RESUMO

OBJECTIVES: Since March 2020, a rapid increase has been observed in the prevalence of the COVID-19 pandemic, which has essentially resulted from increased disease transmission and intensified testing and reporting. The international guidelines for the prevention and treatment of the COVID-19 pandemic have been frequently updated. Such guidelines assist the governmental regulatory bodies in taking optimal measures and safeguarding their citizens against the pandemic. We conducted a short survey with a Saudi cohort to understand the awareness about COVID-19 and estimate the responses for mitigation strategies. METHODS: An electronic survey was conducted, and the first 388 responses were analysed for publishing an initial report. The questionnaire comprised 27 items and was divided into three sections, namely demographic, awareness, and response to mitigation strategies and participants' self-perceived behaviours regarding COVID-19. The perceptions of the participants were compared with their responses to mitigation measures. RESULTS: In our study, 89.7% understood the meaning of pandemic, while 82.2% correctly identified that the elderly belonged to a high-risk group for the COVID-19 infection. As many as 96.1% agreed that staying at home was one of the mitigation strategies. Nearly 35% preferred self-medication. Higher educational level (OR: 2.09, 95% CI: 1.02-4.29) and longer working hours were found to be significantly associated with a positive response to mitigation measures with p < 0.04 and p < 0.02, respectively. CONCLUSIONS: We report better understanding and appropriate response to mitigation measures towards the COVID-19 pandemic among the general population in KSA. Nevertheless, the tendency towards self-medication was reported by one-third of the responders.

The information on the hepatoprotective effect of Bauhinia malabarica Roxb. (Family Leguminosae) used in the folkloric medical practice in Malabar coast and Walayar valley of southern India for the treatment of liver related disorders is completely unknown. Hence, the efficacy of the aqueous methanolic extract of stem bark of B. malabarica (AqMeOH-Ba) was evaluated for liver function serum biochemical markers along with the antioxidant markers in liver tissues of Wistar albino rats. The biochemical observations as well as the histopathological examination of liver sections manifested considerable hepatoprotective activity of B. malabarica stem bark, and thus validated the folkloric claim.

Glycopentalone isolated from Glycosmis pentaphylla (family Rutaceae) has cytotoxic and apoptosis inducing effects in various human cancer cell lines; however, its mode of action is not known. Therefore, target fishing of glycopentalone using a combined approach of inverse docking and reverse pharmacophore mapping approach was used to identify potential targets of glycopentalone, and gain insight into its binding modes against the selected molecular targets, viz., CDK-2, CDK-6, Topoisomerase I, Bcl-2, VEGFR-2, Telomere:G-quadruplex and Topoisomerase II. These targets were chosen based on their key roles in the progression of cancer via regulation of cell cycle and DNA replication. Molecular docking analysis revealed that glycopentalone displayed binding energies ranging from -6.38 to -8.35 kcal/mol and inhibition constants ranging from 0.758 to 20.90 µM. Further, the binding affinities of glycopentalone to the targets were in the order: Telomere:G-quadruplex > VEGFR-2 > CDK-6 > CDK-2 > Topoisomerase II > Topoisomerase I > Bcl-2. Binding mode analysis revealed critical hydrogen bonds as well as hydrophobic interactions with the targets. The targets were validated by reverse pharmacophore mapping of glycopentalone against a set of 2241 known human target proteins which revealed CDK-2 and VEGFR-2 as the most favorable targets. The glycopentalone was well mapped to CDK-2 and VEGFR-2 which involve six pharmacophore features (two hydrophobic centers and four hydrogen bond acceptors) and nine pharmacophore features (five hydrophobic, two hydrogen bond acceptors and two hydrogen bond donors), respectively. The present computational approach may aid in rational identification of targets for small molecules against large set of candidate macromolecules before bioassays validation.

The present study was conducted to test the possible teratogenic and toxic effects of anti-cancer drug heptaplatin (SKI 2053) on developing embryos and fetuses in gestating SWR/J mice. Dose levels of 5.0, 10.0 and 12.5 mg heptaplatin/kg b.wt. were intraperitoneally administered to pregnant mice on days 6-8, 9-11 and 12-14 of gestation. On day 17 of gestation, all fetuses were removed and examined for toxic phenomena (embryo-fetal toxicity) by taking observation on live fetuses and embryonic resorption. Fetuses were also examined for external, internal and skeletal malformations. None of the dams treated with heptaplatin at any of the dose levels used in the present study died during the experimental period. Higher doses of heptaplatin caused greater embryonic resorption and reduced number of live fetuses. However, no loss of body weight was noticed in fetuses at any of the dose levels administered. At highest dose of heptaplatin (12.5 mg kg(-1)), tail deformity was observed in the form of short and curve tails whereas no other anatomical or skeletal malformations were noticed in any of the fetuses. In addition to mild embryo-fetotoxicity, the study indicates mild teratogenic effects of hetaplatin as reflected in fetal abnormalities at low frequency. These results have significant implications for protracted use of this drug.

Over a period of 1 year, from May 1982 to April 1983, the gastrointestinal tracts of 240 camels were examined for the presence of parasitic helminths. The study quantifies the prevalence of gastrointestinal helminths and the seasonal fluctuations in intestinal worm burdens and faecal worm egg counts. Among the three species of cestodes and eight species of nematodes which were recorded for the first time from Kuwait, Trichostrongylus probolurus (93.8%), T. colubriformis (34.2%) and Stilesia vittata (30.0%) were the most prevalent in the small intestine and Camelostrongylus mentulatus (59.6%) in the abomasum. Estimation of the intestinal worm burdens and faecal worm egg counts showed that Trichostrongylus infections were by far the most predominant. The highest worm and egg counts were recorded in June and August, during the hot dry season. This rise is attributed to infections acquired from February to April, during the cool wet season. Possibly the most effective control can be achieved by a critical treatment at the end of the wet season coinciding with the first rise in nematode population.

Infection of indigenous camels, Camelus dromedarius, with hydatid cysts has been recorded for the first time in Kuwait. From February 1982 to April 1983, 293 camels slaughtered for human consumption were examined. The overall rate of infection was 39.6%: 18.5% in animals less than 6 years old, and between 40.1% and 45.2% in older animals. The infection rate in females (44.9%) was significantly higher than in males (24.7%). Females also showed higher susceptibility to infections in multiple organs (22.7%) than males (10.5%). The lung was the most predominant site infected (63.0%). Pulmonary cysts showed a higher fertility rate than hepatic cysts (71.7% vs 29.2%) and were smaller and more numerous. The high infection rate recorded for this animal is probably related to recent shifts in animal maintenance from the traditional free-grazing to the corralling system.
