RESUMO

Microplastics and associated adverse effects have been on the global agenda in recent years. Because of its importance as a model organism for studies on developmental biology, Xenopus laevis has been chosen as the study animal in in vitro teratogenesis studies. FETAX test uses early-stage embryos of X. laevis to measure the potential of substances to cause mortality, malformation, and growth inhibition in developing embryos. The aim of this study was to examine the effects of high molecular weight polyvinyl chloride (HMW-PVC) on parental X. laevis frogs and their embryos using the FETAX test. To this purpose, a HMW-PVC dose of 1% of body weight/twice each week was provided to frogs by oral gavage throughout 6 weeks. After the procedure, oocytes and sperms of HMW-PVC-exposed frogs were fertilized and FETAX was applied to selected embryos. After the completion of a 96-h incubation period, tadpoles were examined, their live/dead status were determined, their lengths were measured, and their anomalies were photographed. Besides, excised organs of the parental frogs were referred to histopathology examination. On the other hand, the mRNA expression levels of Hsp70, Myf5, Bmp4, Pax6, and Esr1 genes were determined by applying real-time quantitative PCR method to cDNA which was synthesized from the total RNA of embryos. The results showed that treatment with HMW-PVC dose of 1% of body weight/twice each week caused malformations and decreased viability. Hsp70 and Pax6 gene expression levels significantly decreased in all assay groups, as compared with controls. Lung and intestine tissues showed normal appearance in histopatological examination. Further research is required to explain the whole effects of HMW-PVC exposure on X. laevis embryos.

RESUMO

Ossification of the mamillo-accessory ligament (MAL) is a misunderstood phenomenon; however, many have posited that it can result in nerve entrapment of the medial branch of the dorsal ramus causing zygapophyseal joint related low back pain. The MAL has been studied anatomically by few, yet the data indicate possible associations between ossification of this ligament and spondylosis. It has been proposed that mechanical stress upon the lumbar spine may also lead to progressive ossification of the MAL into a bony foramen.

Among aquatic organisms, fish are particularly susceptible to ingesting microplastic particles due to their attractive coloration, buoyancy, and resemblance to food. However, in previous experimental setups, fish were usually exposed to unrealistically high concentrations of microplastics, or the microplastics were deliberately contaminated with persistent organic chemicals; also, in many experiments, the fish were exposed only during the larval stages. The present study investigated the effects of virgin microplastics in gilt-head seabream (Sparus aurata) after 45 days' exposure at 0.1 g kg-1 bodyweight day-1 to 6 common types of microplastics. The overall growth, biochemical analyses of the blood, histopathology, and the potential of the microplastics to accumulate in gastrointestinal organs or translocate to the liver and muscles were monitored and recorded. The results revealed that ingestion of virgin microplastics does not cause imminent harm to the adult gilt-head seabream during 45 days of exposure and an additional 30 days of depuration. The retention of virgin microplastics in the gastrointestinal tract was fairly low, indicating effective elimination of microplastics from the body of the fish and no significant accumulation after successive meals. Therefore, both the short- and the long-term retention potential of microplastics in the gastrointestinal tract of fish is close to zero. However, some large particles remained trapped in the liver, and 5.3% of all the livers analyzed contained at least one microplastic particle. In conclusion, the dietary exposure of S. aurata to 6 common types of virgin microplastics did not induce stress, alter the growth rate, cause pathology, or cause the microplastics to accumulate in the gastrointestinal tract of the fish.

Economically important sparid fish species, gilthead (Sparus aurata) and white seabream (Diplodus sargus) (Sparidae) are frequently parasitised by diplectanid monogeneans, known to induce severe losses in farming conditions. We have analysed population dynamic of two diplectanid species, Lamellodiscus echeneis and Lamellodiscus ignoratus (Monogenea: Diplectanidae) collected from two bream species in the Beymelek Lagoon (southwest coast of Turkey), comparing it between different host variables (fish size, age and sex) in order to have insight in parasites' ecology, important for managing parasitosis in the intensive aquaculture system. In seabream (N = 127), L. echeneis prevalence was 46.5 % (exact 95 % confidence limits 38.90-54.14), mean abundance 5.64 (bootstrap 95 % confidence limits 4.20-7.65) and mean intensity 12.14 (bootstrap 95 % confidence limits 9.49-15.59). In white seabream (N = 102), L. ignoratus prevalence was 24.5 % (exact 95 % confidence limits 16.53-34.03), mean abundance 1.73 (bootstrap 95 % confidence limits 0.98-3.21) and mean intensity 7.04 (bootstrap 95 % confidence limits 4.60-11.40). Parasites' parameters differed only between seasons in both hosts and between age categories in gilthead, but not in white seabream.

RESUMO

Two new Dactylogyrus spp., D. angorae and D. turcicus have been described from the gills of the endemic cyprinid fishes, Capoeta angorae and C. mauricii from the Anatolian Region of Turkey. The shape of the anchors, hooks and the copulation organ of both species differ from those of the Dactylogyrus spp. found on other Capoeta species known from the Ponto-Caspian and Tigris-Euphrates regions.

During a survey of freshwater fishes from Turkey two species of Acanthocephala, one of them new, were found. Pomphorhynchus tereticollis (Pomphorhynchidae) is reported at 24% prevalence in 37 Cobitis bilseli (Cobitidae) from Lake Beysehir, Konya, for the first time. The eoacanthoacaphalan Triaspiron aphanii gen. n. et sp. n. (Quadrigyridae), at a prevalence of 90%, is described from 29 Aphanius mento (Cyprinodontidae), from Kirkgöz Springs, Antalya. The new genus most closely resembles Raosentis Datta, 1947, both having a small spindle shaped trunk, and Acanthogyrus Thapar, 1927, both having a proboscis armature of three circles of hooks. Triaspiron differs from Raosentis in proboscis shape, cylindrical not globular, proboscis armature, three circles, a total of 16 hooks in all, not four circles, a total of 26-30 hooks in all, and trunk spination, two fields of spines in the anterior field with spines arranged in up to 40 circular rows, not a single field with 9-17 rows of spines. Triaspiron differs from Acanthogyrus in having fewer proboscis hooks, 16 compared with 18-24, arranged in three circles, one anterior and two posteriorly placed, with an unarmed region between, not three circles of hooks evenly spaced, and two fields of trunk spines, not one.

Branchial epithelium of Pseudophoxinus antalyae was lined by both a thick stratified epithelium lining gill arches, gill rakers and primary filaments and a thin epithelium lining the lamellae. Mucous, chloride and rodlet cells, interspersed between pavement cells, were present in the branchial epithelium. With histochemical procedures for the characterization of glycoconjugates, mucous cells showed a strong positive reaction with Periodic acid-Shiff and Alcian Blue at pH 2.5, although with Alcian Blue at pH 0.5 and pH 1.0 the reaction was much weaker. When the combined Alcian Blue (pH 2.5) - Periodic acid-Shiff reaction was performed, most mucous cells were stained purple, whereas by the combined Aldehyde Fuchsin/Alcian Blue (pH 2.5), most cells showed a positive reaction only to Aldehyde Fuchsin. Methylation/ Alcian Blue (pH 2.5) and Methylation/ Saponification/ Alcian Blue (pH 2.5) methods showed the presence of sulphated and carboxylated glycoconjugates in mucous cells. Mucous cells were also detected to stain all metachromatically with Toluidine Blue.
