Co-evolved natural enemies provide sustainable and long-term control of numerous invasive insect pests, but the introduction of such enemies has declined sharply due to increasing regulations. In the absence of co-evolved natural enemies, native species may attack exotic invasive pests; however, they usually lack adaptations to control novel hosts effectively. We investigated the potential of two native pupal parasitoids, Pachycrepoideus vindemmiae and Trichopria drosophilae, to increase their developmental success on the invasive Drosophila suzukii. Replicated populations of the two parasitoids were subjected to 10 generations of laboratory selection on D. suzukii with Drosophila melanogaster serving as the co-evolved host. We assessed developmental success of selected and control lines in generations 0, 3, and 10. Changes in host preference, sex ratio, development time, and body size were measured to evaluate correlated responses with adaptation. Both parasitoid species responded rapidly to selection by significantly increasing their developmental success on the novel host within three generations, which remained constant for seven additional generations without further improvement. The generalist parasitoid species P. vindemmiae was able to reach similar developmental success as the control populations, while the performance of the more specialized parasitoid T. drosophilae remained lower on the novel than on the co-evolved host. There was no increase in preference towards the novel host over 10 generations of selection nor were there changes in development time or body size associated with adaptation in either parasitoid species. The sex ratio became less female-biased for both parasitoids after three generations of selection but rebounded in P. vindemmiae by generation 10. These results suggest that a few generations of selection may be sufficient to improve the performance of native parasitoids on invasive hosts, but with limits to the degree of improvement for managing invasive pests when exotic co-evolved natural enemies are not available.

Spotted-wing drosophila (SWD), Drosophila suzukii Matsumura (Diptera: Drosophilidae), is an invasive pest of thin-skinned fruits in the United States. Monitoring traps are an integral part of SWD integrated pest management, allowing early detection and timely management of this pest. An ideal monitoring trap should be easy to use, effective in capturing SWD, sensitive and selective to male SWD which are easy to identify due to their spotted wings, and able to predict fruit infestation from trap captures. Deli-cup-based liquid traps (grower standard), which make in-situ observations difficult, were compared with red-panel sticky traps, both baited with commercial lures (Scentry, Trécé Broad-Spectrum (BS), and Trécé High-Specificity (HS)), across several US states in blueberries (lowbush and highbush), blackberry, raspberry, and cherry crops during 2018 and 2021. Results showed that red-panel traps effectively captured SWD, were able to detect male SWD early in the season while also being selective to male SWD all season-long, and in some cases linearly related male SWD trap captures with fruit infestation. Scentry and Trécé BS lures captured similar numbers of SWD, though Trécé BS and Trécé HS were more selective for male SWD in red panel traps than liquid traps in some cases. In conclusion, due to its ease of use with less processing time, red-panel traps are promising tools for detecting and identifying male SWD in-situ and for predicting fruit infestation. However, further research is needed to refine the trap captures and fruit infestation relationship and elucidate the trap-lure interactions in berry and cherry crops.

BACKGROUND: Both innate and adaptive immune responses are important components of anticancer immunity. The CD47-SIRPα interaction could represent an important pathway used by tumour cells to evade immune surveillance. We aimed to evaluate the safety, pharmacokinetics, pharmacodynamics, and anticancer activity of evorpacept (also known as ALX148), a high-affinity CD47-blocking protein with an inactive IgG Fc region in patients with solid tumours. METHODS: We did a first-in-human, open-label, multicentre, phase 1 dose-escalation and dose-expansion study at nine hospitals and one clinic in the USA and Korea. Eligible patients for the dose-escalation and safety lead-in phases were aged 18 years or older with histological or cytological diagnosis of advanced or metastatic solid tumours with no available standard therapy, measurable or unmeasurable disease according to the Response Evaluation Criteria in Solid Tumors version 1.1, and an Eastern Cooperative Oncology Group performance status score of 0 or 1. In the dose-escalation phase, which used a 3 + 3 design, patients received intravenous evorpacept at either 0·3, 1, 3, or 10 mg/kg once per week in 21-day cycles, or 30 mg/kg once every other week in 28-day cycles. In the safety lead-in phase, patients were given the maximum tolerable dose of evorpacept from the dose-escalation phase plus either intravenous pembrolizumab (200 mg administered once every 3 weeks) or intravenous trastuzumab (8 mg/kg loading dose followed by 6 mg/kg once every 3 weeks). In the dose-expansion phase, additional patients aged 18 years or older with second-line or later-line advanced malignancies were enrolled into three parallel cohorts: those with head and neck squamous cell carcinoma (HNSCC) and those with non-small-cell lung cancer (NSCLC) were given the maximum tolerated dose of evorpacept plus intravenous pembrolizumab (200 mg administered once every 3 weeks), and patients with HER2-positive gastric or gastroesophageal junction cancer were given the maximum tolerated dose of evorpacept plus intravenous trastuzumab (8 mg/kg loading dose followed by 6 mg/kg once every 3 weeks) until disease progression, voluntary withdrawal from the study, or unacceptable toxicity. The primary endpoint was the maximum tolerated dose of evorpacept administered as a single agent and in combination with pembrolizumab or trastuzumab, measured by the occurrence of dose-limiting toxicities during the first cycle, and was assessed in all patients who had received at least one dose of evorpacept. Secondary outcomes included the safety, tolerability, and antitumour activity of evorpacept, alone or in combination with pembrolizumab or trastuzumab. The primary outcome, safety, and tolerability were assessed in all patients who had received at least one dose of evorpacept, and antitumour activity was assessed in those who recieved at least one dose of study treatment and underwent at least one post-baseline tumor assessment. This trial is registered with ClinicalTrials.gov, NCT03013218. FINDINGS: Between March 6, 2017, and Feb 21, 2019, 110 patients received single-agent evorpacept (n=28), evorpacept plus pembrolizumab (n=52), or evorpacept plus trastuzumab (n=30), and were included in the safety analysis. Median follow-up was 29·1 months (95% CI not calculable [NC]-NC) in the single-agent cohort, 27·0 months (25·1-28·8) in the evorpacept plus pembrolizumab cohort, and 32·7 months (27·0-32·7) in the evorpacept plus trastuzumab cohort. Two (7%) dose-limiting toxicities in the first cycle were reported in patients who received single-agent evorpacept; neutropenia with an associated infection in one patient with gastroesophageal junction cancer who received 3 mg/kg once per week, and thrombocytopenia with associated bleeding in one patient with pancreatic cancer who received 30 mg/kg once every other week. No maximum tolerated dose was reached; the maximum administered doses were 10 mg/kg once per week or 30 mg/kg once every other week. The 10 mg/kg once per week dose was used in the expansion cohorts in combination with pembrolizumab or trastuzumab. The most common grade 3 or worse treatment-related adverse events were thrombocytopenia with single-agent evorpacept (two [7%] patients) and evorpacept plus pembrolizumab (two [4%]), and thrombocytopenia (two [7%]) and neutropenia (two [7%]) with evorpacept plus trastuzumab. In patients who received single-agent evorpacept, four treatment-related serious adverse events were reported. Five serious treatment-related adverse events related to evorpacept plus pembrolizumab were reported, and one serious adverse event related to evorpacept plus trastuzumab was reported. In response-evaluable patients in the dose-escalation phase (n=15) receiving single-agent evorpacept once per week, four (27%) had a best overall response of stable disease (two received 0·3 mg/kg, one received 3 mg/kg, and one received 10 mg/kg); in the 11 patients who received single-agent evorpacept at the highest dose of 30 mg/kg once every other week, two (18%) had stable disease. In the dose-expansion cohort, overall responses were recorded in four (20·0%; 95% CI 5·7-43·7) of 20 patients with HNSCC who received evorpacept plus pembrolizumab, in one (5·0%; 0·1-24·9) of 20 patients with NSCLC who received evorpacept plus pembrolizumab, and in four (21·1%; 6·1-45·6) of 19 patients with gastric or gastroesophageal junction cancer who received evorpacept plus trastuzumab. INTERPRETATION: The safety findings support the use of evorpacept in combination with pembrolizumab or trastuzumab for patients with advanced solid tumours. Preliminary antitumour activity results support future investigation of evorpacept combined with pembrolizumab or trastuzumab in patients with HNSCC, gastric or gastroesophageal junction cancer, and NSCLC. Drosophila suzukii, or spotted-wing drosophila, is now an established pest in many parts of the world, causing significant damage to numerous fruit crop industries. Native to East Asia, D. suzukii infestations started in the United States a decade ago, occupying a wide range of climates. To better understand invasion ecology of this pest, knowledge of past migration events, population structure, and genetic diversity is needed. In this study, we sequenced whole genomes of 237 individual flies collected across the continental United States, as well as several sites in Europe, Brazil, and Asia, to identify and analyze hundreds of thousands of genetic markers. We observed strong population structure between Western and Eastern US populations, but no evidence of any population structure between different latitudes within the continental United States, suggesting that there are no broad-scale adaptations occurring in response to differences in winter climates. We detect admixture from Hawaii to the Western United States and from the Eastern United States to Europe, in agreement with previously identified introduction routes inferred from microsatellite analysis. We also detect potential signals of admixture from the Western United States back to Asia, which could have important implications for shipping and quarantine policies for exported agriculture. We anticipate this large genomic dataset will spur future research into the genomic adaptations underlying D. suzukii pest activity and development of novel control methods for this agricultural pest.

The invasive spotted-wing drosophila, Drosophila suzukii (Matsumura), is a key insect pest of berries globally, causing lost revenues and increased production costs associated with applications of insecticides. The insecticides utilized are commonly broad-spectrum pyrethroids, organophosphates, or carbamates in conventionally managed fields and spinosad in organically managed fields. Adoption of more selective insecticides has been limited due to their lower residual activity, and the requirement that some must be ingested to be effective. We investigated the use of feeding stimulants for D. suzukii as a method to improve longevity and efficacy in a range of insecticides. In laboratory bioassays, sugar increased the efficacy of all chemical classes tested; however, the inclusion of yeast only showed a benefit with malathion. Feeding stimulants had a limited effect in some cases under field conditions. Similarly, infestation in field plots and a semifield bioassay showed no significant decreases in infestation with the inclusion of feeding stimulants for the insecticides tested in these trials. We discuss the implications of these findings for managing D. suzukii in fruit crops to help ensure the harvest of marketable fruit.

BACKGROUND: Fruit growers largely depend on chemical control to reduce populations of the economically damaging invasive fly, Drosophila suzukii (Matsumura). Drosophila suzukii is susceptible to high temperatures and low humidity; therefore, it may be possible to implement cultural control practices that create microclimates unfavorable for its development and survival. In addition to other fruit production benefits, in-row mulches may impede the development of D. suzukii immatures when larvae leave the fruit to pupate in the soil. This study compared the effects of different mulches (black polypropylene fabric weedmats, sawdust, and wood chips) on temperature and relative humidity (RH), and on adult emergence of D. suzukii from larvae in blueberries and pupae, both above and below the ground surface in blueberry plantings (Vaccinium corymbosum L.). RESULTS: Across regions, both lower larval survival and longer periods with high suboptimal temperatures occurred above the ground in comparison to buried below the ground, regardless of mulch type. Fewer D. suzukii adults emerged from larvae on weedmat mulch at one site, but there was no effect of mulch type on temperature, RH, or D. suzukii emergence at most sites. The relationships between temperature, RH, and the emergence of adults from larvae and pupae varied by region. Natural infestation by D. suzukii in blueberries was lower over weedmat compared to wood-based mulches at one site. Greenhouse experiments showed that larvae burrowed to pupate underneath sawdust mulch, but were unable to pupate underneath a weedmat mulch. CONCLUSIONS: Although weedmats may not modify temperatures or RH enough to consistently affect D. suzukii emergence, they can reduce field suitability for D. suzukii by providing a barrier that prevents larvae from reaching favorable pupation microhabitats underground. © 2019 Society of Chemical Industry.

Vinegar flies (Diptera: Drosophilidae) are well known to be associated with yeasts, which provide important nutrients and emit attractive semiochemicals. Drosophila suzukii (Matsumura) has become a major pest of berries and cherries around the world, requiring intensive management to maintain fruit quality. Although insecticides remain a dominant control approach, disruption of fly-yeast-host interactions remains a promising avenue for reducing the economic impact of this pest. We conducted field and laboratory experiments to explore whether a crop sterilant (peroxyacetic acid and hydrogen peroxide) developed for disease control can affect D. suzukii. In 2 yr of field tests in highbush blueberries, we found significantly lower infestation by D. suzukii in plots treated with the crop sterilant, both alone and in a rotation program with zeta-cypermethrin. When shoots from treated plots were tested in no-choice bioassays, crop sterilant treatments did not affect adult mortality or oviposition, but they reduced infestation. To explore the mechanisms in the laboratory, we found that the crop sterilant did not affect adult mortality, nor oviposition on treated fruit under no-choice settings, but adult flies settled and oviposited less on treated fruit in choice settings. When the crop sterilant was applied to colonies of Hanseniaspora uvarum (Niehaus) (Saccharomycetales: Saccharomycodaceae) and Issatchenkia terricola (Van der Walt) (Saccharomycetales: Saccharomycetacea) yeasts that are attractive and provide nutrition to D. suzukii, there was a dose-dependent inhibition of their growth. We highlight the potential for microbial management as a component of integrated pest management programs and prioritize research needs to incorporate this approach into control programs.

With a high surface to volume ratio, small organisms must carefully regulate their internal water status. Spotted-wing drosophila, Drosophila suzukii (Matsumura), is an invasive frugivorous insect distributed across a wide range of geographical regions that can have periods of dry and hot weather, suggesting that this species has strategies to avoid stressful environments and reduce water loss. It also survives winter as an adult fly, indicating that it has adaptations to the low air humidity of this season. To determine the importance of water stress to D. suzukii, we studied their survival in environments of low humidity, which was manipulated using Drierite, and their survival and water loss in response to desiccation. Survival of both sexes was lower in drier conditions, and while female winter morph D. suzukii had higher mortality early on, remaining flies were able to survive longer in the drier conditions than the summer morphs. A bioassay method was adapted from Enjin et al. (2016) using 48-well plates to videotape the location of flies and quantify their behavioral responses to humidity. Male and female D. suzukii avoided dry conditions within the bioassay system, but only when there was at least 25% differential between humidity extremes. This response was observed for both summer and winter morphs of D. suzukii and our results provide guidance for attempts to manipulate crop environments to reduce the economic impact of this pest.

Drosophila suzukii Matsumura is an invasive species affecting berry crops and cherries throughout North America, South America, and Europe. Previous research suggests that in temperate climates, the overwintering success of D. suzukii is likely dependent on access to food, shelter, and adequate cold hardening. We performed a multi-state study under field conditions for two winters to determine whether D. suzukii sex, phenotype (summer-morphotype, winter-morphotype), and life stage (adults, pupae) affected survival over time while recording naturally-occurring spatial and temporal variation in temperature. Access to food was provided and the flies were buried under leaf litter. Baited traps were deployed to determine whether local populations of D. suzukii were active throughout the winter season. The duration of exposure, mean daily temperature, and cumulative time below freezing significantly affected survival. Below freezing, D. suzukii survival was significantly reduced, particularly in northern locations. In contrast, we observed sustained survival up to 10 wk in southern locations among adults and pupae. Biotic factors also significantly affected survival outcomes: female survival was greater than male survival, winter-morphotype survival was greater than summer-morphotype survival, and adult survival was greater than pupal survival. In the north, wild D. suzukii were captured only in early winter, while in the south they were found throughout the winter. These data suggest that although adult D. suzukii may overwinter in sheltered microclimates, this ability may be limited in regions where the ground temperature, or site of overwintering, falls below freezing for extended durations.

Spotted wing Drosophila, Drosophila suzukii Matsumura (Diptera: Drosophilidae), is an invasive vinegar fly that has become a primary direct pest of berry crops worldwide. We conducted 2 yr of behavioral studies in blueberry plantings to determine how fly activity varied throughout the day. Observations of diurnal activity of adult D. suzukii found the greatest activity in the morning hours between 0600 and 0800 hours, when the majority of flies were on the berries. Flies were also active in the evening mainly between 1800 and 2000 hours; however, this trend was more prominent in 2015, which had cooler and more humid evenings. Experiments examining the effect of irrigation on D. suzukii behavior showed that flies remained active during and after irrigation. The effect of insecticide treatments alone and in combination with irrigation revealed that treatment with spinosad had limited effects on the number of flies per bush, whereas spinetoram reduced the number flying and on the bushes in some cases. Zeta-cypermethrin caused longer and more consistent reduction in D. suzukii flying and on bushes. In all treatments, we observed surviving flies flying near and on treated bushes, indicating that these insecticides do not completely deter fly activity. Irrigation did not influence the effects of zeta-cypermethrin on fly behavior during daily observations up to 3 d after application. Our results highlight that the diurnal patterns of activity of D. suzukii on host plants are flexible and are relatively unaffected by irrigation or insecticide applications.
