RESUMO

The morbidity rates of generalized forms of meningococcal infection in persons immunized with different doses of liquid meningococcal vaccine ABC were compared. The vaccine was introduced subcutaneously by means of a jet injector. Altogether 3,920 males aged 18 years and older were immunized. 1,966 vaccinees received this vaccine in a dose of 200 micrograms and 1,954 vaccinees, in a dose of 400 micrograms. Meningococcal multicomponent vaccine ABC used in these doses showed moderate reactogenicity and did not prevent the development of generalized forms of meningococcal infection.

RESUMO

The comparative study of the composition of immune rabbit sera to N. meningitidis, as well as nonimmune sera, has been made by the methods of HPLC and radial immunodiffusion. The quantitative evaluation of the main serum proteins by the two methods has shown the coincidence of the results yielded by these methods. To study the total level of IgM and IgG in the sera under study, a simple and rapid HPLC technique is proposed. The study of the stability of sera during storage (at 4-6 degrees C) has revealed that immune sera show greater stability during storage under such conditions in comparison with sera obtained from nonimmune animals.

RESUMO

The capacity of heat-killed meningococci and the polysaccharide-protein-lipopolysaccharide fraction ( PPLF ) isolated from the microbial cell wall for changing nonspecific immunological reactivity was studied. In this investigation CBA mice with high response and C57BL/6 mice with low response to sheep red blood cells (SRBC) were used. Heat-killed N. meningitidis, serogroup A, and PPLF , serogroups A and B, were found to enhance and suppress humoral response to the heterologous antigen. The effect of modulation depended on the dose of the antigen, the serogroup of meningococci, the scheme of the experiment and the strain of mice. The immune response of the vaccinated animals to the heterologous antigen was characterized by the following stages: the state of the adjuvant effect was replaced by the state of temporary immunodeficiency and then by enhanced response to SRBC.

RESUMO

The study of meningococcal chemical polyvalent ABC-vaccine, manufactured by the Mechnikov Research Institute for Vaccines and Sera (Moscow), was carried out with the aim of finding the optimum vaccination doses for the immunization of children aged 0.5-15 years. In this study the effectiveness of the vaccine was evaluated by the determination of the titers of specific bactericidal antibodies in paired serum samples obtained before and 30 days after immunization. The analysis of the frequency of positive seroconversions and the levels of the geometric mean antibody titers allowed the authors to reveal the presence of a distinct relationship between the intensity of antibody production and the dose of the vaccine under test, used for immunization, as well as to establish that pronounced immune response to the optimum dose developed in 95-100% of the vaccinees. Taking into account the results obtained in the earlier studies of the safety and reactogenicity of the preparation (see Communication I), the optimum vaccination doses for the immunization of children of different age groups were established. On the basis of the statistical analysis of the characteristics of immune response to different doses of the preparation the following doses were chosen as optimum: 80 micrograms for children aged 0.5-2 years, 100 micrograms for children aged 3-7 years, 120 micrograms for children aged 8-11 years, 160 micrograms for children aged 12-15 years.

RESUMO

The development of protein insufficiency in children with surgical diseases is considered by the authors to be an important problem. Even good appetite does not always meet the protein requirements of the organism. An additional probe feeding is indicated for the reestalishment of passage along the intestine if the natural supply by food products fails to meet the needs of the organism.

The peptide composition and the molecular weight of peptides were determined in meningococci of serogroup A (7 strains), serogroup B (7 strains; of these, 4 strains belonged to serotype 2), serogroup C (4 strains) and serogroups D, X. Y and Z (1 strain each) by means of electrophoresis in polyacrylamide gel with sodium dodecyl sulfate. All serogroup A strains were found to contain 2 main peptides with molecular weights of 39,000 and 45,000 daltons; molecular weights of peptides in other serogroups varied in different strains. In group B and C meningococci, besides the reference strains of serotype 2, 4 more strains with peptide characteristics allowing to consider these strains belonging to serotype 2 were detected. The serotype-specific high-molecular protein preparation, purified to a considerable extent from lipopolysaccharide and hydrocarbons, was obtained from serogroup B strain, serotype 2. This preparation, when used for the immunization of rabbits, induced the formation of serotype-specific antibodies. The high-molecular protein preparation, serotype 2, was found to contain 5 individual antigenic components.

The immunogenicity of 2 meningococcal vaccines, multicomponent vaccine produced at the Mechnikov Research Institute for Vaccines and Sera in Moscow and polysaccharide vaccine obtained from Merck Sharp & Dohme (USA), was evaluated on experimental meningococcal sepsis in mice, produced by the injection of meningococcal culture in mucin suspension. The protective effect of these 2 vaccines, expressed in terms of ED50, was 0.28 +/- 0.12 for the multicomponent vaccine and 0.25 +/- 0.24 for the polysaccharide vaccine; the challenge dose used in the test was 10 LD50 of the culture. The multicomponent vaccine gave the maximum immunological effect in a dose of 8 micrograms, while higher or lower doses induced a lesser increase in antibody titer and thus gave lower protection to mice against infection.

The authors studied a possibility of obtaining experimental meningococcus sepsis model on mice. The use of cyclophosphane, iron compounds, yolk medium produced no significant organism. When 4--5% mucine was injected intraperitoneally together with meningococcus culture mice died with sepsis phenomena. Differences were revealed in the sensitivity of linear and mongrel mice to meningococcus infection--AKR mice proved to be more sensitive. At the same time it was found that mongrel mice weighing from 10 to 12 g could be used to induce meningococcus sepsis.
