OBJECTIVES: The study aimed to evaluate the role of Streptococcus mutans (S. mutans) dexA gene on biofilm structure and microecological distribution in multispecies biofilms. MATERIALS AND METHODS: A multispecies biofilm model consisting of S. mutans and its dexA mutants, Streptococcus gordonii (S. gordonii) and Streptococcus sanguinis (S. sanguinis) was constructed, and bacterial growth, biofilm architecture and microbiota composition were determined to study the effect of the S. mutans dexA on multispecies biofilms. RESULTS: Our results showed that either deletion or overexpression of S. mutans dexA had no effect on the planktonic growth of bacterium, while S. mutans dominated in the multispecies biofilms to form cariogenic biofilms. Furthermore, we revealed that the SmudexA+ group showed structural abnormality in the form of more fractures and blank areas. The morphology of the SmudexA group was sparser and more porous, with reduced and less agglomerated exopolysaccharides scaffold. Interestingly, the microbiota composition analysis provided new insights that the inhibition of S. gordonii and S. sanguinis was alleviated in the SmudexA group compared to the significantly suppressed condition in the other groups. CONCLUSION: In conclusion, deletion of S. mutans dexA gene re-modules biofilm structure and microbiota composition, thereby leading to decreased cariogenicity. Thus, the S. mutans dexA may be an important target for regulating the cariogenicity of dental plaque biofilms, expecting to be a probiotic for caries control.

RESUMO

Objective To construct recombinant lentiviral vector of microRNA-34a and observe the cell viability,cell cycle and apoptosis of hepatocellular carcinoma cells transfected with the vector system and treated with Doxorubicin.Methods Recombinant lentiviral vector containing microRNA-34a gene was constructed and transfected into 3 hepatocellular carcinoma cell lines,and cells were treated with Doxorubicin.The expression of microRNA-34a gene was detected by real-time PCR.The effect of microRNA-34a overexpression on hepatocellular carcinoma cells proliferation were quantified via MTT assay,cell cycle and apoptosis was evaluated by flow cytometry.Western blotting was used to evaluate the expression of cell cycle and apoptosis related protein.Results The successful construction of microRNA-34a recombinant lentiviral vector was confirmed by plasmid enzyme digestion and DNA sequencing.Compared with the control group,relative expression of microRNA-34a gene in hepatocellular carcinoma cells significantly increased ((HepG2:t=15.36,P<0.01;Hep3B:t=36.75,P<0.01;Bel-7402:t=24.17,P<0.01)).Cells viability decreased (HepG2:t =7.12,P < 0.01;Hep3B:t =8.89,P < 0.01;Bel-7402:t =13.62,P <0.01),G1 phase cells increased significantly(HepG2:F =137.65,P < 0.01;Hep3B:F =143.39,P <0.01;Bel-7402:F =1 306.47,P < 0.01) and cell apoptosis increased(HepG2:F =386.14,P < 0.01;Hep3B:F =881.94,P < 0.01;Bel-7402:F =885.89,P < 0.01).Conclusions MicroRNA-34a recombinant lentiviral vector (LV-hsa-mir-34a) transfected hepatocellular carcinoma cells overexpress microRNA-34a,reduce the malignant biological behavior.MicroRNA-34a recombinant lentiviral vector (LV-hsa-mir-34a) enhance the in vitro inhibitory effects of Doxorubicin on hepatocellular carcinoma cell lines.

RESUMO

Objective To analyze the urodynamic data of the patients with benign prostatic hyperplasia(BPH)and provide evidence for the therapy.Methods 452 patients with BPH were selected.The mean age was(65.6 ±5.65)years(51 to 89 years).All patients had intact history documents which included the results of cystoscopy,urodynamics,doppler,DRE and PSA.Results 15 cases were not diagnosed with BOO.430 cases were diagnosed with BOO,among them,65 cases of bladder detrusor muscle weakness,bladder detrusor instability(detrusor,DI,instability)in 365 cases,3 cases were normal,and the other 4 cases had detrusor atony.In 445 patients the mean maximum urinary flow rate(8.32±3.15)ml/s,the average residual urine volume(87.68±79.46)ml,the average maximum flow rate when the detrusor pressure(62.32±7.54)cm H2O,maximum urethral closure pressure was(86.43±18.35)cmH2O,among which 19 cases were not suitable for surgery.At the end of the 426 patients underwent surgical treatment,postoperative follow-up of 3 to 18 months,the International Prostate Symptom Score(IPSS)from 0 to 5,the quality of life index(QOL)score of 0 to 1,Qmax 11 to 21 ml /s,residual urine(45.6±36.2)ml.There were significant differences between the preoperative and postoperative indexes(P<0.05).Conclusion Urodynamic tests can avoid the blindness in the surgery treatment of BPH.

RESUMO

Objective:Give a method for tracing boundary of inner-medium membrane in the ultrasonic image of common carotid artery.Methods: Giving a cost function before the programme begins to search the pixel point set whose cost function have the minimum value. The searching process starts from an initial anchor point given by the precedent programme. The calculation of cost function on each pixel point column on both sides of the initial anchor point will not stop until the terminal conditions are fulfilled. The programme will record the coordinates and pixel values of all the points with the minimum cost function value obtained from the calculation.Results: It can trace the boundary of inner-medium membrane in the CCA image from a clinical exam and can export the processed image with highlighted area. There is a daemon that records the coordinates and pixel values of the point set.Conclusion: This method can trace the boundary of inner-medium membrane in the CCA ultrasonic image when the CCA image is distinct.

<p><b>OBJECTIVE</b>Research for establishing a risk monitoring theoretical model of high risk medical equipment at applying site.</p><p><b>METHODS</b>Regard the applying site as a system which contains some sub-systems. Every sub-system consists of some risk estimating indicators. After quantizing of each indicator, the quantized values are multiplied with corresponding weight and then the products are accumulated. Hence, the risk estimating value of each subsystem is attained. Follow the calculating method, the risk estimating values of each sub-system are multiplied with corresponding weights and then the product is accumulated. The cumulative sum is the status indicator of the high risk medical equipment at applying site. The status indicator reflects the applying risk of the medical equipment at applying site.</p><p><b>RESULT</b>Establish a risk monitoring theoretical model of high risk medical equipment at applying site.</p><p><b>CONCLUSION</b>The model can monitor the applying risk of high risk medical equipment at applying site dynamically and specially.</p>

RESUMO

Objective:To explore the mechanism of grape seed polyphenols (GSP) reverseing mutidrug resistance of GBC-SD cell lines.Methods:GBC-SD cell lines were used to determine the effect of GSP.MTT assay was adopted to evaluate the cytotoxity,RT-PCR was used to determine MDR1 mRNA,P-gp and cellular adriamycin was measured by flow cytometry.Results:IC50 of chemotherapeutic agents was reduced in GSP treated group (P0.05),GSP inhibited the expression of P-gh and MDR1 mRNA (P0.05) ,increased celluar accumulation of adriamycin (P0.05).Conclusions:GSP can reverse mutidrug resistance of GBC-SD cell lines,the potential mechanisms is that GSP decreases expression of P-gp、bcl-2 and MDR1 mRNA.

RESUMO

Objective To study the etiology,diagnosis and treatment of functional delayed gastric emptying(FDGE) resulting from pancreatoduodenectomy.Methods From June,1990~June,2003,136 patients received pancreatoduodenectomy,whose clinical data were retrospectively analyzed.Upper gastrointestinal(radiography) and endoscopy were the main methods of examination.Results Twenty-eight cases were(complicated) with FDGE in the 136 patients(20.6%) after operation.The occurrence of FDGE was(correlated) with hyperbilirubinemia,diabetes,duodenal obstruction,pancreatic fistula and abdominal infection.All patients were cured with conservative treatment.The recovery time of gastric motility was 14-42 days,average time was 28 days.Conclusions Hyperbilirubinemia,diabetes,duodenal obstruction,pancreatic(fistula) and abdominal infection were the main causes of FDGE.

Objective: To investigate effect of exogenous wild-type p53 gene on the cell growth and tumorigenicity of human gallbladder cancer cell lines. Methods: After identification of the genetic status of p53 gene of GBC-SD cell lines with the immunocytochemistry staining and the direct sequencing technique of PCR products, eukaryotic expressing plasmid pCMV-p53 was introduced by lipofectamine-mediated into GBC-SD cell lines. Growing transfected cells were selected by G418. The presence and expression of exogenous p53 gene was detected by PCR, RT-PCR and Western blot. The cellular proliferating ability was assessed using the cell growth curve and cloning assay. The xenograft in nude mice was performed to examine the effect of tumorigenicity. Results: P53 protein overexpression was showed in GBC-SD cell lines. A transversion of TAC→AAC at codon 126 of exon 5 was confirmed. PCR, RT-PCR and Western blot showed exogenous p53 gene had successfully transfected into GBC-SD cells and obtained high expression. The growth and proliferation of the cells were greatly decreased, and the tumorigenicity was significantly inhibited after transfection wtp53. Conclusion: The expression of exogenous wild-type p53 gene could effectively inhibit the growth of gallbladder cancer GBC-SD cells in vitro and in vivo.

RESUMO

Objective To explore an effective treatment for inflammatory hilar bile duct stricture. Methods The clinical data with patients of intra hepatic cholelithiasis complicated with hilar bile duct stricture that received plastic repair of hilar bile duct stricture(PHBDS) with pedicled cholecystic graft or Roux en Y cholangio jejunostomy (RYCJ) in our hospital during recent 10 years were retrospectively analyzed. The peri operative conditions, postoperative morbidity of cholangitis and recurrence of hepaticolithiasis were observed . Results The peri operative morbidity of biliary fistula and inflammatory ileus were not significantly different between the two groups. The postoperative morbidity rate of cholangitis was 5.66% and 21.88%,(P=0.010) and recurrence rate of hepaticolithiasis was 3.77% and 16.67%,(P=0.021) in PHBDS group and RYCJ group respectively . Conclusions PHBDS is an easy and safe operation. RHBDS can preserve the physiologic function of Oddi′s spincter, so the reflux of intestinal contents to bile duct is avoided , and the results are better than the RVCJ.
