RESUMO

Intraneuronal inclusions of alpha-synuclein (α-synuclein, α-syn) are commonly found in the brain of patients with Parkinson's disease (PD). The pathogenesis of the abundant α-syn protein in the blood has been extensively studied to understand its properties better. In recent years, peptidome analysis has received increasing attention. In this study, we identified and analyzed serum peptides from wild-type (WT) and the (Thy-1)-h[A30P] alpha-synuclein transgenic mice (SNCA-A30P mice) using liquid chromatography-tandem mass spectrometry (LC-MS/MS). One thousand eight hundred fifty-six peptides from 771 proteins were analyzed. Among them, 151 peptides from 107 proteins were significantly differentially expressed. The glycoprotein VI platelet pathway (GP6) was the pathway's most significant differentially expressed signaling pathway. Cleavage sites of the differentially expressed peptides may reflect protease distribution and activity. We selected the most significantly differentially expressed peptide, VGGDPI, and found that it contained cathepsin K (Ctsk) and trypsin-1 cleavage sites, suggesting that Ctsk and trypsin-1 may be key peptidases in PD. α-syn is a protein associated with the pathogenesis of PD. mutations in several genes, including SNCA, which encodes α-syn, are associated with the development of PD. Bioinformatics analysis of the physiological pathways related to SNCA genes and apoptosis genes found the five most markedly up-regulated proteins: formin homology 2 domain-containing 1 (FHOD1), insulin receptor substrate 1(IRS1), TRPM8 channel-associated factor 1 (TCAF1), glyceraldehyde 3-phosphate dehydrogenase (GAPDH), and interleukin-16 (IL-16). Therefore, the differentially expressed peptides in the five precursor protein domains may be potential bioactive peptides associated with α-syn and apoptosis. This study provides a validated peptidomics profile of SNCA-A30P mice and identifies potentially bioactive peptides linked to α-syn and apoptosis.

The mode bifurcation between oscillatory motion and no motion of a camphor object floating on surfactant aqueous solution was investigated. The camphor and surfactant were used as the sources of driving and suppressing forces, respectively. A circular plastic sheet was attached to the center of the bottom of a camphor disk, which served as the self-propelled object; further, the contact area between the disk and the solution (S) was varied to control the amount of camphor molecules from the disk that dissolved in the solution. Motion features, that is, the maximum speed, frequency of oscillatory motion, and mode bifurcation, varied depending on S. We discussed the relationship between these motion features and the kinetics of camphor and surfactant molecules around the object. The results suggest that the nature of the oscillatory motion depends on the amount of camphor molecules accumulated at the base of the self-propelled object.

OBJECTIVE: The objective of this study was to explore the association between periodontitis and systemic lupus erythematosus (SLE). METHODS: To identify eligible studies, the PubMed, EMBASE and Web of Science databases were searched from inception to 19 September 2019. Associations of periodontitis, and other periodontal parameters, with SLE were assessed. RESULTS: Ten studies involving 80,633 subjects were included in this meta-analysis. Pooled data showed a significant association between periodontitis and SLE (odds ratio=5.32, 95% confidence interval (CI) 1.69-16.78, p = 0.004). In addition, SLE patients had a higher prevalence of bleeding on probing (mean difference = 0.03, 95% CI 0.00-0.06, p = 0.02) and higher mean clinical attachment loss (mean difference = 0.69, 95% CI 0.39-1.00, p < 0.001). However, there were no significant differences between SLE and reference subjects in mean plaque index, gingival index, pocket depth or decayed, missing or filled teeth. CONCLUSIONS: This study demonstrates a significant association between periodontitis and SLE, which indicates that avoidance of periodontitis by maintaining oral health may be a simple and economical way to prevent SLE.

Fraction of absorbed photosynthetically active radiation (fAPAR) is one of the important remote sensing model parameters of vegetation productivity. However, the crop canopy fAPAR estimation during growing season under different drought conditions has not been reported yet. In this study, the characteristics of summer maize canopy fAPAR and spectral reflectance during growing season under different drought stresses and the relationships of fAPAR with reflectance, the first derivative spectral reflectance and vegetation indices were examined based on the hyperspectral reflectance and fAPAR data from the summer maize drought manipulation experiment with five irrigation levels in 2015. Under mild water stress and sufficient water supply conditions, fAPAR was higher, with the maximum value of 0.7. Under severe water stress and severe persistent drought, fAPAR was lower, with the minimum value of 0.06. Reflectance of visible and shortwave bands increased and near infrared reflectance decreased with increasing drought. The fAPAR was negatively related with visible bands and shortwave bands, but positively correlated with near infrared. Visible and shortwave band reflectance had significant correlation with fAPAR, especially at 383, 680 and 1980 nm, with all the correlation coefficients being more than -0.87. The strong and stable relationship between the first derivative spectral reflectance and fAPAR appeared at 580, 720 and 1546 nm, with the correlation coefficients being -0.91, 0.89 and 0.88, respectively. There were linear or logarithm relationships between fAPAR with nine vegetation indices. Among the nine indices, the enhanced vegetation index (EVI), renormalized difference vegetation index (RDVI), soil adjusted vegetation index (SAVI), and modified soil adjusted vegetation index (MSAVI) performed well with the correlation coefficient being higher than 0.88, and the average relative error (RMAE) 16.6%, 16.6%, 16.7% and 16.2%, respectively. Based on the logarithmic relationship between first derivative spectral reflectance and fAPAR, the simulation effect was best at the band of (720±5) nm, with a correlation coefficient of 0.86. The correlation coefficient of the relationship between fAPAR and reflectance was less than 0.81. The results could provide fAPAR simulation for remote sensing model of vegetation productivity and drought warning.

Crop water deficit status characterized by land surface-air temperature difference (Ts-Ta) has been widely investigated. However, empirical evidence for characteristics and impact factors of Ts-Ta considering the process of crop growth are less yet, which restricts the accurate simulation of Ts-Ta. Here, the data of Ts-Ta during the process of maize growth were obtained from five irrigation water control experiments after the period of summer maize 3-leaf stage in 2014 and jointing stage in 2015. The results showed that Ts-Ta of summer maize cropland was significantly affected by soil water content. Ts-Ta increased with the deficit of soil water. During summer maize water treatments, the normalized difference vegetation index (NDVI) was the main impact factor of Ts-Ta, with a significant linear relationship. However, during different growth stages, some additional factors including meteorological, biological and soil factors could also affect Ts-Ta, including canopy photosynthetic active radiation absorption ratio (fAPAR) after 3-leaf stage, relative soil water content (RSWC), and air relative humidity (RH) from 3-leaf stage to jointing stage. Then, the growth duration simulation model of Ts-Ta, vegetative growth simulation model of Ts-Ta and reproductive growth simulation model of Ts-Ta were established in terms of the data in 2014. Those simulation models were validated based on the experimental data of five irrigation water treatments after summer maize jointing stage in 2015. The results showed that the growth duration simulation mode of Ts-Ta could explain 63% variation of Ts-Ta in 2015. However, 79% variation of Ts-Ta could be explained by the simulation results of the vegetative growth simulation model of Ts-Ta and the reproductive growth simulation model of Ts-Ta. The results provided the basis for the quantitative evaluation of crop drought based on Ts-Ta.

Objective To investigate the effects of bradykinin (BK) on the proliferation of rabbit corneal endothehal ceils (RCECs) and the expression of tight junction-related proteins zonula occludens-1 (ZO-1) and zonula occludens-1-associated nucleic-acid-binding protein (ZONAB),and to explore the underlying mechanisms of BK on cell proliferation in corneal endothelium.Methods RCECs at logarithmic growth phase were treated with different concentrations of BK (0.01,0.1,1,10 μmol · L-1) BK group,with the controls left untreated.Morphological changes of cells in each group were examined under phase-contrast microscope,and MTT assays were used to detect cell proliferation at 24 h,48 h,72 h,96 h after BK treatment.And,at 72 h,the expression levels of ZO-1 and ZONAB protein were determined by Western blot.Results After 72 h of treatment,the cells in each group were fused into pieces and closely linked into a monolayer;but after 96 h,the growth of the cells was restricted,with the intercellular space become larger and the cells exfoliated.Compared with the control group,BK induced a significant increase of absorbance value and cell viability,and the differences were statistically significant (all P < 0.001),and the promoting effects showed a concentration-dependent manner,and 1 μmol · L-1 BK demonstrated the strongest regulative effect (P < 0.001).Western blot results showed that BK upregulated the expression of ZO-1 and ZONAB protein in a concentration-dependent manner.Conclusion BK can stimulate the proliferation of RCECs in a time-and concentration-dependent manner,and the mechanisms are probably associated with ZO-1/ZONAB-mediated signaling pathway.

RESUMO

Idiopathic macular hole is a full - thickness defect of retinal tissue involving the anatomic fovea and affecting central visual acuity and quality of life in elder patients. Recent evidence showed that the alterations of choroidal blood flow and choroidal thickness are associated with the formation of macular holes. Enhanced depth imaging spectral-domain optical coherence tomography (EDI SD-OCT) enables in vivo measurement of choroidal thickness and may provide new insight into the understanding of pathogenesis of idiopathic macular hole. In this article, we reviewed current studies on the relationship between choroidal thickness measured by optical coherence tomography and the pathogenesis of idiopathic macular hole.

OBJECTIVE: To study the physicochemical properties of rhizosphere soil and their influence on appearance characters of semi-natural cultivated Codonopsis pilosula root. METHODS: The comprehensive methods by combining outdoor investigating with indoor analyzing were applied. Rhizosphere soil samples and plant materials were collected from 35 collection points, the physicochemical properties of soil and the appearance characters of plant materials were investigated. RESULTS: The physical property of rhizosphere soil under semi-natural cultivated condition was good, the soil texture was coarse powder of loam, slightly alkaline. The cultivated layer contained abundant nutrient components, the average organic matter content of 35 soil samples was 60.66 g/kg,and 2.99 g/kg for total nitrogen, 115.46 mg/g for available nitrogen, 0.8 g/kg for total phosphorus, 6.43 mg/kg for available phosphorus, 23.69 g/kg for total potassium and 124.48 mg/kg for available potassium, respectively. The result of correlation analysis indicated that the physicochemical properties of rhizosphere soil had significant influence on the appearance characters of Codonopsis pilosula root. CONCLUSION: The semi-natural cultivated model can maintain the structure of soil and hold nutrient components, it is conducive to the growth of Codonopsis pilosula and the standardized production base establishment. To increase the yield of Codonopsis pilosula root, it is recommended to increase the application of nitrogen and phosphorus fertilizer.

BACKGROUND: The oriental fruit moth (OFM) is a worldwide fruit-boring insect pest. In China, OFM monitoring traps use a sex pheromone lure, but their overall design is varied. As such, there is a critical need to develop a standardised OFM trap design. In this field study, ten different trap shapes in varying combinations of colours and sizes (such as trap length and surface area) were examined. RESULTS: The results showed that there was no significant difference in the trapping efficiency between eight colours. The ship-shaped trap could kill more OFM in a short period, whereas the automatic watering basin trap could be more effective in the long run. The optimal trapping diameter of the basin trap was 25 cm. The trapping efficiency of triangle traps with diameters of less than 10 cm was better than that of triangle traps with diameters of over 30 cm. The trapping number of pasteboard traps obviously declined when the surface area increased, and the pasteboard trap with a single board possessed excellent trapping efficiency. CONCLUSION: The results provide useful information for the design of standardised sex pheromone traps for monitoring as well as trapping of OFM in the field.

Background Optic neuropathy is one of the diabetic eye complications.Rosiglitazone,a peroxisome proliferator activated receptor γ(PPARγ) agonist,plays a very important role in arresting the pathogenesis and development of diabetes.However,the role of PPARγ in diabetic optic neuropathy is unclear.Objective This study was to investigate the protective effect of rosiglitazone against diabetic optic neuropathy and its mechanism.Methods Male Sprague-Dawley rats were randomly divided into the control group,diabetic group and rosiglitazone group,with 10 rats for each group.Diabetic models were induced by injecting 50 mg/kg of streptozotocin via the caudal vein,and rosiglitazone(5 ng/[kg· d])was used in the rats of the rosiglitazone group by intragastric administration every day for four weeks.At the end of the experiment,the fasting blood sugar(FBS) was tested in all the animals.The level of vascular endothelial growth factor(VEGF) in the blood plasma was detected by ELISA.Optical neural tissues were obtained from the rats of each group,and Lauck fast Blue myelin stain was used to examine the morphology of the optical myelin.The expression of neural cell adhesion molecule (NCAM) mRNA and protein in the optic nerve was detected by real time PCR and Western blot,respectively.Results The levels of FBS,blood plasma VEGF,NCAM mRNA and protein in the optic nerve were significantly different among the control group,diabetic model group and the rosiglitazone group after the administration of 5 nmg/(kg · d) rosiglitazone for 4 weeks (F =6.12,P<0.01 ; F =5.14,P<0.05 ; F =4.75,P<0.05 ; F =4.87,P<0.05).Compared with the control group,the level of FBS significantly increased in the diabetic model group(t =2.26,P<O.05),and that in the rosiglitazone group significantly declined in comparison with the diabetic model group(t=2.08,P<0.05).The optic nerve exhibited a normal morphology in the control group as revealed by the Lauck fast Blue myelin staining;however,severe demyelination of the optic nerve and proliferation of glial cells were found in the diabetic model group,and mild demyelination of the optic nerve and proliferation of glial cells were seen in the rosiglitazone group.Blood plasma VEGF was(28.76±4.21)ng/L in the control group and(134.28±11.36)ng/L in the diabetic model group,showing a significant difference between them (t=2.36,P < 0.05).Compared with the model group,the blood plasma VEGF was significantly lower in the rosiglitazone group ([42.67 ± 5.83] ng/L) than that in the diabetic model group (t =2.17,P< 0.05).Expression of NCAM mRNA and protein in the optic nerve significantly decreased in the diabetic model group compared with the control group(t =2.21,t =2.58,both at P<0.05);while those in the rosiglitazone group were significantly elevated in comparison with the diabetic model group(t =2.19,t =2.67,both at P<O.05).Conclusions Rosiglitazone can protect optic nerve from damage in diabetic rats mainly by downregulating blood plasma VEGF level and upregulating NCAM expression.

Ionic liquids of an N-alkylethylenediamine-silver(I) complex cation (alkyl=hexyl, 2-ethylhexyl, and octyl) or a protic N-alkylethylenediaminium cation (alkyl=butyl, hexyl, 2-ethylhexyl, octyl, decyl, and dodecyl) with a bis(trifluoromethanesulfonyl)amide counter anion (Ag-ILs and PILs, respectively) were prepared and their physicochemical properties were investigated. The trend of solidification decreased in the order octyl≫hexyl>2-ethylhexyl for the Ag-ILs, and butyl>dodecyl>decyl>octyl>hexyl≫2-ethylhexyl for the PILs. The diffusion coefficients of the cations indicated stronger intermolecular interactions in PILs than in the Ag-ILs because of hydrogen-bonding networks, and it has been revealed that the intermolecular interactions increase in the order, hexyl

OBJECTIVE: To review the treatment of repair gastroschisis with autogenous umbilical cord in 13-year and evaluate its effect. METHODS: Twenty-two newborns who underwent the repair gastroschisis with autogenous umbilical cord between 1992 and 2005. The physical growth, intelligence measuring, area of operation in abdomen in the survived 18 cases were observed and followed-up. RESULTS: Eighteen patients recovered uneventfully, survival rate is 82%, their growth is well. They all developed incisional hernia near the operation, 9 cases recovered himself, 2 cases was operated to repair the abdominal hernia, 7 cases is under observed. CONCLUSIONS: The material is adopted easily in the operating, autogenous umbilical cord is elastic tissue and no toxicity could relax the abdominal press effectively after the operation, the survival rate is high.

AIM: To analyse the main impurity of caderofloxacin. METHODS: The impurity of caderofloxacin was analysed and determinated by RP-HPLC/ESI/MS with a Zorbax SB-C18 (150 mm x 4.6 mm ID, 5 microns) column. The mobile phase was acetonitrile-0.5% acetic acid solution (17:83). A compound was synthesized: 1-cyclopropyl-8-(difluoromethoxy)-6-fluoro-1, 4-dihydro-7-(1-piperazinyl)-4-oxo-3-quinoline carboxylic acid (DMCA). Its HPLC chromatogram, UV and MS spectrum were compared with those of the impurity in caderofloxacin. RESULTS: The molecular weight of the impurity was 14 less than that of caderofloxacin. It means the impurity was a CH2-group less than caderoflixacin. The tR, UV and MS of DMCA were the same as those of the impurity in caderofloxacin. CONCLUSION: Based on the tR (HPLC), UV and MS, the impurity of caderofloxacin is confirmed as DMCA.

AIM: To analyse the impurities of gatifloxacin. METHODS: The impurity of gatifloxacin were analysized and determinated by RP-HPLC/electrospray ionization mass spectrometry with a Zorbax SB-C18(4.6 mm x 150 mm ID, 5 microns). The mobile phase was 3% acetic acid/acetonitrile-3% acetic acid/water (15:85). The two compounds were synthesized: 1-cyclopropyl-6-fluoro-1, 4-dihydro-8-methoxy-7-(1-piperazinyl)-4-oxo-3-quinolinecarboxylic acid (DMP) and 1-cyclopropyl-6-fluoro-1, 4-dihydro-8-hydro-7-(3-methy-1-piperazinyl)-4-oxo-3-quinolinecarboxylic acid (DMO). Their liquid chromatogram, UV, MS were compared with those of the impurity of gatifloxacin. RESULTS: The mass of the impurity was 14 less than that of gatifloxacin. It means the impurity was CH2 less than gatifloxacin. The tR (HPLC), UV and MS of DMP were the same as those of the impurity of gatifloxacin. CONCLUSION: Based on the tR (HPLC), UV and MS, the impurity of gatifloxacin is confirmed as DMP.

RESUMO

<p><b>AIM</b>To analyse the impurities of gatifloxacin.</p><p><b>METHODS</b>The impurity of gatifloxacin were analysized and determinated by RP-HPLC/electrospray ionization mass spectrometry with a Zorbax SB-C18(4.6 mm x 150 mm ID, 5 microns). The mobile phase was 3% acetic acid/acetonitrile-3% acetic acid/water (15:85). The two compounds were synthesized: 1-cyclopropyl-6-fluoro-1, 4-dihydro-8-methoxy-7-(1-piperazinyl)-4-oxo-3-quinolinecarboxylic acid (DMP) and 1-cyclopropyl-6-fluoro-1, 4-dihydro-8-hydro-7-(3-methy-1-piperazinyl)-4-oxo-3-quinolinecarboxylic acid (DMO). Their liquid chromatogram, UV, MS were compared with those of the impurity of gatifloxacin.</p><p><b>RESULTS</b>The mass of the impurity was 14 less than that of gatifloxacin. It means the impurity was CH2 less than gatifloxacin. The tR (HPLC), UV and MS of DMP were the same as those of the impurity of gatifloxacin.</p><p><b>CONCLUSION</b>Based on the tR (HPLC), UV and MS, the impurity of gatifloxacin is confirmed as DMP.</p>
