RESUMO

The aim of this study is to examine the effect of arbuscular mycorrhizal fungi (AMF) colonization on biomass, polyphenol profile, and content of economically important herbs. A pot experiment was performed with marjoram, lemon balm, and marigold applying a commercially available AMF mixture for inoculation. Major polyphenols were identified using HPLC-UV-ESI-qTOFMS on the basis of their UV-vis and mass spectral characteristics, and selected ones were quantified. We showed that AMF can provide different services for each herb. Marjoram had the highest level of fungal colonization (82 M%) followed by lemon balm (62 M%) and marigold (17 M%). AMF inoculation significantly increased the biomass of marjoram (1.5-fold), the number of marigold flowers (1.2-fold), and the yield of rosmarinic acid and lithospermic acid isomers of marjoram (1.5-fold) and lemon balm (1.2-fold). Therefore, the quantity and quality of plant material could be improved by the application of optimized AMF inoculum.

ETHNOPHARMACOLOGICAL RELEVANCE: Oxybaphus nyctagineus (Michx.) Sweet has traditionally been used by several Native American tribes predominantly as a topical anti-inflammatory and analgesic agent. AIM OF THE STUDY: To evaluate the antioxidant, analgesic and anti-inflammatory activity of the extracts prepared from the aerial parts of Oxybaphus nyctagineus and to characterize the major chemical constituents of the bioactive extracts. MATERIALS AND METHODS: Crude polar and apolar extracts (PCE and ACE) of the herb of Oxybaphus nyctagineus were prepared and tested in the models of the CFA-induced hyperalgesia in rat knee and carrageenan-induced paw edema in rat. To identify the active compounds, subfractions were prepared by column chromatography and subjected in vitro assays, such as antioxidant assays (DPPH, peroxynitrite (ONOO-) scavenging), and the LPS-induced IL-1ß release test in human monocytes. Preparative HPLC was employed for the isolation of active substances, while phytochemical analysis was performed by mean of LC-MS/MS and NMR. RESULTS: The topically administered PCE and ACE of Oxybaphus nyctagineus demonstrated a significant analgesic and anti-inflammatory effect in the inflammation animal models. The subfraction A4 of ACE and the subfraction P5 of PCE considerably inhibited the LPS-induced IL-1ß release in human monocytes, while the strongest activity was localized in the subfraction P5 in the antioxidant assays. The HPLC-MS/MS and NMR analysis revealed that 6-methoxyflavonol diglycosides, namely patuletin-3-O-robinobioside (1), 6-methoxykaempferol-3-O-robinobioside (2), spinacetin-3-O-robinobioside (3), and hydroxy-polyenoic fatty acids, namely corchorifatty acid B (4), 9-hydroxy-10E,12Z,15Z-octadecatrienoic acid (9-HOT acid) (5), and 9-hydroxy-10E,12Z-octadecadienoic acid (9-HOD acid) (6) were present in PCE, and in ACE as major compounds. CONCLUSION: The results of this study established a pharmacological evidence for the traditional use of Oxybaphus nyctagineus as an anti-inflammatory agent used topically, and provided data on its phytochemical composition for the first time.

The fruit quality parameters and antioxidant capacity (ferric reducing antioxidant power, FRAP) and total phenolic content (TPC) were determined in 27 apricot cultivars and hybrids of diverse origins. Twenty one- to 35-fold variations were measured among FRAP and TPC values. Besides genotype, harvest year also contributed significantly (P≤0.05) to the variations of TPC presumably due to the climatic differences between years. A subset of genotypes (15) was also analyzed for their antiradical activities (2,2'-diphenyl-1-picrylhydrazyl, DPPH; total radical-scavenging activity, TRSA; water-soluble antioxidant capacity, ACW; and lipid-soluble antioxidant capacity, ACL), and vitamin C contents as well as color indices (CIE H°, L*, and chroma). The hybrid "Preventa" had outstanding FRAP (>10.4 mmol ascorbic acid/L), DPPH (74.45%), TRSA (0.002%), ACW (33587.5 nmol AA/L) and ACL (78.65 nmol Trolox/L), TPC (>2890.0 mg gallic acid/L), and vitamin C (16.17 mg/100 g FW) levels and an average carotenoid content estimated from the hue angle (66.99°). Most antioxidant and antiradical activities correlated significantly except for TRSA; the closest correlation was observed between FRAP and ACW (r=0.952). Only TRSA showed significant correlations with color indices, H° and chroma, suggesting TRSA measures at least a fraction of the antioxidant capacity attributable to apricot carotenoids.
