RESUMO

It is widely acknowledged that we are currently facing a critical tipping point with regards to global extinction, with human activities driving us perilously close to the brink of a devastating sixth mass extinction. As a promising option for safeguarding endangered species, induced pluripotent stem cells (iPSCs) hold great potential to aid in the preservation of threatened animal populations. For endangered species, such as the northern white rhinoceros (Ceratotherium simum cottoni), supply of embryos is often limited. After the death of the last male in 2019, only two females remained in the world. IPSC technology offers novel approaches and techniques for obtaining pluripotent stem cells (PSCs) from rare and endangered animal species. Successful generation of iPSCs circumvents several bottlenecks that impede the development of PSCs, including the challenges associated with establishing embryonic stem cells, limited embryo sources and immune rejection following embryo transfer. To provide more opportunities and room for growth in our work on animal welfare, in this paper we will focus on the progress made with iPSC lines derived from endangered and extinct species, exploring their potential applications and limitations in animal welfare research.

RESUMO

Peripheral arterial disease (PAD) has been historically neglected, which has resulted in a lack of effective drugs in clinical practice. However, with the increasing prevalence of diseases like atherosclerosis and diabetes, the incidence of PAD is rising and cannot be ignored. Researchers are exploring the potential of promoting angiogenesis through exogenous compounds to improve PAD. This paper focuses on the therapeutic effect of natural products (Salidroside, Astragaloside IV, etc.) and synthetic compounds (Cilostazol, Dapagliflozin, etc.). Specifically, it examines how they can promote autocrine secretion of vascular endothelial cells, enhance cell paracrine interactions, and regulate endothelial progenitor cell function. The activation of these effects may be closely related to PI3K, AMPK, and other pathways. Overall, these exogenous compounds have promising therapeutic potential for PAD. This study aims to summarize the potential active compounds, provide a variety of options for the search for drugs for the treatment of PAD, and bring light to the treatment of patients.

The distinction between Keratoacanthoma (KA) and Cutaneous Squamous Cell Carcinoma (cSCC) is critical yet usually challenging to discriminate clinically and histopathologically. One approach to differentiate KA from cSCC is through assessing the immunohistochemical staining patterns of the three indicators, ß-catenin, C-Myc, and CyclinD1, which are critical molecules that play important roles in the Wnt/ß-catenin signaling pathway. Ki-67, as a proliferation biomarker for human tumor cells, was also assessed as an additional potential marker for differentiating KA from cSCC. In this report, these four indicators were analyzed in 42 KA and 30 cSCC cases with the use of the computer automated image analysis system. Computer automated image analysis is a time-based and cost-effective method of determining IHC staining in KA and cSCC samples. We found that C-Myc staining was predominantly localized in the nuclei of basal cells within KA patients, whereas cSCC staining was predominantly localized in the nuclei of diffuse cells. This C-Myc staining pattern has a sensitivity of 78.6% and a specificity of 66.7% for identifying KA. Moreover, positive rates of distinct expression patterns of C-Myc and Ki-67 may also serve as a means to clinically distinguish KA from cSCC. Taken together, our results suggest that these markers, in particular C-Myc, may be useful in differentiating KA from cSCC.

Peroxydisulfate (PDS) is widely used in field-scale remediation of organically polluted soil, yet PDS is detrimental to the soil microbiome. In this work, sludge-derived hydrochar was used to alleviate the oxidative damage of PDS to the soil ecosystem and simultaneously improve atrazine (ATZ) degradation. Response surfaces showed that ATZ degradation was enhanced with an increased dosage of PDS (regression slope of 24.09) and hydrochar (regression slope of 4.19). In contrast, bacterial abundance was negatively related to PDS dosage but positively to hydrochar dosage. At the optimum dosage of PDS (2.21% dry weight of soil) and hydrochar (5% dry weight of soil), ATZ degradation reached 95.31%, and bacterial abundance recovered to 7.72 log gene copy number g-1 soil (versus 8.44 in raw soil). Hydrochar alleviated the negative impacts of PDS on soil fertility such as urease activity. High PDS dosages (3% dry weight of soil) facilitated the proliferation of Halomonas, while moderate dosages (1.5% dry weight of soil) stimulated Alicyclobacillus. Hydrochar facilitated the growth of functional genera like Comamonas, Cloacibacterium, and Terrabacter. ATZ degradation pathway was positively correlated with Bacillus and nitrogen metabolism pathway. Hydrochar mediated intracellular reactive oxidative species scavenger reactions in catalase activity, allowing microbial survival under harsh oxidative conditions due to PDS addition.

The direct effect of CO2 on denitrification has attracted great attention currently. Our previous studies have confirmed that CO2 inhibited heterotrophic denitrification and caused high nitrite accumulation and nitrous oxide emission. Cysteine is a widely reported bio-accelerator; however, its effect on denitrification under CO2 exposure remains unknown. In this paper, the effect of cysteine on heterotrophic denitrification and its mechanisms under CO2 exposure were explored with the model denitrifier, Paracoccus denitrificans. We observed that total nitrogen removal increased from 17.9% to 90.4% as cysteine concentration increased from 0 to 50 µM, probably due to restoration of cell growth and viability. Further study showed that cysteine reduced the inhibition of CO2 on denitrification due to multiple positive influences: (1) regulating glutathione metabolism to eliminate intracellular reactive nitrogen species (RNS), while reducing extracellular polymeric substances (EPS) levels and altering its composition, ultimately restoring cell membrane integrity (2) facilitating the transport and metabolism of carbon sources to increase NADH production, and (3) increasing intracellular iron and up-regulating the expression of key iron transporters genes (AfuA, AfuB, ExbB and TonB) to restore the transport and consumption of electron. This study suggests that cysteine can be added to recover heterotrophic denitrification performance after inhibition by elevated CO2.

The reuse of waste polyvinyl chloride (PVC) has drawn much attention as it can reduce plastic waste and associated pollution, and provide valuable raw materials and products. In this study, sulfonated PVC-derived hydrochar (HS-PVC) was synthesized by two-stage hydrothermal treatment (HT) and sulfonation, and shown to be a versatile adsorbent. The removal of Cu(II) cations and Cr(VI) anions using HS-PVC reached 81.2 ± 1.6% and 60.3 ± 3.8%, respectively. The first stage of HT was crucial for the dichlorination of PVC and the formation of an aromatic structure. This stage guaranteed the introduction of -SO3H onto PVC-derived hydrochar through subsequent sulfonation. HT intensities (i.e., temperature and time) and sulfonation intensity strongly determined the adsorption capacity of HS-PVC. Competitive adsorption between Cu(II) and Cr(VI) onto HS-PVC was demonstrated by binary and preloading adsorption. The proposed Cu(II) cations adsorption mechanism was electrostatic adsorption, while Cr(VI) were possibly complexed by the phenolic -OH and reduced to Cr(III) cations by CC groups in HS-PVC. In addition, HS-PVC derived from PVC waste pipes performed better than PVC powder for Cu(II) and Cr(VI) removal (>90%). This study provides an efficient method for recycling waste PVC and production of efficient adsorbents.

For the time-consuming and stressful body measuring task of Qinchuan cattle and farmers, the demand for the automatic measurement of body dimensions has become more and more urgent. It is necessary to explore automatic measurements with deep learning to improve breeding efficiency and promote the development of industry. In this paper, a novel approach to measuring the body dimensions of live Qinchuan cattle with on transfer learning is proposed. Deep learning of the Kd-network was trained with classical three-dimensional (3D) point cloud datasets (PCD) of the ShapeNet datasets. After a series of processes of PCD sensed by the light detection and ranging (LiDAR) sensor, the cattle silhouettes could be extracted, which after augmentation could be applied as an input layer to the Kd-network. With the output of a convolutional layer of the trained deep model, the output layer of the deep model could be applied to pre-train the full connection network. The TrAdaBoost algorithm was employed to transfer the pre-trained convolutional layer and full connection of the deep model. To classify and recognize the PCD of the cattle silhouette, the average accuracy rate after training with transfer learning could reach up to 93.6%. On the basis of silhouette extraction, the candidate region of the feature surface shape could be extracted with mean curvature and Gaussian curvature. After the computation of the FPFH (fast point feature histogram) of the surface shape, the center of the feature surface could be recognized and the body dimensions of the cattle could finally be calculated. The experimental results showed that the comprehensive error of body dimensions was close to 2%, which could provide a feasible approach to the non-contact observations of the bodies of large physique livestock without any human intervention.

The body dimension measurement of large animals plays a significant role in quality improvement and genetic breeding, and the non-contact measurements by computer vision-based remote sensing could represent great progress in the case of dangerous stress responses and time-costing manual measurements. This paper presents a novel approach for three-dimensional digital modeling of live adult Qinchuan cattle for body size measurement. On the basis of capturing the original point data series of live cattle by a Light Detection and Ranging (LiDAR) sensor, the conditional, statistical outliers and voxel grid filtering methods are fused to cancel the background and outliers. After the segmentation of K-means clustering extraction and the RANdom SAmple Consensus (RANSAC) algorithm, the Fast Point Feature Histogram (FPFH) is put forward to get the cattle data automatically. The cattle surface is reconstructed to get the 3D cattle model using fast Iterative Closest Point (ICP) matching with Bi-directional Random K-D Trees and a Greedy Projection Triangulation (GPT) reconstruction method by which the feature points of cattle silhouettes could be clicked and calculated. Finally, the five body parameters (withers height, chest depth, back height, body length, and waist height) are measured in the field and verified within an accuracy of 2 mm and an error close to 2%. The experimental results show that this approach could be considered as a new feasible method towards the non-contact body measurement for large physique livestock.

BACKGROUND Multiple relevant risk factors for lung cancer have been reported in different populations, but results of previous studies were not consistent. Therefore, a meta-analysis is necessary to summarize these outcomes and reach a relatively comprehensive conclusion. MATERIAL AND METHODS STATA 12.0 software was used for all statistical of the relationship between COX-2 polymorphisms and lung cancer risk. Inter-study heterogeneity was examined with the Q statistic (significance level at P<0.1). The publication bias among studies in the meta-analysis was analyzed with Begg's funnel plot and Egger's test. Hardy-Weinberg equilibrium was tested in all controls of the studies. RESULTS COX-2 rs20417 polymorphism had a significant association with reduced risk of lung cancer under homozygous and recessive models, and similar results were observed in white and population-based subgroups under 2 and 3 contrasts, respectively. Additionally, rs2066826 polymorphism manifested a strong correlation with increased risk of lung cancer under 5 genetic models. CONCLUSIONS In COX-2 gene, rs20417 may have a certain relationship with reduced risk of lung cancer, while rs2066826 may increase the risk of lung cancer.

OBJECTIVE: To explore the clinical significance of multiple radiography of the pelvis in the evaluation of surgical outcomes for patients with slow transit constipation complicated with outlet obstruction. METHODS: Patients with slow transit constipation complicated with outlet obstruction were diagnosed by multiple radiography of the pelvis after screening using colon transit study. Surgery was performed according to the cause of the obstruction. Anorectal angle and the locations of perineum, pelvic peritoneum, and bladder were assessed by multiple radiography of the pelvis one month after surgery. The changes in locations of pelvic organs were assessed and the imaging appearance after the release of obstruction was observed. RESULTS: A total of 48 patients were included. Rectocele repair, partial mucosectomy with rectopexy, and hysteropexy were performed. All the patients were followed up with a mean length of 19(6-58) months. Excluding 2 patients who had no symptomatic improvement, the mean bowel movements was 1.9 times per day in the remaining 46 patients(95.8%). Preoperative anorectal angle at the squeezing phase was(128.09±13.82) degree and the difference between squeezing and resting phase was (11.14±12.58) degree, while the postoperative angle was (180.26±9.98) degree and the difference(20.01±13.11) degree(P<0.05). Preoperative location of the perineum at the squeezing phase was(-2.05±0.83) cm and the difference was(2.23±0.78) cm, while postoperative location was (-0.50±1.13) cm and the difference was (2.18±1.04) cm(P<0.05). Preoperative location of the pelvic peritoneum at the squeezing phase was(4.91±1.32) cm and the difference was (1.32±0.89) cm, while postoperative location was (2.62±2.53) cm and the difference was (3.28±0.68) cm (P<0.05). Preoperative bladder location at the squeezing phase in patients with urological symptoms was (3.92±2.51) cm and the difference was(1.39±1.27) cm, while postoperative location was (2.15±1.55) cm and the difference was (1.98±1.54) cm(P<0.05). CONCLUSION: Multiple imaging of the pelvis provides objective evidence in the evaluation of surgical outcomes for patients with chronic slow transit constipation complicated with outlet obstruction.
