RESUMO

Sulfonamide antibiotics and florfenicol(FFC) are commonly used antibiotics in Zhejiang Province. They have weak adsorption on soil and are easy to migrate, with high environmental risks. In recent years, most of the studies on the potential risk of fecal-derived antibiotics to farmland soil were conducted by adding manure under laboratory conditions; therefore, it is impossible to assess the risk of antibiotic pollution under natural fertilization. Therefore, batch balance experiments were conducted to explore the effects of different soil types and manure types on the adsorption of antibiotics in the soil, in which five types of dryland farmland soils[Lin'an(LA), Jiashan(JS), Longyou(LY), Kaihua(KH), and Jinhua(JH)]in Zhejiang Province that have been used with different fertilizers(chicken manure, pig manure, and chemical fertilizer) for a long time were chosen, and four types of commonly used antibiotics[sulfadiazine(SD), sulfamethazine(SMT), sulfamethoxazole(SMZ), and FFC]were selected. The results showed that the adsorption of the four antibiotics in the experimental soil was weak, and the adsorption capacity decreased in the order of:SMT(1.44-13.23 mg1-(1/n)·L1/n·kg-1)>SMZ(0.73-6.05 mg1-(1/n)·L1/n·kg-1)>SD(0.16-5.57 mg1-(1/n)·L1/n·kg-1)>FFC(0.27-3.81 mg1-(1/n)·L1/n·kg-1). The Freundlich model was superior to the linear model in fitting the isotherm adsorption of SD, SMT, and FFC, in which SD and FFC belonged to "S" type adsorption, and SMT belonged to "L" type adsorption. For SMZ, the fitting effect of the linear model was better than that of the Freundlich model. The contents of total organic carbon(TOC) and dissolved organic carbon(DOC) could better predict the adsorption capacity of the four antibiotics(r=0.548-0.808), and the values of cation exchange capacity(CEC) and electrical conductivity(EC) could better predict the adsorption capacity of SMT and FFC(r=0.758-0.841). Compared with the application of chemical fertilizer, manure application increased the values of TOC, DOC, CEC, and EC in acidic and neutral soils, which was conducive to the adsorption of antibiotics on the soil. Meanwhile, manure application also increased pH in acidic and neutral soils, which was not conducive to the adsorption of antibiotics on the soil. In addition, manure application reduced the values of TOC, DOC, CEC, EC, and pH in alkaline soils. The lower pH was conducive to antibiotic adsorption on the soil, whereas the lower content of the other four was not conducive to antibiotic adsorption on the soil. For the acidic soil with low fertility, the application of manure increased soil fertility and thus increased the adsorption of antibiotics on the soil, such as the LA soil with chicken manure, the LY(1) soil with pig manure, and the JH soil with chicken manure and pig manure. However, for the acidic and neutral soils with high fertility, the application of manure had significantly increased soil pH and thus reduced the adsorption of antibiotics on the soil, such as the JS soil with chicken manure and pig manure and the LY(2) soil with chicken manure. For calcareous soil with high fertility and pH(such as KH soil), the adsorption profiles of the four types of antibiotics on the soil showed diversity after the application of manure:the adsorption capacity of SD increased significantly after the application of chicken manure and pig manure, whereas the adsorption capacity of SMT and SMZ decreased significantly, and the adsorption capacity of FFC declined significantly after the application of chicken manure. Therefore, manure application according to soil fertility could effectively control the environmental risk of fecal antibiotics.

To investigate the ambient pollution caused by volatile organic compounds (VOCs) in a typical industrial city in summer, the characteristics and chemical reactivity from VOCs and the causes of ozone (O3) pollution were analyzed using online VOCs measurements during polluted and non-polluted periods in Zibo city in July 2020. The results showed that the average hourly concentration of total volatile organic compounds (TVOC) during the polluted period[(50.6±28.3)] µg·m-3 was 32.5% higher than that during the non-polluted period[(38.2±24.9) µg·m-3]. The contribution of all VOCs categories were as follows:alkanes>aromatics>alkenes>alkynes, and the diurnal averages of TVOC and O3 concentrations were opposite during the polluted and non-polluted period. Ozone formation potential (OFP),·OH radical loss rate (L·OH), and secondary organic aerosol formation potential (SOAp) during the polluted period were higher than those during the non-polluted period. Alkenes contributed most to OFP and L·OH, whereas aromatics contributed most to SOAp. The tendency of the diurnal average of OFP and SOAp was overall consistent with that of TVOC. The priority species of OFP, L·OH, and SOAp were alkenes and aromatics. The VOCs/NOx method was applied to identify the O3-VOC-NOx sensitivity during the polluted and non-polluted periods, and the results showed that the photochemical regimes were VOCs-limited and transition regions. In addition, the smog production model (SPM) was employed to identify the O3 formation regime, and the results showed that those during the polluted period were identified as VOCs-limited and transition regions from 08:00 to 16:00, whereas the non-polluted period was mainly considered to be VOCs-limited. To mitigate the O3 pollution in summertime, the synergistic control of VOCs (especially alkenes and aromatics) and NOx emissions should be enforced.

To study the differences in volatile organic compound (VOCs) pollution characteristics between an urban site and a background site in summer, ambient VOCs were monitored using an online gas chromatograph (GC) at an urban site and a background site (Mt. Lu) in Zibo in July 2020. The VOCs pollution characteristics and chemical reactivity were analyzed, and the sources of VOCs were identified using the positive matrix factorization model(PMF). The results showed that ρ(TVOC) and ρ(NOx) were higher at the urban site, but ρ(O3) was higher at the background site. Diurnal average characteristics of ρ(TVOC) and ρ(NOx) were high at night and low during the day at the urban site, and there were no obvious variation characteristics at the background site. The diurnal average characteristics of ρ(O3) were consistent at the urban and background sites, showing low level at night and high level during the day; however, the peak in the background site was later than that at the urban site. The average ρ(TVOC) at the urban site and background site were (44.9±27.5) µg·m-3 and (17.3±9.1) µg·m-3, respectively, and the mass fraction of each component was ordered as alkanes>aromatics>alkenes>alkynes in both sites. The average ozone formation potentials(OFP)were (115.5±63.1) µg·m-3 and (38.0±20.2) µg·m-3, and the contribution of each component was ordered as alkenes>aromatics>alkanes>alkynes. The respective average values of·OH radical loss rate(L·OH) were (3.9±2.3) s-1 and (1.0±0.6) s-1, with the highest contribution of alkenes and the lowest contribution of alkynes in both sites. The average values of secondary organic aerosol formation potential(SOAp) were (0.5±0.3) µg·m-3 and (0.2±0.06) µg·m-3, respectively, with aromatic being the most abundant group. According to the source appointment by the PMF model, the main source of VOCs in the urban site was traffic sources (52.4%), followed by petroleum evaporation (19.2%), solvent evaporation (17.3%), and oil and biological sources (11.1%). The source of VOCs in the background site mainly came from traffic sources (40.2%), followed by solvent evaporation (31.3%), combustion sources (19.3%), and biological sources (9.2%). Zibo City should strengthen the management and control of motor vehicle emissions, petroleum evaporation, and the use of industrial solvents.

BACKGROUND: Ewing's sarcoma of the adrenal gland with inferior vena cava (IVC) and right atrium thrombus is extremely rare. Here, we report a case of giant adrenal Ewing's sarcoma with IVC and right atrium tumor thrombus and summarize the anesthesia and perioperative management. CASE SUMMARY: A young female was admitted to the Department of Urology with intermittent pain under the right costal arch for four months. Enhanced abdominal computed tomography revealed a large retroperitoneal mass (22 cm in diameter), which may have originated from the right adrenal gland and was closely related to the liver. Transthoracic echocardiography showed a strong echogenic filling measuring 70 mm extended from the IVC into the right atrium and ventricle. After preoperative preparation with cardiopulmonary bypass, sufficient blood products, transesophageal echocardiography and multiple monitoring, tumor and thrombus resection by IVC exploration and right atriotomy were successfully performed by a multidisciplinary team. Intraoperative hemodynamic stability was the major concern of anesthesiologists and the status of tumor thrombus and pulmonary embolism were monitored continuously. During transfer of the patient to the intensive care unit (ICU), cardiac arrest occurred without external stimulus. Cardiopulmonary resuscitation was performed immediately and cardiac function was restored after 1 min. In the ICU, extracorporeal membrane oxygenation (ECMO) and continuous renal replacement therapy (CRRT) were provided to maintain cardiac, liver and kidney function. Histopathologic examination confirmed the diagnosis of Ewing's sarcoma. After postoperative treatments and rehabilitation, the patient was discharged from the urology ward. CONCLUSION: An adrenal Ewing's sarcoma with IVC and right atrium thrombus is extremely rare, and its anesthesia and perioperative management have not been reported. Thus, this report provides significant insights in the perioperative management of patients with adrenal Ewing's sarcoma and IVC tumor thrombus. Intraoperative circulation fluctuations and sudden cardiovascular events are the major challenges during surgery. In addition, postoperative treatments including ECMO and CRRT provide essential support in critically ill patients. Moreover, this case report also highlights the importance of multidisciplinary cooperation during treatment of the disease.

OBJECTIVE: To investigate the efficacy of self-made arthroscopic single channel in the treatment of carpal tunnel syndrome. METHODS: Sixty patients with primary carpal tunnel syndrome treated from January 2014 to December 2019 were divided into arthroscopic group and traditional open operation group. There were 30 cases in arthroscopic group, including 12 males and 18 females, aged (47.5±4.5) years and the course of disease was (6.6±4.2) months. There were 30 cases in the traditional operation group, including 10 males and 20 females, aged (48.5±3.5) years, and the course of disease was (5.6±4.4) months. Both groups were unilateral. According to the anatomy of wrist joint and the characteristics of transverse carpal ligament and arthroscopy, the instruments including cannula, inner heart and hook knife were designed. The patients in two groups were treated with decompression of transverse carpal ligament using arthroscopy combined with self-made instruments and traditional open sergery. The incision length, operation time, intraoperative bleeding, hospitalization cost, hospitalization time and recovery time of the two groups were observed and compared. Boston Carpal Tunnel Questionnaire (BCTQ) score was used to evaluate the clinical efficacy of arthroscopy combined with self made instruments in the treatment of carpal tunnel syndrome. RESULTS: Compared with the traditional group, the arthroscopic group had significant advantages in incision length, operation time, intraoperative bleeding and hospital stay, but the total cost of hospitalization was increased. The Boston score was significantly higher in the arthroscopic group than that in the traditional group at 1 month after operation, but not at 3 and 6 months after operation. CONCLUSION: Arthroscopy combined with self-made instruments in the treatment of carpal tunnel syndrome is more reliable, minimally invasive and simplified than open surgery, but the patients should be clearly diagnosed and appropriately selected before operation to achieve satisfactory clinical effect.

To study the pollution characteristics, sources, and ecological and health risk of PM2.5-bound metallic elements in road dust in Zibo City, a total of 97 dust samples were collected in eight districts between October 2016 and May 2017, and particles smaller than 2.5 µm were suspended filtered using a resuspension system. Inductively coupled plasma mass spectrometry (ICP-MS) and optical emission spectrometry (ICP-OES) were used to investigate 18 metal elements within the dust samples. The results showed that the mass fraction of Ca[ω(Ca)] was highest with an average of 120307.7 mg·kg-1, which was 7.2 times higher than the soil background values for Shandong Province. The mean values of ω(Zn), ω(Cu), ω(Sb), and ω(Cd) were 13.9, 11.7, 13.3, and 29.6 times higher than the background values, respectively. The geo-accumulation index (Igeo) indicated high levels of Cd, Zn, Cu, and Sb pollution, especially in winter. Enrichment factors (EFs) also indicated high concentrations of Cd, Zn, Sb, and Cu in the road dust, which were notably affected by human activities. Principal component analysis (PCA) showed that biomass combustion, coal burning, vehicle emissions, iron and steel smelting, and soil dust are the five main sources of metal elements in road dust in Zibo City. The potential ecological risk of Cd and the total potential risk were extremely high during three seasons and was highest in winter. Health risk assessment showed that As and Pb had a non-carcinogenic risk for children, while Cr presents a carcinogenic risk. In conclusion, pollution from PM2.5-bound metallic elements in road dust in Zibo City is derived from anthropogenic sources and is most severe during winter. Importantly, the levels of pollution detected represent potential ecological risk as well as some non-carcinogenic and carcinogenic risks for children. Therefore, the source control of road dust requires particular attention.

BACKGROUND: Taurine is an organic amino acid and a major constituent of bile. With its contribution to various cellular functions, it has demonstrated therapeutic effects in a wide range of diseases. Since there is a lack of literature investigating taurine as a treatment for lupus nephritis (LN), here we examined the potential of taurine as a treatment for LN. METHODS: Experiments were carried out using MRL/lpr mice as a model of LN, and C57BL/6 mice were used as negative controls. At 12 weeks old, MRL/lpr mice were divided into four groups and treated with 0, 50 and 100 mg/kg body weight taurine for 5 days. Enalapril is used as a positive control drug. All animals were sacrificed after treatment. LN-induced damage was assessed by proteinuria, blood urea nitrogen (BUN) and serum creatinine (CRE) levels. The degree of inflammation was assessed by inducible nitric oxide synthase (iNOS), interleukin-4 (IL-4), IL-10, and tumor necrosis factor-α (TNF-α) levels. The degree of oxidative stress was assessed by malondialdehyde (MDA), reactive oxygen species (ROS), superoxide dismutase (SOD), and glutathione peroxidase (Gpx) levels. Hematoxylin and eosin (HE) staining and TUNEL staining assessed histopathological damage and apoptosis, respectively. The levels of Bcl-2, Bax, caspase-3, caspase-9, and NF-κB p65 were detected by western blot. RESULTS: The data indicated that taurine administration improved kidney functions, reversed cell death, suppressed oxidative stress, and importantly, adjusted the immune response of LN mice to a more balanced state. CONCLUSIONS: These results provide a novel strategy for LN therapy, which may overcome the disadvantages of traditional immunosuppression and hormone treatments with greater efficacy and fewer side effects.

Recycling waste tires through pyrolysis technology generates refractory wastewater, which is harmful to the environment if not disposed properly. In this study, a combined process of coagulation detoxification and biodegradation was used to treat tire pyrolysis wastewater. Organics removal characteristics at the molecular level were investigated using electrospray ionization (ESI) coupled with Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS). The results showed that nearly 90% of the organic matter from the wastewater was removed through the process. Preference of the two coagulants for different classes of organics in tire pyrolysis wastewater was observed. The covalently bound inorganic-organic hybrid coagulant (CBHyC) used in this work had a complementary relationship with biodegradation for the organics removal: this coagulant reduced toxicity and enhanced the biodegradation by preferentially removing refractory substances such as lignin with a high degree of oxidation (O/C > 0.3). This study provides molecular insight into the organics of tire pyrolysis wastewater removed by a combined treatment process, supporting the advancement and application of waste rubber recycling technology. It also contributes to the possible development of an effective treatment process for refractory wastewater.

INTRODUCTION: This study aimed to develop a new model on the basis of the National Early Warning Score to predict intensive care unit admission and the mortality of patients with acute pancreatitis. METHODS: Patients diagnosed with acute pancreatitis in the emergency department were enrolled. The values of the National Early Warning Score, Modified Early Warning Score, and Bedside Index of Severity in Acute Pancreatitis in predicting intensive care unit admission and mortality of patients with acute pancreatitis were evaluated. RESULTS: A total of 379 patients with acute pancreatitis were enrolled; 77 patients (20.3%) were admitted to the intensive care unit and 14 (3.7%) died. The National Early Warning Score and calcium level were identified as independent risk factors of intensive care unit admission. Serum calcium exhibited a moderate correlation with National Early Warning Score (r = -0.46; P < 0.001), Modified Early Warning Score (r = -0.37; P < 0.001), and Bedside Index of Severity in Acute Pancreatitis (r = -0.39; P < 0.001). A new model called National Early Warning Score-calcium was developed by combining National Early Warning Score and calcium blood test result, which had larger areas under the curve for predicting intensive care unit admission and mortality than the other 3 scoring systems. DISCUSSION: A new model developed by combining National Early Warning Score and calcium exhibited better value in predicting the prognosis of acute pancreatitis than the models involving National Early Warning Score, Modified Early Warning Score, and Bedside Index of Severity in Acute Pancreatitis alone.

BACKGROUND: Knee osteoarthritis (KOA), known as severe degenerative arthritis, commonly occurs in middle-aged and elderly people all over the world. Acupuncture as traditional oriental intervention is getting widely used and several systematic reviews (SRs) have reported the effectiveness of acupuncture on pain relief and functional recovery in patients with KOA. OBJECTIVE: Conducting an overview of SRs to provide more reliable evidence-based medical references for clinical practitioners and researchers of the effectiveness and safety of acupuncture for KOA. DATA SOURCES: EMBASE, Medline, Web of science, the Cochrane library, China National Knowledge Infrastructure, the Chinese Science and Technology Periodical Database, China Biology Medicine, Wan Fang Digital Journals, and PROSPERO databases from inception to December 2018, magazines, websites, and unpublished sources. SELECTION CRITERIA: Potential SRs were independently selected by 2 reviewers following a predetermined protocol. DATA EXTRACTION: Data information of included SRs were independently extracted by 2 reviewers following a predetermined standardized data extraction form. REVIEW APPRAISAL: The risk of bias and reporting quality of included SRs were evaluated by the Risk of Bias in Systematic reviews (ROBIS) tool and the Preferred Reporting Item for Systematic Review and Meta-analysis (PRISMA) statement. The quality of evidence of outcomes was evaluated by the Grading of Recommendations Assessment, Development and Evaluation (GRADE). RESULTS: A total of 12 SRs were included. All the SRs were published in recent 12 years, ranging from 2006 to 2017. According to ROBIS, 4 SRs were in low risk in domain 1 and 7 in domain 3 of phase 2, and 2 SRs were low risk in phase 3. Among 27 items of PRISMA, 19 items were reported over 70% of compliance. Using GRADE assessment, of 34 outcomes, high quality of evidence was found in 5 outcomes, 17 outcomes were rated moderate quality, and 11 outcomes were low quality. According to high-quality outcomes, acupuncture had more total effective rate, short-term effective rate, and less adverse reactions than western medicine in treating KOA. In terms of Lequesne index and Lysholm knee score scale score, the effectiveness of electroacupuncture was better than that of western medicine. LIMITATIONS: There might be missing information. There may be duplicated clinical trials included by each SR that might have impact on the synthetic findings. CONCLUSIONS: According to the high-quality evidence, we concluded that acupuncture may have some advantages in treating KOA. However, there are some risk of bias and reporting deficiencies still needed to be improved.

OBJECTIVE: To compare the clinical efficacy of total arthroscopic surgery and traditional surgery for popliteal cyst. METHODS: From August 2014 to July 2017, 60 cases of popliteal cyst were treated with total arthroscopy or traditional surgery respectively. In total arthroscopy group, there were 30 patients including 7 males and 23 females with an average age of (55.81±8.53) years old; the duration of the disease was (3.52±1.12) years;according to Rauschning-Lingdgren grading, 7 cases were grade I, 19 cases were grade II, 4 cases were grade III. In traditional surgical group, there were 30 patients including 5 males and 25 females with an average age of (57.93±9.84) years old; the duration of the disease was (3.48±1.34) years; according to Rauschning-Lingdgren grading, 5 cases were grade I, 21 cases were grade II, 4 cases were grade III. Preoperative symptoms involved such as arthralgia and swelling after knee joint and limited mobility. MRI confirmed a popliteal cyst. Arthroscopic surgery and traditional surgical was used respectively. Incision length, operation time, average stay, total hospital cost and Lysholm score of two groups were compared. RESULTS: In the total arthroscopic group, 1 case suffered from nerve injury, and all cases' incision healed well. In the traditional surgical group, there was 1 case of incision infection, 2 cases of poor healing, 1 case of nerve injury and 1 case of recurrence. All 60 cases were followed up for 6 to 30 months with an average of (13.3±6.5) months. After operation, the symptoms of knee joint arthralgia and swelling, discomfort were significantly improved in the two groups. There were statistically significant differences in incision length, operation time, average stay, total hospitalcost and Lysholm score 6 months after surgery(P<0.05). CONCLUSIONS: The total arthroscopic resection of popliteal cyst, via anterior approach to having a knee joint cavity exploration and treatment of meniscus and bursa, while combined with posteromedial approach for cyst excision would promise a minimal surgery and less pain for patients. Patients will have a rapider recovery, lower recurrence rate and less complication. Coagulation is an important process to remove organics from water. The molecular composition and structure of organic matter influence water quality in many ways, and the lack of information regarding the organics removed by different coagulants makes it challenging to optimize coagulation processes and ensure reclaimed water safety. In this paper, we investigated coagulation of secondary biological effluent from a municipal sewage treatment plant with different coagulants. We emphasized investigation of organics removal characteristics at the molecular level using Fourier transform-ion cyclotron resonance mass spectrometry (FT-ICR MS) coupled with electrospray ionization (ESI). We found that conventional coagulants can only partially remove condensed polycyclic aromatics and polyphenols with low H/C (H/C < 0.7) and highly unsaturated and phenolic compounds and aliphatic compounds with high O/C (O/C > 0.6). A new coagulant, CBHyC, had better removal efficiencies for all organics with different element compositions and molecular structures, especially organics that are resistant to conventional coagulants such as highly unsaturated and phenolic compounds and aliphatic compounds located in 0.3 < O/C < 0.8 and 1.0 < H/C < 2.0 regions and sulfur-containing compounds with higher O/C (e.g., anionic surfactants and their metabolites or coproducts). This study provides molecular insights into the organics removed by different coagulants and provides data supporting the possible optimization of advanced wastewater treatment processes.

OBJECTIVE: To explore the relationship between different components of fine particulate matter (PM2.5) emitted from coal combustion and their cytotoxic effect in the vascular endothelial cells. METHODS: Coal-fired PM(2.5) was sampled using a fixed-source dilution channel and flow sampler. The sample components were analyzed by ion chromatography and inductively coupled plasma atomic emission spectroscopy (ICP-AES). The PM(2.5) suspension was extracted using an ultrasonic water-bath method and then human umbilical vein endothelial cells (EA.hy926) were treated with various concentrations of the PM(2.5) suspension. Cell proliferation, oxidative DNA damage, and global DNA methylation levels were used to measure the cellular toxicity of PM(2.5) emitted from coal combustion. RESULTS: Compared to other types of coal-fired PM(2.5) preparations, the PM2.5 suspension from Yinchuan coal had the highest cytotoxicity. PM(2.5) suspension from Datong coal had the highest toxic effect while that from Yinchuan coal had the lowest. Exposure to coal-fired PM(2.5) from Jingxi coal resulted in lower 8-hydroxy-2'-deoxyguanosine (8-OHdG) levels. At the same dose, PM(2.5) emitted from coal combustion could produce more severe DNA impairment compared to that produced by carbon black. Cell survival rate was negatively correlated with chloride and potassium ions content. The 5-methylcytosine (5-mC) level was positively correlated with Mn and negatively correlated with Zn levels. The 8 OHdG% level was positively correlated with both Mn and Fe. CONCLUSION: PM(2.5) emitted from coal combustion can decrease cell viability, increase global DNA methylation, and cause oxidative DNA damage in EA.hy926 cells. Metal components may be important factors that influence cellular toxicity.

This study was conducted to compare the effect of active dry yeasts (ADY) and yeast cultures (YC), two typical products of yeast preparations, on growth performance, carcass characteristics, meat quality and blood indexes in finishing bulls fed a high-concentrate diet. Forty-five finishing bulls (mean body weight (BW) ± standard deviation: 505 ± 29 kg BW) were allocated to three groups of 15 bulls and assigned randomly to one of three diets which were CON diet (basal diet), ADY diet (basal diet + Levucell SC) and YC diet (basal diet + Diamond V XP), respectively. After 98 days of trial, all bulls were slaughtered. The result showed that ADY rather than YC improved growth performance and carcass traits of bulls compared to CON. Moreover, both ADY and YC improved beef tenderness and changed blood indexes related to fat metabolism. In conclusion, ADY had more pronounced effect on growth performance of bulls fed high-concentrate diet, and both ADY and YC improved the beef quality by intensive fat metabolism.

OBJECTIVE: Randomised controlled trials (RCTs) have given contradictory results about the efficacy and safety of ibandronate in treating metastatic bone disease (MBD) or multiple myeloma. This review meta-analysed the literature to gain a more comprehensive picture. DESIGN: Systematic review and meta-analysis of ibandronate compared with placebo or zoledronate. DATA SOURCES: PubMed, EMBASE and the Cochrane Library databases were systematically searched to identify RCTs published up to March 2015 evaluating ibandronate to treat MBD or multiple myeloma. REVIEW METHOD: 10 RCTs involving 3474 patients were included. Six RCTs were placebo-controlled and four compared ibandronate with zoledronate. The studies included in this review were mainly from European countries. RESULTS: Intravenous ibandronate (6 mg) or oral drug (50 mg) decreased the risk of skeletal-related events compared to placebo (risk ratio (RR) 0.80, 95% CI 0.71 to 0.90, p=0.002). It also reduced the bone pain score below baseline significantly more than did placebo at 96 weeks (weighted mean difference -0.41, 95% CI -0.56 to -0.27, p<0.001). The incidence of diarrhoea, nausea and adverse renal events was similar between the ibandronate and placebo groups, but ibandronate was associated with greater risk of abdominal pain. Ibandronate was associated with similar risk of skeletal-related events as another bisphosphonate drug, zoledronate (RR 1.02, 95% CI 0.82 to 1.26, p=0.87). The incidence of nausea, jaw osteonecrosis and fatigue was similar for the two drugs, but the incidence of adverse renal events was significantly lower in the ibandronate group. CONCLUSIONS: Ibandronate significantly reduces the incidence of skeletal-related events and bone pain in patients with MBD or multiple myeloma relative to placebo. It is associated with a similar incidence of skeletal-related events as zoledronate.

RESUMO

Here we demonstrate that protein enzymes captured in the solvent channels of three-dimensional DNA crystals are catalytically active. Using RNase A as a model enzyme system, we show that crystals infused with enzyme can cleave a dinucleotide substrate with similar kinetic restrictions as other immobilized enzyme systems. This new vehicle for immobilized enzymes, created entirely from biomolecules, opens possibilities for developing modular solid-state catalysts that could be both biocompatible and biodegradable.

The mitochondrial tyrosyl tRNA synthetase from Neurospora crassa (CYT-18 protein) is a bifunctional group I intron splicing cofactor. CYT-18 is capable of splicing multiple group I introns from a wide variety of sources by stabilizing the catalytically active intron structures. CYT-18 and mt TyrRSs from related fungal species have evolved to assist in group I intron splicing in part by the accumulation of three N-terminal domain insertions. Biochemical and structural analysis indicate that the N-terminal insertions serve primarily to create a structure-stabilizing scaffold for critical tertiary interactions between the two major RNA domains of group I introns. Previous studies concluded that the primarily α-helical N-terminal insertion, H0, contributes to protein stability and is necessary for splicing the N. crassa ND1 intron but is dispensable for splicing the N. crassa mitochondrial LSU intron. Here, we show that CYT-18 with a complete H0 deletion retains residual ND1 intron splicing activity and that addition of the missing N-terminus in trans is capable of restoring a significant portion of its splicing activity. The development of this peptide complementation assay has allowed us to explore important characteristics of the CYT-18/group I intron interaction including the stoichiometry of H0 in intron splicing and the importance of specific H0 residues. Evaluation of truncated H0 peptides in this assay and a re-examination of the CYT-18 crystal structure suggest a previously unknown structural role of the first five N-terminal residues of CYT-18. These residues interact directly with another splicing insertion, making H0 a central structural element responsible for connecting all three N-terminal splicing insertions.

OBJECTIVE: To investigate the effects of hepatitis B virus (HBV) in semen on human semen parameters and sperm DNA integrity. METHODS: We detected HBV DNA in the semen samples of 153 HBsAg-seropositive patients by real-time fluorescence quantitative PCR and calculated the sperm nuclear DNA fragmentation index (DFI) by sperm chromatin dispersion (SCD) assay. We compared the semen parameters between the HBV DNA-positive group (A, n = 43) and HBV DNA-negative group (B, n = 110) and analyzed the correlation of sperm DFI with the number of HBV DNA copies in the semen. RESULTS: HBV DNA was detected in 43 (28.1%) of the 153 semen samples. No statistically significant differences were observed in age, semen volume and sperm concentration between groups A and B (P >0.05). Compared with group B, group A showed significantly decreased sperm viability ([58.0 +/- 18.8]% vs [51.4 +/-17.1]%, P<0.05), progressively motile sperm ([29.6 +/- 13.3]% vs [24.5 +/- 10.1]%, P<0.05), average straight-line velocity ([23.7 +/- 4.0] microm/s vs [19.9 +/- 4.5 ] microm/s, P<0.01) and average path velocity ([26.5 +/- 7.0] microm/s vs [23.4 +/- 5.3] microm/s, P<0.01), but remarkably decreased sperm DFI ([19.3 +/- 8.0]% vs [24.2 +/- 9.4]%, P<0.01). The number of HBV DNA copies in semen exhibited a significant positive correlation with sperm DFI (r = 0.819, P < 0.01). CONCLUSION: HBV DNA in semen is not significantly associated with the number of sperm, but may affect sperm viability, velocity and DFI. There is a load-effect relationship between the number of HBV DNA copies in semen and sperm nuclear DNA integrity.

Inhalatory and dermal exposures of on-duty vehicle inspection workers to polycyclic aromatic hydrocarbons (PAHs) in Beijing were investigated from April 18 to May 17, 2011. Exposure levels to particulate PAHs for the vehicle inspection workers at gasoline, bus and diesel lines were found to be 56.07 ng m(-3), 111.72 ng m(-3) and 199.80 ng m(-3), respectively. A probabilistic risk assessment framework was integrated with the toxic equivalence factors (TEFs) and the incremental lifetime cancer risk (ILCR) approaches to quantitatively estimate the exposure risk for vehicle inspection workers of the three work lines. The median values of inhalation risk were estimated to be 3.7 × 10(-7), 5.0 × 10(-7) and 1.37 × 10(-6), respectively, while the median dermal ILCR values were 7.05 × 10(-6), 6.98 × 10(-6) and 1.28 × 10(-5), respectively for gasoline, bus, and diesel inspection workers. Total ILCR was higher than the acceptable risk level of 10(-6), indicating unacceptable potential cancer risk.

The structures and energies of the electronic ground states of the FeS(0/-), FeS2(0/-), Fe2S2(0/-), Fe3S4(0/-), and Fe4S4(0/-) neutral and anionic clusters have been computed systematically with nine computational methods in combination with seven basis sets. The computed adiabatic electronic affinities (AEA) have been compared with available experimental data. Most reasonable agreements between theory and experiment have been found for both hybrid B3LYP and B3PW91 functionals in conjugation with 6-311+G* and QZVP basis sets. Detailed comparisons between the available experimental and computed AEA data at the B3LYP/6-311+G* level identified the electronic ground state of (5)Δ for FeS, (4)Δ for FeS(-), (5)B2 for FeS2, (6)A1 for FeS2(-), (1)A1 for Fe2S2, (8)A' for Fe2S2(-), (5)A'' for Fe3S4, (6)A'' for Fe3S4(-), (1)A1 for Fe4S4, and (1)A2 for Fe4S4(-). In addition, Fe2S2, Fe3S4, Fe3S4(-), Fe4S4, and Fe4S4(-) are antiferromagnetic at the B3LYP/6-311+G* level. The magnetic properties are discussed on the basis of natural bond orbital analysis.
