RESUMO

In order to diagnose patients with pregnancy and antiphospholipid antibody syndrome, provide early treatment, and effectively reduce the pregnancy outcome of the abnormal pregnancy, the effect of antiphospholipid syndrome (APS) immunotherapy on the incidence of abortion was discussed based on clustering algorithm. We selected 62 cases of APS leading to recurrent miscarriage patients, in the early pregnancy injection of low molecular heparin, intravenous drip proprocyclin, and oral tactics, using B-ultrasound images to observe the pregnancy ending. The results show that the hormone levels in the two groups were different before treatment (P > 0.05); after treatment, the HCG, E2, and P hormone levels in the two groups were significantly improved, and the HCG, E2, and P hormone levels in the observation group were significantly higher than those of the control group (P < 0.05); the abortion rate of patients in the observation group was significantly lower than that of the control group (P < 0.05); the antiphosphorus antibody of the study group was significantly higher than that of the control group. For APS patients, immunotherapy is effective. Antiphospholipid syndrome causes remarkable immunotherapy effect in patients with recurrent miscarriage, effectively improves the clinical symptoms of patients, improves antiprochemical antibody rosion, and improves the patient's pregnancy outcomes, which is worth promoting.

@#[Abstract] Objective: To investigate the effect of Beclin1 knockdown on cisplatin resistance in ovarian cancer A2780 cells and its related mechanisms. Methods: The mRNA and protein expressions of Beclin1 in A2780 cells and drug resistant A2780/DDP cells were determined by qPCR and Western blotting. After transfection with Beclin1 siRNA, the sensitivity of A2780/DDP cells to cisplatin was detected by MTT assay; Cell clone formation and apoptosis were detected by the Colony formation assay and Flow cytometry assay, respectively; cell autophagy was monitored by monodansylcadaverin (MDC) staining. Furthermore, the protein levels of cell autophagy related proteins, lysosomal associated membrane protein Lamp-2 and Cathepsin B were detected by Western blotting. Results: The mRNA and protein expression levels of Beclin1 in cisplatin-resistant A2780/DDP cells were significantly higher than those in A2780 cells (all P<0.05). The expression of Beclin1 was significantly increased in A2780 cells after treated with cisplatin (P<0.05). Beclin1 knockdown promoted cisplatin induced apoptosis of A2780/DDP cells (P<0.05), inhibited autophagy and cell colony formation (all P<0.05), and increased cell sensitivity to cisplatin (P<0.05). Meanwhile, Western blotting showed that Beclin1 knockdown increased the protein levels of cleaved-caspase 3 and Cathepsin B in A2780/DDP cells, while down-regulated the protein expressions of Atg3, Atg7, LC3Ⅱ/Ⅰand Lamp-2 (all P<0.05). Conclusion: Beclin1 knockdown can improve the sensitivity of A2780/DDP cells to cisplatin, and the mechanism may be related to the inhibition of protective autophagy of cells by regulating the expressions of autophagy related proteins, and the regulation of lysosomes, thus further promoting cisplatin-induced apoptosis of drug-resistant cells.

Vaginal hysterectomy (VH) is generally considered the surgical approach of choice for treatment of benign uterine disease. Laparoscopically assisted vaginal hysterectomy (LAVH) has been introduced as a surgical alternative to the standard methods of VH. A meta-analysis of randomized controlled trials was conducted to compare LAVH vs VH. Medline, Embase, ISI Web of Knowledge CPCI-S, and the Cochrane Library were searched, and the methodologic quality of trials included was evaluated. Outcomes evaluated were complications, conversion rate, operative time, hospital stay, blood loss, paralytic ileus duration, and weight of the surgically treated uterus. Results were pooled in meta-analyses as odds ratio and weighted mean difference. Of 629 patients in 9 randomized controlled trials, 312 were allocated to the VH group, and 317 to the LAVH group. LAVH required longer operating time than VH did (weighted mean difference, 39.59; 95% confidence interval, 20:00-59.18 minutes; p < .001; χ(2) p < .001; I(2) = 97%), with evidence of significant heterogeneity (χ(2) = 209.14; p < .001). Complications, conversion rate, hospital stay, blood loss, paralytic ileus duration and weight of the surgically treated uterus were similar in both groups. Clinical outcomes with LAVH were comparable to those with VH; however, operative time was longer with LAVH. Currently, VH should be routinely used.

OBJECTIVE: To confirm the long-term outcomes of stroke patients and determine predicting factors for death, recurrence of vascular events and poor outcome (either recurrence or death) after the use of combined TCM therapy. METHODS: This was a retrospective hospital-based cohort study and was performed in the First Affiliated Hospital of Tianjin University of Traditional Chinese Medicine in Tianjin. All subjects with stroke consecutively admitted to an inpatient ward of the Acupuncture Department from January 1, 2008, to December 31, 2008 were retrospectively followed through one year. The main outcomes were either a recurrence of vascular events, mortality or both. Risk factors were recorded from medical records. Multivariate regression models were used to analyze predictors. The following independent variables were used: age, gender, hypertension, ischemic heart disease, atrial fibrillation, diabetes mellitus, carotid arterial lesions and history of stroke. RESULTS: Four-hundred and five patients were included. The 1-year mortality rate was 11.11%. 23.70% of the patients had a recurrent vascular event, and 30.86% suffered a poor outcome. Multiple logistic regression analysis found that previous stroke, and advanced age were predictors of death within one year, Recurrence of vascular events was associated with carotid arterial lesions, history of diabetes and previous stroke. Long-term poor outcome was predicted by advanced age, history of diabetes, and previous stroke. CONCLUSION: Age, previous stroke, carotid arterial lesions and diabetes history seem to have different impacts on the three outcomes within one year. Our findings provide important data for planning future hospital register studies of stroke patients in TCM hospitals.

Differences in design and implementation of clinical research on hypertension treatment with acupuncture and moxibustion were analyzed in this article so as to provide experiences to the design of clinical research on its treatment. Through document retrieval, it can be acquired that results of high quality clinical researches on treatment of hypertension with acupuncture and moxibustion have already been published. However, limitations in terms of sample size calculation, stochastic method, implementation of the blind method, selection of the control group and report on implementation process still existed in domestic documents. Therefore, high quality design proposals which not only in compliance with the features of acupuncture-moxibustion, but also accepted by modern medicine should be established with the combination of the advantages of acupuncture-moxibustion and clinical features of the disease. Moreover, standardizations on measuring the blood pressure, sample size calculation, selection of the control measurements, stochastic method and implementation of the blind methods should be established as well.

The aim of this study was to clarify the function of non-homologous end-joining (NHEJ) in tumorigenesis and chemoresistance, and to explore the potential of DNA-PK as a target of reversal of chemoresistance and enhancing the sensitivity of cells to chemotherapeutic agents. Plasmid vectors pSIREN-Ku70shRNA and pSIREN-DNA-PKcssh-RNA, which coded small interfering RNA of Ku70 and DNA-PKcs, were constructed and transfected into human cervical cancer cell line HeLa. The relationship between the down-regulation of Ku70 or DNA-PKcs and tumor cell proliferation and the sensitivity of cells to chemotherapeutic agents were analyzed. Down-regulation of Ku70 and DNA-PKcs expression inhibited cell proliferation, and increased cell apoptosis in DDP-treated HeLa cells. DNA-PK might play an important role in drug resistance, and inhibition of the DNA-PK expression suppressed the growth of tumor cells and enhanced the sensitivity of cells to chemotherapeutic agents.
