The relationship between fruit position, ripening stage, and variety has not been well studied in pepper plants. To understand the interaction of these factors, a diversity of phytochemical traits as well as antioxidant activity were investigated with agronomic traits in eleven hot pepper varieties collected from the upper and lower parts of the plant and harvested at three maturity stages (green, orange, and red). Capsaicin content (CAP) showed a relatively high genetic effect; on the contrary, total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activity were more affected by the ripening stage and fruit position. The CAP values ranged from 0.29 ('FKbM') to 0.77 ('Bka') mg CAP equivalents g-1 DW. The ripening stage was the predominant factor for TPC, TFC, DPPH, and FRAP. There was no significant interaction between A × FP, A × RS, and FP × RS for all agro-morphological fruit traits. Variety, fruit position, and ripening stage effects are more significant than all interactions calculated. Lower fruit positions in all samples showed a maximum fruit size, whereas phytochemical traits and yield per plant were relevant in the upper parts, and Phytochemical traits and yield per plant were significantly correlated. From PCA and cluster analysis, all varieties showed the highest biochemical and antioxidant levels with moderate fruit size, except the 'Bel' variety that showed the smallest fruit traits with high yields, and the 'FKbM' and 'FKbK' varieties that showed the highest fruit size but low yields. This study supplies information to identify interesting cultivars with considerable levels of bioactive and phytochemical metabolites, which is useful for breeding programs of novel varieties.

Genetic diversity and differences among durum-wheat cultivars evolved in various regions of the world are important for sustainable production in the current climate change scenario. Information regarding genetic differences was also important for the correct choice of parental material for the selection of high quality cultivars. Two elite and six obsolete cultivars of durum-wheat were characterized with 25-simple sequence repeats (SSR) markers. All accessions were evaluated for 2-agronomic-traits (Yield (Y) and Thousand-Kernel-Weight (TKW)) and 11 grain quality-traits (grain protein content (GPC), grain moisture contents (H), carotene content (CT), sedimentation test (SDS), gluten content (GC), gluten index (GI), semolina color index (L*, a*, b*) and alveographic parameters (W and P/L)) under randomized complete block design with three replication for two crop seasons (2015-2017). Genetic characterization through SSR markers revealed 126 alleles with an average of 5.04 alleles locus-1 and had average 0.79 polymorphism information content (PIC). The comparisons revealed that elite accessions were more productive in terms of grain yield and TKW, whereas obsolete accessions showed high GPC and end-use quality-traits. The generated dendrogram based on SSR markers, agronomic, seed quality-traits clearly differentiate the genotypes in two main groups obsolete and elite accessions. Analysis of correlation revealed a significant association between the traits TKW, Y, b*, a*, GPC, GC, SDS and H. High genetic diversity found between elite and obsolete cultivars for parameters such as yield, end-use quality and their correlation with SSR markers could help breeders for an eventual breeding program on durum-wheat.

Background: Consanguinity increases the incidence of recessive diseases such as beta-thalassaemia major (ßTM), one of the most prevalent lethal inherited diseases in the world.Aim: This study aims to identify the frequency of endogamy and consanguinity in two Mediterranean ßTM populations and to study the implication of socio-economic factors.Subjects and methods: A trans-sectional study was conducted in 203 Tunisian families and 75 Italian families. Data were collected using a questionnaire completed by patients and parents.Results: Complete endogamy and consanguinity were observed in 82.75% and 62.56% of Tunisian families, respectively. Complete endogamy was found in 90.67% of Italian families, no consanguinity was noted. The low occupation status of Tunisian mothers was associated with an increasing frequency of consanguinity (p = .01) and endogamy (p = .0003). Consanguinity was associated with low education level (p = .012) and low occupation status (p=.047) of fathers. No significant association was found between endogamy and socio-economic factors in the Italian sample.Conclusions: High consanguinity and endogamy rates in Tunisian families may explain the frequency of ßTM in Tunisia. The high endogamy rate in Italian families could also increase the frequency of ßTM. Identification of geographical distribution and socio-economic factors leading to endogamy and consanguinity in these populations might help to improve ßTM prevention.

This study aimed to characterize eight accessions of Vicia narbonensis L. originated from different Mediterranean countries. The cytology of these species is rarely known despite the fact of its great socio-economical and ecological interest in these arid and semi-arid zones. This work aimed mainly to characterize the karyotype, morphological pod and seed traits of the species. Karyotypes of all accessions were similar to a diploid number of 2n = 2x = 14. All the accessions have submetacentric chromosomes with a secondary constriction attached to the long arm of pair VII close to the centromere. Variation in chromosome size was observed; it ranged from 5.86 µm to 7.62 µm. Indices of karyotype asymmetry were calculated as the total form percentage (TF%) and symmetric indices (Syi) which ranged from 33.75% to 35.42% and from 51.01% to 54.85%, respectively. The predominance of submetacentric chromosomes indicated that the karyotype is symmetrical and can be considered as primitive. However, the analysis of quantitative parameters measured on pods and seeds showed a significant variation between accessions. A relationship between centromeric index and the pod beak length was found. Estimation of phenotypic diversity using the Shannon diversity index (H') showed that the length, the seed color and the number of seeds per pod are the most polymorphic traits with respectively, H' = 0.92, 0.80 and 0.83. Cluster analysis of karyological, pod and seed traits showed four groups of accessions. This clustering is partially due to the geographical origin of the studied accessions. The variation in chromosome size, pod and seed traits could offer potentially valuable genetic resources for the improvement of V. narbonensis which is considered as neglected and underutilized crop species (NUCS).

The human gyrovirus derived protein Apoptin (HGV-Apoptin) a homologue of the chicken anemia virus Apoptin (CAV-Apoptin), a protein with high cancer cells selective toxicity, triggers apoptosis selectively in cancer cells. In this paper, we show that HGV-Apoptin acts independently from the death receptor pathway as it induces apoptosis in similar rates in Jurkat cells deficient in either FADD (fas-associated death domain) function or caspase-8 (key players of the extrinsic pathway) and their parental clones. HGV-Apoptin induces apoptosis via the activation of the mitochondrial intrinsic pathway. It induces both mitochondrial inner and outer membrane permebilization, characterized by the loss of the mitochondrial potential and the release into cytoplasm of the pro-apoptotic molecules including apoptosis inducing factor and cytochrome c. HGV-Apoptin acts via the apoptosome, as lack of expression of apoptotic protease-activating factor 1 in murine embryonic fibroblast strongly protected the cells from HGV-Apoptin-induced apoptosis. Moreover, QVD-oph a broad-spectrum caspase inhibitor delayed HGV-Apoptin-induced death. On the other hand, overexpression of the anti-apoptotic BCL-XL confers resistance to HGV-Apoptin-induced cell death. In contrast, cells that lack the expression of the pro-apoptotic BAX and BAK are protected from HGV-Apoptin induced apoptosis. Furthermore, HGV-Apoptin acts independently from p53 signal but triggers the cytoplasmic translocation of Nur77. Taking together these data indicate that HGV-Apoptin acts through the mitochondrial pathway, in a caspase-dependent manner but independently from the death receptor pathway.

Cell death is a fundamental ingredient of life. Thus, not surprisingly more than one form of cell death exists. Several excellent reviews on various forms of cell death have already been published but manuscripts describing interconnection and interdependence between such processes are uncommon. Here, what follows is a brief introduction on all three classical forms of cell death, followed by a more detailed insight into the role of p53, the master regulator of apoptosis, and other forms of cell death. While discussing p53 and also the role of caspases in cell death forms, we offer insight into the interplay between autophagy and apoptosis, or necrosis, where autophagy may initially serve pro-survival functions. The review moves further to present some details about less researched forms of programmed cell death, namely necroptosis, necrosis and mitoptosis. These "mixed" forms of cell death allow us to highlight the interconnected nature of cell death forms, particularly apoptosis and necrosis. The interdependence between apoptosis, autophagy and necrosis, and their significance for cancer development and treatment are also analyzed in further parts of the review. In the concluding parts, the afore-mentioned issues will be put in perspective for the development of novel anti-cancer therapies.

Ten varieties and eight generations (2F1, 2F2, 2B1 and 2B2) of durum wheat derived from two crosses were evaluated for resistance to natural infection by Stagonospora nodorum blotch (SNB) at the 2-3 and 6-7 leaf stages at two sites over two years. There were significant differences in the incidence of SNB between leaf stages in most of the wheat varieties, with resistance being most evident at the 6-7 leaf stage. Separate analyses of the mean values for each generation showed that the genetic mechanism of defense against the pathogen depended upon the leaf stage. At the 2-3 leaf stage, only additive and dominance effects were implicated in the control of SNB for the two crosses at the two sites and for the two replications. For the 6-7 leaf stage, inheritance was more complicated and an epistatic effect was involved. Narrow-sense heritability values (range: 0.63-0.67) were consistent between crosses and leaf stages. These findings indicate a lack of resistance to SNB at the 2-3 leaf stage whereas resistance was observed at the 6-7 leaf stage and involved the genetic mechanisms of plant defense such as epistasis.

This study evaluated the types of gene action governing the inheritance of resistance to Phytophthora nicotianae necrosis in populations derived from two crosses involving two susceptible (Beldi and Nabeul II) and one resistant (CM334) cultivars of pepper (Capsicum annuum L.). Populations, composed of Pr, Ps, F1,F2,BC1Pr, and BC1Ps generations, were inoculated with six P. nicotianae isolates. Generation means analysis indicated that an additive-dominance model was appropriate for P. nicotianae isolates PnKo1,PnKo2 and PnKr1, which showed low aggressiveness in the two crosses. For the more aggressive isolates PnBz1,PnBz2 and PnKr2, epistasis was an integral component of resistance in the two crosses. The presence of epistasis in the resistance of pepper to P. nicotianae was dependent on the level of aggressiveness of the isolates. Selection in pepper with less aggressive isolates was efficient, but not with more aggressive isolates; on the other hand, selection with more aggressive isolates was more stable. The minimum number of genes controlling resistance was estimated at up to 2.71. In the majority of cases, the additive variance was significant and greater than the environmental and dominance variance.

Thirteen Aegilops geniculata Roth (geniculate goat grass) accessions from collection of the North and Central Tunisian (Cap-Bon, Mogodses, Kroumiry and the Dorsal areas) were used to assess its genetic diversity by morphological and Random Amplified Polymorphic DNA (RAPD) data and to evaluate relationship between morphological and RAPD markers. Nineteen morphological traits were analyzed on all accessions using Principal Analysis Component (PCA) and clusters were constrained based on median joining distances. Nineteen arbitrary universal primers were used for the amplification of random DNA sequences and generated 212 bands ranging from 0.5 to 3 kb with 71.27% polymorphism across the 13 accessions. Both RAPD and morphological data classified accessions in two main groups. Both methods were used to compare how morphological traits and RAPD molecular markers described accessions relationship and showed a high degree of variation among analyzed accessions, indicating an important source of genetic diversity that can be used in future breeding programs. Morphological PCA traits and cluster indicated climatic stage. In fact, they grouped Ae. geniculata accessions according to genetic criteria such as earliness and high kernel yield. Comparison of morphological and molecular data using the Mantel test indicated a non significant correlation (r = -0.268). Nevertheless, RAPD and selected morphological characters appear as useful and complementary techniques for evaluation of genetic diversity in Ae. geniculata.

This study aims at characterization four cultivars of the olive trees, Chétoui, Chemlali, Gerboui, and Chaïbi, cultivated in three different geographical locations, from pomological and technological points of view. The pomological characters of the fruit are influenced by the geographical location. Each individual of the same cultivar expresses different pomological characters. We have noted a significant fluctuation of the flush percentage in three Chaïbi individuals according to their geographical site; it varies from 49.06 to 82.19%. The three Gerboui individuals showed a significant variability of the fruit weight (from 1.13 to 3.17 g). Fluctuations of olive oil contents were also observed. Several fatty acid compositions showed some variation. The oleic and linoleic acid contents varied among individuals from Chétoui and Chaïbi. Moreover, the individuals of the cultivar Chemlali showed a variation of their content in palmitic and palmitoleic acids. Indeed, each individual of a cultivar showed its own potentialities, which are reflected by its pomological and technological characters. According to their geographical location, individuals from a given cultivar displayed diverse potentialities.

This study presents a new approach to obtain dominance estimates without using the full Henderson's mixed model equations (MMEs) related to an additive plus dominance animal model. This reduction could decrease substantially the computing time and hence its cost. In contrast to a procedure that we proposed before, the method developed in this paper does not require D(-1) and provides best linear unbiased prediction (BLUP) of genetic values that is close to that given by processing the full MMEs. In the previous study, we also elaborated an algorithm (denoted xi-REML) in order to approximate restricted maximum likelihood estimation of variance components via the expectation maximization (EM) algorithm. The xi-REML algorithm has been modified to be adapted to our new resolution approach. Through a numerical example, we show that there is a good agreement between REML-(EM), xi-REML and modified xi-REML estimates and that the latter algorithm is more efficient than our first proposition in terms of computing time and memory conservation.

In the case of noninbred and unselected populations with linkage equilibrium, the additive and dominance genetic effects are uncorrelated and the variance-covariance matrix of the second component is simply a product of its variance by a matrix that can be computed from the numerator relationship matrix A. The aim of this study is to present a new approach to estimate the dominance part with a reduced set of equations and hence a lower computing cost. The method proposed is based on the processing of the residual terms resulting from the BLUP methodology applied to an additive animal model. Best linear unbiased prediction of the dominance component d is almost identical to the one given by the full mixed model equations. Based on this approach, an algorithm for restricted maximum likelihood (REML) estimation of the variance components is also presented. By way of illustration, two numerical examples are given and a comparison between the parameters estimated with the expectation maximization (EM) algorithm and those obtained by the proposed algorithm is made. The proposed algorithm is iterative and yields estimates that are close to those obtained by EM, which is also iterative.
