RESUMO

The effect of rosuvastatin (Ros) on cognitive function and anxiety-like behaviour in ovariectomized rats were evaluated. Eighteen female Wistar rats (218-310 g, 6-8 months old) were allocated into sham (n = 6), ovariectomy (Ovx, n = 6) or Ovx + Ros (up to eighth week n = 6, then n = 4) groups. Ros was administered at 20 mg/kg/day by oral gavage for 12 weeks. Behavioural tests were performed at 4, 8 and 12 weeks following Ovx. At 12 weeks, Ovx group had significantly longer escape latency than the sham group at the first day of the four-day training period of the Morris Water Maze test (p < .01). In the Elevated Plus Maze test, Ovx group spent significantly more time in the closed arms than the sham group (p < .01), and this anxiety-like behavioural effect of Ovx was prevented by 12-weeks Ros treatment (p < .05). In conclusion, Ros prevents memory deficit and anxiety-like behaviour in the ovariectomized rats, a model for human surgical menopause. Impact StatementWhat is already known on this subject? Reduced levels of oestrogen in surgical postmenopausal period has been linked to an increased risk of cognitive dysfunction. Although statins have been shown to improve cognitive function in experimental and clinical studies, there are limited studies evaluating the effect of statins on the cognitive decline and anxiety-like behaviour associated with surgical menopause.What do the results of this study add? Rosuvastatin, a long-acting statin, prevents learning and memory deficit and anxiety-like behaviour in the ovariectomized rat model.What are the implications of these findings for future clinical practice and/or future clinical research? These findings will form the basis for further experimental and clinical research on the effects of statins on cognitive functions and anxiety-like behaviour in the surgical menopause.

AIM: To evaluate the possible neuroprotective effects of ketamine and dantrolene on the hippocampal apoptosis and spatial learning in rats exposed to repeated electroconvulsive seizures (ECS) as a model of status epilepticus (SE). MATERIAL AND METHODS: Twenty-four rats were assigned to 4 groups. 1st Group was Sham. 2nd Group was ECS: ECS was induced by ear electrodes via electrical stimulation. The same ECS protocol was applied to the 3th and 4th Groups which received ketamine (40 mg/kg s.c.) or dantrolene (5 mg/kg i.p.) 1 h before each ECS, respectively. Following 30 days of recovery, the cognitive status of the animals was evaluated via Morris Water Maze (MWM). The same experimental protocol was repeated 14 days afterward to evaluate the retention of the memory. Hippocampal apoptosis was examined in corresponding experimental groups. RESULTS: All the animals in four groups learned the task with no significant difference between groups in MWM. The ECS+ketamine group showed memory impairment 14 days afterward. ECS+dantrolene group was not different from controls. ECS caused long term apoptotic processes in dentate gyrus (DG) and non-apoptotic neuronal injury in CA1 and CA2. CONCLUSION: Dantrolene and ketamine inhibited apoptosis and showed neuroprotective effects. Although ketamine and dantrolene inhibited ECS-induced apoptosis and non-apoptotic injury, they did not produce similar effects on memory retention. It will be warranted to evaluate cognitive dysfunction by taking into consideration the other factors in addition to apoptosis and neurodegenerative changes.

Objective: Bisphosphonate-related osteonecrosis of the jaw (BRONJ) is a challenging complication of chronic bisphosphonate (BP) use. The hormone relaxin is able to induce the multistep differentiation process of human osteoclastogenesis, exhibits antifibrotic and anti-inflammatory actions, and promotes vasodilatation, wound healing, and angiogenesis. The present study aimed to evaluate the effects of relaxin in the prevention and management of BRONJ. Material and Methods: Thirty-six male Sprague Dawley rats were randomly divided into four groups. Rats in group 1 (n = 10) received relaxin and BP simultaneously for 12 weeks. Rats in group 2 (n = 10) received injections of BP for 12 weeks, followed by relaxin for another 12 weeks. Rats in group 3 (n = 10) received only BP injections, and those in group 4 (control, n = 6) received only saline. Necrosis and inflammation in the rats' mandibles were evaluated as indicators of BRONJ. Results: Necrosis and inflammation were not detected in group 1 (BP + relaxin). In group 3 (BP only), incidence rates of necrosis and inflammation were 90% and 60%, respectively. Conclusions: Our findings suggest that relaxin may be potently effective in preventing BRONJ and have some benefit in the treatment of existing BRONJ (AU)


Objetivo: A osteonecrose da mandíbula relacionada ao bisfosfonato (BRONJ) é uma desafiadora complicação do uso crônico de bisfosfonato (BP). O hormônio relaxina é capaz de induzir o processo múltiplo de diferenciação da osteoclastogênese humana, exibe ações anti-fibróticas e anti-inflamatórias e promove vasodilatação, cicatrização de feridas e angiogênese. O presente estudo teve como objetivo avaliar os efeitos da relaxina na prevenção e tratamento do BRONJ. Material e Métodos: Trinta e seis ratos Sprague Dawley machos foram divididos aleatoriamente em quatro grupos. Os ratos do grupo 1 (n = 10) receberam relaxina e BP simultaneamente por 12 semanas. Os ratos do grupo 2 (n = 10) receberam injeções de BP por 12 semanas, seguidos de relaxina por mais 12 semanas. Os ratos do grupo 3 (n = 10) receberam apenas injeções de BP e os do grupo 4 (controle, n = 6) receberam apenas solução salina. Necrose e inflamação nas mandíbulas dos ratos foram avaliadas como indicadores de BRONJ. Resultados: Necrose e inflamação não foram detectadas no grupo 1 (BP + relaxina). No grupo 3 (somente BP), as taxas de incidência de necrose e inflamação foram de 90% e 60%, respectivamente. Conclusões: Nossos resultados sugerem que a relaxina pode ser potentemente eficaz na prevenção do BRONJ e ter algum benefício no tratamento do BRONJ existente.(AU)

OBJECTIVE: To test the protective effect of metformin against noise-induced hearing loss. METHODS: 24 rats were included in the study. The first group was exposed to noise only, the second group took metformin, the third group was exposed to noise and took metformin, and the fourth group was neither exposed to noise nor took metformin as control group. After measurement of baseline DPOAE and ABR of rats, the metformin group and the metformin + noise group received 300 mg/kg/day metformin via gavage for 10 days. On the 11th day, group 1 and group 3 were exposured to white noise at 105 dB SPL for 15 h. After noise exposure, DPOAE and ABR measurements of all rats were repeated on days 1st, 7th, and 21st. At the end of the study, all animals were sacrificed and cochlear tissues were separated for immunohistochemical assessments. RESULTS: ABR threshold values and DPAOE measurements of groups 1 and 3 were deteriorated on the 1st day after noise, while deterioration in group 1 continued on 7th and 21st days, but normalized on 7th day in group 3. After immune staining, a significant immunoreaction was observed in the noise group, while the reaction in the noise + metformin group was close to the control group. CONCLUSION: Metformin has a protective effect on noise-induced hearing loss in rats. As a conclusion, it is determined that metformin protects from permanent threshold shift in rats. It can be considered a good alternative for protecting noise-induced hearing loss.

Alternative treatment approaches to improve the regeneration capacity of damaged peripheral nerves are currently under investigation. The objective of the present study was to evaluate the effects of platelet-rich fibrin (PRF) membrane after sciatic nerve crush injury in rabbits by histomorphometric and electromyographic analysis. The left sciatic nerves of 20 male Vienna rabbits were clamped for 30 seconds to induce crush injuries. Animals were randomly divided into 2 groups: PRF and control. For each animal in the PRF group, a PRF membrane was wrapped around the injured part of the sciatic nerve to form a tube. No additional treatment was performed in the control group. After a 12-week healing period, tissue samples from the injured nerve region were harvested and the g-ratio of axons, axon density, and impulse transmission changes were evaluated. Analysis revealed that axon density differences were not statistically significant between groups (P = 0.139). The rate of nerve fibers with optimum g-ratio was significantly lower in the PRF group than in the control group (P = 0.02). Conduction velocity differences between groups were not statistically significant. Although PRF application has previously shown positive regeneration effects on maxillofacial tissues, local PRF membrane application in tube form did not show any histomorphometric or functional improvement in peripheral nerve crush injury recovery.

OBJECTIVE: To investigate the level of correlation and direction of linearity between academic motivation and subjective workload. METHODS: The study was conducted at Baskent University School of Medicine, Ankara, Turkey, from December 2013 to February 2014, and comprised Phase 5 Phase 6 medical students. Subjective workload level was determined by using National Aeronautics and Space Administration Task Load Index scale that was adapted to Turkish. Academic motivation values were obtained with the help of Academic Motivation Scale university form. SPSS 17 was used for statistical analysis. RESULTS: Of the total 105 subjects, 65(62%) students were in Phase 5 and 40(38%) were in Phase 6. Of the Phase 5 students, 18(27.7%) were boys and 47(72.3%) were girls, while of the Phase 6 students, 16(40%) were boys and 24(60%) were girls. There were significant differences in Phase 5 and Phase 6 students for mental effort (p=0.00) and physical effort (p=0.00). The highest correlation in Phase 5 was between mental effort and intrinsic motivation (r=0.343). For Phase 6, highest correlation was between effort and amotivation (r= -0.375). CONCLUSIONS: Subjective workload affected academic motivation in medical students.
