RESUMO

OBJECTIVE: To observe the effect of Nailifu Spray on the treatment of premature ejaculation. METHODS: A total of 90 patients were included in this study from January 1, 2022 to January 1, 2023. Nailifu spray was used to spray the surface of penile skin once a day, 2 sprays per session for 4 weeks.And the patients' premature ejaculation diagnostic tool (PEDT) scores, intravaginal ejaculation latency time (IELT), and international index of erectile function-5 (IIEF-5) scores were collected before and after treatment, respectively. RESUTS: The median (P25,P75) PEDT scores was 16.0(15.0,18.0) scores before treatment and 10.0(10.0,10.0) scores after treatment. The median (P25,P75) of IELT was 20.0 (10.0,30.0) s before treatment and 240.0 (180.0,300.0) s after treatment. The median (P25,P75) of IIEF-5 scores was 21.0 (21.0,22.0) scores before treatment and 21.0 (21.0,21.0) scores after treatment. Compared with baseline levels, IELT was significantly longer and PEDT scores were significantly lower, with statistically significant differences. No significant changes in IIEF-5 scores were seen. CONCLUSION: Nailifu spray treatment of premature ejaculation is accurate and effective, worthy of clinical promotion.

OBJECTIVE: Chronic nonbacterial prostatitis (CNP) has remained one of the most prevalent urological diseases, particularly in older men. Dihydroartemisinin (DHA) has been identified as a semi-synthetic derivative of artemisinin that exhibits broad protective effects. However, the role of DHA in inhibiting CNP inflammation and prostatic epithelial cell proliferation remains largely unknown. MATERIALS AND METHODS: CNP animal model was induced by carrageenan in C57BL/6 mouse. Enzyme linked immunosorbent assay (ELISA), Real-time quantitative polymerase chain reaction (RT-qPCR) and Western blot were used to examine inflammatory cytokines and proliferation genes expression. Immunofluorescence and immunochemistry staining were used to detect and E2F7 expression. Human prostatic epithelial cells (HPECs) and RWPE-1 was induced by lipopolysaccharide (LPS) to mimic CNP model in vitro. Cell proliferation was determined using MTS assay. RESULTS: DHA significantly alleviated the rough epithelium and inhibited multilamellar cell formation in the prostatic gland cavity and prostatic index induced by carrageenan. In addition, DHA decreased the expression of TNF-α and IL-6 inflammatory factors in prostatitis tissues and in LPS-induced epithelial cells. Upregulation of transcription factor E2F7, which expression was inhibited by DHA, was found in CNP tissues, human BPH tissues and LPS-induced epithelial cells inflammatory response. Mechanically, we found that depletion of E2F7 by shRNA inhibited epithelial cell proliferation and LPS-induced inflammation while DHA further enhance these effects. Furthermore, HIF1α was transcriptional regulated by E2F7 and involved in E2F7-inhibited CNP and cellular inflammatory response. Interestingly, we found that inhibition of HIF1α blocks E2F7-induced cell inflammatory response but does not obstruct E2F7-promoted cell growth. CONCLUSION: The results revealed that DHA inhibits the CNP and inflammation by blocking the E2F7/HIF1α pathway. Our findings provide new evidence for the mechanism of DHA and its key role in CNP, which may provide an alternative solution for the prevention and treatment of CNP.

Phytochemical investigation of ethanol extract from the flowers of Trollius chinensis Bunge resulted in the isolation of two new flavone C-glycosides (1-2), along with 10 known compounds (3-12). The structures of the new compounds were established as 6‴-(3-hydroxy-3-methylglutaroyl)-2″-O-ß-d-galactopyranosyl orientin (1) and 6‴-(3-hydroxy-3-methylglutaroyl)-2″-O-ß-d-galactopyranosyl vitexin (2) on the basis of various spectroscopic analysis (including different 1D and 2D NMR spectroscopies, high-resolution electrospray ionization mass spectrometry) and chemical evidences. Bioassay showed that eight flavonoids inhibited complement activation on the classic pathway in vitro, with their IC50 values ranging from 0.88 to 4.02 mM, which may contribute to the applications of the herb in treatment of acute respiratory distress syndrome, etc.

To explore structure-activity relationships with respect to light-harvesting behavior, a family of bis-cyclometalated iridium complexes [Ir(C^N)(2)(Hbpdc)] 2-5 (where C^N = 2-phenylbenzothiazole and its functionalized derivatives, and H(2)bpdc =2,2'-bipyridine-4,4'-dicarboxylate) was synthesized using a facile method. The photophysical and electrochemical properties of these complexes were investigated and compared to those of analogue 1 (C^N = (4-trifluoromethyl)-2-phenylbenzothiazole); they were also investigated theoretically using density functional theory. The molecular structures of complexes 2-4 were determined by X-ray crystallography, which revealed typical octahedral coordination geometry. The structural modifications involved in the complexes were accomplished through the attributes of electron-withdrawing CF(3) and electron-donating NMe(2) substituents. The UV-vis spectra of these species, except for that of 5, displayed a broad absorption in the low-energy region, which originated from metal-to-ligand charge-transfer transitions. These complexes were found to exhibit visible-light-induced hydrogen production and light-to-electricity conversion in photoelectrochemical cells. The yield of hydrogen production from water using these complexes was compared, which revealed substantial dependences on their structures, particularly on the substituent of the cyclometalated ligand. Among the systems, the highest turnover number of 1501 was achieved with complex 2, in which the electron-withdrawing CF(3) substituent was connected to a phenyl ring of the cyclometalated ligand. The carboxylate anchoring groups made the complexes highly suitable for grafting onto TiO(2) (P25) surfaces for efficient electron transfer and thus resulted in an enhancement of hydrogen evolution compared to the unattached homogeneous systems. In addition, the combined incorporation of the electron-donating NMe(2) group and the electron-withdrawing CF(3) substituent on the cyclometalated ligand caused complex 5 to not work well for hydrogen production. Their incorporation, however, enhanced the performance of 5 in the light-harvesting application in nanocrystalline TiO(2) dye-sensitized solar cells, which was attributed to the intense absorption in the visible region.

OBJECTIVE: To investigate the optimal condition for complete removal of the template molecules from vinblastine (VLB)-imprinted polymer. METHOD: The prepared polymers were packed into the cartridges of solid-phase extraction column and washed by methanol-glacial acetic acid mixture with different proportions. The contents and recoveries of VLB in the effluents were determined. RESULTS: Polymer extraction with methanol-glacial acetic acid (9:1, V/V) resulted in VLB recovery of 91.73%, but template bleeding was observed because of incomplete VLB removal. Using methanol-glacial acetic acid (6:4, V/V) as the extraction solvent, the recovery of VLB reached 98.03% with less solvents and extract times. The polymers could selectively adsorb VLB through non-covalent interactions and still exhibited strong affinity for the template molecule but not for the structural analogue vincristine after extraction with methanol-glacial acetic acid (6:4, V/V). CONCLUSION: Methanol-glacial acetic acid (6:4, V/V) is an ideal extract solvent for complete template molecule removal from the polymers, and the processed polymers possess stable capacity of specific recognition and selectivity to the template.

OBJECTIVE: To optimize the polymerization conditions of vinblastine (VLB) imprinted polymer. METHODS: The conditions were optimized by the method of uniform design. The major factors investigated included the amount of functional monomer (MAA) and the cross-linker (EDMA) and the progenic solvent (toluene or acetonitril). The adsorption rate of VLB on the solid-phase extraction (SPE) column packed with MIP was adopted as the response value. RESULTS: The optimal conditions were MAA 0.4 mmol, EDMA 1.6 mmol and using acetonitrile as the solvent. Under the conditions,the VLB imprinted polymer was synthesized and the absorption rate of VLB was 88.20%. The characterizations of the optimal MIP were determined by IR spectrometry and scanning electron microscope (SEM) analysis. CONCLUSION: It is possible to furtherly improve the nature of the polymer by optimizing the polymerization parameters with uniform design. The polymer synthesized under the optimal conditions exhibited high affinity to the target molecule VLB.
