This study aimed to evaluate the effects of different nutritional plans on the productive, physiological and metabolic parameters of F1 ½ Holstein x ½ Zebu cows in different stages of lactation. Sixty lactating cows were allotted to a completely randomized 5 x 3 factorial design with five feed allowances and three lactation periods. The dry matter intake, milk yield and heart rate were reduced by 5.69kg, 2.41kg and 10.36 beats/min (morning) and 10.25 beats/min (afternoon) for each 1% feed restriction, respectively. There was no difference in the concentration of glucose, total protein, albumin, cholesterol and non-esterified fatty acids for cows subjected to different feed allowances, with means of 95.25, 7.98, 2.95, 121.68 and 0.45mg/dL, respectively. Feed restriction of up to 2.50% BW is a cost reduction strategy that does not alter milk yield, regardless of the stage of lactation.(AU)


Objetivou-se avaliar os efeitos de diferentes planos nutricionais sobre as características produtivas, fisiológicas e metabólicas de vacas F1 ½ Holandês x ½ Zebu. Foram utilizadas 60 vacas em lactação, seguindo-se o delineamento inteiramente ao acaso, em esquema fatorial 5 x 3, com cinco níveis de oferta de dieta e três períodos de lactação. À medida que se aumentou 1% na restrição da oferta da dieta, houve redução linear de 5,69kg no consumo de matéria seca pelos animais, 2,41kg na produção de leite, bem como de 10,36bat/min (manhã) e 10,25 bat/min (tarde) na frequência cardíaca dos animais. Não houve diferença para a concentração de glicose, proteínas totais, albumina, colesterol e NEFA com a restrição na oferta da dieta dos animais, sendo a média de 95,25, 7,98, 2,95, 121,68 e 0,45mg/dL, respectivamente. Recomenda-se a restrição de até 2,50% de peso corporal como estratégia de redução dos custos em todos os estágios em lactação, visando não alterar, economicamente, a produção de leite.(AU)

Foot-and-mouth disease (FMD) is a major constraint to transboundary trade in animal products, yet much of its natural ecology and epidemiology in endemic regions is still poorly understood. To address this gap, a multidisciplinary, molecular and conventional epidemiological approach was applied to an investigation of endemic FMD in Vietnam. Within the study space, it was found that 22.3% of sampled ruminants had previously been infected with FMD virus (FMDV), of which 10.8% were persistent, asymptomatic carriers (2.4% of the total population). Descriptive data collected from targeted surveillance and a farm questionnaire showed a significantly lower prevalence of FMDV infection for dairy farms. In contrast, farms of intermediate size and/or history of infection in 2010 were at increased risk of FMD exposure. At the individual animal level, buffalo had the highest exposure risk (over cattle), and there was spatial heterogeneity in exposure risk at the commune level. Conversely, carrier prevalence was higher for beef cattle, suggesting lower susceptibility of buffalo to persistent FMDV infection. To characterize virus strains currently circulating in Vietnam, partial FMDV genomic (VP1) sequences from carrier animals collected between 2012 and 2013 (N = 27) and from FMDV outbreaks between 2009 and 2013 (N = 79) were compared by phylogenetic analysis. Sequence analysis suggested that within the study period, there were two apparent novel introductions of serotype A viruses and that the dominant lineage of serotype O in Vietnam shifted from SEA/Mya-98 to ME-SA/PanAsia. FMDV strains shared close ancestors with FMDV from other South-East Asian countries indicating substantial transboundary movement of the predominant circulating strains. Close genetic relationships were observed between carrier and outbreak viruses, which may suggest that asymptomatic carriers of FMDV contribute to regional disease persistence. Multiple viral sequences obtained from carrier cattle over a 1-year period had considerable within-animal genetic variation, indicating within-host virus evolution.

Three experiments were conducted to evaluate the effect of ß-mannanase BM: supplementation on the performance, metabolizable energy, amino acid digestibility, and immune function of broilers. A total of 1,600 broilers were randomly distributed in a 4 × 2 factorial arrangement (4 nutritional levels × 0 or 500 g/ton BM), with 10 replicates and 20 broilers per pen. The same design was used in the energy and digestibility experiments with 8 and 6 replicates, respectively, and 6 broilers per pen. The nutritional levels : NL : were formulated to meet the nutritional requirements of broilers : NL1 : ; reductions of 100 kcal metabolizable energy : NL2 : ; 3% of the total amino acids (NL3); and 100 kcal metabolizable energy and 3% total amino acids (NL4) from NL1. The serum immunoglobulin (Ig) concentration was determined in two broilers per pen, and these broilers were slaughtered to determine the relative weight of spleen, thymus, and bursa of Fabricius. Throughout the experiment, the lower nutritional levels reduced (P < 0.05) body weight gain : BWG : and increased (P < 0.05) feed conversion : FCR : for the NL4 treatment. The BM increased (P < 0.05) the BWG values and improved (P < 0.05) the FCR of the broilers. The apparent metabolizable energy corrected for nitrogen balance (AMEn) values were reduced (P < 0.05) for NL2 and NL3. The BM increased (P < 0.05) the AMEn values and reduced (P < 0.05) the excreted nitrogen. NL3 and NL4 reduced (P < 0.05) the true ileal digestibility coefficients (TIDc) of the amino acids cystine and glycine, and BM increased (P < 0.05) the TIDc for all amino acids. The addition of BM reduced (P < 0.05) the relative weights of the spleen and bursa. NL2 increased (P < 0.05) the Ig values, whereas BM reduced (P < 0.05) the serum IgA, IgG, and IgM values of the broilers. This study indicates that using suboptimal nutrient levels leads to losses in production parameters, whereas BM-supplemented diets were effective in improving performance, energy values, and TIDc levels of amino acids and immune response of broilers.

The objective of this study was to evaluate the quality of pellets from the end of the pelletizer machine until the end of the line feed plate, using two methods of analysis: pellet durability index (PDI) and EMBRAPA pelletizing method (MEP). Furthermore, the effect of the discharging methods at the broiler farm on the physical quality of pellets using MEP as a method of analysis was evaluated. The first phase of the experiment was a randomized block with six treatments: A - pellets at the end of the pelletizer machine; B - pellets inside the truck; C - truck's discharging at the feed silo; D - place inside the warehouse after the feed silo; E - the middle of the feed plate line and F - the end of the feed plate line. The experiment was carried out in seven replicates (blocks). The second phase of the experiment was a randomized block with two treatments: A - smaller discharge opening (for pellets feed); B - bigger discharge opening (for mashed feed). The experiment was performed with twelve replicates (blocks). The discharge of the pellets feed at broilers farm is the point that worsens the pellets quality. The discharge of pellets feed by the truck should be done with the bigger discharge opening. Moreover, MEP showed to be a better method than PDI to evaluate the integrity of the pellets for broiler chickens.(AU)

A challenging aspect of ASFV control in wild boar populations is the design and implementation of effective surveillance and monitoring programmes, both for early warning, and to determine the ongoing epidemiological situation in an infected population. Testing blood samples requires invasive sampling strategies like hunting or capture of wild boar. Besides being biased towards healthy animals, such strategies are also linked to further spread of the virus. Non-invasive sampling strategies would increase the reliability of surveillance of ASFV in wild boar populations, without the negative side effects. This study evaluates the potential of faeces and tissue samples as a basis for non-invasive sampling strategies for ASFV in wild boar. In the acute phase (0-21 days after infection), in comparison with virus detection in blood, virus can be detected in faeces 50-80% of the time. This percentage decreases to below 10% for the subacute/chronic phase. ASFV DNA is quite stable in faeces. Half-lives range from more than 2 years at temperature up to 12°C, to roughly 15 days at temperatures of 30°C. In tissue samples, stored at 20°C, half-lives mostly range from 1.7 to 7.4 days. The sample of preference is the spleen, where the highest titres and highest half-life of ASFV DNA are observed. The level and duration of excretion of ASFV in the faeces, combined with the stability of the DNA, suggest that sampling of faeces could be the basis for a non-invasive sampling strategy to monitor ASFV in wild boar.

Infection caused by canine distemper virus (CDV) is a highly contagious disease with high incidence and lethality in the canine population. Antiviral activity of flavonoids quercetin, morin, rutin and hesperidin, and phenolic cinnamic, trans-cinnamic and ferulic acids were evaluated in vitro against the CDV using the time of addition assay to determine which step of the viral replicative cycle was affected. All flavonoids displayed great viral inhibition when they were added at the times 0 (adsorption) and 1h (penetration) of the viral replicative cycle. Both quercetin and hesperidin presented antiviral activity at the time 2h (intracellular). In the other hand, cinnamic acid showed antiviral activity at the times 0 and 2h while trans-cinnamic acid showed antiviral effect at the times -1h (pre-treatment) and 0 h. Ferulic acid inhibited CDV replicative cycle at the times 0 and 1h. Our study revealed promising candidates to be considered in the treatment of CDV. Structural differences among compounds and correlation to their antiviral activity were also explored. Our analysis suggest that these compounds could be useful in order to design new antiviral drugs against CDV as well as other viruses of great meaning in veterinary medicine.

African swine fever (ASF) is a highly lethal, viral disease of swine. No vaccine is available, so controlling an ASF outbreak is highly dependent on zoosanitary measures, such as stamping out infected herds and quarantining of affected areas. Information on ASF transmission parameters could allow for more efficient application of outbreak control measures. Three transmission experiments were carried out to estimate the transmission parameters of two ASF virus isolates: Malta'78 (in two doses) and Netherlands'86. Different criteria were used for onset of infectiousness of infected pigs and moment of infection of contact pigs. The transmission rate (ß), estimated by a Generalized Linear Model, ranged from 0.45 to 3.63 per day. For the infectious period, a minimum as well as a maximum infectious period was determined, to account for uncertainties regarding infectiousness of persistently infected pigs. While the minimum infectious period ranged from 6 to 7 days, the average maximum infectious period ranged from approximately 20 to nearly 40 days. Estimates of the reproduction ratio (R) for the first generation of transmission ranged from 4.9 to 24.2 for the minimum infectious period and from 9.8 to 66.3 for the maximum infectious period, depending on the isolate. A first approximation of the basic reproduction ratio (R0) resulted in an estimate of 18.0 (6.90-46.9) for the Malta'78 isolate. This is the first R0 estimate of an ASFV isolate under experimental conditions. The estimates of the transmission parameters provide a quantitative insight into ASFV epidemiology and can be used for the design and evaluation of more efficient control measures.

Knowledge on African Swine Fever (ASF) transmission routes can be useful when designing control measures against the spread of ASF virus (ASFV). Few studies have focused on the airborne transmission route, and until now no data has been available on quantities of ASF virus (ASFV) in the air. Our aim was to validate an air sampling technique for ASF virus (ASFV) that could be used to detect and quantify virus excreted in the air after experimental infection of pigs. In an animal experiment with the Brazil'78, the Malta'78 and Netherlands'86 isolates, air samples were collected at several time points. For validation of the air sampling technique, ASFV was aerosolised in an isolator, and air samples were obtained using the MD8 air scan device, which was shown to be suitable to detect ASFV. The half-life of ASFV in the air was on average 19 min when analysed by PCR, and on average 14 min when analysed by virus titration. In rooms with infected pigs, viral DNA with titres up to 10(3.2) median tissue culture infective dose equivalents (TCID50eq.)/m(3) could be detected in air samples from day 4 post-inoculation (dpi 4) until the end of the experiments, at dpi 70. In conclusion, this study shows that pigs infected with ASFV will excrete virus in the air, particularly during acute disease. This study provides the first available parameters to model airborne transmission of ASFV.

Avaliou-se o efeito do flavonoide quercetina na replicação do parvovírus canino in vitro por meio do ensaio de determinação da atividade virucida (ensaio 1), ensaio de determinação da atividade sobre a célula (ensaio 2) e ensaio de tempo de adição das drogas em diferentes etapas do ciclo replicativo viral (ensaio 3). A quercetina apresentou significante atividade antiviral, com valores máximos de redução do título viral de 96,3% no ensaio 1, 90% no ensaio 2 e 90% no ensaio 3. Os efeitos mais expressivos ocorreram nas etapas de adsorção e penetração viral. Os resultados deste trabalho sugerem a importância da quercetina para a medicina veterinária.


The in vitro effect of the flavonoid quercetin against canine parvovirus was evaluated. The antiviral activity of quercetin was evaluated by determining the virucidal activity (assay 1), determining the activity on the cell (assay 2) and using the time of addition assay to test the inhibition of the viral replication cycle (assay 3). Quercetin showed a significant antiviral activity, with maximum viral titer reduction of 96.3% in assay 1, 90% in assay 2 and 90% in assay 3. The most expressive effects occurred in the stages of viral adsorption and penetration. The results show the importance of quercetin for veterinary medicine.

The continuing circulation of African swine fever (ASF) in Russia and in the Trans-Caucasian countries has led to increased efforts in characterizing the epidemiology of ASF. For a better insight in epidemiology, quantitative data on virus excretion is required. Until now, excretion data has mainly focused on the initial stages of the disease. In our study we have studied ASF virus (ASFV) excretion dynamics in persistently infected animals. For this purpose, virus excretion through different routes was quantified over 70 days after infection. Three virus isolates of moderate virulence were used: the Brazil'78, the Malta'78 (a low and a high inoculation dose) and the Netherlands'86 isolate. For each isolate or dose, 10 animals were used. All (Brazil'78 group), or three animals per group were inoculated and the other animals served as contact animals. It was shown that dose (Malta'78 low or high) or infection route (inoculated or naturally infected) did not influence the ASFV excretion (p>0.05). Nasal, ocular and vaginal excretions showed the lowest ASFV titres. Virus was consistently present in the oropharyngeal swabs, showing two peaks, for up to 70 days. Virus was occasionally present in the faeces, occasionally with very high titres. Viral DNA persisted in blood for up to 70 days. The results presented in this study show that a high proportion of persistently infected animals shed virus into the environment for at least 70 days, representing a possible risk for transmission and that should be considered in future epidemiological analysis of ASF.

The vasodilator effect of the ethanolic extract of leaves from Hancornia speciosa Gomes (HSE) was evaluated in superior mesenteric artery rings. HSE produced a concentration-dependent vasodilation (IC50 = 10.8 +/- 4.0 microg/mL) in arterial rings pre-contracted with phenylephrine, which was completely abolished in endothelium-denuded vessels. Endothelium-dependent vasodilation induced by HSE was strongly reduced by L-NAME (100 microM), a nitric oxide (NO) synthase inhibitor, but neither by atropine, a muscarinic receptor antagonist (1 microM), nor by indomethacin (10 microM), a cyclooxygenase inhibitor. In rings pre-contracted with 80 mM KCl, the vasodilator effect of HSE was shifted to the right and was completely abolished in the presence of L-NAME (100 microM). Similar effects were obtained in mesenteric rings pre-contracted with phenylephrine in the presence of KCl 25 mM alone or in addition to 100 microM L-NAME. In addition, BaCl2 (1 mM) dramatically reduced the vasodilation induced by HSE. Together, these findings led us to conclude that HSE induces an endothelium-dependent vasodilation in rat mesenteric artery, by a mechanism dependent on NO, on the activation of potassium channels and endothelium-derived hyperpolarizing factor release. Rutin, identified as a major peak in the HPLC fingerprint obtained for HSE, might contribute for the observed vasodilator effect, since it was able to induce an endothelium-dependent vasodilation in rat superior mesenteric arteries.

The effects of LASSBio596, a phosphodiesterase type-4 and -5 inhibitor, were tested in Escherichia coli lipopolysaccharide (LPS)-induced acute lung injury. Twenty-four BALB/c mice were randomly divided into four groups. In the control group, saline (0.05 mL) was injected intratracheally (i.t.). The LPS group received LPS (10 microg i.t., 0.05 mL). In the LASSBio596 groups, LASSBio596 (10 mg x kg(-1), 0.2 mL) was injected intraperitoneally 1 h before or 6 h after LPS administration. After 24 h, in vivo (lung resistive and viscoelastic pressures, and static and dynamic elastances) and in vitro (tissue resistance, elastance and hysteresivity) pulmonary mechanics, lung morphometry and collagenous fibre content were computed. Neutrophils and tumour necrosis factor (TNF)-alpha levels were evaluated in the bronchoalveolar lavage fluid. LASSBio596 prevented the changes in lung mechanics, and inhibited neutrophilic recruitment, TNF-alpha release, bronchoconstriction, alveolar collapse and the increment of collagen fibre content induced by LPS, independently of the moment of injection. In conclusion, LASSBio596 modulated the lung inflammatory process and had the potential to block fibroproliferation. Thus, agents that inhibit phosphodiesterase 4 and 5 simultaneously may be a useful adjunct therapy for acute lung injury.

UNLABELLED: A simplified gravimetric method is proposed for fibrinogen estimation in plasma. Technic: 0.5 ml plasma (citrated) are diluted to up 5 ml with saline. A stick, 5 to 6 cm long, made of a glass capillary tube, or, aluminium foil, with ends bended as a hock, are weighted up to 0.001 g. Introduce one stick previously weighted in diluted plasma. Place two drops of thrombin (10 U/ml), and mix. In seconds, all fibrinogen is transformed in fibrin, deposited around the stick. Remove the stick with the fibrin, and press gently with the fingers, expelling the liquid in excess. Hang the stick with the fibrin, on the rim of a dry centrifuge tube. Centrifuge at 3.000 rpm for 10 minutes (dry centrifugation). Centrifugal forces acting on the fibrin clot expell all liquid particles. Fibrin consist of a very thin layer adhered on the stick. The weight difference (initial and final), represents the solid fibrin. CALCULATION: difference of weight added by 0.1 of the difference (anticoagulant dilution) x 200 to convert in dl. The method is highly reproductible, and takes about 20 minutes to be completed.
