RESUMO

The MeOH extract of the flower heads of Coreopsis lanceolata L. (Asteraceae) exhibited aldose reductase (AR) inhibitory activity (IC50 8.36 µg/mL). Bioassay-guided fractionation of the extract resulted in the isolation of a new biflavanone-named Lanceolanone A (1) and a chalcone glucoside (6), along with 12 known compounds (2-5 and 7-14), of which 4, 7, 9, 10, and 12 were isolated from C. lanceolata for the first time. The structures of the new compounds (1 and 6) were determined by extensive spectroscopic analysis, including two-dimensional (2D) NMR, and ECD calculation method. Compounds 2, 4, 11, 13, and 14 exhibited AR inhibitory activities with IC50 values between 2.40 and 9.99 µM. Furthermore, 8-13 at 1.0 mM activated AMPK expression in HepG2 human hepatoma cells compared to the control.

RESUMO

The electron conductivity of an extended metal atom chain (EMAC) that consisted of penta-nickel(II) ions bridged by oligo-α-pyridylamino ligands was examined by density functional theory (DFT) and elastic scattering Green's functions (ESGF) calculations. The calculated results revealed that an intramolecular ferromagnetic (FM) coupling state showed a higher conductivity in comparison with an anti-ferromagnetic (AFM) coupling state. The present results suggest the potential of the complex as a molecular switch as well as a molecular wire.

Although the mechanical properties and compositions of lipid bilayer membranes can change upon deformation, the fundamental relations between the composition, membrane tension and fluidity of membranes with little curvature have not yet been studied. In the current study, the membrane tension and the diffusion coefficients of free-standing black lipid membranes (BLMs), based on 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC), were observed by systematic control of the cholesterol concentration and the osmotic pressure with the laser-induced surface deformation (LISD) and fluorescence recovery after photobleaching (FRAP) techniques. When the osmotic pressure was raised and, therefore, the curvature became larger, both the membrane tension and the diffusion coefficients increased as well. On the other hand, when the cholesterol concentration was raised, the membrane tension increased whereas the diffusion coefficient decreased. The importance of the present results goes beyond this quantitative evaluation of the relation between the membrane tension and the fluidity, as it clarifies the changes in the fundamental properties of lipid bilayers upon natural fluctuations and perturbative deformation that were hitherto unknown.

A sample containing nitrated aromatic hydrocarbons (NPAHs) was injected into the sample inlet port of a gas chromatograph (GC), along with hydrazine, a reducing reagent. The analytes that eluted from the GC column were measured by mass spectrometry (MS) using an ultraviolet femtosecond laser as the ionization source. When no reducing reagent was used, large numbers of polycyclic aromatic hydrocarbons (PAHs) including NPAHs were observed in the two-dimensional GC/MS display. In contrast, when hydrazine was present, reduced forms of NPAHs, which included amino PAHs, were detected. When a palladium or platinum catalyst was placed in the GC inlet port, the compounds were further reduced to non-aromatic hydrocarbons. The present approach would be useful for studies to evaluate the chemical reaction that converts the constituents contained in exhaust emitted from a diesel engine.

RESUMO

Non-invasive measurement of the membrane tension of free-standing black lipid membranes (BLMs), with sensitivity on the order of µN m(-1), was achieved using laser-induced surface deformation (LISD) spectroscopy. A BLM was vertically formed via the folding method and aqueous phases with different refractive indices were added on each side in order to induce radiation pressure by a laser beam. The dynamic response of the deformed BLMs was measured under periodic intensity modulation and their tensions could be estimated. The dependence of membrane tension on the cholesterol concentration of BLMs composed of phosphatidylcholine and phosphatidylethanolamine was investigated, with the membrane tension increasing from 1.3 µN m(-1) to 68.1 µN m(-1) when the cholesterol concentration increased from zero to 33%. These tension values are much smaller than some of those previously reported, because this method does not suppress membrane fluctuation unlike other conventional methods. Our LISD system can be a promising tool for the measurement of membrane tension in BLMs.

RESUMO

A method was developed for determination of inorganic anions, including nitrite (NO(2)(-)), nitrate (NO(3)(-)), bromide (Br(-)), and iodide (I(-)), in seawater by ion chromatography (IC). The IC system used two dilauryldimethylammonium bromide (DDAB)-coated monolithic ODS columns (50 × 4.6 mm i.d. and 100 × 4.6 mm i.d.) connected in series for separation of the ions. Aqueous NaCl (0.5 mol/L; flow rate, 3 mL/min) containing 5 mmol/L phosphate buffer (pH 5) was used as the eluent, and detection was with a UV detector at 225 nm. The monolithic ODS columns were coated and equilibrated with a 1-mmol/L DDAB solution (in H(2)O/methanol, 90:10 v/v). The hydrophilic ions (NO(2)(-), NO(3)(-), and Br(-)) were separated within 3 min and the retention time of I(-) was 16 min. No interferences from matrix ions, such as chloride and sulfate ions, were observed in 35 ‰ artificial seawater. The detection limits were 0.6 µg/L for NO(2)(-), 1.1 µg/L for NO(3)(-), 70 µg/L for Br(-), and 1.6 µg/L for I(-) with a 200-µL sample injection. The performance of the coated columns was maintained without addition of DDAB in the eluent. The IC system was successfully applied to real seawater samples with recovery rates of 94-108 % for all ions.

Using a comparative genetics approach, one or more of the BtrA, BtrL, BtrP, and BtrV proteins encoded in the butirosin biosynthetic gene cluster (btr) from Bacillus circulans SANK72073 were identified as being responsible for an O-ribosylation process leading to the formation of ribostamycin, a key intermediate in this, and related antibiotic biosynthetic pathways. Functional analysis of the recombinantly expressed proteins revealed that both BtrL and BtrP were responsible for the ribosylation of neamine, using 5-phosphoribosyl-1-diphosphate (PRPP) as the ribosyl donor. Further detailed analysis indicated that this process occurs via two discrete steps: with BtrL first catalyzing the phosphoribosylaion of neamine to form 5''-phosphoribostamycin, followed by a BtrP-catalyzed dephosphorylation to generate ribostamycin. To the best of our knowledge, this is the first time that the functional characterization of a glycosyltransferase from an aminoglycoside biosynthetic gene cluster has been reported.

RESUMO

We comprehensively investigated the effects of substitution of the benzyl group in S-benzylisothiourea derivatives on antibacterial activity, because we found previously that some substitutions enhanced it. A 2,4-Cl2-derivative was found to be the most effective compound, it was stronger than the original one in Gram-negative rod shaped-bacteria such as Escherichia coli and Salmonella typhimurium.
