RESUMO

The effect of a short-term creatine supplementation on hindlimb suspension (HS)-induced muscle atrophy was investigated. Creatine monohydrate (5 g/kg b.w. per day) or placebo, divided in 2 daily doses, was given by oral gavage for 5 days. Rats were maintained in HS with dietary supplementation concomitantly for 5 days. Body weight, soleus and EDL muscle masses, and cross-sectional areas (CSA) of the muscle fibers were measured. Signaling pathways associated with skeletal muscle mass regulation (FST, MSTN, FAK, IGF-1, MGF, Akt, mTOR, atrogin-1, and MuRF1 expressions, and Akt, S6, GSK3B, and 4EBP1 proteins) were evaluated in the muscles. Soleus muscle exhibited more atrophy than the EDL muscle due to HS. Creatine supplementation attenuated the decrease of wet weight and increased p-4EBP1 protein in the EDL muscle of HS rats. Also, creatine increased mTOR and atrogin-1 expressions in the same muscle and condition. In the absence of HS, creatine supplementation increased FAK and decreased MGF expressions in the EDL muscle. Creatine attenuated the increase in FST expression due to HS in the soleus muscle. MuRF1 expression increased in the soleus muscle due to creatine supplementation in HS animals whereas atrogin-1 expression increased still further in this group compared with untreated HS rats. In conclusion, short-term creatine supplementation changed protein metabolism signaling in soleus and EDL muscles. However, creatine supplementation only slightly attenuated the mass loss of both muscles and did not prevent the CSA reduction and muscle strength decrease induced by HS for 5 days.

The tumorigenesis of pituitary adenomas is poorly understood. Mutations of the PIK3CA proto-oncogene, which encodes the p110-α catalytic subunit of PI3K, have been reported in various types of human cancers regarding the role of the gene in cell proliferation and survival through activation of the PI3K/Akt signaling pathway. Only one Chinese study described somatic mutations and amplification of the PIK3CA gene in a large series of pituitary adenomas. The aim of the present study was to determine genetic alterations of PIK3CA in a second series that consisted of 33 pituitary adenomas of different subtypes diagnosed by immunohistochemistry: 6 adrenocorticotropic hormone-secreting microadenomas, 5 growth hormone-secreting macroadenomas, 7 prolactin-secreting macroadenomas, and 15 nonfunctioning macroadenomas. Direct sequencing of exons 9 and 20 assessed by qPCR was employed to investigate the presence of mutations and genomic amplification defined as a copy number ≥4. Previously identified PIK3CA mutations (exon 20) were detected in four cases (12.1%). Interestingly, the Chinese study reported mutations only in invasive tumors, while we found a PIK3CA mutation in one noninvasive corticotroph microadenoma. PIK3CA amplification was observed in 21.2% (7/33) of the cases. This study demonstrates the presence of somatic mutations and amplifications of the PIK3CA gene in a second series of pituitary adenomas, corroborating the previously described involvement of the PI3K/Akt signaling pathway in the tumorigenic process of this gland.

The reduction of hepatic microsomal transfer protein (MTP) activity results in fatty liver, worsening hepatic steatosis and fibrosis in chronic hepatitis C (CHC). The G allele of the MTP gene promoter, -493G/T, has been associated with lower transcriptional activity than the T allele. We investigated this association with metabolic and histological variables in patients with CHC. A total of 174 untreated patients with CHC were genotyped for MTP -493G/T by direct sequencing using PCR. All patients were negative for markers of Wilson’s disease, hemochromatosis and autoimmune diseases and had current and past daily alcohol intake lower than 100 g/week. The sample distribution was in Hardy-Weinberg equilibrium. Among subjects with genotype 1, 56.8 percent of the patients with fibrosis grade 3+4 presented at least one G allele versus 34.3 percent of the patients with fibrosis grade 1+2 (OR = 1.8; 95 percentCI = 1.3-2.3). Logistic regression analysis with steatosis as the dependent variable identified genotypes GG+GT as independent protective factors against steatosis (OR = 0.4, 95 percentCI = 0.2-0.8; P = 0.01). The results suggest that the presence of the G allele of MTP -493G/T associated with lower hepatic MTP expression protects against steatosis in our CHC patients.

BACKGROUND: Pituitary tumors account for approximately 10-15% of intracranial neoplasms. AIM: Using the cDNA microarray method, we have previously compared expression under two distinct conditions: a pool of 4 clinically non-functioning pituitary adenomas (NFPA) and a spinal cord metastasis of a non-functioning pituitary carcinoma, in order to gain biological insights into genomic changes of pituitary neoplasias. In the present study, we further investigated the mRNA expression of 3 selected genes previously described as being involved in other neoplasias based on a series of 60 pituitary adenomas: CRABP1 (cellular retinoic acid binding protein 1), GRP (gastrin-releasing peptide), and RERG (Ras-related, estrogen- regulated, growth inhibitor). MATERIAL AND METHODS: The expression of CRABP1, GRP, and RERG was determined by quantitative RT-PCR. RESULTS: A significantly higher content of CRABP1 mRNA was observed in NFPA compared to functioning adenomas, and PRL-secreting adenomas showed a lower expression of this gene compared to normal pituitary. A lower expression of GRP mRNA was detected in NFPA compared to normal pituitary and also to functioning adenomas. RERG mRNA was overexpressed in NFPA in comparison to functioning adenomas and to normal pituitary. Among the functioning adenomas, only the ACTH-secreting adenomas presented a higher expression of RERG mRNA compared to normal pituitary. CONCLUSIONS: The findings of differential expression of CRABP1 in prolactinomas and of RERG in NFPA compared to normal pituitary suggests that retinoic acid and estrogen receptor, respectively, could be involved in the tumorigenesis of these adenomas subtypes. Additional studies are required to further confirm this hypothesis.

The neurotensin (NT) produced in the hypothalamus and in pituitary gonadotrophs and thyrotrophs participates in neuroendocrine regulation. Recently, the involvement of this peptide in normal and neoplastic cell proliferation has been postulated. In the present study, we evaluated the expression of NT and its receptors (NTR1, 2 and 3) in a series of 50 pituitary adenomas [11 growth hormone (GH)-, eight prolactin (PRL)-, four adrenocorticotrophic hormone (ACTH)- and 27 nonfunctioning adenomas]. NT mRNA expression was significantly higher in functioning compared to nonfunctioning adenomas and with normal pituitary. Nonfunctioning pituitary adenomas showed lower expression of NT mRNA than normal pituitary. In the immunohistochemical study of functioning adenomas, NT was colocalised with GH, PRL and ACTH secreting cells. In nonfunctioning adenomas, the NT immunoreactivity intensity was variable among the samples. NTR3 mRNA expression was observed in all examined samples and was higher in the adenomas, both functioning and nonfunctioning, compared to normal pituitary. By contrast, NTR1 and NTR2 mRNA were not detected in either pituitary adenomas or normal tissue. The higher expression of NTR3, as well as the expression of NT by tumoural corticotrophs, lactotrophs and somatotrophs, which are cells types that do not express this peptide in the normal pituitary, suggests that NT autocrine and/or paracrine stimulation mediated by NTR3 may be a mechanism associated with the tumourigenesis of functioning adenomas.

We have calculated the shapes of flat liquid films, and of the transition region to the associated Plateau borders (PBs), by integrating the Laplace equation with a position-dependent surface tension γ(x), where 2x is the local film thickness. We discuss films in either zero or non-zero gravity, using standard γ(x) potentials for the interaction between the two bounding surfaces. We have investigated the effects of the film flatness, liquid underpressure, and gravity on the shape of films and their PBs. Films may exhibit 'humps' and/or 'dips' associated with inflection points and minima of the film thickness. Finally, we propose an asymptotic analytical solution for the film width profile.

RESUMO

Instabilities play a central role in the physics of foams. Some that change the topology of a dry foam are indicated by the laws promulgated by Plateau in his 1873 book. Their occurrence is less clearly predictable in wet foams. Various other instabilities are related to gravitational loading and gas compressibility. We gather up many examples as a guide to future research and identify problems that remain, including what we call instabilities, which occur before they are expected on the basis of Plateau's laws.

RESUMO

We show that if the triple line around a three-dimensional double bubble or lens bubble is decorated with another bubble or with a Plateau border, then the film prolongations into the decoration no longer meet at 2pi/3. These deviations can be accounted for in terms of a line tension that equals half the excess surface energy associated with the decoration.

We have experimentally studied the dissociation/coalescence of internal Plateau borders (PBs) in simple monolayer bubble clusters, as a result of changing the liquid fraction. At large liquid content, the clusters consist of n bubbles of the same size, symmetrically placed around an internal n-sided PB (n-PB). On decreasing the liquid fraction we observed symmetry-breaking transitions in the 4- and 5-bubble clusters (but not in the 3-bubble cluster), followed by dissociation of the PBs. We used the Surface Evolver to determine the various equilibrium configurations of the corresponding two-dimensional wet clusters and their surface energies. The sequence of 4-bubble cluster configurations observed on varying the liquid fraction correlates qualitatively with that predicted on the basis of Surface Evolver calculations. The same is not true of the 5-bubble cluster.

We have carried out a systematic study of buckling-like mechanical instabilities in simple two- (2D) and three-dimensional (3D) symmetric foam clusters sandwiched between parallel planar walls. These instabilities occur when the wall separation w is reduced below a critical value, w*, for which the foam surface energy E reaches its minimum, E*. The clusters under investigation consist of either a single bubble, or of "twin bubbles" of fixed equal sizes (areas A in 2D or volumes V in 3D), which are either free to slide or pinned at the confining walls. We have numerically obtained w* for both free and pinned 2D and 3D clusters. Furthermore, we have calculated the buckled configurations of 2D twin bubbles, either free or pinned, and of 3D free twin bubbles, whose energy is independent of w and equal to the minimum energy E* of the unbuckled state. Finally, we have also predicted the critical w(t)* at which the terminal configurations under extension of 2D and 3D single and twin bubbles are realised. Experimental illustrations of these transitions under compression and extension are presented. Our results, together with others from the literature, suggest that a bubble cluster bounded by two parallel walls is stable only if the normal force it exerts on the walls is attractive, i.e., if dE/dw > 0; clusters that cause repulsion between the walls are unstable. We correlate this with the distribution of film orientations: films in a stable cluster cannot be "too parallel" to the confining walls; rather, their average tilt must be larger than for a random distribution of film orientations.

In addition to a high coarsening rate, foams at low pressures may show special properties. These include a bulk modulus going to zero, a Poisson's ratio approaching zero, and a zero binding energy, implying foam separation into individual bubbles at a critical pressure. We provide examples of calculations made for particular two-dimensional clusters with an ideal gas, which illustrate these features. We also discuss the difficulties in performing experiments at the low pressures at which the unusual properties show up.

We reinterpret an instability in a two-dimensional free foam cluster previously discussed by Weaire et al. (Eur. Phys. J. E 7, 123 (2002)) in terms of the excess pressure in the bubbles. We conclude that in a free foam cluster no bubble can have a pressure below that of the surrounding gas.

Amongst the two-dimensional cellular patterns that fill a plane, dry foams at stable equilibrium typify a particular subset for which the total perimeter P of cell boundaries ( i.e., films between bubbles) has a local minimum. For a given set of bubble areas Ai (i=1,..., N), P can be written in the form P=R(SigmaN(i=1) square root Ai)/2, where R is topology dependent. We seek the set of areas Ai and the cluster topology that minimise R, and propose lower bounds for R that set lower bounds for the surface energy of i) individual bubbles, with circular edges meeting at 2pi/3 angles at vertices (Plateau cells), and ii) infinite periodic bubble clusters.

We have examined a number of candidates for the minimum-surface-energy arrangement of two-dimensional clusters composed of N bubbles of area 1 and N bubbles of area lambda (lambda< or =1). These include hexagonal bubbles sorted into two monodisperse honeycomb tilings, and various mixed periodic tilings with at most four bubbles per unit cell. We identify, as a function of lambda, the minimal configuration for N-->infinity. For finite N, the energy of the external (i.e., cluster-gas) boundary and that of the interface between honeycombs in "phase-separated" clusters have to be taken into account. We estimate these contributions and find the lowest total energy configuration for each pair (N, lambda). As lambda is varied, this alternates between a circular cluster of one of the mixed tilings, and "partial wetting" of the monodisperse honeycomb of bubble area 1 by the monodisperse honeycomb of bubble area lambda.

We show that the distribution of film orientations in a free (i.e., unstrained) foam cluster, though in general not uniform, has the same second moments as a uniform distribution. This is, however, in general not true of its other moments, which reflects the internal order of film orientations imposed by Plateau's laws. For a strained cluster, we relate the deviations of the second moments of the orientation distribution from their values for an unstrained cluster, to the stresses acting on the cluster. Our predictions are corroborated by experimental results for free and strained two-dimensional clusters, allowing us to find the force on a cluster bounded by two parallel walls.

This property is calculated for two- and three-dimensional ordered and disordered foams using a broken bond approach. The contact angle of a foam with a solid or a liquid substrate is also calculated from the energies of the bubbles in contact with the substrate. Experiments were carried out in which the contact angles of foams were measured, with reasonable agreement with the predictions. Copyright 2001 Academic Press.
