RESUMO

This study evaluated the effects of different roasting profiles (time/temperature) and brewing methods on the physicochemical and sensory characteristics of coffee brews. Cold brewing (dripping and immersion) and hot brewing (French press) methods were studied to understand the effects of water temperature and technical brewing method conditions on the chemical compound extraction in coffees roasted at high-temperature short time (HTST) and low-temperature long time (LTLT). The results showed that coffee beverages were clearly differentiated concerning the roasting profile when hot water was used (90 ± 3 °C) in brewing. Separation of beverages according to the water temperature used in brewing was observed. Notably, hot brewing coffees were distinguished from cold brewing (19 ± 2 °C) based on a higher titratable acidity and abundance of some furan compounds. The non-volatile extraction rate increased at higher brewing temperatures. At the same brewing temperature, dripping exhibited a higher extraction rate than immersion brewing, which suggests that the coffee extraction process is affected by the design and operation of the cold brewing system. Coffee beverages brewed with HTST and cold dripping displayed the highest value in total dissolved solids (TDS), extraction yield, as well as the highest caffeine, trigonelline, 4- and 5-caffeoylquinic acids (CQAs) contents. Regardless of the roasting profile, coffees brewed by cold dripping were perceived with more bitter and roasted flavors. In contrast, cold immersion and hot coffee beverages showed remarkable sweetness, nutty, caramel, and malt attributes. In turn, these attributes showed an inverse correlation with caffeine concentration, trigonelline, CQAs, and TDS. The findings of this study demonstrate that volatile and non-volatile compounds present in roasted coffee depend on time-temperature roasting conditions; in turn, their presence in the resulting beverages are related to the extraction of the operational conditions of coffee brewing methods.

RESUMO

We present the complete genome sequences of three Helicobacter pylori strains isolated from patients who resided in Tolima Department, Colombia, diagnosed with chronic gastritis. The genomes present an average length of 1.6 Mbp and 1,546 genes and correspond to different H. pylori subpopulations.

The objective of this study was to evaluate the pasture performance of different cultivars of perennial ryegrass, two "high sugar" and two standard cultivars, under two contrasting agronomic managements (aimed at either decreasing or increasing water soluble carbohydrates concentration), and their effects on the grazing preference of dairy cows. Eight treatments arising from the factorial combination of four cultivars and two managements were randomly applied to 31-m2 plots in three blocks. Pasture dry matter production and growth rate were measured for one year. Three grazing assessments were performed to establish the grazing preferences of six dairy cows in spring, summer and autumn. High sugar cultivars produced less dry matter per hectare than the standard cultivars. Cows consumed more grass and harvested a greater proportion of the pasture under the agronomic management aimed at decreasing sugar concentration, i.e., with a greater nitrogen fertilization rate and under a more frequent defoliation regime, which could be explained by the greater crude protein concentration achieved under this management. The results suggest that the genetic selection for greater levels of sugars was at the expense of herbage yield, and that cows preferred to graze herbage with a greater crude protein level instead of a greater sugar concentration.

BACKGROUND: Several recent studies have shown a strong association between non-alcoholic steatohepatitis (NASH) and chronic kidney disease. AIM: To examine the relationship between changes in liver histology and renal function in patients with NASH. METHODS: The present analysis represents a post hoc analysis of a recently published trial that included 261 patients with NASH who were treated with lifestyle modifications during 52 weeks. Kidney function was evaluated through Chronic Kidney Disease Epidemiology Collaboration estimated glomerular filtration rates (eGFR, mL/min/1.73 m2 ) overtime. We explored correlations between the kidney function and improvement in histological outcomes at 52 weeks. RESULTS: Interestingly, a one-stage reduction in fibrosis (r = 0.20, P < 0.01) and resolution of NASH (r = 0.17, P < 0.01) were significantly correlated with an improvement in the kidney function. The eGFR values significantly increased in patients with fibrosis improvement (+7.6 ± 6.5 mL/min/1.73 m2 ), compared to those without fibrosis improvement (-1.98 ± 6.4 mL/min/1.73 m2 ) (P < 0.01) at end of treatment (EOT). Likewise, NASH resolution was associated with an increase in eGFR compared with patients without NASH resolution (2.32 ± 7.8 mL/min/1.73 m2 vs. -1.04 ± 5.9 mL/min/1.73 m2 , P = 0.04) at EOT. After controlling for the confounders, the association between fibrosis improvement, NASH resolution and eGFR change remained significant (P < 0.05 for both). CONCLUSIONS: Improvement in liver histology due to lifestyle modification is independently associated with improved kidney function in NASH. As new drugs for NASH emerge, studies should address whether improvement in histology in response to pharmacotherapies yield the same improvement in kidney function as weight loss.

RESUMO

Adult neurogenesis in the dentate gyrus (DG) in the hippocampus is a process that involves proliferation, differentiation, maturation, migration, and integration of young neurons in the granular layer of DG. These newborn neurons mature in three to four weeks and incorporate into neural circuits in the hippocampus. There, these new neurons play a role in cognitive functions, such as acquisition and retention of memory, which are consolidated during sleep period. In this review, we describe recent findings that associate sleep deprivation with changes in hippocampal neurogenesis and cognitive processes. In addition, we describe possible mechanisms implicated in this deterioration such as circadian rhythm, melatonin receptors, and growth factors.

Background: cryopreservation decreases sperm viability by approximately 50%. Objective: the objective of this study was to determine the effect of the addition of seminal plasma proteins on post-thawing sperm viability in Sanmartinero and Zebu semen. Methods: semen samples from 10 bulls of each breed were used, and seminal plasma was subjected to two-dimensional electrophoresis to establish the relationship between the relative amount of each protein spot and sperm viability. Then, seminal plasma was subjected to exclusion chromatography to separate the fraction containing these proteins. This fraction was added in doses of 0.5, 1.0, 1.5 and 2.0 mg, to 1 x 10(6). Sperm was thawed and incubated at 37 °C for 1 h to determine its effect on postthaw viability. Sperm were frozen using two media (citrate-fructose-yolk and Bioxcell®). Results: we found one protein spot (16.20 kDa, PI 5.5) in Sanmartinero seminal plasma that correlated (r = 0.64 p<0.001) with viability. This spot was found in 21-25 chromatography fractions. The percentage of post-thaw viable sperm increased 20% (p<0.05) at 1.0 and 1.5 mg of the fraction when sperm was frozen using citrate-fructose-yolk; it increased 25% (p<0.01) with 0.5 mg when it was frozen with Bioxcell® media. Addition of 0.5 mg of the fraction to semen cryopreserved with Bioxcell® resulted in a greater (p<0.05) percentage increase of viable sperm in Sanmartinero semen (23 ± 8.3%) compared with Zebu semen (6.0 ± 2.0%). Conclusions: these results show that seminal plasma proteins decrease cryopreservation damage in sperm. The effect depends on the cryoprotectant dose as well as the breed of bull.


Antecedentes: la criopreservación disminuye la viabilidad espermática por debajo del 50%. Objetivo: el objetivo de esta investigación fue determinar el efecto de la adición de proteínas del plasma seminal sobre la viabilidad espermática post-descongelación de semen de toros Sanmartinero y Cebú. Métodos: se colectó semen de 10 toros de cada raza, y el plasma seminal se sometió a electroforesis bidimensional, para establecer la relación entre la cantidad relativa de cada punto de proteína y la viabilidad espermática. Identificados dichos puntos, el plasma seminal se sometió a cromatografía de exclusión para separar la fracción que contenía estas proteínas. Esta se adicionó en dosis de 0,5, 1,0, 1,5 y 2,0 mg, a muestras de 1 x 10(6) espermatozoides, descongelados e incubados a 37 °C durante 1 hora, para determinar su efecto en la viabilidad post-descongelación. Los espermatozoides se congelaron usando dos medios (citrato-fructosa-yema y Bioxcell®). Resultados: se encontró un punto de proteína (16,20 kDa, punto Isoeléctrico 5,5) en plasma de toros Sanmartinero, que correlacionó (r = 0,64 p<0,001) con la viabilidad. Este punto de proteína se encontró en la fracción 21-25 de la cromatografía. El porcentaje de espermatozoides viables post-descongelación aumentó 20% (p<0,05) con dosis de 1 y 1,5 mg de la fracción, cuando los espermatozoides se congelaron en medio citrato-fructosa-yema; y 25% (p<0,01) con dosis de 0,5 mg cuando se congelaron en medio Bioxcell®. La adición de 0,5 mg de la fracción a semen descongelado previamente criopreservado en medio Bioxcell®, evidenció un incremento mayor (p<0,05) en el porcentaje de espermatozoides viables de semen de toros Sanmartinero (23 ± 8,3 %), que en semen de toros Cebú (6,0 ± 2,0%). Conclusiones: los resultados anteriores demuestran que las proteínas del plasma seminal disminuyen el daño en los espermatozoides por la criopreservación, y que el efecto de estas proteínas depende del medio de congelación, la dosis adicionada y la raza de los toros.

Objetivo: o objetivo desta pesquisa foi determinar o efeito da adição de proteínas do plasma seminal na viabilidade espermática pós-descongelamento de sêmen de touros das raças Sanmartinero y Zebú. Métodos: coletou-se sêmen de 10 touros de cada raça, as amostras do plasma seminal foram submetidas à eletroforese bidimensional, para estabelecer a relação entre a quantidade relativa de cada ponto de proteína e a viabilidade espermática. Ao serem identificados os pontos, o plasma seminal também foi submetido ao processo de cromatografia por exclusão para separar a fração que continha as proteínas. A fração foi adicionada nas doses de 0,5, 1,0, 1,5 y 2,0 mg, amostras de 1 x 106 espermatozoides, em descongelamento e incubados à temperatura de 37 ° C durante 1 hora, para determinar o efeito na viabilidade pós-descongelamento. Os espermatozoides foram congelados utilizando dois meios (Citrato- frutose-gema e Bioxcell®). Resultados: encontrou-se um ponto de proteína (16,20 kDa, ponto Isoelétrico 5,5) no plasma de touro Sanmartinero, que correlacionou (r=0,64 p<0,001) com a viabilidade. Esse ponto de proteína foi encontrado na fração 21-25 da cromatografia. O percentagem de espermatozoides viáveis pós-descongelamento aumentou em 20% (p<0,05) nas doses de 1 y 1,5 mg da fração, quando os espermatozoides foram congelados em meio de citrato-frutose-gema; e 25% (p<0,01) com doses de 0,5 mg congelados em meio Bioxcell®. A adição de 0,5 mg da fração ao sêmen descongelado e previamente criopreservado em meio Bioxcell®, evidenciou um incremento maior (p<0,05) no percentagem de espermatozoides viáveis do sêmen de touros Sanmartinero (23 ± 8,3 %), do que em sêmen de touros zebu (6,0 ± 2,0%). Conclusões: os anteriores resultados demonstram que as proteínas do plasma seminal diminuem o dano nos espermatozoides pela criopreservação e que o efeito destas proteínas depende do meio de congelação, a dose adicionada e a raça dos touros.

En el laboratorio de fisiología de la Facultad de Medicina (Quito- Ecuador) se procedió a determinar los parámetros hemostáticos: tiempo de ssangría, tiempo de coagulación, tiempo de retraccción dle coïgulo: en adultos y jóvenes residnetes a 2.800mts sobre el nivel del mar (Quito) De una muestra de 203 jóvenes adultos en 104 hombres y 99 mujeres, se determinó, el tiempo de sangría (n=93) en 49 hombres y 44 mujeres: el tiempo de coagulación (n=71) en 36 hombres y 35 mujeres, el tiempo de retracción del coágulo (n=39) en 19 hombres y 20 mujeres. Se aplicaron técnicas convencionales para el timpo de sangría, punción a nivel del pulpejo de un dedo, para tiempo de coagulación y tiempo de tretracción del coágulo gota fresca en placa de vidrio. Se encontró que los valores promedios de la población total del tiempo de ssangría, tiempo de coagulación y tiempo de retracción del coagúlo fueron 3:37 más o menos1.85;8.55 más o menos2.76; 44.10 más o menos 12.31 respectivamente. Se observó que los valores de Ts y Tc fueron similares entre ambos sexos en tanto que TCR fue superior en el sexo masculino (p 0.03); sin influencia de los cambios crónicos de la altitud. Los valores de referencia local en adultos jóvenes de ambos sexos residentes a 2820 mts sobre el nivel del mar se muestran simialres con los parámetros de referencia internacional...
