RESUMO

Sickle cell disease (SCD) is an inherited red blood cell disorder that occurs worldwide. Acute vaso-occlusive crisis is the main cause of hospitalization in patients with SCD. There is growing evidence that inflammatory vasculopathy plays a key role in both acute and chronic SCD-related clinical manifestations. In a humanized mouse model of SCD, we found an increase of von Willebrand factor activity and a reduction in the ratio of a disintegrin and metalloproteinase with thrombospondin type 1 motif, number 13 (ADAMTS13) to von Willebrand factor activity similar to that observed in the human counterpart. Recombinant ADAMTS13 was administered to humanized SCD mice before they were subjected to hypoxia/reoxygenation (H/R) stress as a model of vaso-occlusive crisis. In SCD mice, recombinant ADAMTS13 reduced H/R-induced hemolysis and systemic and local inflammation in lungs and kidneys. It also diminished H/R-induced worsening of inflammatory vasculopathy, reducing local nitric oxidase synthase expression. Collectively, our data provide for the firsttime evidence that pharmacological treatment with recombinant ADAMTS13 (TAK-755) diminished H/R-induced sickle cell-related organ damage. Thus, recombinant ADAMTS13 might be considered as a potential effective disease-modifying treatment option for sickle cell-related acute events.

The analysis of high molecular weight (HMW) proteins from complex mixtures is still a challenge in proteomics. This work introduces a novel hydrogel obtained by the copolymerization of an allyl-PVA derivative with acrylamide and bisacrylamide and applies this matrix to the electrophoretic separation of HMW proteins. By inducing gelation of polyacrylamide in the presence of variable amounts of allyl-PVA, it is possible to control and vary the average gel porosity. This gel is easy to produce and handle and offers the advantage of being highly mechanically resistant and macroporous. The new matrix was tested in mono-dimensional separations of complex protein mixtures extracted from red cell membranes with different detergents. The improved performance of this macroporous matrix allowed to identify new proteins by MS and immunoblot analysis using specific antibodies. In particular, the resolution of proteins ranging in size between 97 and 279 kDa was greatly improved here compared to standard polyacrylamide gels, suggesting that this matrix can be a useful tool in routine analysis of HMW proteins in cell biology.

The scope of this work was to rescue and document the traditional knowledge regarding the medicinal plants used by Passo da Ilha rural community, in Pato Branco, Paraná State, Southern Brazil (26° 11' S, 52°36' W and 760 m high). Structured interviews were made in field research with 16 informants who had the traditional knowledge about medicinal plants. The research was carried out from October to December 2000. The plants were collected in the field, identified and their vouchers were housed at the Herbarium "Irina Delanova De Gemtchjnicov" (BOTU) of São Paulo State University, in Botucatu. A total of 47 botanical families and 114 species of medicinal plants were identified. These plants ere suitable for ore than 30 different edicinal uses. The residents are mainly of European descent, which justify the presence of many exotic plants. The knowledge on how to use medicinal plants properly is held mainly by the females, and decreases in the newer generations, denoting "cultural erosion" in progress.


O objetivo deste trabalho foi resgatar e documentar o conhecimento popular referente às plantas medicinais utilizadas na comunidade rural do Passo da Ilha, em Pato Branco-PR (26° 11' S, 52°36' W e 760 m de altitude). Optou-se por utilizar o modelo de entrevista estruturada com 16 informantes, os quais possuíam o conhecimento sobre o uso das plantas medicinais em suas famílias. O período da realização das entrevistas foi de outubro a dezembro de 2000. As plantas foram coletadas no campo, identificadas e "vouchers" foram depositados no Herbário "Irina Delanova De Gemtchjnicov" (BOTU) da Universidade Estadual Paulista, em Botucatu-SP. Foram identificadas 47 famílias botânicas e 114 espécies de plantas medicinais. Estas plantas foram indicadas para mais de 30 usos medicinais diferentes. A comunidade é composta na maioria por descendentes de europeus, justificando a presença de muitas plantas exóticas. O domínio do uso das plantas é feminino, porém o conhecimento é menor nas gerações mais novas, denotando um processo de erosão cultural.

RESUMO

A aptidão tecnológica representa uma oportunidade de agregar valor de mercado ao trigo, principalmente em face do mercado internacional e do setor industrial, o qual busca diferencial de qualidade aos seus produtos. Os fatores genéticos, meteorológicos e de manejo são determinantes para a obtenção da qualidade desejada. Nesse sentido, a adequada escolha da cultivar, o conhecimento das limitações climáticas da região de cultivo e da fertilidade do solo e a execução dos tratos culturais recomendados pela pesquisa podem contribuir substancialmente para a obtenção das características físicas, químicas e biológicas que conferem qualidade à farinha e aos produtos derivados de farinha, conforme abordado nesta revisão.


Technological performance represents an opportunity to add market value to wheat, especially considering international market and industrial sector which seek to achieve distinctive quality of their products. Genetic, metereological, and management factors are crucial to obtain the desired quality. With respect to the selection of an adequate cultivar, knowledge about the climatic limitations of the wheat-growing region and soil fertility, and accomplishing the management practices recommended by research can substantially contribute toward obtaining the physical, chemical, and biological characteristics that lend quality to flour and flour-based products, as discussed in this review.
