BACKGROUND AND PURPOSE: Diffuse midline gliomas with histone H3 K27M mutation are biologically aggressive tumors with poor prognosis defined as a new diagnostic entity in the 2016 World Health Organization Classification of Tumors of the Central Nervous System. There are no qualitative imaging differences (enhancement, border, or central necrosis) between histone H3 wildtype and H3 K27M-mutant diffuse midline gliomas. Herein, we evaluated the utility of diffusion-weighted imaging to distinguish H3 K27M-mutant from histone H3 wildtype diffuse midline gliomas. MATERIALS AND METHODS: We identified 31 pediatric patients (younger than 21 years of age) with diffuse gliomas centered in midline structures that had undergone assessment for histone H3 K27M mutation. We measured ADC within these tumors using a voxel-based 3D whole-tumor measurement method. RESULTS: Our cohort included 18 infratentorial and 13 supratentorial diffuse gliomas centered in midline structures. Twenty-three (74%) tumors carried H3-K27M mutations. There was no difference in ADC histogram parameters (mean, median, minimum, maximum, percentiles) between mutant and wild-type tumors. Subgroup analysis based on tumor location also did not identify a difference in histogram descriptive statistics. Patients who survived <1 year after diagnosis had lower median ADC (1.10 × 10-3mm2/s; 95% CI, 0.90-1.30) compared with patients who survived >1 year (1.46 × 10-3mm2/s; 95% CI, 1.19-1.67; P < .06). Average ADC values for diffuse midline gliomas were 1.28 × 10-3mm2/s (95% CI, 1.21-1.34) and 0.86 × 10-3mm2/s (95% CI, 0.69-1.01) for hemispheric glioblastomas with P < .05. CONCLUSIONS: Although no statistically significant difference in diffusion characteristics was found between H3-K27M mutant and H3 wildtype diffuse midline gliomas, lower diffusivity corresponds to a lower survival rate at 1 year after diagnosis. These findings can have an impact on the anticipated clinical course for this patient population and offer providers and families guidance on clinical outcomes.

BACKGROUND AND PURPOSE: The 2016 World Health Organization Classification of Tumors of the Central Nervous System includes "diffuse midline glioma with histone H3 K27M mutation" as a new diagnostic entity. We describe the MR imaging characteristics of this new tumor entity in pediatric patients. MATERIALS AND METHODS: We retrospectively reviewed imaging features of pediatric patients with midline gliomas with or without the histone H3 K27 mutation. We evaluated the imaging features of these tumors on the basis of location, enhancement pattern, and necrosis. RESULTS: Among 33 patients with diffuse midline gliomas, histone H3 K27M mutation was present in 24 patients (72.7%) and absent in 9 (27.3%). Of the tumors, 27.3% (n = 9) were located in the thalamus; 42.4% (n = 14), in the pons; 15% (n = 5), within the vermis/fourth ventricle; and 6% (n = 2), in the spinal cord. The radiographic features of diffuse midline gliomas with histone H3 K27M mutation were highly variable, ranging from expansile masses without enhancement or necrosis with large areas of surrounding infiltrative growth to peripherally enhancing masses with central necrosis with significant mass effect but little surrounding T2/FLAIR hyperintensity. When we compared diffuse midline gliomas on the basis of the presence or absence of histone H3 K27M mutation, there was no significant correlation between enhancement or border characteristics, infiltrative appearance, or presence of edema. CONCLUSIONS: We describe, for the first time, the MR imaging features of pediatric diffuse midline gliomas with histone H3 K27M mutation. Similar to the heterogeneous histologic features among these tumors, they also have a diverse imaging appearance without distinguishing features from histone H3 wildtype diffuse gliomas.

One hundred thirty-seven spring-born yearling beef heifers of British breed types were used to determine the relationships between residual feed intake (RFI) and growth rate, body composition, mature size, and fertility. Heifers were housed in a dry lot facility during the experimental period, and data were collected over a 2-yr period (yr 1, n = 67; yr 2, n = 70). Individual feed intake, BW, BCS, hip height, and ultrasonic measurements [subcutaneous rib fat (UBF), rump fat (URF), LM area (LMA), and intramuscular fat (IMF)] of body composition were recorded. Individual feed intakes (kg of TDN consumed/d) were used to calculate RFI combining both years of data. Heifers averaged 387.0 ± 19.4 d of age and 337.1 ± 29.9 kg of BW at initiation of the experiment. Mean ADG was 1.14 ± 0.21 kg/d during the trial. Based on RFI, with year of test and farm of origin included in the model as covariates, heifers were classified into groups: positive (POS; 0.74 kg of TDN/d) or negative (NEG; -0.73 kg TDN/d) for first analysis and high (HI), medium (MED), or low (LO; mean RFI = 1.06, -0.01, and -1.13 kg of TDN/d, respectively) subsequently. An initial phenotypic relationship (P < 0.05) between RFI and both UBF and URF (r = 0.19 and 0.17, respectively) was sustained (P < 0.01) with UBF (r = 0.27) and URF (r = 0.24) to trial conclusion. No other correlations with RFI were significant. Heifers classified as POS reached puberty earlier than those classified as NEG (414 ± 3.83 vs. 427 ± 4.67 d of age, P = 0.03), and possessed greater LMA per 100 kg of BW (LMACWT) at conclusion of the trial (P < 0.01). Medium heifers exhibited less URF (P < 0.05) compared with either HI or LO heifers at trial initiation. Low heifers possessed less LM area (cm(2)) per 100 kg of BW (P < 0.05) than HI but did not differ (P > 0.10) from MED heifers at either the beginning or the end of test. Additionally, a negative linear relationship was observed between RFI and age at puberty (P < 0.05). Each 1-unit increase in RFI corresponded to a decrease of 7.5 d in age at puberty, but did not affect pregnancy or conception rates (P > 0.10). Differences in body fat and rate of metabolism associated with RFI could delay reproductive maturity.

A brain-computer interface (BCI) is a communication system that takes recorded brain signals and translates them into real-time actions, in this case movement of a cursor on a computer screen. This work applied Fitts' law to the evaluation of performance on a target acquisition task during sensorimotor rhythm-based BCI training. Fitts' law, which has been used as a predictor of movement time in studies of human movement, was used here to determine the information transfer rate, which was based on target acquisition time and target difficulty. The information transfer rate was used to make comparisons between control modalities and subject groups on the same task. Data were analyzed from eight able-bodied and five motor disabled participants who wore an electrode cap that recorded and translated their electroencephalogram (EEG) signals into computer cursor movements. Direct comparisons were made between able-bodied and disabled subjects, and between EEG and joystick cursor control in able-bodied subjects. Fitts' law aptly described the relationship between movement time and index of difficulty for each task movement direction when evaluated separately and averaged together. This study showed that Fitts' law can be successfully applied to computer cursor movement controlled by neural signals.

The effect of shifting calf-weaning age on profiles of energy status (BW, BCS, and rib and rump fat) and reproductive performance of beef cows was evaluated in a 3-yr study. Pregnant and lactating crossbred beef cows (n = 408), mainly of Angus and Hereford breeding, were stratified by age and by sex and BW of their calves and assigned randomly into 2 treatments: weaning at approximately 180 d (early weaning) and normal weaning 45 d later (control). Cows were managed together on native range pastures and supplemented with harvested forage during the winter months. Cow BW, BCS, rib fat, and rump fat were measured periodically from early weaning through the next breeding. Reproductive performance was evaluated by calving intervals (CI), days from initiation of breeding to calving (BCI), retention in the herd, and adjusted 205-d weaning BW of the subsequent calf. Early weaned cows had greater (P < 0.001) BW at normal weaning than control cows, but the overall pattern of cow BW did not differ (P > 0.05) among treatments. Peak and nadir BCS occurred at precalving and postcalving periods, respectively and were greater (P < 0.001) at each period in early weaned than in control cows and in cows > or =5-yr-old than in younger cows. Patterns for rib fat and rump fat were nearly identical to those of BCS except for the 3-way interaction (P < 0.001) of treatment, age, and period on rump fat. Mean CI (372.4 +/- 2.1 d) and BCI (299.7 +/- 1.9 d) were not affected (P = 0.42) by treatment but varied (P < 0.001) with age of the cow. Age of cow accounted for 16% of total variation in CI and 12% of total variation in gestation length (P < 0.001). The intervals were longer (P < 0.001) in primiparous cows than in older cows. Early weaning decreased risk of culling in cows and thereby increased (P < 0.05) overall persistence by 11% over control cows. Earlier weaning of cows in the previous year increased (P < 0.001) weaning weight of the subsequent calf by 8.6 kg per cow per yr. Shifting weaning time increased storage of consumed energy as evidenced by increased rump fat, for use later during high-energy demand, ultimately improving overall productivity of the cow-calf system.

Angus-crossbred steers (n = 216) were used in a 3-yr study to assess the effects of winter stocker growth rate and finishing system on finishing performance and carcass characteristics. During winter months (December to April) steers were randomly allotted to 3 stocker growth rates: low (0.23 kg x d(-1)), medium (0.45 kg x d(-1)), or high (0.68 kg x d(-1)). Upon completion of the winter phase, steers were randomly allotted within each stocker treatment to a corn silage-concentrate or pasture finishing system. All steers regardless of finishing treatment were finished to an equal-time endpoint to eliminate confounding of treatments with animal age or seasonal factors. Upon completion of the finishing period, steers were slaughtered in 2 groups (one-half of pasture and one-half of feedlot cattle each time) and carcass data were collected. Winter data were analyzed as a completely randomized design, with winter treatment, pen replicate, year, and the winter x year interaction in the model. Finishing performance and carcass data were analyzed in a split-plot design with finishing system in the whole plot, and winter growth rate and winter x finish in the split-plot. Winter treatment mean within finishing replication was the experimental unit, and year was considered a random effect. Winter stocker phase treatments resulted in differences (P < 0.001) in final BW, ADG, and ultrasound LM area between all treatments for that phase. Pasture-finished cattle had lower (P < 0.001) final BW, ADG, HCW, LM area, fat thickness, KPH, dressing percent, USDA yield grade, and USDA quality grade. Winter stocker treatment influenced (P < 0.05) final BW and HCW, with low and medium being less than high. Steers with low stocker gain had greater (P < 0.05) finishing ADG. Dressing percent was greater (P < 0.001) for high than low, and USDA quality grade was greater (P < 0.05) for high than low and medium. Carcass LM area, fat thickness, KPH, and USDA yield grade were not influenced (P > 0.05) by winter rate of gain. Cattle on low during winter exhibited compensatory gain during finishing but were unable to catch the high group regarding BW or HCW. The USDA quality grade was greater for high than low or medium. Animal performance during the winter stocker period clearly impacts finishing performance, carcass quality and beef production in both pasture- and feedlot-finishing systems, when cattle were finished to an equal-time endpoint.

The hypothesis of this experiment was that increasing dietary fat through the use of whole oilseeds and altering the dietary ratio of PUFA:saturated fatty acids would alter carcass composition of finishing steers. Seventy-two steers (443.6 +/- 1.0 kg) were fed for 76 d one of four dietary treatments: a corn/ soybean meal-based diet (NOFAT); two diets containing 16% (DM basis) whole raw soybeans; and a corn/soybean meal-based diet containing choice white grease (CWG) equal to the fat addition supplied by the soybeans. Soybeans used in the diets were either a standard variety (NORM-SB) or a variety high in oleic acid content (HO-SB). The fatty acid profile of diets differed (P < 0.05) in the degree of saturation and content of palmitic, stearic, oleic, linoleic, and linolenic acids. There were no differences in ADG (1.73 kg/d), hot carcass weight (347 kg), longissimus muscle area (79.4 cm2), yield grade (3.31), or percentage of boneless retail cuts (48.8%). Contrasts revealed differences (P < 0.05) in G:F and marbling score with the addition of fat (0.126 vs. 0.137 and 4.66 vs. 4.91, respectively, for NOFAT vs. fat). The addition of fat tended (P < 0.10) to increase backfat, and feeding NORM-SB increased (P < 0.01) dressing percent compared with the HO-SB treatment. Loin samples taken from steers fed NOFAT, NORM-SB, and HO-SB did not differ in alpha-tocopherol content. Loins from the CWG treatment tended (P < 0.10) to have lower alpha-tocopherol content than did the soybean treatments (0.79 vs. 0.99 ppm, respectively). From main-effects analysis, HO-SB loin samples had the highest (F3,8 = 32.91; P < 0.01) concentration of gamma-tocopherol (0.33 ppm); this resulted in differences (P < 0.05) in gamma-tocopherol when comparing all contrasts. When comparing loin samples from NORM-SB-fed steers with those from HO-SB-fed steers, NORM-SB samples had a greater (P < 0.05) percentage oflinoleic acid and PUFA and a lower (P < 0.05) percentage of oleic acid and monounsaturated fatty acids. Furthermore, loin samples from soybean-fed steers tended (P < 0.10) to have a greater concentration of conjugated linoleic acid than samples from CWG-fed steers. These data suggest that the source of added dietary fat may affect overall carcass composition. Furthermore, dietary addition of soybeans or CWG can improve feed efficiency and marbling, whereas the addition of whole raw soybeans compared with CWG may increase unsaturation and total vitamin E content of beef.

Two experiments were conducted to determine the efficacy of whole raw soybeans as a partial or whole replacement for soybean meal in a corn/soybean meal-based feedlot diet. In Exp. 1, 80 crossbred steers (average BW = 441.3 kg) and, in Exp. 2, 96 Angus-sired steers (average BW = 413.7 kg) were blocked by weight and assigned randomly to one of four dietary treatments. Treatments were 0, 8, 16, and 24% dietary inclusions of whole raw soybeans. Diets within experiments were isonitrogenous. Across experiments, diets were similar, differing only in amount of corn silage (8 vs. 15% DM) at the expense of whole, shelled corn for Exp. 1 and Exp. 2, respectively. No treatment differences were observed for ADG or final BW. Dry matter intake from d 0 to d 58 decreased linearly (P < 0.05) with increased inclusion of whole raw soybeans in Exp. 1, with no effect on feed efficiency. In Exp.2 from d 0 to 72, whole raw soybean inclusion had no effect on DMI or feed efficiency. There tended (P < 0.10) to be a linear reduction in hot carcass weight when whole raw soybeans were included in Exp. 1. Unexpectedly, longissimus muscle area tended (P < 0.10) to respond quadratically (P < 0.10) to the increased inclusion of whole raw soybeans in Exp.1. No differences were detected in marbling score, 10th-rib backfat, or yield grade for Exp. 1 and 2 steers. In Exp. 2, inclusion of whole raw soybeans had no effect on hot carcass weight or longissimus muscle area. Incrementally increasing the inclusion of whole raw soybeans in the diet of feedlot steers had little overall effect on weight gain, feed efficiency, or carcass quality in Exp. 1 and 2. There were subtle differences in the treatment responses observed for hot carcass weight and longissimus muscle area between Exp. 1 and Exp. 2 for the 24% inclusion level. These noted differences may indicate that inclusion levels above 24% might not be beneficial.

Anisotropic particles suspended in a nematic liquid crystal disturb the alignment of the liquid crystal molecules and experience small forces that depend on the particles' orientation. We have measured these forces using magnetic nanowires. The torque on a wire and its orientation-dependent repulsion from a flat surface are quantitatively consistent with theoretical predictions based on the elastic properties of the liquid crystal. These forces can also be used to manipulate submicrometer-scale particles. We show that controlled spatial variations in the liquid crystal's alignment convert the torque on a wire to a translational force that levitates the wire to a specified height.

The effects of pH, acetate, propionate, or butyrate concentration, and diet on acid resistance of fecal Escherichia coli and E. coli O157:H7 were determined by in vitro and in vivo experiments. The pH tested was from 4.0 to 8.0, and the VFA concentrations tested were 0 to 100 mM. The E. coli O157:H7 used was strain 505B. In an in vivo study, cattle were fed a grain-based diet, then either not switched or switched to a grain-based diet with 3% added calcium carbonate or two fiber-based diets (soybean hulls or hay). Acid resistance was expressed as viability after acid-shock at pH 2.0 for 1 h and 4 h for fecal E. coli and E. coli O157:H7, respectively. Enumeration methods used were multitube fermentation, agar plate, and petri-film methods. The E. coli O157:H7 was not found in continuous culture inocula or in vivo samples. The viability of fecal E. coli decreased linearly (P < 0.01) as the culture pH increased, and viability of E. coli O157:H7 was highest (P < 0.01) when cultivated at pH 6.0. The viability of fecal E. coli and E. coli O157:H7 showed quadratic responses (P < 0.05) as acetate and butyrate concentrations increased at pH 7.2, with maximal acid resistance at 20 and 12 mM, respectively. As propionate concentration increased, the acid resistance was not different (P > 0.05) for fecal E. coli. Acid resistance of E. coli was induced by acetate and butyrate, even though the environmental pH was near neutral. Similar results were measured in the in vivo study, where viability after acid shock was more dependent on VFA concentration than on pH. Increasing the dietary calcium carbonate concentration also increased (P < 0.05) acid resistance of fecal E. coli. Results from these studies demonstrated that culture pH and VFA affect acid resistance of E. coli.

Experimentation, with a single-phase continuous culture system operated at fractional dilution rates of 0.03 and 0.09 per hour and four cannulated crossbred steers (260 +/- 20 kg) used in a 4 x 4 Latin square design, was conducted to determine the level of ruminally degradable protein (RDP) that maximized microbial efficiency (MOEFF), microbial nitrogen flow, and nutrient digestibility in the rumen. Treatments consisted of increasing diet RDP levels (from 3.5 to 17.5% RDP on a DM basis). The basal diet was corn with casein or soybean meal used as the source of RDP in the continuous culture experiment and animal study, respectively. Dietary nonstructural carbohydrate (NSC) ranged from 52 to 75% on a dry matter basis. Urea was added to ensure an adequate ammonia nitrogen source for microbial growth. The RDP levels did not affect true digestibility of dry matter and organic matter, ruminal pH, particulate passage rate, or liquid passage rate in the animal study. As RDP increased, bacterial nitrogen production increased linearly (P < 0.05) only for the 0.09/h dilution rate. Microbial efficiency was not influenced by RDP level. Ruminal peptide and NH3 N concentration increased linearly (P < 0.05) as RDP increased. Based on this experimentation, 1.8 mM peptide maximized MOEFF when ammonia nitrogen was not limiting (> 2 mg/dL). We concluded from this research that the RDP requirement of NSC-fermenting bacteria was lower than that currently fed in many diets.

RESUMO

The delivery of healthcare relies on the sharing of patient information between those who are providing for the care of the patient and this information is increasingly being expressed in terms of a 'record'. Further, it is desirable that these records are available in electronic form as Electronic HealthCare Records. As it is likely that patient records or parts of records will be stored in many different information systems and in the form of disparate record architectures, uniform access to patient records would be problematic. This paper presents an overview of the Synapses computing environment in which a Federated Healthcare Record Server provides uniform access to patient information stored in connected heterogeneous autonomous information systems and other Synapses servers. The Synapses record architecture is based on the architecture proposed by the Technical Committee 251 of the European Committee for Standardisation and the interfaces to the Synapses server are specified in the ISO standard Interface Definition Language. Synapses is a pan-European project involving a number of hospitals, software companies, universities and research institutes and is partly funded by the EU Health Telematics Programme. The overview is described in terms of the Open Distributed Processing Reference Model.

RESUMO

The ability to exchange in a meaningful, secure, and simple fashion relevant healthcare data about patients is seen as vital in the context of efficient and cost-effective shared or team-based care. The electronic healthcare record (EHCR) lies at the heart of this information exchange, and it follows that there is an urgent need to address the ability to share EHCR's or parts of records between carers and across distributed health information systems. This paper presents the Synapses approach to sharing based on a standardized shared record, the Federated Healthcare Record, which is implemented in an open and flexible manner using the Common Object Request Broker Architecture (CORBA). The architecture of the Federated Healthcare Record is based on the architecture proposed by the Technical Committee 251 of the European Committee for Standardization.

A diverse terrestial biota inhabited polar latitudes during the Cretacous, 105 to 130 Ma (million years ago), along what is now the southeast coast of Australia This biota, from rocks in the Otway and Strzelecki groups, cnsisted of more than 150 taxa of vertebrates, invertebrates, and plants. Oxygen isotope ratios in diagenetic calcite suggest that mean annual temperatures were most likely less than 5 degrees C, and rings present in the fossil araucarian-podocarp-ginko woods indicate saonality. Southeastern Austalia, thus, seems to have had a cool, seasonal, nontropical climate. Dinosaurs that have been recovered are up to five species and three genera of hypsilophodontids, all of which were endemic, and three species of theropods. The occurrence of Allosaurus sp. and labyrinthodont amphibians, which had become extinct elsewhere in the Jurassic, indicate that isolation may have allowed extended surival of these taxa in Australia. In that dinosaurs coped with high latitude for at least 65 million years [Valaginian to Albian time in Australia and Campanian to Maastrictian time (80 to 65 Ma) in Alaska] suggests that cold and darkness may not have been prime factors bringing about the extinction of dinosaurs and some other groups at the Cretaceous-Tertiary boundary, unless they were prolonged.
