RESUMO

The contamination of sediments by toxic metals poses a significant threat to both river ecosystems and human health. In this study, the geo-accumulation index (Igeo), biotoxicity evaluation method, and potential ecological risk index (RI) were employed to analyze the contamination level, biotoxicity risk, and potential ecological risk of toxic metals in surface sediments of the Xiaoqing River. To identify toxic metal sources, Spearman correlation and principal component analysis with multiple linear regression analysis (PCA-MLR) were employed. Additionally, redundancy analysis (RDA) was utilized to investigate potential driving factors affecting toxic metal accumulation in sediments. The results revealed that the levels of the five investigated metals (Cr, Pb, As, Hg, and Cd) showed constant fluctuations during the period 1996-2020. The midstream was found to be more polluted than the upstream and downstream. In the research area, Hg was identified as the primary contaminant with high levels of contamination, posing a biotoxicity risk and potential ecological risk. Pollution sources were identified for two periods: A (1996-2010) and B (2011-2020), with industrial, agricultural, traffic, and natural sources being the main contributors. During period A, industrial sources accounted for the highest proportion (40.8%), followed by agricultural sources (36.6%), and geological natural sources (22.6%). During period B, agricultural sources accounted for the highest proportion (42%), followed by industrial and traffic sources (32.4%), and geological natural sources (25.6%). The distribution of toxic metals in the basin was significantly influenced by water pH, sediment organic matter, population density, and per capita GDP. The study results provide fundamental data for preventing pollution and managing water resources contaminated with toxic metals in the sediments of the Xiaoqing River in Jinan. Additionally, it serves as a reference for analyzing related ecological and environmental issues in the basin.

RESUMO

Adrenocortical carcinoma (ACC) is a malignancy with a poor prognosis and high mortality rate. A high tumor mutational burden (TMB) has been found to be associated with poor prognosis in ACC. Thus, exploring ACC biomarkers based on TMB holds significant importance for patient risk stratification. In our research, we utilized weighted gene coexpression network analysis and an assay for transposase-accessible chromatin with high-throughput sequencing to identify genes associated with TMB. Through the comprehensive analysis of various public datasets, Lamin B1 (LMNB1) was identified as a biomarker associated with a high TMB and low chromatin accessibility. Immunohistochemical staining demonstrated high expression of LMNB1 in ACC compared to noncancerous tissues. Functional enrichment analyses revealed that the function of LMNB1 is associated with cell proliferation and division. Furthermore, cell assays suggested that LMNB1 promotes tumor proliferation and invasion. In addition, mutation analysis suggested that the high expression of LMNB1 is associated with TP53 mutations. Additionally, LMNB1 was highly expressed in the vast majority of solid tumors across cancers. In our immune analysis, we discovered that the high expression of LMNB1 might suppress the infiltration of CD8+ T cells in the ACC microenvironment. In summary, LMNB1 is a predictive factor for the poor prognosis of adult and pediatric ACC. Its high expression in ACC is positively associated with high TMB and lower chromatin accessibility, and it promotes ACC cell proliferation and invasion. Therefore, LMNB1 holds promise as a novel biomarker and potential therapeutic target for ACC.

Glioma is the most common primary brain tumor. Filamin-binding LIM protein 1 (FBLIM1) has been identified in multiple cancers and is suspected of playing a part in the development of tumors. However, the potential function of FBLIM1 mRNA in glioma has not been investigated. In this study, the clinical information and transcriptome data of glioma patients were, respectively, retrieved from the TCGA and CGGA databases. The expression level of FBLIM1 mRNA was shown to be aberrant in a wide variety of malignancies. Significantly, when glioma samples were compared to normal brain samples, FBLIM1 expression was shown to be significantly elevated in the former. A poor prognosis was related to high FBLIM1 expression, which was linked to more advanced clinical stages. Notably, multivariate analyses demonstrated that FBLIM1 expression was an independent predictor for the overall survival of glioma patients. Immune infiltration analysis disclosed that FBLIM1 expression had relevance with many immune cells. The results of RT-PCR suggested that FBLIM1 expression was markedly elevated in glioma specimens. Functional experiments unveiled that the knockdown of FBLIM1 mRNA suppressed glioma cell proliferation. In general, we initially discovered that FBLIM1 mRNA might be a possible prognostic marker in glioma.

The accumulation of heavy metals in river sediment poses a major threat to ecological safety. The Xiaoqing River originates in western Jinan, with higher population density and per capita gross domestic product (GDP) in its basin compared to the Shandong province average. This study analyzed the spatial characteristics, ecological risk, human health risk, and contamination sources of heavy metals by collecting sediment samples from Xiaoqing River. We use the methods such as geo-accumulation index (Igeo), ecological risk assessment based on the interval number sorting method, and health risk assessment to evaluate the risk of heavy metals in sediments. The research finding suggests heavy metals including Pb, As, Ni, and Cr are low ecological risks, while Hg and Cd have reached high and extreme ecological risks. Correlation analysis and principal component analysis were used to analyze the correlation and sources of different heavy metals. The six heavy metals were categorized into three groups. Factor 1, comprising Hg, Cr, and Pb, was identified as a mixed source with a contribution rate of 37.76%. Factor 2 is an agricultural source and comprises Ni, Cd, and As with a contribution rate of 27.05%. Factor 3 includes Pb and Ni contributing to 15.30% as a natural source. This study offers valuable insights for the prevention of heavy metal pollution, as well as promoting sustainable urban development.

Background: Metastatic castration-resistant prostate cancer (mCRPC) is a highly aggressive stage of prostate cancer, and non-mutational epigenetic reprogramming plays a critical role in its progression. Super enhancers (SE), epigenetic elements, are involved in multiple tumor-promoting signaling pathways. However, the SE-mediated mechanism in mCRPC remains unclear. Methods: SE-associated genes and transcription factors were identified from a cell line (C4-2B) of mCRPC by the CUT&Tag assay. Differentially expressed genes (DEGs) between mCRPC and primary prostate cancer (PCa) samples in the GSE35988 dataset were identified. What's more, a recurrence risk prediction model was constructed based on the overlapping genes (termed SE-associated DEGs). To confirm the key SE-associated DEGs, BET inhibitor JQ1 was applied to cells to block SE-mediated transcription. Finally, single-cell analysis was performed to visualize cell subpopulations expressing the key SE-associated DEGs. Results: Nine human TFs, 867 SE-associated genes and 5417 DEGs were identified. 142 overlapping SE-associated DEGs showed excellent performance in recurrence prediction. Time-dependent receiver operating characteristic (ROC) curve analysis showed strong predictive power at 1 year (0.80), 3 years (0.85), and 5 years (0.88). The efficacy of his performance has also been validated in external datasets. In addition, FKBP5 activity was significantly inhibited by JQ1. Conclusion: We present a landscape of SE and their associated genes in mCPRC, and discuss the potential clinical implications of these findings in terms of their translation to the clinic.

PURPOSE: To investigate the clinical relevance and biological function of the kinesin super-family protein 4A (KIF4A) expression in prostate cancer (PCa). METHODS: We examined 1) the relationship between the expression of KIF4A and clinico-pathological characteristics of PCa patients using a tissue microarray and the Cancer Genome Atlas database, 2) the prognostic value of KIF4A expression in patients using Kaplan-Meier plots and 3) the functions of KIF4A in LNCaP and DU145 cells, such as cell proliferation, cell cycle and cell apoptosis. RESULTS: Compared with normal prostate, the mRNA and protein expressions of KIF4A were up-regulated in PCa. Metformin is a classic type II diabetes drug which possesses anti-tumor properties for various cancers. However, different cancers do not respond to metformin with the same effectiveness or acquire resistance. Thus, searching for vulnerabilities of metformin-resistant prostate cancer is a promising strategy to improve the therapeutic efficiency of the drug. A genome-scale CRISPR-Cas9 activation library search targeting 23,430 genes was conducted to identify the genes that confer resistance to metformin in prostate cancer cells. Candidate genes were selected by total reads of sgRNA and sgRNA diversity, and then a CCK8 assay was used to verify their resistance to metformin. Interestingly, we discovered that the activation of ECE1, ABCA12, BPY2, EEF1A1, RAD9A, and NIPSNAP1 contributed to in vitro resistance to metformin in DU145 and PC3 cell lines. Notably, a high level of RAD9A, with poor prognosis in PCa, was the most significant gene in the CCK8 assay. Furthermore, we discerned the tumor immune microenvironment with RAD9A expression by CIBERSORT. These results suggested that a high level of RAD9A may upregulate regulatory T cells to counterbalance metformin in the tumor immune microenvironment.

Ordered arrays of metallic nanopillars embedded in a ceramic matrix have recently attracted considerable interest for their multifunctionality in advanced devices. A number of hurdles need to be overcome for achieving practical devices, including selections of metal-ceramic combination, creation of tunable and ordered structure, and control of strain state. In this article, we demonstrate major advances to create such a fine nanoscale structure, i.e., epitaxial self-assembled vertically aligned metal-ceramic composite, in one-step growth using pulsed laser deposition. Tunable diameter and spacing of the nanopillars can be achieved by controlling the growth parameters such as deposition temperature. The magnetic metal-ceramic composite thin films demonstrate uniaxial anisotropic magnetic properties and enhanced coercivity compared to that of bulk metal. The system also presents unique anisotropic electrical transport properties under in-plane and out-of-plane directions. This work paves a new avenue to fabricate epitaxial metal-ceramic nanocomposites, which can simulate broader future explorations in nanocomposites with novel magnetic, optical, electrical, and catalytical properties.

OBJECTIVE: To identify a specific hypermethylated molecular biomarker for human malignant glioblastoma prognosis. MATERIALS AND METHODS: Genome-wide methylation profiling was performed on 33 tumors and 3 normal glioblastoma samples using the Infinium HumanMethylation450 microarray. Cluster analysis was carried out in these samples according to the differentiated methylated genes. DNA methylation of selected significant candidates was subsequently validated to analyze the association of methylation status of these genes with overall survival as well as gene expression. RESULTS: We found 217 hypermethylated CpG sites located in 210 respective genes with significant differences in short- and long-term survival (STS and LTS) samples (P < 0.01). Cluster analysis revealed fine clustering of genes with LTS and STS. Of these, we selected 10 most hypermethylated genes, including IL11, RRAD, MS4A6A, SNAPC2, ALDH1A3, ADCY1, MMS19L, NDUFB8, POMC, and THSD4, to perform cluster analysis. It came out with the same fine classification and with survival time of these patients. The top ranking genes were further examined to compare their methylation status with the overall survival rate of patients, as well as with gene expression levels. CONCLUSION: We obtained a featured global profiling of DNA methylation in glioblastoma. Our findings strongly indicate that epigenetic silencing of IL11, RRAD, MS4A6A, SNAPC2, and ALDH1A3 are common events in glioblastoma which could be used as novel biomarkers for the prognosis of glioblastoma.

Mixed ionic-electronic conductors are widely used in devices for energy conversion and storage. Grain boundaries in these materials have nanoscale spatial dimensions, which can generate substantial resistance to ionic transport due to dopant segregation. Here, we report the concept of targeted phase formation in a Ce0.8Gd0.2O2-δ-CoFe2O4 composite that serves to enhance the grain boundary ionic conductivity. Using transmission electron microscopy and spectroscopy approaches, we probe the grain boundary charge distribution and chemical environments altered by the phase reaction between the two constituents. The formation of an emergent phase successfully avoids segregation of the Gd dopant and depletion of oxygen vacancies at the Ce0.8Gd0.2O2-δ-Ce0.8Gd0.2O2-δ grain boundary. This results in superior grain boundary ionic conductivity as demonstrated by the enhanced oxygen permeation flux. This work illustrates the control of mesoscale level transport properties in mixed ionic-electronic conductor composites through processing induced modifications of the grain boundary defect distribution.

Hoxa5 is preferentially expressed in haematopoietic stem cells (HSCs) and multipotent progenitor cells (MPPs), and is more highly expressed in expanding HSCs. To date, little is known regarding the role of Hoxa5 in HSCs and downstream progenitor cells in vivo. In this study, we show that increased expression of Hoxa5 in haematopoietic stem cells leads to aberrant erythropoiesis in vivo. Hoxa5 differentially modifies the cell cycle of HSCs and lineage committed progenitor cells, depending on the cellular context. Hoxa5 drives HSCs, but not MPPs, through the cell cycle and arrests erythroid progenitor cells in G0 phase. Although the HSC pool shrinks after overexpression of Hoxa5, HSCs sustain the abilities of self-renewal and multipotency. In vivo, Hoxa5 has two effects on erythropoiesis: it causes a predominance of mature erythroid lineage cells and the partial apoptosis of erythroid progenitors. RNA-seq indicates that multiple biological processes, including erythrocyte homeostasis, cell metabolism, and apoptosis, are modified by Hoxa5. The results of this study indicate that Hoxa5 is a key regulator of the HSC cell cycle, and the inappropriate expression of Hoxa5 in lineage-committed progenitor cells leads to aberrant erythropoiesis.

NiO-yttria-stabilized zirconia (YSZ) hollow fiber anode support with different microstructures was prepared using a phase-inversion method. The effect of the solid loading of the phase-inversion suspensions on the microstructure development of the NiO-YSZ anode support was investigated. Solid loading in the suspension was found to have an important influence on the microstructure of the NiO-YSZ anode support and viscosity-related viscous fingering mechanism can be adopted to explain the pore formation mechanism of the as-prepared hollow fibers. NiO-YSZ anode-supported microtubular solid oxide fuel cells (SOFCs) with different anode microstructures were fabricated and tested, and the correlation between the anode support microstructures, porosity, gas permeability, electrical conductivity, and the cell electrochemical performance was discussed. Microtubular SOFCs with a cell configuration of Ni-YSZ/YSZ/YSZ-LSM (LSM = (La(0.8)Sr(0.2))(0.95)MnO(3-x)) and optimized anode microstructure show cell output power density of 833.9 mW cm(-2) at 750 °C using humidified H2 as fuel and ambient air as oxidant.

The distribution of fine roots of Pinus tabuliformis, Populus tomentosa, Prunus armeniaca, Robinia pseudoacacia, Hippophae rhamnoides, and Caragana korshinskii was investigated by using soil core method and the fine root was defined as root with diameter less than 2 mm. The soil moisture and soil properties were measured. The results showed that in the horizontal direction, the distribution of fine root biomass of P. tabuliformis presented a conic curve, and the fine root biomass of the other species expressed logarithm correlation. Radial roots developed, the fine root biomass were concentrated within the scope of the 2-3 times crown, indicating that trees extended their roots laterally to seek water farther from the tree. In the vertical direction, the fine root biomass decreased with the increasing soil depth. Fine root biomass had significant negative correlation with soil water content and bulk density, while significant positive correlation with organic matter and total N contents.

Conventional Ni-BaCeO3-based membranes possess high hydrogen permeation flux but suffer serious flux degradation in CO2-containing atmosphere because of the formation of BaCO3 insulating layer. In this work, we report a novel Ni-BaZr0.1Ce0.7Y0.1Yb0.1O(3-δ) (Ni-BZCYYb) membrane, capable of both high hydrogen permeation flux and stable performance in CO2-containing atmosphere at 900 °C. Most importantly, the flux is found to be promoted rather than being diminished by CO2 normally observed for other high temperature proton conductors. The flux enhancement in Ni-BZCYYb membrane is attributed to the increase of moisture content in feed gas. When CO2 is introduced, the reverse water-gas shift reaction takes place generating H2O and CO. This work demonstrates that CO2 can be beneficial rather than detrimental for hydrogen permeation membranes that possess high chemical stability.

Taking the Pinus tabulaeformis plantation in the Anjiagou catchment on Longzhong Loess Plateau as test object, an observation was made on the characteristics of throughfall, stemflow, interception, and canopy structure of P. tabulaeformi during its growth season (from May to September) in 2011. Based on the observed data, the revised Gash analytical model was adopted to simulate the canopy interception, aimed to understand the ecological hydrological processes of Pinus tabulaeformis plantation and related mechanisms. In the observation period, a total of 19 precipitation events were observed, with a total precipitation of 215.80 mm. The throughfall, stemflow, and canopy interception were 165.24 mm, 2.29 mm, and 48.27 mm, occupying 76.7%, 1.1%, and 22.4% of the total precipitation, respectively. The simulated canopy interception was 41.24 mm, being 7.13 mm lower than the observed value and with a relative error of 14.7%. There were 33.8% and 60.0% of interception were evaporated from the canopy during and after precipitation, respectively. The revised Gash analytical model was highly sensitive to the canopy storage capacity, forest coverage, rainfall intensity, and evaporation, but less sensitive to the stemflow rate and stem water holding capacity.

From May to October 2011, an investigation was conducted on the effects of rainfall and its intensity on the canopy interception, throughfall, and stemflow of Caragana korshinskii and Hippophae rhamnoides, the main shrub species commonly planted to stabilize soil and water in the Anjiagou catchment of Loess Plateau. A total of 47 rainfall events were observed, most of which were featured with low intensity, and the total amount and average intensity of the rainfalls were 208.9 mm and 2.82 mm x h(-1), respectively. As a whole, the rainfall events of 2-10 mm and 0.1-2 mm x h(-1) had the highest frequency. The canopy interception, throughfall, and stemflow of C. korshinski were 58.5 mm (28%), 124.7 mm (59.7%), and 25.7 mm (12.3%), while those of H. rhamnoides were 17.6 mm (8.4%), 153. 1 mm (73.3%), and 38.2 mm (18.3%), respectively. Regression analysis showed that the canopy interception, throughfall, and stemflow of the two shrub species all had significant positive correlations with the rainfall amount, and had exponent or power correlations with the rainfall amount and the maximum rainfall intensity in 10 minutes.
