RESUMO

Immune checkpoint blockade therapy (ICB) is ineffective against cold tumors and, although it is effective against some hot tumors, drug resistance can occur. We have developed a Plasmodium immunotherapy (PI) that can overcome these shortcomings. However, the specific killing effect of PI on tumor cells is relatively weak. Radiotherapy (RT) is known to have strong specific lethality to tumor cells. Therefore, we hypothesized that PI combined with RT could produce synergistic antitumor effects. We tested our hypothesis using orthotopic and subcutaneous models of mouse glioma (GL261, a cold tumor) and a subcutaneous model of mouse non-small cell lung cancer (NSCLC, LLC, a hot tumor). Our results showed that, compared with each monotherapy, the combination therapy more significantly inhibited tumor growth and extended the life span of tumor-bearing mice. More importantly, the combination therapy could cure approximately 70 percent of glioma. By analyzing the immune profile of the tumor tissues, we found that the combination therapy was more effective in upregulating the perforin-expressing effector CD8+ T cells and downregulating the myeloid-derived suppressor cells (MDSCs), and was thus more effective in the treatment of cancer. The clinical transformation of PI combined with RT in the treatment of solid tumors, especially glioma, is worthy of expectation.

The effect of potassium ferrate/ultrasonic (K2FeO4/US) treatment on the physicochemical features of textile dyeing sludge was studied. The soluble chemical oxygen demand (SCOD), deoxyribonucleic acid (DNA), sludge volume index (SVI), sludge viscosity, capillary suction time (CST) and particle size were measured to understand the observed changes in the sludge physicochemical features. The results showed that the combined K2FeO4/US treatment presented great advantages for disrupting the sludge floc structure over K2FeO4 or ultrasonic treatments alone. The optimal parameters of sludge disintegration were found to be a K2FeO4 treatment time of 60 min, a K2FeO4 dosage of 0.5936 g/g SS, an ultrasonic time of 15 min and an ultrasonic intensity of 0.72 W/mL. The initial median diameter of the sludge particles was 15.24 µm, and this value decreased by 35.89%. The CST was initially 59.6 s and increased by 231%, whereas the SVI was 97.78 mL/g and decreased by 25.89%. Scanning electron microscope (SEM) images indicated that the sludge surface was irregular and loose with a large amount of channels or voids during K2FeO4/US treatment. K2FeO4/US treatment synergistically enhanced the sludge solubilization and reached 668.67 mg/L SCOD, which is 31.81% greater than the additive value obtained with K2FeO4 treatment alone (215.95 mg/L) or with ultrasonic treatment alone (240 mg/L).

Sawdust was used as a filter aid for the textile dyeing sludge dewatering in this study. Results showed that sawdust conditioning in conjunction with cationic polyacrylamide (CPAM) presented much better dewaterability than CPAM alone. The optimal sawdust and CPAM dosage for the best dewaterability was found to be 60 wt.% (mass percent) and 15 kg/t DS (dry solid), the time to filter (TTF) and the yield at 90% degree of the filtration completion (YN90) were 5s and 15.6 kg/m(2)h under the conditions, respectively. TTF and YN90 were more appropriate parameters than specific resistance to filtration (SRF) for assessing the sludge dewaterability as affected by physical conditioners. The moisture content of various filter cake layers gradually became the same with the increase of sawdust dosage. The flocculated sludge cake became relatively incompressible after sawdust conditioning. Sawdust acts to maintain the permeability during the compressed filtration by resisting cake compression.
