RESUMO

An analysis of the amino acid sequence of influenza A/Alma-Ata/1417/84 (H1N1-Hsw1N1 serovariant) virus hemagglutinin heavy chain deduced from the nucleotide sequence of cloned full-size DNA complementary to the 4th segment of genome RNA was carried out. Unlike A/New Jersey/8/76 virus, the hemagglutinin of the virus under study was found to be more similar in the rate of HA1 homology, amino acid sequence of the signal peptide, antigenic sites Sa, Ca, and the receptor-binding site to human influenza viruses isolated in the 1930-1980-ies, in particular to influenza A/Taiwan/1/86 virus. It is assumed that an influenza virus more adapted to the human population like the strain A/Alma-Ata/1417/84 may be an etiological factor of a new influenza pandemic.

RESUMO

The full-length copy of the hemagglutinin gene RNA of the influenza virus A/Alma Ata/1417/84 (Hsw1 N1-serovariant) has been synthesized and cloned on Escherichia coli plasmid pBR327. The complete nucleotide sequence of the cloned DNA copy was determined by the Maxam-Gilbert procedure. The predicted amino acid, sequence of HA1 hemagglutinin subunit was compared with the sequences of HA1 subunits from other H1N1-subtype influenza virus strains. It has been found that the structure of the HA1-subunit of the studied strain is most similar to the structure of the identical region of the A/New Jersey/18/76 hemagglutinin.

The induction of the synthesis of interleukin-1 (IL-1) in human monocytes under the influence of the endotoxic preparations (LPS) and Y. pestis basis somatic antigen has been experimentally studied. The results obtained in this study make it possible to come to the conclusion that the capacity of the endotoxin of Y. pestis cell wall, consisting of LPS of type R, for inducing the synthesis of IL-1 in human monocytes is not different from the corresponding capacity of Salmonella and Shigella LPS, type S. Y. pestis O-specific polysaccharide in a discrete state has considerably greater IL-1-inducing activity in comparison with other preparations used in this experimental study. Such typical changes, characteristic of the initial stage of Y. pestis infection, as a sharp rise in temperature, transitory neutropenia, significant primary suppression of the oxygen-dependent metabolism of polymorphonuclear neutrophils are probably due to the induction of the synthesis of IL-1 by the polysaccharide-containing antigen of Y. pestis cell wall (LPS, basic somatic antigen) in cells of the mononuclear phagocytizing system.

The kinetic component composition of wheat embryo nuclear DNA was studied. It was shown that 32% of the genome consist of highly repetitive sequences. Intermediate repetitive sequences with repetitive frequency 1300 per genome constitute the bulk (52%) of the wheat embryo nuclear genome. The unique sequences constitute 12% of wheat embryo DNA. The individual families of intermediate repetitive and unique sequences were isolated; their reassociation kinetics were investigated and their kinetic complexity and repetition frequency were evaluated. Intermediate repetitive sequences 600-800 nucleotides in length were interspersed with unique sequences 800-1000 nucleotides long in the nuclear genome of wheat embryos. The linear relationship between the fragment length and the increasing amount of the zero-time binding DNA implies that 4% of the wheat embryos genome consists of palindromic sequences, which are clustered into groups.

RESUMO

A preparation of purified polyribosomes was isolated from wheat embryos germinating for 1 h with [3H]adenosine. Treatment of polyribosomes with pancreatic ribonuclease leads to the liberation of particles containing poly(A) sequences. Poly(A)-containing complexes show a heterogeneous distribution in the region 8--15 S. The ability of 8--15 S material to be absorbed to membrane filters under the conditions of low ionic strength and its sensitivity to pronase indicate that poly(A) sequences in polyribosomes are associated with protein. Analysis of this protein by electrophoresis in a polyacrylamide gel demonstrates the presence of two polypeptides with the molecular weights equal to 86,000 and 52,000.

RESUMO

A purified polyribosome preparation has been isolated from sprouting wheat embryos. After treatment of the polyribosomes with pancreatic RNase the poly-A-sequences are found in the fractions sedimenting in the region of 8--14 S. The ability of 8--14 S poly-A-containing complexes to bind to the membrane filters at high ionic strength and their sensitivity to pronase indicate that in sprouting wheat embryo polyribosomes the poly-A-sequences occur as poly-A-protein complexes.

RESUMO

Rapidly labelled mRNAs were isolated from informosomes and polyribosomes of imbibed wheat embryos. The distribution of poly(A) sequences in these fractions were studied by poly(U) Sepharose chromatography. It was shown that informosomes contain 11% polyadenylated mRNA while polyribosomes--38%. This fact suggests the important role of poly(A) sequences for translation of mRNA.
