RESUMO

Iron bioavailability in unleavened white and wholegrain bread made from two commercial wheat varieties was assessed by measuring ferritin production in Caco-2 cells. The breads were subjected to simulated gastrointestinal digestion and the digests applied to the Caco-2 cells. Although Riband grain contained a lower iron concentration than Rialto, iron bioavailability was higher. No iron was taken up by the cells from white bread made from Rialto flour or from wholegrain bread from either variety, but Riband white bread produced a small ferritin response. The results probably relate to differences in phytate content of the breads, although iron in soluble monoferric phytate was demonstrated to be bioavailable in the cell model. Nicotianamine, an iron chelator in plants involved in iron transport, was a more potent enhancer of iron uptake into Caco-2 cells than ascorbic acid or 2'-deoxymugineic acid, another metal chelator present in plants.

Increasing nitrogen supply can increase Fe and Zn concentrations in wheat grain, but the underlying mechanisms remain unclear. Size-exclusion chromatography coupled with inductively coupled plasma mass spectrometry was used to determine Fe and Zn speciation in the soluble extracts of grain pearling fractions of two wheat cultivars grown at two N rates (100 and 350 kg of N ha(-1)). Increasing N supply increased the concentrations of total Fe and Zn and the portions of Fe and Zn unextractable with a Tris-HCl buffer and decreased the concentrations of Tris-HCl-extractable (soluble) Fe and Zn. Within the soluble fraction, Fe and Zn bound to low molecular weight compounds, likely to be Fe-nicotianamine and Fe-deoxymugineic acid or Zn-nicotianamine, were decreased by 5-12% and 4-37%, respectively, by the high N treatment, whereas Fe and Zn bound to soluble high molecular weight or soluble phytate fractions were less affected. The positive effect of N on grain Fe and Zn concentrations was attributed to an increased sink in the grain, probably in the form of water-insoluble proteins.

This study aimed to determine differences among wheat cultivars in the distribution and speciation of Fe and Zn in grain milling fractions. Cultivars with higher Fe and Zn concentrations in the wholemeal flour were found to contain higher concentrations in the white flour. Soluble Fe and Zn were extracted and analyzed by size exclusion-inductively coupled plasma mass spectrometry. Fe speciation varied between milling fractions with a low molecular weight (LMW) complex likely to be Fe-deoxymugenic acid/nicotianamine being the predominant extractable Fe species in white flour, accounting for approximately 85% of the extractable Fe. Bran fractions had a lower amount of LMW-Fe form but more as soluble Fe-phytate and an unidentified high molecular weight peak. In the white flour fraction soluble Zn was found to be present mainly as a LMW peak likely to be Zn-nicotianamine complex. Soluble Fe-phytate was found in the white flour fraction of a high-Fe cultivar but not in a low-Fe cultivar.

Most research on micronutrients in maize has focused on maize grown as a monocrop. The aim of this study was to determine the effects of intercropping on the concentrations of micronutrients in maize grain and their acquisition via the shoot. We conducted field experiments to investigate the effects of intercropping with turnip (Brassica campestris L.), faba bean (Vicia faba L.), chickpea (Cicer arietinum L.), and soybean (Glycine max L.) on the iron (Fe), manganese (Mn), copper (Cu) and zinc (Zn) concentrations in the grain and their acquisition via the above-ground shoots of maize (Zea mays L.). Compared with monocropped maize grain, the grain of maize intercropped with legumes showed lower concentrations of Fe, Mn, Cu, and Zn and lower values of their corresponding harvest indexes. The micronutrient concentrations and harvest indexes in grain of maize intercropped with turnip were the same as those in monocropped maize grain. Intercropping stimulated the above-ground maize shoot acquisition of Fe, Mn, Cu and Zn, when averaged over different phosphorus (P) application rates. To our knowledge, this is the first report on the effects of intercropping on micronutrient concentrations in maize grain and on micronutrients acquisition via maize shoots (straw+grain). The maize grain Fe and Cu concentrations, but not Mn and Zn concentrations, were negatively correlated with maize grain yields. The concentrations of Fe, Mn, Cu, and Zn in maize grain were positively correlated with their corresponding harvest indexes. The decreased Fe, Mn, Cu, and Zn concentrations in grain of maize intercropped with legumes were attributed to reduced translocation of Fe, Mn, Cu, and Zn from vegetative tissues to grains. This may also be related to the delayed senescence of maize plants intercropped with legumes. We conclude that turnip/maize intercropping is beneficial to obtain high maize grain yield without decreased concentrations of Fe, Mn, Cu, and Zn in the grain. Further research is required to clarify the mechanisms underlying the changes in micronutrient concentrations in grain of intercropped maize.

Food can be an important source of inorganic As for human intake. Recent studies have focused on rice, while little information is available on As accumulation, distribution, and speciation in wheat, which is the second most important food grain cereal. Grain samples of 26 wheat cultivars grown in five field trials located in productive farming regions in Europe were therefore analyzed for As concentration and speciation. Grain from four trials contained low concentrations of total As (7.7 +/- 5.4 microg kg(-1)), reflecting low levels of As in the soils (1.3-11 mg kg(-1)). In contrast, at one of the trial sites the As level in the soil was greater (29 mg kg(-1)), and much higher As concentrations (69 +/- 17 microg kg(-1)) were present in the wheat grain. Milling of wheat grain into bran and white flour fraction showed the concentration of As in the bran, with a 3.8-4.7-fold higher As concentration than in the white flour. Two methods (a phosphate buffer solution and 1% HNO(3)) were used to extract As species from wholemeal, bran, and white flour of wheat, with average extraction efficiencies of 65% and 88%, respectively. Only inorganic As was found in the extracts, with no methylated As being detected. The contribution of wheat to human intake of inorganic As is small for wheat crops grown in uncontaminated soils but becomes significant for those grown in soils with elevated As. In the latter case, milling can be used to reduce the As concentration in the white flour.
