RESUMO

Bacterial-associated wound infections are an obstacle for individuals and the medical industry. Developing versatile, antibiotic-free therapies helps heal wounds more quickly and efficiently. In the current study, fungal metabolites were employed as a reducing agent in fabricating selenium nanoparticles (SeNPs) for improved antibacterial and wound healing properties. Utilizing UV-visible spectroscopy, dynamic light scattering (DLS), zeta potential, X-ray diffraction (XRD), and electron microscopic examination, the properties of the synthesized nanoparticles were extensively evaluated. Myco-synthesized SeNPs demonstrated strong antibacterial activity against Staphylococcus aureus ATCC 6538 with a minimum inhibitory concentration of 0.3125 mg/mL, reducing cell number and shape distortion in scanning electron microscope (SEM) images. SeNPs' topical administration significantly reduced wound area and healing time, exhibiting the least bacterial load after six days compared to controls. After six and 11 days of treatment, SeNPs could decrease proinflammatory cytokines IL-6 and TNF-α production. The histopathological investigation showed a healed ulcer with moderate infiltration of inflammatory cells after exposing mice's skin to SeNPs for six and 11 days. The docking interaction indicated that SeNPs were highly efficient against the IL-6 and TNF-α binding receptors. These findings imply that myco-fabricated SeNPs might be used as topically applied antimicrobial agents for treating skin infections and wounds.

In this research, Orobanche aegyptiaca extract was utilized as an eco-friendly, and cost-effective green route for the construction of bimetallic silver-selenium nanoparticles (Ag-Se NPs). Bimetallic Ag-Se NPs were characterized by XRD, EDX, FTIR, HR-TEM, DLS, SEM/mapping and EDX studies. Antimicrobial, and antibiofilm potentials were tested against some selected pathogenic bacteria and unicellular fungi by ZOI, MIC, effect of UV exposure, and inhibition %. Reaction mechanism was assessed through membrane leakage assay and SEM imaging. HRTEM analysis confirmed the spherical nature and was ranged from 18.1 nm to 72.0 nm, and the avarage particle size is determined to be 30.58 nm. SEM imaging prove that bimetallic Ag-Se NPs presents as a bright particles, and both Ag and Se were distributed equally across O. aegyptiaca extract and Guar gum stabilizers. ZOI results showed that, bimetallic Ag-Se NPs have antimicrobial activity against S. aureus (20.0 nm), E. coli (18.5 nm), P. aeruginosa (12.6 nm), and C. albicans (18.2 nm). In addition, bimetallic Ag-Se NPs were able to inhibit the biofilm formation for S. aureus by 79.48%, for E. coli by 78.79%, for P. aeruginosa by 77.50%, and for C. albicans by 73.73%. Bimetallic Ag-Se NPs are an excellent disinfectant once it had excited by UV light. It was observed that the quantity of cellular protein discharged from S. aureus is directly proportional to the concentration of bimetallic Ag-Se NPs and found to be 244.21 µg/mL after the treatment with 1 mg/mL, which proves the antibacterial characteristics, and explains the creation of holes in the cell membrane of S. aureus producing in the oozing out of the proteins from the S. aureus cytoplasm. Based on the promising properties, they showed superior antimicrobial potential at low concentration (to avoid toxicity) and continued-phase durability, they may use in pharmaceutical and biomedical applications.

Thymoquinone (TQ) is an active constituent in Nigella sativa (black cumin) and is extensively reported for its distinguished antioxidant and anti-inflammatory bioactivities. Despite the local protective response of acute inflammation, it contributes to the development of various disease conditions such as cell death, organ damage, or carcinogenesis. Hence, in this study, the effects of orally administered TQ (50 mg/kg and 100 mg/kg) for 14 days against edema development, oxidative stress, and inflammation were investigated in paw edema induced by carrageenan in mice. Indomethacin (10 mg/kg) was used as a reference drug. The results revealed that TQ reduced the paw edema volume in a time-dependent manner, attenuated acetic acid-provoked writhing movements, and reduced xylene-triggered ear edema. Hematological findings revealed marked normalization of altered counts of WBCs, and platelets. Furthermore, paw tissue levels of malondialdehyde and nitric oxide showed marked decreases together with increases in nuclear factor erythroid 2-related factor 2, glutathione, superoxide dismutase, catalase, glutathione peroxidase, and glutathione reductase after TQ administration. Additionally, TQ decreased pro-inflammatory mediators, such as interleukin-1 beta, tumor necrosis factor-alpha, interleukin-6, monocyte chemoattractant protein-1, C-reactive protein, myeloperoxidase, and nuclear factor kappa-B in the inflamed paw tissue. Moreover, appreciable decreases were recorded in cyclooxygenase-2 and its product prostaglandin E2 and the immune reaction of tumor necrosis factor-alpha in TQ-treated mice. Histopathological findings further validated the potential antiedematous, anti-inflammatory power of TQ in inflamed tissues. Conclusively, the results encourage the potent application of TQ to subside acute inflammatory events because of its striking antioxidant and anti-inflammatory properties in inflamed paw tissue.

In this study, the mitigative impact of bradykinin potentiating factor (BPF) and low doses of γ-irradiation (LDR) were evaluated against doxorubicin (DOX) hepatotoxicity through Ang II/AMPK crosstalk. Rats have received a single dose of DOX (10 mg/kg, i.p.). BPF administration at a dose of 1 µg/g (b.wt./twice a week) was started one week before the administration of DOX and followed throughout the study for another consecutive week where LDR rats were subjected to two low fractions of γ-irradiation; 0.5 Gy/fraction/week up to the cumulative dose of 1 Gy at 7 days before and after doxorubicin administration. DOX produced a remarkable disturbance in serum hepatic enzymes activities, hepatic oxidative stress indices, as well as hepatic inflammatory and fibrotic markers in response to a marked elevation in hepatic angiotensin II (Ang II) together with marked depression in hepatic AMP-activated protein kinase (AMPK) expressions. The combination of BPF and LDR produced a significant improvement in all examined parameters as well as mitigates hepatic toxicity through inhibition of Ang II induced by DOX, which might also be mediated by AMPK activation. Furthermore, histopathological and immunohistochemical examination reinforced the previous results. In conclusion, these findings shed new light on the mechanism underlying the anti-inflammatory and anti-fibrosis consequence of our remedy and support the potential use of it as a preventive and therapeutic candidate against hepatic toxicity through Ang II/AMPK crosstalk.

In this study, electrospun cellulose acetate - poly(ethylene oxide) nanofibrous membrane was found to be unique in immobilizing bacterial cells. Here, removal of methylene blue in aqueous media was achieved by using isolated species of bacteria (Bacillus paramycoides) from industrial wastewater and immobilized on cellulose acetate- poly(ethylene oxide) nanofibers using DMSO as a solvent. The decolorization time was varied from 0 to 72 h, different dye concentrations from 20 to 200 mg/L and bacterial cells count was investigated to achieve the maximum MB removal by bacteria-immobilized CA/PEO nanofibrous membrane. The effective dye decolorization was achieved within 48 h and MB removal % was around 93%. Furthermore, reusability of the bacteria-immobilized CA/PEO nanofibrous membrane was tested. It was found that after the 4th usage, 44% of the dye decolorization capacity still could be achieved. These results are promising and suggest that bacteria-immobilized CA/PEO nanofibrous membrane could be economically feasible and eco-friendly when used in MB removal from industrial wastewater. Combination of both adsorption and biodegradation methods was found to be effective in MB removal from aqueous media.
