RESUMO

Optogenetics has been a powerful scientific tool for two decades, yet its integration with non-human primate (NHP) electrophysiology has been limited due to several technical challenges. These include a lack of electrode arrays capable of supporting large-scale and long-term optical access, inaccessible viral vector delivery methods for transfection of large regions of cortex, a paucity of hardware designed for large-scale patterned cortical illumination, and inflexible designs for multi-modal experimentation. To address these gaps, we introduce a highly accessible platform integrating optogenetics and electrophysiology for behavioral and neural modulation with neurophysiological recording in NHPs. We employed this platform in two rhesus macaques and showcased its capability of optogenetically disrupting reaches, while simultaneously monitoring ongoing electrocorticography activity underlying the stimulation-induced behavioral changes. The platform exhibits long-term stability and functionality, thereby facilitating large-scale electrophysiology, optical imaging, and optogenetics over months, which is crucial for translationally relevant multi-modal studies of neurological and neuropsychiatric disorders.

RESUMO

Deciphering the function of neural circuits can help with the understanding of brain function and treating neurological disorders. Progress toward this goal relies on the development of chronically stable neural interfaces capable of recording and modulating neural circuits with high spatial and temporal precision across large areas of the brain. Advanced innovations in designing high-density neural interfaces for small animal models have enabled breakthrough discoveries in neuroscience research. Developing similar neurotechnology for larger animal models such as nonhuman primates (NHPs) is critical to gain significant insights for translation to humans, yet still it remains elusive due to the challenges in design, fabrication, and system-level integration of such devices. This review focuses on implantable surface neural interfaces with electrical and optical functionalities with emphasis on the required technological features to realize scalable multimodal and chronically stable implants to address the unique challenges associated with nonhuman primate studies.

Optogenetics is a powerful neuroscientific tool which allows neurons to be modulated by optical stimulation. Despite widespread optogenetic experimentation in small animal models, optogenetics in non-human primates (NHPs) remains a niche field, particularly at the large scales necessary for multi-regional neural research. We previously published a large-scale, chronic optogenetic cortical interface for NHPs which was successful but came with a number of limitations. In this work, we present an optimized interface which improves upon the stability and scale of our previous interface while using more easily replicable methods to increase our system's availability to the scientific community. Specifically, we (1) demonstrate the long-term (~3 months) optical access to the brain achievable using a commercially-available transparent artificial dura with embedded electrodes, (2) showcase large-scale optogenetic expression achievable with simplified (magnetic resonance-free) surgical techniques, and (3) effectively modulated the expressing areas at large scales (~1 cm2) by light emitting diode (LED) arrays assembled in-house.

BACKGROUND: Stair descent analysis has been typically limited to laboratory staircases of 4 or 5 steps. To date there has been no report of gait parameters during unconstrained stair descent outside of the laboratory, and few motion capture datasets are publicly available. RESEARCH QUESTION: We aim to collect a dataset and perform gait analysis for stair descent outside of the laboratory. We aim to measure basic kinematic and kinetic gait parameters and foot placement behavior. METHODS: We present a public stair descent dataset from 101 unimpaired participants aged 18-35 on an unconstrained 13-step staircase collected using wearable sensors. The dataset consists of kinematics (full-body joint angle and position), kinetics (plantar normal forces, acceleration), and foot placement for 30,609 steps. RESULTS: We report the lower limb joint angle ranges (30° and 8° for hip flexion and extension, 85° and -11° for knee flexion and extension, and 31° and 28° for ankle dorsi- and plantar-flexion). The self-selected speed was 0.79 ± 0.16 m/s, with cycle duration of 0.97 ± 0.18 s. Mean foot overhang as a percentage of foot length was 17.07 ± 6.66 %, and we calculate that foot size explains only 6% of heel placement variation, but 79% of toe placement variation. We also find a minor but significant asymmetry between left and right maximum hip flexion angle, though all other measured parameters were symmetrical. SIGNIFICANCE: This is the first quantitative observation of gait data from a large number (n = 101) of participants descending an unconstrained staircase outside of a laboratory. This study enables analysis of gait characteristics including self-selected walking speed and foot placement to better understand typical stair gait behavior. The dataset is a public resource for understanding typical stair descent.

Onsite wastewater disposal systems (OWDS) can introduce bacterial and chemical contaminants, via groundwater, into aquifers and adjacent waterways. We evaluated the concentration of fecal indicator bacteria (FIB) and antibiotic resistant bacteria (ARB) in the shallow groundwater of Eastern Long Island, New York, downgradient of OWDS using cultivation approaches and analysis of 16 S rRNA genes. While FIB and ARB were detected in 80% and 67% of groundwater samples, respectively, concentrations were low, suggesting that, at least at the time of sampling, groundwater was not a large-scale source of fecal bacterial contamination to adjacent embayments. ARB isolates did not include common fecal associated genera and the concentration of FIB and ARB did not correlate well with the concentration of pharmaceutical contaminants, suggesting that bacterial contaminants were poorly linked to OWDS discharge. Concentrations of FIB in the studied embayments were significantly greater in nearshore compared to mid-channel environments, suggesting that land-based sources are likely to be the major contributors of bacterial contamination.

Coastal onsite wastewater disposal systems (OWDS) were inundated by Hurricane Sandy's storm tide. This study compares the shallow groundwater quality (nutrients, pharmaceuticals, and hormones) downgradient of OWDS before and after Hurricane Sandy, where available, and establishes a baseline for wastewater influence on groundwater in coastal communities inundated by Hurricane Sandy. Nutrients and contaminants of emerging concern (CECs) were detected in shallow groundwater downgradient of OWDS in two settings along the New Jersey and New York coastlines: 1) a single, centralized OWDS in a park; and 2) multiple OWDS (cesspools) in low-density residential and mixed-use/medium density residential areas. The most frequently detected pharmaceuticals were lidocaine (40%), carbamazepine (36%), and fexofenadine, bupropion, desvenlafaxine, meprobamate, and tramadol (24-32%). Increases in the number and total concentration of pharmaceuticals after Hurricane Sandy may reflect other factors (seasonality, usage) besides inundation, and demonstrate the importance of analyzing for a wide variety of CECs in regional studies.

Bed sediment samples from 79 coastal New York and New Jersey, USA sites were analyzed for 75 compounds including wastewater associated contaminants, PAHs, and other organic compounds to assess the post-Hurricane Sandy distribution of organic contaminants among six regions. These results provide the first assessment of wastewater compounds, hormones, and PAHs in bed sediment for this region. Concentrations of most wastewater contaminants and PAHs were highest in the most developed region (Upper Harbor/Newark Bay, UHNB) and reflected the wastewater inputs to this area. Although the lack of pre-Hurricane Sandy data for most of these compounds make it impossible to assess the effect of the storm on wastewater contaminant concentrations, PAH concentrations in the UHNB region reflect pre-Hurricane Sandy conditions in this region. Lower hormone concentrations than predicted by the total organic carbon relation occurred in UHNB samples, suggesting that hormones are being degraded in the UHNB region.

Changes in bed sediment chemistry of Hempstead Bay (HB) have been evaluated in the wake of Hurricane Sandy, which resulted in the release of billions of liters of poorly-treated sewage into tributaries and channels throughout the bay. Surficial grab samples (top 5cm) collected before and (or) after Hurricane Sandy from sixteen sites in HB were analyzed for 74 wastewater tracers and steroid hormones, and total organic carbon. Data from pre- and post-storm comparisons of the most frequently detected wastewater tracers and ratios of steroid hormone and of polycyclic aromatic hydrocarbon concentrations indicate an increased sewage signal near outfalls and downstream of where raw sewage was discharged. Median concentration of wastewater tracers decreased after the storm at sites further from outfalls. Overall, changes in sediment quality probably resulted from a combination of additional sewage inputs, sediment redistribution, and stormwater runoff in the days to weeks following Hurricane Sandy.

The abundant iron sulfide mineral pyrite has been shown to catalytically produce hydrogen peroxide (H2O2) and hydroxyl radical (.OH) in slurries of oxygenated water. Understanding the formation and fate of these reactive oxygen species is important to biological and ecological systems as exposure can lead to deleterious health effects, but also environmental engineering during the optimization of remediation approaches for possible treatment of contaminated waste streams. This study presents the use of the amino acid phenylalanine (Phe) to monitor the kinetics of pyrite-induced .OH formation through rates of hydroxylation forming three isomers of tyrosine (Tyr) - ortho-, meta-, and para-Tyr. Results indicate that about 50% of the Phe loss results in Tyr formation, and that these products further react with .OH at rates comparable to Phe. The overall loss of Phe appeared to be pseudo first-order in [Phe] as a function of time, but for the first time it is shown that initial rates were much less than first-order as a function of initial substrate concentration, [Phe]o. These results can be rationalized by considering that the effective concentration of .OH in solution is lower at a higher level of reactant and that an increasing fraction of .OH is consumed by Phe-degradation products as a function of time. A simplified first-order model was created to describe Phe loss in pyrite slurries which incorporates the [Phe]o, a first-order dependence on pyrite surface area, the assumption that all Phe degradation products compete equally for the limited supply of highly reactive .OH, and a flux that is related to the release of H2O2 from the pyrite surface (a result of the incomplete reduction of oxygen at the pyrite surface). An empirically derived rate constant, Kpyr, was introduced to describe a variable .OH-reactivity for different batches of pyrite. Both the simplified first-order kinetic model, and a more detailed numerical simulation, yielded results that compare well to the observed kinetic data describing the effects of variations in concentrations of both initial Phe and pyrite. This work supports the use of Phe as a useful probe to assess the formation of .OH in the presence of pyrite, and its possible utility for similar applications with other minerals.

Cellular exposure to particulate matter with concomitant formation of reactive oxygen species (ROS) and oxidization of biomolecules may lead to negative health outcomes. Evaluating the particle-induced formation of ROS and the oxidation products from reaction of ROS with biomolecules is useful for gaining a mechanistic understanding of particle-induced oxidative stress. Aqueous suspensions of pyrite particles have been shown to form hydroxyl radicals and degrade nucleic acids. Reactions between pyrite-induced hydroxyl radicals and nucleic acid bases, however, remain to be determined. Here, we compared the oxidation of adenine by Fenton-generated (i.e., ferrous iron and hydrogen peroxide) hydroxyl radicals to adenine oxidation by hydroxyl radicals generated in pyrite aqueous suspensions. Results show that adenine oxidizes in the presence of pyrite (without the addition of hydrogen peroxide) and that the rate of oxidation is dependent on the pyrite loading. Adenine oxidation was prevented by addition of either catalase or ethanol to the pyrite/adenine suspensions, which implies that hydrogen peroxide and hydroxyl radicals are causing the adenine oxidation. The adenine oxidation products, 8-oxoadenine and 2-hydroxyadenine, were the same whether hydroxyl radicals were generated by Fenton or pyrite-initiated reactions. Although nucleic acid bases are unlikely to be directly exposed to pyrite particles, the formation of ROS in the vicinity of cells may lead to oxidative stress.
