RESUMO

We have developed methods for detecting the genetic diversity of grapevine rupestris stem pitting-associated virus (GRSPaV) based on restriction fragment length polymorphism (RFLP) and single stranded conformational polymorphism (SSCP) in the 905 nt 3' sequence. The amplicons were cloned from six grapevine cultivars, and colony polymerase chain reaction (colony PCR) using recombination bacteria was subsequently analyzed by RFLP and SSCP. Four haplotypes of SSCP and six haplotypes of Sac I RFLPs were defined. The two methods had a 40% discrepancy rate in showing the degree of diversity. All clones were sequenced and were used to construct a phylogenetic tree with seven previously reported GRSPaV sequences. In the tree, all the newly acquired sequences were divided into three clusters, I, II, and III, which corresponded to haplotypes I, II, and III of SSCP, respectively. Haplotype IV of SSCP was grouped into cluster II. A recombination analysis showed that haplotype IV has undergone a recombination event. Together, these results indicate that the SSCP assay is useful for the rapid identification of genetic diversity of GRSPaV. This is the first report of an analysis of the large fragment of GRSPaV by colony PCR-SSCP. Phase shifts from scattering are used to analyze and engineer acoustic radiation forces. With the aid of phase shifts, analytical results for acoustic radiation forces can be simplified into compact and physically meaningful expressions, which can be used to develop a simplified procedure for the engineering of the radiation force. The desired radiation force can be fulfilled by a specific set of phase shifts up to a certain order, and then the required phase shifts can be fulfilled by engineering object and beam parameters. As an example, the phase shift approach is used to engineer the inner-to-outer radius ratio and the outer radius of a spherical shell to show how to use the phase shift method for the design of acoustic radiation forces. The example here is a force that is desired to pull particles against the propagation of a Bessel beam. A small paraxial parameter to pull a spherical shell is satisfied by in-phase scattering of monopole, dipole, quadrupole, octupole, and beyond. The example presented here is relatively simple yet reveals the advantages of the phase shift approach. The phase shift method can provide a simplified route for the design of acoustic tweezers using either traveling beams or standing waves.

Nanomedicine has attracted increasing attention and emerged as a safer and more effective modality in cancer treatment than conventional chemotherapy. In particular, the distinction of tumor microenvironment and normal tissues is often used in stimulus-responsive drug delivery systems for controlled release of therapeutic agents at target sites. In this study, we developed mesoporous silica nanoparticles (MSNs) coated with polyacrylic acid (PAA), and pH-sensitive lipid (PSL) for synergistic delivery and dual-pH-responsive sequential release of arsenic trioxide (ATO) and paclitaxel (PTX) (PL-PMSN-PTX/ATO). Tumor-targeting peptide F56 was used to modify MSNs, which conferred a target-specific delivery to cancer and endothelial cells under neoangiogenesis. PAA- and PSL-coated nanoparticles were characterized by TGA, TEM, FT-IR, and DLS. The drug-loaded nanoparticles displayed a dual-pH-responsive (pHe = 6.5, pHendo = 5.0) and sequential drug release profile. PTX within PSL was preferentially released at pH = 6.5, whereas ATO was mainly released at pH = 5.0. Drug-free carriers showed low cytotoxicity toward MCF-7 cells, but ATO and PTX co-delivered nanoparticles displayed a significant synergistic effect against MCF-7 cells, showing greater cell-cycle arrest in treated cells and more activation of apoptosis-related proteins than free drugs. Furthermore, the extracellular release of PTX caused an expansion of the interstitial space, allowing deeper penetration of the nanoparticles into the tumor mass through a tumor priming effect. As a result, FPL-PMSN-PTX/ATO exhibited improved in vivo circulation time, tumor-targeted delivery, and overall therapeutic efficacy.

Invisibility or unhearability cloaks have been made possible by using metamaterials enabling light or sound to flow around obstacle without the trace of reflections or shadows. Metamaterials are known for being flexible building units that can mimic a host of unusual and extreme material responses, which are essential when engineering artificial material properties to realize a coordinate transforming cloak. Bending and stretching the coordinate grid in space require stringent material parameters; therefore, small inaccuracies and inevitable material losses become sources for unwanted scattering that are decremental to the desired effect. These obstacles further limit the possibility of achieving a robust concealment of sizeable objects from either radar or sonar detection. By using an elaborate arrangement of gain and lossy acoustic media respecting parity-time symmetry, we built a one-way unhearability cloak able to hide objects seven times larger than the acoustic wavelength. Generally speaking, our approach has no limits in terms of working frequency, shape, or size, specifically though we demonstrate how, in principle, an object of the size of a human can be hidden from audible sound.

The past decade witnesses considerable efforts to design acoustic illusion cloak that produces the desired scattered field for a specific object illuminated by an external field. Yet the possibility of generating acoustic illusion directly for a sound source still remains unexplored despite the great fundamental and practical significance, and previous transformation acoustics-based designs need to have bulky sizes in terms of working wavelength. Here we propose to produce arbitrary illusion for an airborne sound source with no need to resort to coordinate transformation method. Based on an inherently different mechanism that uses acoustic metasurface to provide azimuthally-dependent local phase delay to the radiated wavefront, we shrink the thickness of the single layer enclosing the source to subwavelength scale without modulating the shape of layer. The performance of our scheme is demonstrated via distinct phenomena of virtually shifting the source location and introducing angular momentum. Numerical results verify our theoretical predictions, showing the extraordinary capability of the presented device to freely manipulate the radiation pattern of a simplest point source, making it acoustically appearing like another arbitrarily complicated source. Our findings open new avenues to the design and application of acoustic illusion devices and may have deep implications in many diverse fields such as architectural acoustics and biomedical engineering.

Two primer pairs were used to detect apple stem pitting virus (ASPV) using a reverse transcription (RT)-PCR test. 82 out of the 141 randomly collected samples, from ten orchards in five provinces and regions of China, tested positive. In the positive samples forty-five (55%) were infected by ASPV and two other viruses. The full coat protein (CP) and the triple gene block (TGB) gene 1, 2 and 3 of partial ASPV isolates were subsequently cloned. The nucleotide and amino acid identities of 39 CP sequence variants from 31 Chinese apple samples were compared with that of previously reported ASPV isolates and were 67.4-96.0% and 68.4-97.7%, respectively. All ASPV sequence variants from Chinese apples separated into two clades with CP- and TGB-based phylogenetic trees, whilst the grouping of TGB2 and TGB3 trees was the same. Three recombinants (FS06-2, X5-2, and XLF-C-2) for CP and six (TH2-5, X8-2, FS05-2, X6-2 and XLF-A-1) recombinants for TGB were identified from the Chinese apple isolates. Two recombinants were found in the TGB sequence of isolate XLF-A-1. The results presented here may assist in the development of a more comprehensive screening tool for apple viruses.

Non-blind invisibility cloaks allowing the concealed object to sense the outside world have great application potentials such as in high-precision sensing or underwater camouflage. However the existing designs based on coordinate transformation techniques need complicated spatially-varying negative index or intricate multi-layered configurations, substantially increasing the difficulty in practical realization. Here we report on the non-blind acoustic invisibility for a circular object in free space with simple distribution of cloak parameters. The mechanism is that, instead of utilizing the transformation acoustics technique, we develop the analytical formulae for fast prediction of the scattering from the object and then use an evolutionary optimization to retrieve the desired cloak parameters for minimizing the scattered field. In this way, it is proven possible to break through the fundamental limit of complementary condition that must be satisfied by the effective parameters of the components in transformation acoustics-based cloaks. Numerical results show that the resulting cloak produces a non-bflind invisibility as perfect as in previous designs, but only needs two layers with homogenous single-negative parameters. With full simplification in parameter distribution and broken symmetry in complementary relationship, our scheme opens new route to free-space non-blind invisibility, taking a significant step towards real-world application of cloaking devices.

ETHNOPHARMACOLOGICAL RELEVANCE: The ripe seeds of Herpetospermum caudigerum have been used in Tibetan folk medicine for treatment of bile or liver diseases including jaundice, hepatitis, intumescences or inflammation. Previously reports suggested that the seed oil and some lignans from H. caudigerum exhibited protective effects against carbon tetrachloride (CCl4)-induced hepatic damage in rats, which may be related to their free radical scavenging effect. However, the protective effect of H. caudigerum against cholestasis is still not revealed. The aim of the present study was to investigate the pharmacological effect and the chemical constituents of the petroleum ether extract (PEE) derived from H. caudigerum against α-naphthylisothiocyanate (ANIT)-induced acute cholestasis in rats. MATERIALS AND METHODS: Male cholestatic Sprague-Dawley (SD) rats induced by ANIT (60mg/kg) were orally administered with PEE (350, 700 and 1400mg/kg). Levels of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), γ-Glutamyl transpeptidase (γ-GTP), total bilirubin (TBIL), direct bilirubin (DBIL) and total bile acid (TBA), as well as bile flow, and histopathological assay were evaluated. Hepatic malondialdehyde (MDA), myeloperoxidase (MPO), superoxide dismutase (SOD), glutathione S-transferase (GST), and nitric monoxide (NO) in liver were measured to explore the possible protective mechanisms. Phytochemical analysis of PEE was performed by gas chromatography-mass spectrometer (GC-MS). RESULTS: PEE have exhibited significant and dose-dependent protective effect on ANIT-induced liver injury by reduce the increases in serum levels of ALT, AST, ALP, γ-GTP, TBIL, DBIL and TBA, restore the bile flow in cholestatic rats, and reduce the severity of the pathological tissue damage induced by ANIT. Hepatic MDA, MPO and NO contents in liver tissue were reduced, while SOD and GST activities were elevated in liver tissue. 49 compounds were detected and 39 of them were identified by GC-MS analysis, in which long-chain fatty acids were the main constituents. CONCLUSIONS: PEE exhibited a dose-dependently protective effect on ANIT-induced liver injury in cholestatic rats with the potential mechanism of attenuated oxidative stress in the liver tissue, and the possible active compounds were long-chain fatty acids.

RESUMO

The chemical constituents of Herpetospermum caudigerum were investigated using chromatographic methods, including silica gel column chromatography, Sephadex LH-20 and semi-preparative HPLC. Four compounds were isolated and their structures were elucidated by spectral data and physicochemical properties, which were identified as 2,11-dimethoxy-3,9-dihydroxy-7 H-dibenzo[c,e]oxepin-5-one (1),7,8'-didehydroherpetotriol(2), herpetotrio l(3) and kaempferitrin (4). Among those, compound 1 is one new 7H-dibenzo[c,e]oxepin-5-one, named as herpetolide C.

The complete nucleotide sequences of two isolates of grapevine rupestris stem pitting-associated virus (LSL and JF) collected from grapevine of Xingcheng in Liaoning Province, China, were determined. The genomes of both LSL and JF were found to contain five open reading frames (ORFs). Sequence alignments showed that the genomic sequences of JF were 76.1 %-83.5 % identical to the other ten GRSPaV isolates that have been reported previously and that the nucleotide sequence identity of isolate LSL to other isolates was no more than 78 %. Phylogenetic analysis based on the complete genome sequence indicated that JF belongs to group III and that LSL belongs to a new group (group IV). The average genetic distances of the new genetic lineage from groups I, II and III were 0.34, 0.32 and 0.33, respectively.

Phytochemical investigation on the seeds of Herpetospermum caudigerum afforded two new coumarin glycosides A and B (1 and 2), together with a known compound herpetolide A (3). Their structures were determined by spectroscopic methods, including 2D NMR techniques. Compound 3 exhibited cytotoxicity against HepG2 cells with IC50 value of 6.47 µg/ml. Compound 2 showed anti-hepatitis B virus activity with HBsAg secretion by 33.1% at 200 µg/ml.

Citrus tristeza virus (CTV) isolates collected from southern China were characterized by biological indexing on citrus indicators, restriction fragment length polymorphism (RFLP), and bi-directional reverse transcription-polymerase chain reaction (BD-PCR) analysis of their coat protein (CP) genes. Of the 30 isolates, only two isolates, N3 and N4, did not induce visible symptoms. Twenty-eight other isolates induced stem pitting and vein clearing, plant stunting, and leaf yellowing symptoms on Mexican lime, Duncan grapefruit, and sour orange seedlings. In BD-PCR analysis, a 392-bp fragment specific for the mild strains was amplified from isolates N3 and N4, and a 320-bp fragment specific for the severe strains was produced from the other 28 isolates. The RFLP analysis for RT-PCR products of the CP gene with restriction enzyme HinfI identified seven groups representing groups I-VI and a new group, which was not involved in the seven groups defined by Gillings (J Virol Methods 44:305-317, 1993). The sequences of the CP genes from 12 Chinese CTV isolates showed a high divergence, with 91.5-99.7% identities at the nucleotide level and 94.2-99.6% identities at the amino acid level. Our results suggest that the composition of CTV populations from China has a high genetic diversity in the CP gene.
