RESUMO

Thirty-seven nonalcoholic individuals (22 women, 15 men), ages 26-76, and 36 abstinent alcoholic individuals (11 women, 25 men), ages 31-74, participated in a cued-detection task that assessed right hemisphere (RH) functioning associated with aging and alcoholism. Young controls were less reliant on cues following RH activation, which is consistent with the view that the RH has an advantage because it has the ability to attend to a broader spatial array than does the left hemisphere (LH). This RH advantage was not obtained in older controls or alcoholic participants. The pattern of results for the older nonalcoholic participants indicated that they neither benefited from valid cues following LH activation nor exhibited enhanced processing on invalid cue trials following RH activation. The results for the alcoholic participants were consistent with RH functional decline, but did not support the view that alcoholism and aging have synergistic effects.

Investigated were the effects of 2-week exposure of four Macaca mulattas to the head-end tilted downward at 10 degrees on contractility of the left myocardium and its controls basing on the data of functional loading tests on the tilting table and at two regimes of centrifugation. The experiment included registration of the left heart pressure, volumetric blood flow in the aorta, and EKG in order to determine stroke volume, maximal isovolumetric pressure, end-systolic and end-diastolic pressures, maximal contraction and relaxation velocities, end-systolic myocardial elasticity, and a constant of the isovolumetric relaxation. As was stated, HDT does not impair contractility of the left myocardium; however, sensitivity of the HR baroreflex control wakens.

Chronic heavy drinking and alcoholism can have serious repercussions for the functioning of the entire nervous system, particularly the brain. These effects include changes in emotions and personality as well as impaired perception, learning, and memory. Neuropathological and imaging techniques have provided evidence of physical brain abnormalities in alcoholics, such as atrophy of nerve cells and brain shrinkage. At the cellular level, alcohol appears to directly affect brain function in a variety of ways, primarily by interfering with the action of glutamate, gamma-aminobutyric acid, and other neurotransmitters. Neurological disorders also can result from vitamin deficiency and liver disease, two health problems that commonly occur with alcoholism. Other hypotheses, based on factors such as aging, gender, and genetics, have been developed to explain various alcohol-related neurological consequences. Many pharmacological treatments to improve neuropsychological functioning in alcoholics have been tested, but none has proved entirely successful. With prolonged abstinence, however, slow recovery of cognitive functioning can occur in some cases.

Dynamics of the left heart ventricular muscle contractility and compliance was studied in 4 monkeys in the head down position (antiorthostatic hypokinesia) with the body angle 10 during 2 weeks. Functional tests on a tilt table and under two conditions of centrifuge rotation were performed prior to and after the antiorthostatic hypokinesia. No changes in the left heart ventricular muscle contractility was found. However, the sensitivity level of the baroreflex control decreased. Compliance of the left heart myocardial fibre increased in the first hours and days of the antiorthostatic hypokinesia.

Bioactivity-directed fractionation of the ethanol extract of Asimina longifolia led to the isolation of four novel bioactive annonaceous acetogenins: longimicins A-D (1-4). Compounds 1-4 represent the asimicin type of acetogenins; however, the locations of the adjacent bis-tetrahydrofuran (THF) ring moieties are shifted along the aliphatic chains compared to the known compounds of this type. They are the first examples among this type of acetogenins with the placements of the ring systems altered. Compounds 1-4 showed bioactivities in several bioassays, but they are less active than their structural isomers. Study of their structure-activity relationships (SAR) reveals that the position of the adjacent bis-THF ring moiety is essential for maximization of the bioactivities among these asimicin type annonaceous acetogenins.

RESUMO

Longifolicin (1), longicoricin (2) and (2,4-cis and trans)-gigantetroneninone (3), three novel bioactive mono-tetra-hydrofuran (THF) gamma-lactone acetogenins, were isolated from the leaves and twigs of Asimina longifolia (Annonaceae) by directing the fractionation with the brine shrimp lethality test (BST). The structures were elucidated based on spectroscopic and chemical methods. Compounds 1-3 showed selective and potent cytotoxicities to certain human tumor cell lines.

The serial pattern found for conjunction visual-search tasks has been attributed to covert attentional shifts, even though the possible contributions of target location have not been considered. To investigate the effect of target location on orientation x color conjunction searches, the target's duration and its position in the display were manipulated. The display was present either until observers responded (Experiment 1), for 104 msec (Experiment 2), or for 62 msec (Experiment 3). Target eccentricity critically affected performance: A pronounced eccentricity effect was very similar for all three experiments; as eccentricity increased, reaction times and errors increased gradually. Furthermore, the set-size effect became more pronounced as target eccentricity increased, and the extent of the eccentricity effect increased for larger set sizes. In addition, according to stepwise regressions, target eccentricity as well as its interaction with set size were good predictors of performance. We suggest that these findings could be explained by spatial-resolution and lateral-inhibition factors. The serial self-terminating hypothesis for orientation x color conjunction searches was evaluated and rejected. We compared the eccentricity effect as well as the extent of the orientation asymmetry in these three conjunction experiments with those found in feature experiments (Carrasco & Katz, 1992). The roles of eye movements, spatial resolution, and covert attention in the eccentricity effect, as well as their implications, are discussed.

Longicin [1] and (2,4-cis and trans)-goniothalamicinone [2], two new monotetrahydrofuran Annonaceous acetogenins, have been isolated from the leaves and twigs of Asimina longifolia (the long leaf paw paw) by the use of the brine shrimp lethality test for bioactivity-directed fractionation. The structures were elucidated based on spectroscopic and chemical methods. Compound 1 was converted to its ketolactone isomer, (2,4-cis and trans)-longicinone [3], to aid the stereochemical elucidation of 1. Compounds 1-3 showed selective and potent cytotoxicities to certain human tumor cell lines, with the potency of 1 against pancreatic carcinoma (PaCa-2) over one million times that of adriamycin. Nine known cytotoxic acetogenins, annonacin, xylomaticin, isoannonacin, gigantetrocins A and B, muricatetrocins A and B, gigantetrocin-A-one and goniothalamicin, were also isolated for the first time from this species.

Precuing an observer as to where a target is more likely to occur in a subsequent visual array can increase the detectability (d') of a target at that location. This is often attributed to the observer's increased allocation of some limited cognitive resource ("attention") to the cued location. Two experiments are reported which are difficult to interpret in this way even though they involve similar cue effects. The first involves postcuing a location well after the array but before the observer responds, so that the cue can influence the response but not the observation. The second involves precuing, but with slow sequential presentation of array elements prior to the response, so the observer need not share any limited resource while processing each element in turn. Enhanced detectability similar to that produced with precues and simultaneous presentation of elements is shown to occur in each experiment. An alternative data-limited (rather than resource-limited) interpretation of these effects is provided by a mathematical model in which the observer integrates equally noisy or degraded internal representations of the array elements, but gives more weight to cued elements in selecting a response. Theoretical parameters of the model are shown to provide separate measures of both an observer's overall sensitivity and precue effects in cost-benefit analyses of cuing data.

RESUMO

The EtOH extract of Asimina parviflora (Annonaceae), when subjected to activity-directed fractionation using lethality to brine shrimp, led to the isolation and identification of five bioactive compounds: asimicilone [1], which is a new 2-quinolone alkaloid, 6-cis-docosenamide [2], which is a new amide of a long hydrocarbon chain fatty acid, and three known compounds, asimicin, (+)-syringaresinol, and beta-sitosterol-beta-D- glucopyranoside. The structure determination of the new alkaloid was performed by extensive nmr analyses, including HMQC and HMBC. Selective cytotoxic activities of these compounds in three human solid tumor cell lines are also reported.

A sample holder was designed and built to facilitate measuring the magnitude and phase angle of the electrical impedance of internodal stem sections from Cornus stolonifera Michx. A nonpolarizing, electrically conducting manganese dioxidecarbon paste used between the stem sample and the electrodes of the sample holder allowed measurement of impedance at frequencies from 50 hertz to 500 kilohertz without electrode polarization or electrical interference. The impedance magnitude was linearly dependent on the sample length, but this dependence was minimized by computing a normalized impedance magnitude. The normalized impedance magnitude (Z(nf)) was calculated using the impedance magnitude (Z) at any specified frequency (f) and the impedance magnitude at 500 kilohertz (Z(500 khz)) in the following formula: Z(nf) = (Z - Z(500 khz))/Z(500 khz). The normalized impedance magnitude was sensitive to injury produced by boiling and peeling the sample. Electrical impedance measurements on the bark and wood separately demonstrated that they have different electrical properties.

The ratio of electrical conductance measured at two frequencies can be used to predict the cold hardiness of stem sections of Cornus stolonifera Michx. during the first stage of cold acclimation. Electrical conductance at 50 hertz divided by electrical conductance at 100 kilohertz gave a better estimate of hardiness than measurements at either frequency alone. The observed increase in the electrical conductance ratio as hardiness increased is consistent with an increase in membrane permeability. After plants were exposed to nonlethal frost, hardiness increased rapidly, and the relation between the conductance ratio and hardiness changed. This change indicates that ice crystallization induces a significant physiological alteration in the plants. Contrary to expectations, stem sections exposed to lethal temperatures could not consistently be separated from sections exposed to nonlethal temperatures by electrical conductance ratio measurements made immediately after thawing.

A warm (20 to 15 Celsius day or night) preconditioning treatment enhanced cold acclimation of Cornus stolonifera bark under short-day conditions when plants were preconditioned for at least 4 weeks. Warm preconditioning inhibited the acclimation of plants subjected to long photoperiods. Removing leaves from plants exposed to low temperatures and short days inhibited acclimation. Removal of buds did not affect acclimation. Plants did not acclimate unless they were exposed to at least 4 weeks of short photoperiods prior to defoliation. Plants began to acclimate to cold at the time of growth cessation but not before. When half of the leaves were removed from plants, the defoliated and foliated branches both acclimated as well as branches on completely foliated plants. Girdling the phloem between foliated and defoliated branches prevented acclimation of the latter regardless of the position of the girdle in relation to the root system and the defoliated branch. When all of the leaves of plants were covered with aluminum foil to exclude light after 0 or 4 weeks of exposure to short days, the results resembled a defoliation study, i.e., plants with leaves covered at the start of the experiment failed to acclimate, and those covered after 4 weeks acclimated to some extent but less than uncovered control plants. Under longday conditions plants with all leaves covered failed to acclimate, and plants with none or half of their leaves covered acclimated equally and to a limited extent. Under short-day conditions, however, the covered branches of partially covered plants acclimated more than their uncovered counterparts or branches of totally uncovered plants.
