Candida control (5.86 × 104 ± 1.21 CFU/Petri), p ≤ 0.001. In addition, CMJ significantly induced neutrophil production (32.92% ± 1.29 and 35.68% ± 1.77) compared to the Candida control level of 26.50% ± 2.44. An immunomodulatory effect of CMJ at two doses appeared, with a considerable elevation in INF-γ (103.88 and 115.91%), IL-2 (143.50, 182.33%), and IL-17 (83.97 and 141.95% Pg/mL) compared with the Candida group. LC-MS/MS analysis operated in negative mode was used for tentative identification of secondary (SM) metabolites based on their retention times and fragment ions. A total of 42 phytoconstituents were tentatively identified. Finally, CMJ exhibited a potent antifungal effect. CMJ fought Candida through four strategies: (i) promotion of classical phagocytosis of neutrophils; (ii) activation of T cells that activate IFN-γ, IL-2, and IL-17; (iii) increasing the production of cytotoxic NO and H2O2 that can kill Candida; and (iv) activation of SOD, which converts superoxide to antimicrobial materials. These activities could be due to its active constituents, which are documented as anti-fungal, or due to its richness in flavonoids, especially the active compounds of kaempferol glycosides and aglycone, which have been documented as antifungal. After repetition on another type of small experimental animal, their offspring, and an experimental large animal, this study may lead to clinical trials.

OBJECTIVE: To discover a drug from natural triterpenes that has no side effects and is effective in treating Alzheimer's disease. We predict that the drug will be put on the market soon and achieve success. METHODS: The methanolic extract of M. leucodendron leaves was fractionated and subjected to different chromatographic techniques to isolate two new triterpene glycosides alongside five known compounds kaempferol 3, quercetin 4, quercetin3-O-ß-D-glucopyranoside 5, kaempferol3- O-ß-D-glucopyranoside 6 and kaempferol3-O-α-L-rhamnoside 7. The structures of compounds 1 and 2 were elucidated by spectroscopic analysis and chemical means. RESULTS: Two new triterpene glycosides, 21-O-α-L-rhamnopyranosyl-olean-12-ene-3-O-[α-Lrhamnopyranosyl (1-4) ß-D-galactopyranosyl (1-4) ß-D-glucouronopyranoside]1 and 21-O-α-Lrhamnopyranosyl- olean-12-ene-3-O-[α-L-rhamnopyranosyl (1→4) ß-D-galactopyra-nosyl (1→4) ß-D-galactopyranoside] 2, were isolated for the first time from 70% aqueous methanolic extract (AME) of M. leucodendron leaves. The inhibitory activities of the said compounds toward acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) were then assayed. Both compounds exhibited significant inhibitory activities toward the two enzymes, and evidence indicated that compound 2 was a more effective inhibitor than compound 1. CONCLUSION: Compounds 1 and 2 have a significant role in inhibiting the enzymes acetylcholinesterase and butyrylcholinesterase.

Copper is an essential metal that occurs chronically in the environment and affects the development and physiology of aquatic insects. In excess amounts, it can impair their nervous system and behaviour. We tested the anti-predatory behaviour of Cx. pipiens larvae after seven days exposure with several concentrations of copper up to 500 mg L-1. We measured responses to non- consumptive (predation cues) and consumptive predation (dragonfly larvae) across two generations. We also tested the accumulated effect of copper on AChE enzyme activity. We exposed half of treated and control larvae to predation cues (water with predator odour and crushed conspecifics) and the other half to water without predation cues. We evaluated total distance moved and velocity. Copper reduced the distance moved and velocity, with stronger effects in the second generation. Copper had no significant effect on larvae eaten by dragonflies. Copper inhibited the AChE enzyme across both generations at 500 µg L-1. Copper can affect the nervous system directly by inhibiting AChE activity, and possibly also by impairing the olfaction sensors of the larvae, resulting in larval inability to detect predation cues.

The goal of this research is to determine what chemicals are present in two different extracts (hexane and acetone) of Vicia faba (family Fabaceae, VF) peels and evaluate their effectiveness as a corrosion inhibitor on mild steel in a saline media containing 3.5% sodium chloride. Gas chromatography-mass spectrometry (GC/MS) was used to determine the composition of various extracts. It was determined that fourteen different chemicals were present in the hexane extract, the most prominent of which were octacosane, tetrasodium tetracontane, palmitic acid, and ethyl palmitate. Heptacosane, lauric acid, myristic acid, ethyl palmitate, and methyl stearate were some of the 13 chemicals found in the acetone extract. Using open circuit potential, potentiodynamic polarisation, and electrochemical impedance spectroscopic techniques, we can approximate the inhibitory effects of (VF) extracts on mild steel. The most effective inhibitory concentrations were found to be 200 ppm for both the hexane and acetone extracts (97.84% for the hexane extract and 88.67% for the acetone extract). Evaluation experiments were conducted at 298 K, with a 3.5% (wt/v) NaCl content and a flow velocity of about 250 rpm. Langmuir adsorption isotherm shows that the two extracts function as a mixed-type inhibitor in nature. Docking models were used to investigate the putative mechanism of corrosion inhibition, and GC/MS was used to identify the major and secondary components of the two extracts. Surface roughness values were calculated after analyzing the morphology of the metal's surface with and without (VF) using a scanning electron microscope (SEM). The results showed that throughout the surface of the mild steel, a thick adsorbate layer was formed. Quantum chemical calculations conducted on the two extracts as part of the theoretical research of quantum chemical calculation demonstrated a connection between the experimental analysis results and the theoretical study of the major chemical components.

CONTEXT: The roots of Tagetes lucida Cav. (Asteraceae) have antioxidant and antimicrobial properties. OBJECTIVE: This study aimed to examine the hepatoprotective effects of T. lucida roots ethanol extract (TLRE) using carbon tetrachloride (CCl4)-induced hepatotoxicity in rats. MATERIALS AND METHODS: The active ingredients of TLRE were identified by high-performance liquid chromatography, infra-red spectrum, and mass spectrometric procedures. Ninety rats were distributed into four main groups: positive, therapeutic, protective, and negative group. The therapeutic group was implemented using CCl4 (a single dose of 2 mL/kg) before TLRE or silymarin administration. Meanwhile, the protective group was implemented by administering CCl4 (a single dose of 2 mL/kg) after force-feeding TLRE or silymarin. Each therapeutic and protective group was divided into three subgroups: force-fed with saline, TLRE (500 mg/kg), and silymarin (25 mg/kg). The positive group was split into two subgroups that were force-fed TLRE and silymarin. Positive, therapeutic, and protective groups were compared to the negative group (untreated rats). CCl4, TLRE, and silymarin were orally administrated using a gastric tube. RESULTS: In the therapeutic and protective groups, TLRE significantly reduced liver enzymes, i.e., aspartate aminotransferase (12.47 and 6.29%), alanine aminotransferase (30.48 and 11.39%), alkaline phosphatase (17.28 and 15.90%), and cytochrome P450-2E1 (39.04 and 48.24%), and tumour necrosis factor-α (53.72 and 53.72%) in comparison with CCl4-induced hepatotoxicity controls. CONCLUSIONS: TLRE has a potent hepatoprotective effect with a good safety margin. After a repeated study on another type of small experimental animal, their offspring, and an experiment with a large animal, this study may lead to clinical trials.

Aquatic invertebrates can be exposed to copper from various sources, including agricultural applications. For example, concentrations up to 1000 µg L-1 are found within rice fields, where copper-containing formulations are used as fungicides and algaecides. We conducted toxicity tests to study lethal and sublethal effects of copper sulfate pentahydrate on all immature stages across two generations of Culex pipiens mosquitoes as our model organism. Mortality was dose-dependent at concentrations of 500 µg L-1 and above in the first generation, and 125 µg L-1 and above in the second generation. The median lethal concentrations (LC50) of copper sulfate pentahydrate for larval Cx. pipiens were 476 ± 30.60 µg L-1 and 348.67 ± 23.20 µg L-1 for the first and second generations, respectively. Generation one pupation decreased from 96% in controls to 48% at 500 µg L-1, while the second-generation pupation decreased from 96% in controls to 17.5% at 500 µg L-1. Mortality during the pupal stage varied from 2 to 10% at 500 µg L-1 of first and second generations, respectively. Higher levels also delayed development to adulthood in both generations. The duration of the immature period was 14.8 days in controls in both generations, but when exposed at 500 µg L-1 it increased to 18.8 days in the first generation and to 20.5 days in the second generation. The chronic, multi-generation exposures in this study showed greater toxicity than reported for shorter exposures of Cx. pipiens, and confamilial taxa like Culex hortensis and Anopheles hispaniola.

Tropospheric ozone (O3) is a naturally occurring gas in the atmosphere. However, the concentration of O3 increased in the twentieth century. Although the effects of O3 on vegetation have been extensively studied since the 1950s, limited information exists regarding the effects of O3 on insect herbivores. In particular, evidence is lacking regarding the effects of O3 on the biology of insect herbivores. Agelastica coerulea Baly (1874) is a coleopteran species that grazes on Betulaceae plants. In this study, to investigate the effects of O3 on A. coerulea biology for the first time, female adult insects were collected from Japanese white birch trees grown in a Free Air Controlled Exposure System (FACE) in Sapporo, Japan. These beetles inhabited trees exposed either to ambient or to elevated O3 for 23 days. After collection, the enzyme activities in the beetles were measured. Elevated O3 led to a greater total antioxidant activity and lower α- and ß-esterase activities, a phenomenon that may suggest an increased resistance of the beetles to stress. Our results are further discussed with regard to biological and toxicological aspects. Collectively, our findings indicate that total antioxidants and α- and ß-esterase activities can serve as effective O3 biomarker systems in this beetle species. This adaptive response of the beetle, which was induced by moderate O3 exposure, should be further tested across generations and for its protection against greater exposure.
