RESUMO

Terrestrial gamma-ray flashes (TGFs) are transient gamma-ray emissions from thunderstorms, generated by electrons accelerated to relativistic energies in electric fields. Elves are ultraviolet and optical emissions excited in the lower ionosphere by electromagnetic waves radiated from lightning current pulses. We observed a TGF and an associated elve using the Atmosphere-Space Interactions Monitor on the International Space Station. The TGF occurred at the onset of a lightning current pulse that generated an elve, in the early stage of a lightning flash. Our measurements suggest that the current onset is fast and has a high amplitude-a prerequisite for elves-and that the TGF is generated in the electric fields associated with the lightning leader.

Zymoseptoria tritici causes Septoria leaf blotch of wheat. The prevailing paradigm of the Z. tritici-wheat interaction assumes fungal ingress through stomata within 24-48h, followed by days of symptomless infection. This is extrapolated from studies testing the mode of fungal ingress under optimal infection conditions. Here, we explicitly assess the timing of entry, using GFP-tagged Z. tritici. We show that early entry is comparatively rare, and extended epiphytic growth possible. We test the hypotheses that our data diverge from earlier studies due to: i. random ingress of Z. tritici into the leaf, with some early entry events; ii. previous reliance upon fungal stains, combined with poor attachment of Z. tritici to the leaf, leading to increased likelihood of observing internal versus external growth, compared to using GFP; iii. use of exceptionally high humidity to promote entry in previous studies. We combine computer simulation of leaf-surface growth with thousands of in planta observations to demonstrate that while spores germinate rapidly on the leaf, over 95% of fungi remain epiphytic, growing randomly over the leaf for ten days or more. We show that epiphytic fungi are easily detached from leaves by rinsing and that humidity promotes epiphytic growth, increasing infection rates. Together, these results explain why epiphytic growth has been dismissed and early ingress assumed. The prolonged epiphytic phase should inform studies of pathogenicity and virulence mutants, disease control strategies, and interpretation of the observed low in planta growth, metabolic quiescence and evasion of plant defences by Zymoseptoria during symptomless infection.

RESUMO

A modified quasi-quantum treatment (MQQT) of molecular scattering has been developed to account for the softness of the repulsive part of the anisotropic atom-molecule PES. A contour of the PES is chosen such that the barrier height is just large enough to reflect the incoming kinetic energy, directed anti-parallel to the hard shell normal at the site of impact. The resulting rotationally inelastic quantum state resolved DCSs and ICSs of He + NO(X) at Ecol = 508 cm(-1) are compared to those obtained from regular QQT and from quantum mechanically exact calculations performed on the full highest quality ab initio Vsum PES. The MQQT parity changing DCSs for Δj ≤ 4 exhibit much better agreement with the QM DCSs than is obtained using regular QQT, particularly in the forward scattered direction. The improvements upon the remaining MQQT DCSs with respect to the regular QQT were minor, due to the near incompressible hard shell character of the n ≠ 1 or 3 anisotropic Legendre polynomial terms of the PES.

The metal hyperaccumulator plant Noccaea caerulescens is protected from disease by the accumulation of high concentrations of metals in its aerial tissues, which are toxic to many pathogens. As these metals can lead to the production of damaging reactive oxygen species (ROS), metal hyperaccumulator plants have developed highly effective ROS tolerance mechanisms, which might quench ROS-based signals. We therefore investigated whether metal accumulation alters defence signalling via ROS in this plant. We studied the effect of zinc (Zn) accumulation by N. caerulescens on pathogen-induced ROS production, salicylic acid accumulation and downstream defence responses, such as callose deposition and pathogenesis-related (PR) gene expression, to the bacterial pathogen Pseudomonas syringae pv. maculicola. The accumulation of Zn caused increased superoxide production in N. caerulescens, but inoculation with P. syringae did not elicit the defensive oxidative burst typical of most plants. Defences dependent on signalling through ROS (callose and PR gene expression) were also modified or absent in N. caerulescens, whereas salicylic acid production in response to infection was retained. These observations suggest that metal hyperaccumulation is incompatible with defence signalling through ROS and that, as metal hyperaccumulation became effective as a form of elemental defence, normal defence responses became progressively uncoupled from ROS signalling in N. caerulescens.

The quasi-quantum treatment (QQT) (Gijsbertsen et al., J. Am. Chem. Soc., 2006, 128, 8777) provides a physically compelling framework for the evaluation of rotationally inelastic scattering, including the differential cross sections (DCS). In this work the QQT framework is extended to treat the DCS in the classically forbidden region as well as the classically allowed region. Most importantly, the QQT is applied to the collision energy dependence of the angular distributions of these DCSs. This leads to an analytical formalism that reveals a scaling relationship between the DCS calculated at a particular collision energy and the DCS at other collision energies. This scaling is shown to be exact for QM calculated or experimental DCSs if the magnitude of the (kinematic apse frame) underlying scattering amplitude depends solely on the projection of the incoming momentum vector onto the kinematic apse vector. The QM DCSs of the NO(X)-He collision system were found to obey this scaling law nearly perfectly for energies above 63 meV. The mathematical derivation is accompanied by a mechanistic description of the Feynman paths that contribute to the scattering amplitude in the classically allowed and forbidden regions, and the nature of the momentum transfer during the collision process. This scaling relationship highlights the nature of (and limits to) the information that is obtainable from the collision-energy dependence of the DCS, and allows a description of the relevant angular range of the DCSs that embodies this information.
